Abstract
L’invention concerne le domaine technique de l'industrie biochimique. L'invention concerne plus particulièrement un procédé de préparation de bêta-galactosidase, comprenant les étapes suivantes : (1) ensemencer un milieu Luria-Bertani (LB) avec le Bacillus circulans pour la culture et l'activation, ensuite, ensemencer un réservoir d'ensemencement avec le Bacillus circulans cultivé et activé pour la culture, afin d'obtenir une solution de culture d'ensemencement ; (2) ajouter la solution de culture d'ensemencement obtenue à un fermenteur contenant un milieu de culture de fermentation, pour la fermentation, afin d’obtenir une liqueur de fermentation de bacillus circulans ; (3) filtrer ou centrifuger la liqueur de fermentation de bacillus circulans obtenue, pour éliminer des thallus, afin d’obtenir une solution de bêta-galactosidase, dans lequel le milieu de culture de fermentation contient du lactose, du galactose, de la peptone végétale, de la poudre de maïs, de l'extrait de levure, du phosphate, du carbonate et de l'eau. Le procédé présente les avantages tels qu’un temps de fermentation, un cycle de de production raccourcie et une activité spécifique élevée du liquide enzymatique produit. L'invention concerne également un procédé de préparation d'enzyme immobilisée de bêta-galactosidase sans utiliser d'agent de réticulation. L'enzyme immobilisée préparée en adoptant le procédé a une bonne stabilité et peut être utilisée en continu pendant plus de 264 heures, de plus, le coût de préparation de galactose en utilisant l'enzyme immobilisée préparée est considérablement réduit.
