Abstract

1,259,578. Orotic acid. LONZA Ltd. 27 May, 1970 [28 May, 1969], No. 25353/70. Heading C2C. Orotic acid or a salt thereof is prepared by chlorinating diketene in an inert organic solvent, reacting the resulting solution of γ- chloroacetoacetic acid chloride with urea, said chlorination and reaction being carried out at a temperature of from - 10‹ to - 30‹ C., the urea being added as a pyridine/glacial acetic acid solution in an amount such that it is immediately consumed, isolating the 4-chloromethyluracil so formed and oxidizing it by means of alkaline hydrogen peroxide, and optionally, liberating the free orotic salt from the salt so formed by the addition of an aqueous mineral acid. The chloromethyl-uracil is preferably dried before being reacted with the hydrogen peroxide which has preferably been heated to from 50‹ to 60‹ C. The γ-chloroacetoacetic acid chloride is preferably reacted immediatey with the urea.
