Abstract

Al menos un aminoácido de los péptidos de citoquina de la invención y preferiblemente dos aminoácidos consecutivos, posee(n) uno de sus átomos separado por menos 5 angstroms de un átomo del receptor correspondiente a dicha citoquina. Está ventajosamente separado menos de 4,5 angstroms, principalmente separado menos de 4 angstroms y particularmente separado menos de 3,5 angstroms de un átomo del receptor
5 correspondiente a dicha citoquina.
En general el átomo afectado del aminoácido se encuentra en la cadena lateral de dicho aminoácido.
En condiciones preferidas de aplicación de la invención 2, en particular 3, y preferiblemente 4 aminoácidos consecutivos del péptido de citoquina responden a estemismo criterio de separación.
Esta separación se evalúa por datos estructurales, por ejemplo, de cristalografía, o incluso por RMN que da 10 resultadossimilaresalasmedidascristalográficas.
Los péptidos de citoquina anteriores constan ventajosamente de más de 8, principalmente más de 10, particularmentemás de 12 ymás particularmentemás de 15aminoácidos.
En otras condiciones preferidas de aplicación de la invención, los péptidos de citoquina anteriores constan demenos de 35, ventajosamente menos de 30, principalmente menos de 25 y particularmente menos de 20 aminoácidos. 15 Generalmente, las secuencias más cortas corresponden a péptidos que contienen un solo grupo de al menos dos aminoácidos consecutivos que constan de al menos uno de sus átomos separados por menos de 5 angstroms de un átomo del receptor correspondiente a dicha citoquina, mientras que las secuencias más largas corresponden en general a s péptidos según la invención que contienen dos o incluso tres grupos o más de tales aminoácidos consecutivos. En efecto, estos grupos pueden estar separados por varios, por ejemplo, 10 aminoácidos como en el
20 caso de IL1.

Como es conocido por los expertos en la inmunología, son posibles modificaciones de las cadenas peptídicas naturales sin por ello modificar la naturaleza de las propiedades inmunológicas de los péptidos inmunógenos. Por tanto, se pueden citar igualmente los derivados de péptidos de citoquina que son altamente homólogos de estas secuencias naturales, por ejemplo que tienen más de 80% de homología o incluso más de 90% de homología con el péptido natural correspondiente, conservando al mismo las propiedades inmunológicas de este sitio epitópico del péptido natural. Su zona de homología puede variar de 5 a 40 residuos, por ejemplo de 8 a 40 residuos, o incluso de 8 a 35 residuos, preferiblemente de 10 a 35 residuos, pero también de 12 a 35 residuos, principalmente de 12 a 30 residuos, en particular de 15 a 30residuos ymás particularmente de 15 a 25 residuos.
Los derivados de los péptidos de citoquina pueden contener residuos modificados, con la condición de que las modificaciones no disminuyan sensiblemente la inmunogenicidad, bien por adición de radicales químicos (metilo, acetilo, etc.) o bien por modificación estereoquímica (uso de aminoácidos de la serie D). Los derivados peptídicos de citoquina deben inducir, como los péptidos de citoquina, anticuerpos queinteractúan con la citoquina.
Los derivados peptídicos de citoquina según la invención pueden constar de una o varias modificaciones en los aminoácidos de los que están constituidos, tales como deleciones, sustituciones, adiciones o funcionalizaciones (tales como acilación) de uno o varios aminoácidos, en la medida en la que estas modificaciones permanecen en el ámbito precisado antes (caracteres inmunológicos). Por ejemplo, en general la sustitución de un residuo de leucina por un residuo de isoleucina no modifica dichas propiedades; las modificaciones deben afectar en general a menos del 40% de los aminoácidos, principalmente a menos de 30%, preferiblemente a menos de 20% y más particularmente a menos de 10% de los aminoácidos del péptido natural. Es importante que los anticuerpos inducidos por los péptidos modificados sean activos frente a la citoquina natural.
Estas modificaciones están al alcance de los expertos en la técnica que pueden verificar la incidencia de las modificaciones por ensayos sencillos. La inmunogenicidad de dichos derivados modificados se puede evaluar por ELISA después de la inmunización de ratones, siendo el antígeno ensayado por ELISA la citoquina completa o el péptido de citoquina inmunizante, o por ensayos de bloqueo de la unión citoquina-receptor. Las eventuales modificaciones afectan preferiblemente a menos de 8 aminoácidos, ventajosamente a menos de 6 aminoácidos, principalmente a menos de 4 aminoácidos, y particularmentea 3 aminoácidos o menos, como 2 o 1 solo aminoácido.

Ejemplo 3:
Se inmunizaron 5 ratones con el péptido de síntesis procedente de TNF humano que es: KYQAEGQLQWLNRRANALLANGVELRDNQL (SEQ ID Nº 39). Este péptido se acopló a la proteína portadora KLH por reacción con diazobencidina. A la secuencia natural se añadieron los residuos K e Y para permitir una unión a la KLH por acoplamiento con ayuda de glutaraldehído. Después de la inmunización en las mismas condiciones que en el Ejemplo 1, se extrajo suero el D60 y se realizaron ensayos ELISA con la citoquina natural para revelar la presencia de anticuerpos.
La absorbancia media obtenida para cada ratón se presenta en la tabla siguiente:

10 La existencia de anticuerpos que neutralizan la citoquina natural pudo ser evaluada de la manera siguiente: 50 unidades de citoquina TNF natural se pre-incubaron con suero de ratón (inmunizado o de control) diluido a 1/200 durante 2 horas a 37ºC. El conjunto se añadió luego a células EL929 dispuestas, 25.000 células por micropocillo. La lisis de las células se evaluó después de 24 horas por coloración con naftol azul negro (Sigma). El porcentaje de inhibición de la lisis indica el porcentaje de neutralización del TNF, y como se puede ver, se observan respuestas
15 para los ratones inmunizados pero no para los controles.

Ejemplo 4:
Se inmunizaron 5 ratones con el péptido procedente de IFN humano que es: KKYFNAGHSDVADNGTLFLGILKN (SEQ ID Nº 40). Este péptido se sintetizó químicamente en forma de MAPS. Este péptido MAPS se acopló luego a la
20 proteína portadora toxoide del tétano por reacción con glutaraldehído. Después de inmunización en las mismas condiciones que en el Ejemplo 1, se extrajo suero el D60 y se realizaron ensayos ELISA con la citoquina natural para revelar la presencia de anticuerpos.
La absorbancia media obtenida para cada ratón se presenta en la tabla siguiente:

25 La existencia de anticuerpos que neutralizan la citoquina natural pudo ser evaluada de la manera siguiente: 100 unidades de citoquina natural se pre-incubaron con suero deratón (inmunizado o de control), diluido 1/250 durante 2 horas a 37ºC. El conjunto se añadió luego a células RAW264.7, dispuestas 300.000 células por pocillo de 2 mL. Después de 24 horas se evaluó la expresión de CMH de clase II sobre las células por citometría de flujo, por marcaje con anticuerpos anti-CMH de clase II acoplados a fluoresceína. El porcentaje de inhibición de la expresión
30 de CMH de clase II indica el porcentaje de neutralización de IFN, y como se puede ver, se observan inhibiciones para los ratones inmunizados pero no para los controles. El umbral de significación para estos experimentos es 30%.
