Abstract

Anhydrotetracycline is prepared by treating chlortetracycline or anhydrochlortetracycline (see Specification 744,019), or acid addition salts thereof, with hydriodic acid. Aqueous solutions containing up to 57 per cent hydriodic acid may be used, 48 per cent acid being preferred. The reaction temperature is up to 100 DEG C. At the higher temperatures red phosphorus is preferably added to remove free iodine liberated during the reaction. Anhydrotetracycline may be crystallized from ethyl alcohol. It melts at 224 DEG to 226 DEG C. and [a ]D25 DEG = +24 DEG (in 2-ethoxyethanol). Acid addition salts specified are those with hydrochloric, hydriodic, hydrobromic and acetic acids.
