Abstract

The invention comprises O-acyl-N-alkyl-heparinamides, and non-toxic salts, e.g. alkali metal salts, thereof. The acyl group may be derived from an aliphatic carboxylic acid containing 2 to 7 carbon atoms, and the alkyl group may contain 1 to 6 carbon atoms, the preferred compounds being the sodium or potassium salts of O - acetyl - N - methyl - heparinamide. The heparinamides of the invention may be prepared by subjecting an alkali-metal N-alkyl heparinamide salt (Specification 973,893) to acylation. The acylation may be effected by an acid anhydride, e.g. acetic anhydride, in presence of a mixture of formamide and pyridine and the product may be obtained in free acid form by contacting it with an appropriate ion-exchange resin. Compositions containing the heparinamides of the invention have antilipaemic activity (Division A5). Specification 993,453 also is referred to.ALSO:A pharmaceutical composition having antilepaemic activity contains an o-acyl-n-alkyl heparinamide or a non-toxic salt thereof, in association with a carrier. Preferred heparinamides are sodium or potassium salts of o-acetyl-n-methyl-heparinamide (see Division C3). The composition may be in the form of a tablet, coated tablet, glosset, syrup or elixir, when the carrier is an ingestible base; a suppository; or in the form of an injectable solution, when the carrier is a sterile aqueous medium. The preferred modes of administration are orally, perlingually, transcutaneously and rectally. Specifications 973,893 and 993,453 are referred to.
