Abstract

15 La dosificación y frecuencia (dosis únicas o múltiples) de la administración o trasplante de las células a un ser humano, incluyendo la cantidad real de células trasplantadas en el ser humano, pueden variar dependiendo de diversos factores, incluyendo la afección particular que se esté tratando (por ejemplo, afección neurológica, afección cardiaca, tal como un infarto de miocardio, afección degenerativa, lesión aguda), tamaño, edad, sexo, salud, peso corporal, índice de masa corporal, dieta del ser humano, naturaleza y grado de los síntomas de la afección neurológica (por ejemplo, enfermedad de Parkinson de aparición temprana frente a enfermedad de Parkinson avanzada; traumatismo de médula ósea frente a seccionamiento parcial o completo de la médula ósea), o afección cardiaca (por ejemplo, fallo cardiaco congestivo, infarto de miocardio); tipo de tratamiento concurrente (por ejemplo, esteroides); complicaciones de la afección neurológica o cardiaca; grado de tolerancia al tratamiento u otros

Los seres humanos con una afección neurológica o cardiaca pueden tratarse durante días (por ejemplo, 30) con células obtenidas por los métodos de la invención (por ejemplo, aproximadamente 106 células), por varias vías de administración (por ejemplo, intra-tecal, intravenosa).
También se prevé que los métodos descritos en este documento puedan emplearse para tratar afecciones neurológicas en mamíferos diferentes al ser humano. Por ejemplo, un mamífero no humano en necesidad de tratamiento veterinario, por ejemplo, animales de compañía (por ejemplo, perros, gatos), animales de granja (por ejemplo, vacas, ovejas, cerdos, caballos) y animales de laboratorio (por ejemplo, ratas, ratones, cobayas).

Ejemplificación
Se aspiraron células de médula ósea de la cresta ilíaca de voluntarios humanos adultos sanos. El componente de
45 glóbulos rojos del aspirado se lisó mezclando el aspirado con un tampón de cloruro de amonio que consistía en cloruro de amonio 155 mM, bicarbonato de potasio 10 mM y EDTA (ácido etilendiaminatetraacético) 0,1 mM, pH 7,2, a una proporción 1:20 de aspirado de médula a tampón. La suspensión celular resultante se agitó en vórtice durante 2 segundos, se incubó durante 2 minutos a temperatura ambiente y después se centrifugó (10 minutos a 500 x g). El sedimento celular mononuclear resultante se resuspendió en medio completo y se centrifugó (10 minutos a 500 x g). El medio completo es medio esencial mínimo-alfa (Gibco BRL, Rockville, MD) suplementado con glutamina 4 mM y suero bovino fetal de lote de suero seleccionado al 10 % (FBS, Gibco BRL, Rockville, MD). El sedimento celular después se resuspendió en el medio completo y se centrifugó una segunda vez (10 minutos a 500 x g).
El sedimento resultante se resuspendió en el medio completo y se determinó la cantidad de células viables por
55 exclusión de azul de tripano. La suspensión celular mononuclear después se sembró en matraz T75 tratado con cultivo tisular a una densidad de 50.000 células/cm2 y se incubó a 37 ºC en una atmósfera que consistía en 5 % de dióxido de carbono, 5 % de oxígeno, y 90 % de nitrógeno. En el quinto día de cultivo, las células no adherentes y el medio condicionado (también mencionado en este documento como "medio gastado") se aspiraron de los matraces y a las células adherentes se les volvió a administrar medio completo fresco. Las unidades formadoras de colonias (CFU) adherentes se expandieron durante 3-5 días adicionales.
La generación de CFU se controló en placas de 6 pocillos iniciadas de forma concurrente en condiciones idénticas a los matraces T75.
