Abstract
Un método para la producción de metabolitos secundarios, comprendiendo el método: (a) transformar las bacterias Synechocystis sp. con uno o más de un gen policétido sintasa, un gen péptido sintetasa, o un gen ácido graso sintasa requeridos para la producción de metabolitos secundarios; (b) cultivar las bacterias Synechocystis sp. en condiciones adecuadas para la expresión del uno o más genes requeridos para la producción de los metabolitos secundarios; y (c) purificar los metabolitos secundarios de las bacterias, en el que dichas bacterias Synechocystis sp. expresan una fosfopanteteinil transferasa exógena (PPT) de Nodularia spumigena.

- Conservación de secuencias en la familia tipo Sfp
Los alineamientos resultantes revelan ejemplos adicionales de secuencias divergentes de los aminoácidos considerados críticos para la función de PPT. Por ejemplo, se aprecia que H90 del Motivo 2, asociado a la unión del 3'-fosfato de CoA, está ausente en varias secuencias, incluyendo todas las Methanosarcina, algunos Staphylococcus sp., y un Streptomyces sp. Se muestra un alineamiento de PPT tipo Sfp representativas en la figura 3. Se observaron dos subfamilias distintas. El Motivo 4 de la primera subfamilia se representa como F(S/C)KES (en lo sucesivo aquí denominado como "la subfamilia F/KES"). La segunda subfamilia incluyó la secuencia Sfp de B. subtilis. Este grupo muestra la secuencia peptídica W(T/C)KEA como el motivo 4 (en lo sucesivo aquí denominado como "la subfamilia W/KEA").

- Análisis filogenético
Los árboles filogenéticos presentados muestran distintos clados novedosos y soportan a las subfamilias observadas en los análisis de alineamiento. Las subfamilias de PPT tipo Sfp se separaron y se soportaron por los datos muestreados de forma repetitiva (figura 4). La PPT AcpS se utilizó para servir como un valor atípico.
Los representantes que albergan múltiples PPT tipo Sfp demostraron tener PPT incluidos en ambas ramificaciones F/KES y W/KEA de la filogenia, incluyendo Streptomyces, Escherichia, Microbacterium, Pseudomonas, Xanthomonas y Salmonella (figura 4). Las PPT tipo Sfp de organismos sin una PPT tipo AcpS también están presentes en ambos clados. Por ejemplo, se observaron Pseudomonas aeruginosa y Haemophilus influenzae (AAC21831) en las subfamilias F/KES y W/KEA respectivamente. Las PPT encontradas en agrupamientos biosintéticos híbridos (PKS-NRPS) se representan también en ambas subfamilias.
La subfamilia F/KES incluía la mayor parte de PPT asociadas a las péptido sintetasas y la síntesis de sideróforos, incluyendo todas las enzimas enterobactina EntD y el subconjunto de PPT de Streptomyces que se describen en Weissman y col., (2004) Identification of a phosphopantetheinyl transferase for erythromycin biosynthesis in Saccharopolyspora erythraea", ChemBioChem 5: 116-25.
La segunda subfamilia del subgrupo W/KEA incluía la PPT de B. subtilis Sfp. Este filotipo incluía las diversas PPT biosintéticas de glucolípidos de heterocistos, las PPT de biosíntesis de lisina y las secuencias de PPT de invertebrados y eucariotas. Las enzimas de PPT implicadas en la biosíntesis de policétido son predominantes en el grupo W/KEA, tal como MupN (AAM12928) asociado a la producción de mupirocina en Pseudomonas fluorescens.
Sppt se inactivó por inserción para determinar si fue la única enzima capaz de la fosfopanteteinilación en Synechocystis sp. PCC6803. El ADN genómico para la amplificación por PCR se extrajo como se ha descrito previamente (Moffitt, M. C. y B. A. Neilan. 2004. Characterization of the nodularin synthetase gene cluster and proposed theory of the evolution of cyanobacterial hepatotoxins. Appl Environ Microbiol 70: 6353 -62). Todas las enzimas de restricción se suministraron por New England Biolabs (Ipswich, MA) o Promega (Madison, WI).
Un fragmento de 2,5 kb que incluía Sppt (slr0495, número de acceso a Swissprot Q55185) se amplificó con los cebadores slrup (5'-GTAAACTCCATTAACGCTGGC-3') (SEQ ID NO: 7) y slrdn (5'-GGTGCAAATCCGTTACATGGA3') (SEQ ID NO: 8). Este fragmento se clonó en pGEM-T-Easy (Promega) y se digirió con la enzima de restricción AvaI. Un casete de resistencia a cloranfenicol (ChlR) se ligó a este sitio y el plásmido resultante, pGCSlr, se transformó de forma natural en Synechocystis sp. PCC6803 para inactivar por inserción Sppt usando los métodos descritos en Eaton-Rye, J., 2004 The construction of gene knockouts in the cyanobacterium Synechocystis sp. PCC 6803, Methods Mol Biol.; 274: 309-24. La Synechocystis sp. PCC6803 de tipo silvestre (WT) también se transformó con un plásmido de desactivación con resistencia a cloranfenicol como un control positivo para la viabilidad de la cepa. En resumen, se centrifugaron aproximadamente 2 ml de células de Synechocystis sp. PCC6803 y se lavaron en medio BG-11. Después de la resuspensión en 300 µl de BG-11, se añadieron 3 µg ml-1 de plásmido y las células se incubaron a 25 ºC en 30 µmol m-2 s-1 de luz constante durante 6 horas. Las células se extendieron sobre filtros de nitrocelulosa estériles (0,45 µm, Millipore, Billerica MA) en placas BGTS no selectivas (BG-11, TES 10 mM, tiosulfato sódico al 0,3 %, agar al 1 %). Después de 36 horas, los filtros se transfirieron a placas BGTS selectivas con 10 µg ml-1 de cloranfenicol.
