555,450. Glucosides of senna. SANDOZ, Ltd. Feb. 6, 1942, No. 1599. Convention date, May 13, 1941. [Class 2 (iii)] Glucosides are extracted from senna leaves and pods by a process which comprises making an alcoholic extract and treating this with an alcohol soluble alkaline-earth metal salt and adding basic substances until neutral or nearly neutral, or treating the extract with an alcoholsoluble basic alkaline earth metal derivative, separating the precipitated alkaline earth salt of the glucoside, and liberating the free glucosides by treatment with an acid. By the above treatment two glucosides which are described as Sennoside A and Sennoside B are obtained. These differ greatly in their solubility in methyl alcohol by means of which they may be separated. The glucosides may be purified by recrystallization from methyl alcohol or a mono-ether of a glycol. Both the above glucosides are acidic in nature and form salts. In examples (1) Senna leaves are extracted with benzene containing a little methyl alcohol to remove resins etc., and then with methyl alcohol. To the alcoholic extract an alcohol solution of calcium chloride is added followed by methyl alcoholic ammonia. The precipitated salts are. filtered, suspended in methanol and mixed with gluconic acid, heated to 40‹C. and filtered, the insoluble part containing Sennoside A remaining behind. Sennoside B crystallizes from the filtrate. Sennoside A is isolated from the.insoluble portion by treatment of a methanol suspension with ammonia, filtration of the calcium gluconate and isolation of Sennoside A by pouring into a hydrobromic acid solution of methanol. (2) Senna leaves or pods are extracted with aqueous acetone containing sodium carbonate, the extract neutralized with acetic acid and evaporated to dryness. A methanol solution of the product is treated with hydrobromic acid, and the liberated glucosides are precipitated in the form of their salts with barium bromide which precipitate is then worked up as in example (1).
