Abstract
The invention comprises the alkali metal and alkaline earth metal bisulphite addition products of D 4-3,11,20-triketo-17a -hydroxypregnene-21-aldehyde (cortisone-21-aldehyde) and D 4 - 3,20 - diketo - 11b ,17a - dihydroxypregnene - 21 - aldehyde (hydrocortisone - 21 - aldehyde) and the preparation thereof by reacting an alkali metal or alkaline earth metal bisulphite with the required 21-aldehyde. The reaction is preferably effected by reacting the 21-aldehyde with an aqueous solution of the bisulphite salt, e.g. the aldehyde, or the monohydrate salt thereof, dissolved in an inert organic solvent, may be added to an aqueous solution of the bisulphite-a water miscible organic solvent such as a lower alcohol, acetone or dioxane being preferably used for the aldehyde. The desired product may be isolated by distilling off the organic solvent and then evaporating the water, or after removal of the organic solvent the aqueous solution may be freeze dried. The alkaline earth metal bisulphite addition products may be prepared also by reacting an alkali metal bisulphite addition product with an alkaline earth metal salt in an aqueous medium. In the examples the sodium, potassium, magnesium and calcium bisulphite addition products of cortisone-21-aldehyde and the sodium bisulphite addition product of hydrocontisone-21-aldehyde are prepared. The potassium, calcium and magnesium bisulphite addition products of hydrocortisone-21-aldehyde are also referred to.ALSO:An injectable therapeutic composition comprises a solution of an alkali metal or alkaline earth metal bisulphite addition product of D 4-3, 11, 20 - triketo - 17 - a - hydroxypregnene - 21 - aldehyde (cortisone - 21 - aldehyde) or D 4 - 3, 20 - diketo - 11b , 17a - dihydroxypregnene - 21 - aldehyde (hydrocortisone - 21 - aldehyde) in a sterile liquid, e.g. water.
