Abstract
La présente invention concerne une composition comprenant :- au moins une molécule conjuguée de façon covalente à une poly-lysine, et- au moins une molécule hydrophobe et non conjugable de façon covalente à une poly-lysine, ladite molécule étant encapsulée dans une micelle, et/ou au moins une molécule conjuguée à une poly-lysine par polymérisation, ladite molécule et la poly-lysine formant un copolymère. L’invention a également pour objet son procédé de fabrication et ses utilisations.
Description
L’invention concerne des compositions de molécules, certains étant conjuguées à des polymères spécifiques et d’autres, non conjugables directement à ces polymères, sont sous forme de copolymères ou encapsulées dans des micelles.
Etat de l’art
Pour augmenter l’efficacité et la biodisponibilité de certaines molécules dans des compositions thérapeutiques, nutritionnelles ou cosmétiques, il est connu de conjuguer lesdites molécules à des polymères spécifiques, comme la poly-lysine, et en particulier la poly-L-lysine.
La poly-lysine est un homopolypeptide, qui peut être linéaire, appartenant au groupe des polymères cationiques. À pH neutre, la poly-lysine contient un groupe amino hydrophile chargé positivement. L'acide aminé précurseur, la lysine, contient deux groupes amino, l'un sur le carbone α et l'autre sur le carbone ε. Au cours du développement du projet PNOS, PTS a produit iPr-PLys.HBr, où le contre-ion de l'ammonium terminal est un ion bromure.
La poly-lysine renforce l'interaction électrostatique entre les ions à charge négative de la membrane cellulaire et les ions à charge positive des facteurs de fixation à la surface de la culture. Lorsqu'elle est adsorbée à la surface de la culture, elle augmente le nombre de sites à charge positive disponibles pour la fixation des cellules. La poly-lysine présente une densité de charge positive élevée qui lui permet de former des complexes solubles avec des macromolécules chargées négativement (Park, Tae Gwan; Jeong, Ji Hoon; Kim, Sung Wan (2006-07-07). La poly-lysine, en tant que macromolécule fortement cationique, est efficacement trans-portée dans les cellules par endocytose (Hugues J.‐P. Ryser, Iain Drummond, Wei‐Chiang Shen. The cellular uptake of horseradish peroxidase and its poly(lysine) conjugate by cultured fibroblasts is qualitatively similar despite a 900‐fold difference in rate,1982, https://doi.org/10.1002/jcp.1041130126). Cette absorption est précédée d'une forte adsorption à la surface des cellules, qui est due à une interaction non spécifique des charges positives du polymère avec les charges négatives présentes à la surface de la plupart des cellules mammaliennes. Le transport membranaire de la poly-lysine peut donc être décrit comme une endocytose absorptive non spécifique, par opposition à l'endocytose en phase liquide ou à l'endocytose médiée par les récepteurs.
Étant donné sa nature polypeptidique, la poly-lysine est censée se biodégrader physiologi-quement sous l'action hydrolytique de plusieurs enzymes protéolytiques, comme l'endopeptidase tripsin, la chimotripsine ou les exopeptidases aminopeptidases (Hudecz F, Kutassi-Kovács S, Mezö G, Szekerke M. Avantageusement, la poly-lysine augmentation la demi-vie des molécules auxquelles elle est associée. De plus, du fait de son caractères amphiphile elle permet le passage de la Barrière Hémato Encéphalique.
Une molécule peut être conjuguée à la poly-lysine si des groupes fonctionnels appropriés sont présents dans la structure de ladite molécule de telle sorte que la conjugaison à la poly--lysine puisse être réalisée d'une manière orthogonale. En particulier il convient que les molécules comportent des groupes fonctionnels appropriés pour la conjugaison à des résidus de groupe amino au sein de la poly-L-lysine (amine primaire disponible dans les poly-lysine), tels que par exemple un groupe alcool, acide carboxylique, aldéhyde, amine, etc, et qu’elles ne comportent pas de groupes fonctionnels incompatibles pour une conjugaison à la poly-lysine.
Toutefois, certaines molécules n'incluent pas de groupes fonctionnels appropriés pour permettre la conjugaison orthogonale au squelette poly-lysine et/ou comportent des groupes fonctionnels incompatibles pour effectuer une conjugaison de façon covalente avec la poly-lysine. C’est le cas notamment de certaines molécules très hydrophobes, comme par exemple le cholestérol, ou c’est le cas également des acides aminés. Ces molécules ne peuvent pas être conjuguées à la poly-lysine, et lorsqu’elles sont intégrées dans des compositions avec de la poly-lysine, lesdites molécules restent libres, ne sont pas transportées par la poly-lysine, si bien que leur biodisponibilité est faible et identique à celle qu’elles présentent lorsqu’elles sont administrées seules.
Pour répondre à cette problématique, l’invention propose :
- d’encapsuler dans des micelles les molécules hydrophobes non conjugables à la poly-lysine, en particulier de les encapsuler, de les piéger, dans des structures micellaires préférentiellement formées par des conjugués poly-lysine, et
- d’intégrer au squelette poly-lysine les autres molécules non conjugables de façon covalente comme les acides aminés, via une copolymérisation des monomères Ncarboxyanhydrides (NCA).
- d’encapsuler dans des micelles les molécules hydrophobes non conjugables à la poly-lysine, en particulier de les encapsuler, de les piéger, dans des structures micellaires préférentiellement formées par des conjugués poly-lysine, et
- d’intégrer au squelette poly-lysine les autres molécules non conjugables de façon covalente comme les acides aminés, via une copolymérisation des monomères Ncarboxyanhydrides (NCA).
Ainsi l’invention a pour objet une composition sous forme liquide comprenant :
- au moins une molécule (i) conjuguée de façon covalente à une poly-lysine, et
- au moins une molécule (ii) hydrophobe et non conjugable de façon covalente à une poly-lysine, ladite molécule (ii) étant encapsulée dans une micelle, et/ou au moins une molécule (iii) conjuguée à une poly-lysine par polymérisation, ladite molécule (iii) et la poly-lysine formant un copolymère.
- au moins une molécule (i) conjuguée de façon covalente à une poly-lysine, et
- au moins une molécule (ii) hydrophobe et non conjugable de façon covalente à une poly-lysine, ladite molécule (ii) étant encapsulée dans une micelle, et/ou au moins une molécule (iii) conjuguée à une poly-lysine par polymérisation, ladite molécule (iii) et la poly-lysine formant un copolymère.
Avantageusement, l’invention permet ainsi que bénéficier des avantages de la poly-lysine pour toutes les molécules, y compris celles qui ne sont chimiquement pas conjugables à la poly-lysine par les méthodes connues de l’homme du métier. Ainsi l’efficacité, la solubilité et la biodisponibilité de toutes les molécules sont améliorée.
Cette configuration permet en particulier :
- d’augmenter la demi-vie des molécules actives de la composition dans l’organisme,
- de cibler les tissus ou les cellules sur lesquelles les molécules de composition doivent agir,
- de permettre aux molécules actives de la composition de passer la Barrière Hémato Encéphalique et de pénétrer dans les cellules, notamment dans les neurones et les motoneurones,
- d’augmenter la stabilité et la biodisponibilité de l’actif.
- d’augmenter la demi-vie des molécules actives de la composition dans l’organisme,
- de cibler les tissus ou les cellules sur lesquelles les molécules de composition doivent agir,
- de permettre aux molécules actives de la composition de passer la Barrière Hémato Encéphalique et de pénétrer dans les cellules, notamment dans les neurones et les motoneurones,
- d’augmenter la stabilité et la biodisponibilité de l’actif.
L’efficacité de la composition est donc plus importante et il est possible d’administrer des doses plus faibles et de réduire la toxicité aigüe ou chronique des molécules actives contenues dans la composition.
L’invention a également pour objet un procédé de fabrication des compositions selon l’invention.
Les compositions selon l’invention peuvent être utilisées comme médicament seules ou combinées à l’utilisation d’au moins une autre composition, et l’invention vise aussi les compositions pour leur utilisation comme médicament.
Enfin, les compositions selon l’invention peuvent être utilisées comme complément alimentaire ou composition cosmétique, seules ou combinées à l’utilisation d’une autre composition, et l’invention vise aussi l’utilisation de ces compositions comme complément alimentaire ou composition cosmétique.

Claims (19)
- Composition sous forme liquide comprenant :
- au moins une molécule conjuguée de façon covalente à une poly-lysine, et
- au moins une molécule hydrophobe et non conjugable de façon covalente à une poly-lysine, ladite molécule étant encapsulée dans une micelle, et/ou au moins une molécule conjuguée à une poly-lysine par polymérisation, ladite molécule et la poly-lysine formant un copolymère. - Composition selon la revendication 1, caractérisée en ce qu’elle comprend au moins une molécule hydrophobe et non conjugable de façon covalente à une poly-lysine est encapsulée dans au moins une micelle formée par des conjugués amphiphiles constitués chacun par au moins une molécule hydrophobe conjuguée de façon covalente à une poly-lysine.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la ou les molécule(s) hydrophobe(s) et non conjugable(s) de façon covalente à une poly-lysine sont choisies parmi le cholestérol, le co-enzyme Q10, le farnésyl cystéine, la curcumine, et leurs mélanges.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la ou les molécule(s) conjuguée(s) de façon covalente à une poly-lysine sont choisies parmi les acides gras, les antioxydants, les vitamines et leurs mélanges.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la ou les molécule(s) conjuguée(s) de façon covalente à une poly-lysine sont choisies parmi l’acide myristique, l’acide linoléique, l’acide palmitique, l’acide palmitoléique, l’acide laurique, l’acide oléique, l’acide orotique, la spermine, la taurine, l’acide patothénique, l’acide azélaïque, GABA, la biotine, l’acide thioctique, l’acide ascorbique, acide acétique, acide propionique, acide butyrique, acide pyruvique, acide lactique, l’alpha-tocophérol, l’acide pantothénique, le glutathion et leurs mélanges.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la ou les micelle(s) sont formées au moins par un ou plusieurs conjugués d’alpha-tocophérol conjugué de façon covalente à une poly-lysine et/ou par un ou plusieurs conjugués d’acide rétinoïque conjugué de façon covalente à une poly-lysine.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la ou les micelle(s) sont formées au moins par un ou plusieurs conjugués d’acide(s) gras conjugué(s) de façon covalente à une poly-lysine.
- Composition selon l’une des précédentes revendications, caractérisée en ce qu’elle comprend au moins une molécule conjuguée à une poly-lysine par polymérisation choisie parmi les acides aminés.
- Composition selon la précédente revendication, caractérisée en ce que la molécule conjuguée à une poly-lysine par polymérisation est choisie parmi la cystéine et la méthionine.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la poly-lysine est la poly-L-lysine.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la poly-lysine est linéaire et a un poids moléculaire compris entre 12 000 à 20 000 Da.
- Composition selon l’une des précédentes revendications, caractérisée en ce que la poly-lysine a un contre ion bromure, ou chlore ou TFA acide trifluoroacétique.
- Procédé de fabrication d’une composition selon l’une des précédentes revendications comprenant au moins une molécule hydrophobe et non conjugable de façon covalente à une poly-lysine, encapsulée dans une micelle formée par des conjugués amphiphiles constitués chacun par au moins une molécule hydrophobe conjuguée de façon covalente à une poly-lysine, caractérisé en ce qu’il comprend les étapes suivantes :
- a. créer un premix ampliphile : mélanger des conjugués de molécules hydrophobes conjuguées de façon covalente à une poly-lysine dans une solution aqueuse,
- b. ajouter dans le premix amphiphile, une ou plusieurs molécule(s) hydrophobe(s) non conjugable(s) à une poly-lysine, et mettre sous agitation de façon à former une micelle de molécules hydrophobes conjuguées de façon covalente à une poly-lysine encapsulant le ou les molécules hydrophobe(s). - Procédé de fabrication selon la précédente revendication, caractérisé en ce que l’agitation à l’étape b. est réalisée pendant entre 5 et 20 minutes à une vitesse d’agitation comprise entre 50 et 800 tours par minute.
- Procédé de fabrication selon l’une des revendications 13 ou 14, caractérisé en ce qu’il comprend également une étape de d. de séparation des phases soluble et insoluble, pour récupérer la phase soluble.
- Procédé de fabrication selon la revendication 13 à 15, caractérisé en ce que le premix de l’étape a. comprend également un ou plusieurs copolymère(s) de molécule conjuguée à une poly-lysine par polymérisation et/ou à une étape c. qui est mise en œuvre après l’étape b. ou après l’étape d., un ou plusieurs copolymère(s) de molécule conjuguée à une poly-lysine sont ajouté(s) dans le mélange.
- Procédé de fabrication selon la revendication 13 à 16, caractérisé en ce que le premix de l’étape a. comprend également une ou plusieurs autres molécules conjuguées de façon covalente à une poly-lysine et/ou qu’après l’étape b. ou c., une ou plusieurs autres molécules conjuguées de façon covalente à une poly-lysine sont ajoutée(s) dans le mélange.
- Procédé de fabrication d’une composition selon l’une des revendications 1 à 12, comprenant au moins une molécule conjuguée à une poly-lysine par polymérisation, ladite au moins une molécule et la poly-lysine formant un copolymère, caractérisé en ce qu’il comprend le mélange dudit au moins un copolymère avec au moins une molécule conjuguée de façon covalente à une poly-lysine.
- Composition sous forme solide, caractérisée en ce qu’elle est obtenue par lyophilisation ou atomisation ou une déshydratation lente d’une composition selon l’une des revendications 1 à 11.
