Abstract
Un roedor o célula de roedor modificado(a) genéticamente, caracterizado por que el gen que codifica el polipéptido MASP-2 endógeno y/o el gen que codifica el polipéptido MAp19 endógeno está(n) ausente(s) o parcialmente ausente(s) o desorganizados de tal modo que no se produce el polipéptido MASP-2 endógeno de roedor y/o MAp19 endógeno de roedor.

Un gen exógeno es un gen que es extraño, es decir no natural, al mamífero o célula no humano hospedador.
Así pues, el método puede utilizarse para producir un mamífero no humano, que es preferiblemente un roedor, más preferiblemente un ratón, que es capaz de expresar uno o más genes MASP-2 extraños, preferiblemente humanos por reproducción del mamífero no humano modificado genéticamente como se define en esta memoria que es incapaz de expresar genes MASP-2 funcionalmente activos (endógenos), con un mamífero no humano compatible, precisamente un roedor, más preferiblemente un ratón, que es capaz de expresar uno o más genes MASP-2 extraños funcionalmente activos, preferiblemente humanos. Esto permite el intercambio interespecies gen/locus para producir progenie seleccionada (heterocigótica u homocigótica) con uno más genes exógenos funcionalmente activos, preferiblemente humanos de las características deseadas en un fondo en el que los genes correspondientes del mamífero no humano están silenciados o eliminados. Un mamífero no humano de este tipo puede ser útil en la producción de anticuerpos para el gen exógeno, y la presente invención proporciona adicionalmente un método para la producción de tales anticuerpos, ylos anticuerpos producidos en dicho método.
Como se utiliza esta memoria, el término “anticuerpo” abarca anticuerpos y fragmentos de anticuerpo de los mismos derivados de cualquier mamífero productor de anticuerpos (v.g., ratón, rata, conejo, y primate, con inclusión de humano) que se fija específicamente a polipéptidos MASP-2 o porciones de los mismos. Anticuerpos ilustrativos incluyen anticuerpos policlonales, monoclonales y recombinantes, anticuerpos multiespecíficos (v.g., anticuerpos biespecíficos), anticuerpos humanizados, anticuerpos murinos, anticuerpos quiméricos, anticuerpos ratón-humano, ratón-primate, anticuerpos monoclonales humanos, y anticuerpos anti-idiotipo, y pueden ser una molécula intacta o un fragmento de la misma.
Como se utiliza en esta memoria, el término “fragmentos de anticuerpo” se refiere a una porción derivada de o afín a un anticuerpo anti-MASP-2 de longitud total, que incluye generalmente la región de fijación de antígeno o la región variable del mismo. Ejemplos ilustrativos de fragmentos de anticuerpo incluyen fragmentos Fab, Fab’, F(ab)2, F(ab’)2, y Fv, fragmentos scFv, diacuerpos, anticuerpos lineales, moléculas de anticuerpo monocatenarias y anticuerpos multiespecíficos formados a partir de fragmentos de anticuerpo.
Como se utilizan esta memoria, los fragmentos de anticuerpo “Fv monocatenario” o “scFv” comprenden los dominios VH y VL de un anticuerpo, en donde estos dominios están presentes en una sola cadena de polipéptido. Por regla general, el polipéptido Fv comprende además un enlazador polipeptídico entre los dominios VH y VL, lo que permite que elscFv forme la estructura deseada para fijación del antígeno.
Como se utiliza en esta memoria, un “anticuerpo quimérico” es una proteína recombinante que contiene los dominios variables y las regiones determinantes de la complementariedad derivadas de un anticuerpo de especie no humana (v.g., roedor), mientras que el resto de la molécula de anticuerpo se deriva de un anticuerpo humano.
Como se utiliza en esta memoria, un “anticuerpo humanizado” es un anticuerpo quimérico que comprende una secuencia mínima que se ajusta a las regiones específicas determinantes de la complementariedad derivadas de una inmunoglobulina no humana que se trasplanta a un entramado de anticuerpo humano. Los anticuerpos humanizados son típicamente proteínas recombinantes en las cuales únicamente las regiones determinantes de la complementariedad del anticuerpo son de origen no humano.
Anticuerpos policlonales contra MASP-2 pueden prepararse por inmunización de un animal con el polipéptido MASP2 ounaporción inmunógena delmismoutilizandométodos bienconocidosporquienesposeenexperienciaordinaria en la técnica. Véase, por ejemplo, Green et al., "Production of Polyclonal Antisera," en Immunochemical Protocols (Manson, ed.), página 105. La inmunogenicidad de un polipéptido MASP-2 puede aumentarse por el uso de un adyuvante, con inclusión de geles minerales, tales como hidróxido de aluminio o adyuvante de Freund (completo o incompleto), sustancias tensioactivas tales como lisolecitina, polioles Pluronic, polianiones, emulsiones de aceite, hemocianina de lapa bocallave y dinitrofenol. Los anticuerpos policlonales se generan típicamente en animales tales como caballos, vacas, perros, pollos, ratas, ratones, conejos, cobayos, cabras, u ovejas. Alternativamente, un anticuerpo anti-MASP-2 útil en la presente invención puede derivarse también de un primate subhumano. Técnicas generales para anticuerpos diagnóstica y terapéuticamente útiles en babuinos pueden encontrarse, por ejemplo, en Goldenberg et al., Publicación de Patente Internacional No. WO 91/11465, y en Losman et al., Int. J. Cancer 46:310, 1990. Sueros que contienen anticuerpos inmunológicamente activos se producen luego a partir de la sangre de tales animales inmunizados utilizando procedimientos estándar bien conocidos en la técnica.
En algunos casos, el anticuerpo MASP-2 puede ser un anticuerpo monoclonal. Los anticuerpos monoclonales anti-MASP-2 son altamente específicos, estando dirigidos contra un solo epítopo de MASP-2. La tercera variedad de murino direccionada por el gen MASP2 se estableció utilizando el constructo de desorganización de genes descrito en la Figura 3, de igual manera que se ha descrito arriba para el ratón deficiente en MASP-2. Para generar una variedad de murino deficiente en MAp19/sMAP y MASP-2, el exón 5, que es el exón responsable de la generación del transcrito de mRNA específico de MAp19/sMAP del gen MASP2 se reemplazó por una casete de neomicina. A diferencia del constructo descrito con referencia a la Figura 2, el constructo de la Figura 3 no incluye secuencias loxP. Por tanto, el gen marcador utilizado no puede escindirse, por lo que la variedad de ratón resultante carece del exón 5 de MASP-2, pero los exones de MASP-2 restantes están desorganizados por la presencia delgenmarcador.Elratónresultante es porconsiguientedeficientetanto en MAp19 como en MASP-2. El linaje de murino direccionado por el gen creado utilizando este constructo se denominará de ahora en adelante MASP-2/MAp19 -/-.
Con objeto de reemplazar el MASP-2 murino deficiente y/o el MAp19 murino deficiente con MASP-2 humano o MAp19 humano, se establecieron dos constructos de minigénconstructos de minigén como se describe en la Figura
4. La Figura 4a describe el constructo de minigén que codifica MASP-2 humano que utiliza la región promotora del gen MASP-2 humano, incluyendo los 3 primeros exones (exón 1 a exón 3) seguidos por la secuencia de cDNA que representa la secuencia codificante de los 8 exones siguientes, codificando por tanto la longitud total de la serinaproteasa de MASP-2. Este constructo de minigén, denominado mini hMASP-2 se inyectó en huevos fertilizados de MASP-2-/-CS a fin de reemplazar el gen MASP-2 murino deficiente por MASP-2 humano expresado transgénicamente. La secuencia del constructo de minigén MASP-2 (SEQ ID NO 1) se da en la Figura 10.
La Figura 4B describe el constructo de minigén que codifica MAp19/sMAP humano que utiliza la región promotora del gen MASP-2 humano, con inclusión de los 3 primeros exones (exón 1 a exón 3) seguido por la secuencia de cDNA que representa la secuencia codificante de los dos exones siguientes, codificando por tanto la longitud total de MAp19. Este constructo de minigén, denominado mini hMAp19, se inyectó en huevos fertilizados de MASP-2-/-CS a fin de reemplazar MAp19 murino deficiente por MAp19 humano expresado transgénicamente. La secuencia del constructo MAp19 (SEQ ID NO 2) se da en la Figura 11.
La primera parte de la secuencia dada en ambas Figuras 10 y 11 representa el promotor de MASP-2 humano y los tres primeros exones, y es idéntica en ambos constructos. La segunda parte de cada secuencia representa la secuencia codificante de cDNA de los 8 exones siguientes (Figura 10) o los dos exones siguientes (Figura 11). La secuencia peptídica relevante se da también en la Figura 11, en tanto que la parte codificante de la Figura 10 se muestra en mayúsculas.
Un ratón desactivado MASP2-/-que expresa MASP2 humano para uso como modelo para someter a cribado con respecto a agentes inhibidores de MASP-2 puede producirse como sigue. Un ratón MASP2-/-como se ha descrito arriba y un ratón MASP2-/-que expresa un constructo transgénico de MASP2 humano (MASP2 humano desbloqueado) como se describe arriba se cruzan, y la progenie que son MASP-2-/-murino, MAp19+ murino, y MASP-2+ humano se utilizan para identificar agentes inhibidores de MASP-2 humano.
Tales modelos animales pueden utilizarse como sustratos de test para la identificación y eficacia de agentes inhibidores de MASP-2 tales como anticuerpos anti-MASP-2 humanos, péptidos y sustancias no peptídicas inhibidoras de MASP-2, y composiciones que comprenden agentes inhibidores de MASP-2. Por ejemplo, el modelo animal se expone a un compuesto o agente que se sabe desencadena la activación del complemento dependiente de MASP-2, y se administra al modelo animal un agente inhibidor de MASP-2 durante un tiempo y a una concentración suficientes para provocar una reducción de los síntomas de enfermedad en el animal expuesto.
Adicionalmente, pueden utilizarse ratones MASP-2-/-, MAp19+ murino, y MASP-2+ humano para generar linajes de células que contienen uno o más tipos de células implicados en una enfermedad asociada a MASP-2 que pueden utilizarse como modelo de cultivo de células para dicho trastorno. La generación de linajes de células continuas a partir de animales transgénicos es bien conocida en la técnica, por ejemplo, véase Small, et al., Mol. Cell Biol., 5: 642-48 (1985).
Los resultados de un experimento que demuestra que el sistema de activación del complemento de MASP-2 dependiente de lectina se activa en la fase de reperfusión después de reparación de un aneurisma aórticoabdominal se presentan en la Figura 8. Éste es un gráfico que ilustra el cambio en los niveles de MBL en pacientes que tienen lesiones isquémicas de reperfusión. Pacientes que sufren reparación de aneurisma aórtico abdominal (AAA) se someten a lesión isquémica de reperfusión, que está mediada en gran parte por activación del complemento. Los inventores investigaron el papel de la vía de lectina del complemento en la lesión isquemia-reperfusión en pacientes que se sometieron a reparación AAA.
Los pacientes a reparación AAA infrarrenal electiva se sometieron a extracción de muestras de sangre sistémica tomada de su arteria radial (por una vía arterial) en cuatro momentos definidos durante el procedimiento (momento 1: inducción de la anestesia, momento 2: inmediatamente antes del pinzamiento aórtico, momento 3: inmediatamente antes de la retirada del pinzamiento aórtico y momento 4: durante la reperfusión).
