Abstract
Aqueous solutions of kanamycin are stabilized by the addition of a non-toxic salt of sulphurous acid, a non-toxic salt of hydrosulphurous acid, a non-toxic aldehyde addition product of sulphurous acid or of hydrosulphurous acid, monothioglycerol, monothiosorbitol, monothioglucose, beta, beta1-thiodipropionic acid, a non-toxic water-soluble ester of b ,b 1-thiodipropionic acid or a mixture thereof. The kanamycin may be a fermentation mixture of kanamycin A and B, kanamycin A, kanamycin B or a prepared mixture of kanamycin A and B. Specification 817,385 is referred to.ALSO:An aqueous injectable antibacterial composition comprises 100 to 700 mg. per ml. calculated as karamycin base, of a non-toxic water-soluble karamycin salt, 0.5 to 15 mg. per ml. of a stabilizing agent as hereinafter specified and from 2 to 50 mg. per ml. of buffering agent, the solution being adjusted to a pH of 4.0 to 8.2. A preferred composition contains 250 to 333 mg. per ml. of the karamycin (as base) and is adjusted to pH 4.0 to 6.5. The karamycin may be a fermentation mixture of karamycin A and B, karamycin A, karamycin B or a prepared mixture of karamycin A and B, especially a 1 : 1 mixture thereof. Specified stabilizing agents are a non-toxic salt of sulphurous acid, a non-toxic salt of hydro-sulphurous acid, a non-toxic aldehyde addition product of sulphurous acid, a non-toxic aldehyde addition product of hydrosulphurous acid, monothioglycerol, monothiosorbitol, monothioglucose, b ,b 1-thiodipropionic acid, a non-toxic water-soluble ester of b ,b 1-thiodipropionic acid or a mixture thereof. Preservatives such as phenol, chlorobutanol and methyl and/or butyl p-hydroxybenzoic acid may be present. The composition is prepared by adding the non-toxic karamycin salt to sterile water containing a buffering agent, adjusting the pH to 4.0 to 8.2, contacting the solution with activated charcoal, removing the charcoal and adding the stabilizing agent.
