Abstract

Die Erfindung betrifft Hydrogele, die aus Polyurethan(meth)acrylaten in Wasser mit wasserlöslichen Redoxinitiatoren hergestellt werden.

Description

Hvdrogele aus hydrophilen Polvurethan(meth)acrylaten
Die Erfindung betrifft Hydrogele, die aus Polyurethan(meth)acrylaten in Wasser mit wasserlöslichen Redoxinitiatoren hergestellt werden.
Hydrogele dienen der medizinischen Behandlung von Wunden und werden in Form von entsprechenden Wundauflagen insbesondere dort eingesetzt, wo das Feuchthalten einer Wunde zu einer Verbesserung der Wundheilung fuhrt (feuchte Wundbehandlung). Zur Herstellung von Hydrogelen werden typischerweise synthetische Polymere auf Basis von Poly(meth)acrylaten, Polyvinylpyrro- lidon oder Polyvinylalkohol eingesetzt. In der Regel zeichnen sich solche Hydrogele durch eine gute Verträglichkeit mit lebendem Gewebe aus.
Darüber hinaus bekannt sind Hydrogele aus Polyurethanen, welche durch Reaktion von hydrophilen, isocyanatfunktionellen Präpolymeren und einem großen Überschuss von Wasser hergestellt werden können, wie beispielsweise beschrieben in der EP-A 426 422, EP-A 455324, WO 9817215, WO 9913923 und WO2002060501. Der große Überschuss von Wasser ist notwendig, um eine Schaumbildung durch das freiwerdende Kohlendioxid, aus der Reaktion von Isocyanatgruppen mit Wasser, zu vermeiden. Umgekehrt heißt dies, dass Polyurethan-Hydrogele mit einem geringen Gehalt an anfänglich zugesetztem Wasser (blasenfrei) nicht zugänglich sind. Auf diese Weise hergestellte Polyurethan-Hydrogele können daher nur Wasser an eine (trockene) Wunde abgeben, wohingegen Wundflüssigkeit nur in sehr begrenztem Maße aufgenommen werden kann.
Die Herstellung von Polyurethan-Hydrogelen ist nach dem Stand der Technik bislang überdies ein langsamer Prozess, in welchen häufig drei Komponenten eingesetzt werden müssen, und bei welchem das resultierende Gel noch große Mengen an ungebundenem Polyol enthält. Bei der Herstellung werden zumeist aliphatische, isocyanatfunktionelle Präpolymere auf Basis eines Polyethylen- glykols, Polypropylenglykols oder von Glycerin als Polyol, teilweise in Gegenwart eines Be- schleunigers, z.B. Oligoalkylenoxide mit primären Aminoendgruppen, eingesetzt, d.h. dass für schneller reagierende Systeme drei Komponenten notwendig sind. Das Polyol wird dabei in deutlich überstöchiometrischen Mengen eingesetzt. Daraus folgt, dass die beschriebenen Hydrogele noch überschüssiges Polyol und Aktivator enthalten. Ferner sind die Reaktionszeiten sehr langsam, so dass der Gelpunkt oft erst nach 90 Minuten erreicht wird. In wie weit sich solche Gele mit an- timikrobiell wirksamen Substanzen additivieren lassen, ist bislang nicht beschrieben..
Femer sind Hydrogele bekannt, die durch radikalische Vernetzung hergestellt werden. Die GB-A 2086927 beschreibt semi-IPN-Hydrogele, die, durch Peroxide initiiert, bei erhöhter Temperatur durch Vernetzung niedermolekularer Polyacrylate in Gegenwart einer ethanolischen Lösung eines linearen Polyurethans hergestellt werden. Anschließend wird das Hüfslösungsmittel Ethanol entfernt.
In der GB-A 2131442 werden niedermolekulare Polyallylverbindungen und in der GB 2150938 Hydroxyethylmethacrylat und anderen Monoacrylate als externe, radikalisch polymerisierende Monomere beschrieben. Auch hier wurde mit einem Hilfslösungsmittel, wie Ethanol, und Peroxiden bei erhöhter Temperatur gearbeitet.
In der EP-A 351 364 werden Hydrogele beschrieben, die aus N,N-Dimethylacrylamid, fluorhalti- gen Polymeren und Vernetzern hergestellt werden, wobei auch mit ungesättigten Isocyanaten, wie MOI (Methacryloyloxyethylisocyanat) oder TMI (α,α-Dimethyl-3-isopropenyl-benzylisocyanat) umgesetzte Polyole, wie Polyvinylalkohol oder Triethylenglykol als Vernetzer eingesetzt werden können. Die Vernetzung erfolgte durch eine Initiierung mit UV oder Radikalstartern. Es gelten dieselben Nachteile, wie oben geschildert. Auch eine UV-Vernetzung ist im Körperkontakt kritisch (Reaktion mit der Haut, Augenschutz des Patienten und des medizinischen Personals).
US 2005 0271727 beschreibt über Redoxpolymerisation hergestellte Hydrogele, die aus Polyvinylalkohol und einem aufwendig nach Furch et al, Polymer,(1998) 39 (10), 1977-1982 aus HEMA und Glycolid hergestellten Vernetzer hergestellt werden.
Es bestand daher Bedarf an einem Polyurethan-Hydrogel, welches bei Bedarf unter Verwendung von nur geringen Mengen Wasser hergestellt werden kann und somit in Kontakt mit einer Wunde zusätzlich Wundflüssigkeit aufnehmen kann. Selbstverständlich soll das Polyurethan-Hydrogel weiterhin auch Wasser an (trockene) Wunden abgeben können. Da das Polyurethan-Hydrogel gegebenenfalls erst in einer Wunde oder einer anderen Körperöffnung (z.B. bei endoskopischen Eingriffen) gebildet werden soll, muß die Vernetzung ohne wesentliche Exothermie verlaufen. Der Einsatz von Strahlungshärtung (z.B. UV- Vernetzung) soll aufgrund des hohen Aufwandes vermie- den werden. Alle flüssigen Vorstufen wie auch das Polyurethan-Hydrogel selbst sollen eine gute Biokompatibilität aufweisen, dazu soll auf die Verwendung von organischen Lösungsmitteln verzichtet werden.
Es wurde nun gefunden, dass wässerige Lösungen von hydrophilen Polyurethanacrylaten mit Molmassen von mindestens 2000 g/mol durch ein Redoxsystem bereits bei Temperaturen von 10 bis 42°C vernetzbar sind, wobei der Wassergehalt des hergestellten Polyurethan-Hydrogels in weiten Grenzen eingestellt werden kann. Die Verwendung organischer Lösungsmittel oder monomerer Acrylate ist nicht notwendig; die erhaltenen Polyurethan-Hydrogele zeigen eine gute Biokompatibilität. Gegenstand der Erfindung sind daher ein Verfahren zur Herstellung von Polyurethan-Hydrogelen, bei dem
A) hydrophile Polyurethane mit olefϊnisch ungesättigten Gruppen in Gegenwart von
B) Wasser und
C) einem Redox-System, umfassend ein wasserlösliches Oxidationsmittel und wasserlösliches
Reduktionsmittel, wobei das Oxidationsmittel vom Redox-Potential her in der Lage ist mit dem wasserlöslichen Reduktionsmittel unter Radikalbildung zu reagieren,
radikalisch vernetzt werden.
Neben dem Verfahren zur Herstellung sind auch die so erhältlichen Hydrogele ein Gegenstand der Erfindung.
Die erfindungswesentlichen hydrophilen Polyurethane mit olefinisch ungesättigten Gruppen sind durch Umsetzung von Polyisocyanaten mit hydroxyfunktionellen Polyalkylenoxiden erhältlich.
Bevorzugt haben diese Polyalkylenoxide einen Ethylenoxidgehalt von mindestens 50 % bezogen auf die enthaltenen Oxalkyleneinheiten, besonders bevorzugt mindestens 60 %. Wenn nicht an- ders angegeben, beziehen sich alle Prozentangaben auf das Gewicht (hier und im folgenden).
Besonders bevorzugt handelt es sich bei diesen Polyalkylenoxiden um Copolymere aus Ethylen- oxid und Propylenoxid mit einem Ethylenoxidgehalt von 50 bis 100 %, bevorzugt 60 bis 82 %, gestartet auf Polyolen oder Aminen, wie Wasser (als Diol aufgefasst), Ethylenglykol, Propylengly- kol, Butylenglykol, Glycerin, TMP, Sorbit, Pentaerythrit, Triethanolamin, Ammoniak oder Ethy- lendiamin.
Die Molmassen dieser Polyalkylenoxide liegen bevorzugt bei Mn = 2.000 bis 20.000 g/mol, besonders bevorzugt 4000 bis 8500 g/mol .
Die OH-Funktionalität beträgt bevorzugt 2 bis 6, besonders bevorzugt 3 bis 6, ganz besonders bevorzugt 3 bis 4.
Als Polyisocyanate können zum einen olefinisch ungesättigte Isocyanate, wie MOI (Methacrylsäu- reisocyanatoethyleste, TMI (3-Isopropenyl-α,α-dimethyl-benzylisocyanat) oder Allylisocyanat eingesetzt werden.
Ebenfalls möglich ist, statt oder neben den olefinisch ungesättigten Di- oder Polyisocyanaten, gesättigte Di- oder Polyisocyanate einzusetzen und anschließend die olefinisch ungesättigten Grup- pen durch Umsetzung mit Verbindungen, die neben der ungesättigten Gruppe wenigstens eine isocyanatreaktive Gruppe aufweisen, einzuführen.
Dazu wird durch Reaktion einer 1,6 bis 30-fach äquivalenten Menge Diisocyanat bezogen auf die OH-Gruppen des Polyalkylenoxids ein Prepolymer hergestellt. Bevorzugt beträgt das NCO/OH- Verhältnis 4: 1 bis 12: 1 , besonders bevorzugt 2 : 1.
Gegebenenfalls können während der Herstellung Katalysatoren wie Amine oder Zinnverbindungen und/oder Stabilisatoren wie Benzoylchlorid, Isophthaloylchlorid, Dibutylphosphat, 3- Chlorpropionsäure oder Methyltosylat zugesetzt werden.
Die Reaktionstemperatur beträgt dabei 20 bis 1200C, bevorzugt 60 bis 1000C.
Überschüssiges, unreagiertes Isocyanat kann anschließend, bevorzugt mittels Dünnschichtdestillation, entfernt werden.
Geeignete gesättigte Diisocyanate entsprechen der allgemeinen Formel (I)
OCN-R-NCO (I),
wobei R für einen C4-C22-Alkylenrest, einen C3-C22-Cycloalkylenrest, einen C8-C22-Aralkylenrest, bei dem die mit der Isocyanatgruppe verknüpften Kohlenstoffatome sp3 -hybridisiert sind, oder einen C6-C18-Arylrest steht.
Beispiele für solche Diisocyanate sind HDI, IPDI, Bisisocyanatocyclohexylmethan, 2,2,4- Trimethylhexarnethylendiisocyanat, Bisisocyanatomethylcyclohexan, Bisisocyanatomethyltricyc- lodecan, Xylylendiisocyanat, Tetramethylxylylendiisocyanat, Norbornandiisocyanat, Cyclohexan- diisocyanat, Diisocyanatododecan, 2,4-TDI, 2,6-TDI, 2,2-, 2,4- oder 4,4-MDI, 4,4'-Diisocyanato- 3,3'-dimethylbiphenyl, 3,4'-Diisocyanatodiphenylether, 1,5-Naphthylendiisocyanat oder deren Gemische.
Jedoch sind die aliphatischen Diisocyanate der vorstehend genannten Art wegen der weitgehenden toxikologischen Unbedenklichkeit der Hydrolyseprodukte der aus ihnen resultierenden Polyu- rethane bevorzugt.
Anschließend werden die auf diese Weise erthaltenden isocyanatfunktionellen Präpolymere mit Verbindungen umgesetzt, die neben der ungesättigten Gruppe wenigstens eine isocyanatreaktive Gruppe aufweisen. Bevorzugte isocyanatreaktive Gruppen sind Amino- oder Hydroxyfunktionen, bevorzugt Hydroxyfunktionen. Das Equivalentverhältnis von NCO-Gruppen zu NCO-reaktiven Gruppen beträgt dabei bevorzugt 1,5 : 1 bis 1,0 : 1. Ein Überschuss an NCO-reaktiven Gruppen ist zu vermeiden, da sonst ungesättigte Monomere übrig bleiben. Bei einem Unterschuss an solchen Verbindungen werden verbliebene freie NCO-Gruppen durch Reaktion mit Wasser oder Monoolen, wie Methanol, Ethanol oder Isopropanol vor Applikation entfernt.
Bevorzugte Verbindungen mit olefϊnisch ungesättigten Gruppen und wenigstens einer isocya- natreaktiven Gruppe sind Hydroxyacrylate und -methacrylate wie Hydroxyethylacrylat, Hydroxye- thylmethacrylat, Hydroxypropylacrylat, Hydroxypropylmethacrylat, Hydroxybutylacrylat, Hydro- xybutylmethacrylat, Glycerindiacrylat, Glycerindimethacrylat, Glycerinmonoacrylatmonomethac- rylat, Glycerinmonoallylethermethacrylat, TMP = Trimethylolopropan-Diacrylat, TMP = Tri- methylolpropan-Dimethacrylat, Pentaerythrittriacrylat, Pentaerythrittrimethacrylat und deren beliebige Mischungen.
Die Umsetzung zu hydrophilen Polyurethanen mit olefϊnisch ungesättigten Gruppen erfolgt bei Temperaturen von 20 bis 800C, vorzugsweise in Gegenwart von Polymerisationsinhibitoren, wie Hydrochinon, Hydrochinonmonoalkylethern, Di-tert-butylkresol oder Methylen-bis-(tert-butyl- kresol) und von Katalysatoren der OH-NCO-Reaktion, wie Zinnverbindungen oder Aminen.
Unter wasserlöslichen Oxidationsmitteln werden Stoffe verstanden, die in der Lage sind, die unter wasserlösliche Reduktionsmittel aufgeführten Stoffe oder andere Stoffe unter Bildung von Radikalen zu oxidieren.
Als wasserlösliche Oxidationsmittel können Wasserstoffperoxid und seine anorganischen Salze, Peroxide, Hydroperoxide, wie tert-Butylhydroperoxid, Percarbonsäuren, wie Peressigsäure, Per- benzoesäure, Per-m-chlorbenzoesäure, Phthalmonopersäure, und deren Ammonium-, Alkali- oder Erdalkalisalze, anorganische Persäuren, vorzugsweise in Form ihrer Ammonium- oder Alkalisalze, wie Perborsäure, Perkohlensäure, Peroxodischwefelsäure, Peroxodiphosphorsäure, Carosche Säu- re, Periodsäure, Permangansäure, Perrheniumsäure, Halogene oder halogenabgebende Substanzen, Hypohalogenide, Natriumchlorit, Cer(IV)-Verbindungen, wie Cerammoniumnitrat, oder Metallionen in einer höheren Oxidationsstufe als der in Wasser stabilsten, wie Vanadium(V)- Verbindungen, Mangan(m)salze, Eisen(iπ)salze, Hexacyano(IH)ferrate, Kobalt(πi)salze, SiI- ber(I)salze, Kaliumbromat oder Oxone eingesetzt werden. Bevorzugt sind Ammonium-, Natrium- oder Kaliumperoxodisulfat.
Unter wasserlöslichen Reduktionsmitteln werden Stoffe verstanden, die in der Lage sind, die unter wasserlösliche Oxidationsmittel genannten Stoffe unter Bildung von Radikalen zu reduzieren. AIs wasserlösliche Reduktionsmittel können Ascorbinsäure, Isoascorbinsäure, Verbindungen, die vicinale OH-Gnippen enthalten, wie Glycerin oder Zuckeralkohole oder Zucker oder Oligo- oder Polysaccharide, reduzierende Zucker, wie Glukose, Formaldehyd, Glyoxal, Glyoxylsäure, Natriumsulfit, Ammoniumsulfit, Kaliumsulfit oder die entsprechenden Hydrogensulfite oder Bisulfϊte oder Metabisulfite, Sulfit-Aldehyd-Addukte, Natriumthiosulfat, Kaliumthiosulfat, Thioharnstoff, Hydrochinon, Pyrogallol, Gallussäure, Oxalsäure und deren Salze, Weinsäure und deren Salze, Malonsäure und deren Salze, Ameisensäure und deren Salze, Milchsäure und deren Salze, Ti- tan(πi)salicylaldoxim, Natriumhypophosphit, Derivate der Phosphorigen, Phosphonigen oder Phosphinigen Säure, Natriumtoluolsulfinat, Derivate der Sulfinigen Säure, wie Rongalit (Natrium- formaldehydsuloxylat), Natriumdithionit, Mercaptoethanol, Cystein und Cystein enthaltende Peptide, Cysteinamin, Thioglykolsäure, 3-Thiopropionsäure, Schwefeldioxid, NADH, Iodide, Ko- balt(ü)salze oder Ethylenthioharstoff eingesetzt werden. Bevorzugt ist Ascorbinsäure.
Zusätzlich zu den Oxidations- und Reduktionsmitteln können auch ein oder mehrere Aktivatoren mit verwendet werden. Bevorzugte Aktivatoren sind Übergangmetallsalze, die, wie Eisen, ihre Oxidationsstufe in ungeraden Schritten ändern können. Besonders bevorzugte Aktivatoren sind Eisensalze.
Bevorzugte Eisensalze sind Eisen(II)- oder Eisen(HT)-Salze, wie Eisen(II)chlorid, Ammoniumei- sen(E)sulfat, Eisen(II)sulfat, Eisen(m)sulfat, Eisen(IH)chlorid, Eisen(m)nitrat, Ei- sen(II)acetylacetonat, Kalium- oder Natrium- oder AmmoniumhexacyanoQOQferrat, Kalium- oder Natrium- oder Ammoniumhexacyano(III)ferrat, Natriumnitroprussiat, Eisen(II)-D-Gluconat, Ei- sen(II)lactat, Eisen(II)iodid, Eisen(II)perchlorat, Eisen(III)perchlorat, Eisen(II)tetrafluoroborat oder Eisen(III)tosylat, wobei zusätzlich Komplexliganden, wie Nitrilotriessigsäure oder EDTA zugesetzt werden können.
In einer bevorzugten Ausfuhrungsform werden in A) 1,0 Gewichtsteil des hydrophilen, ungesättig- ten Polyurethans, in B) 0,2 bis 19 Gewichtsteile Wasser sowie in C) als Redox-System jeweils bezogen auf die Summe der Mengen von A) und B) 0,05 bis 5 Gew.-%, bevorzugt 0,1 bis 2 Gew.- % des wasserlöslichen Oxidations- bzw. Reduktionsmittels eingesetzt.
Sofern ein Aktivator mit verwendet wird, so wird dieser in Mengen von 0,00001 bis 0,01 Gew.-%, bevorzugt 0,0001 bis 0,001 Gew.-% Übergangmetallsalz bezogen auf die Summe der Mengen von A) und B) zugesetzt.
Im erfindungsgemäßen Verfahren wird bevorzugt derart vorgegangen, dass das Oxidations- und das Reduktionsmittels jeweils getrennt von einander als wässrige Lösung bereit gestellt wird. In wenigstens einer der beiden Lösungen ist das zu vernetzende hydrophile Polyurethan dispergiert oder gelöst. Die Vernetzung wird durch Vereinigung beider Lösungen ggf. unter Zusatz eines Aktivators, der gelöst sein kann, initiiert.
Besonders bevorzugt enthält sowohl die Lösung des Oxidationsmittels als auch die des Reduktionsmittels hydrophiles Polyurethan in gelöster oder dispergierter Form.
Eine befriedigende Reaktionsgeschwindigkeit der Ausbildung des Hydrogels ist bereits unterhalb der Raumtemperatur zu beobachten. Bevorzugt wird die Hydrogelherstellung bei 5 bis 1000C, besonders bevorzugt 10 bis 42°C durchgeführt.
Zur Vermischung beider Lösungen können die an sich bekannten Mischtechniken verwandt werden.
Zusätzlich zu den Bildnern des Hydrogels können auch antibiotisch wirksame Stoffe zugegeben werden, wie sie in der WO 2002060501 beschrieben sind, wobei wasserlösliche Stoffe bevorzugt sind. Dabei ist auf die mögliche Wechselwirkung dieser Stoffe mit den Oxidations- oder Reduktionsmitteln zu achten, wobei eine gegenseitige Zerstörung der Komponenten möglich ist, die eine langfristige Lagerung vor der Applikation unmöglich macht. Jedoch können antibiotisch wirksame Stoffe auch die Rolle von Oxidations- oder Reduktionsmitteln ganz oder teilweise übernehmen, wie Peroxodisulfate, Wasserstoffperoxid, Permanganate, Silber(I)-Verbindungen, Sulfite oder Aldehyde.
Es können auch Verdicker zugesetzt werden, wie Polyvinylalkohol, Methyl-, Hydroxyethyl-, Hydroxypropyl- oder Carboxymethylether von Polysacchariden, wie Cellulose oder Stärke, PoIy- vinylpyrrolidon, Polyacrylsäure, Methylvinylether-MSA-Copolymere, anorganische Verdicker, wie Kieselsäuren, Aluminosilikate oder Aluminiumhydroxide, Polypeptide, Polysaccharide, wie Gummiarabikum oder Agar, Chitosan, Hyaluronsäure oder Polyurethanverdicker, wobei die Verdicker auch die Rolle von Reduktionmitteln ganz oder teilweise übernehmen können. Die Verdicker können schon vorab in die wässrigen Lösungen des hydrophilen Urethanacrylats eingebracht werden oder aber erst kurz vor der Reaktion zugesetzt werden.
Die erfϊndungsgemäßen Polyurethan-Hydrogele eignen sich z.B. als Wundauflagen. Dabei können die Polyurethan-Hydrogele entweder durch Vernetzung von einer oder mehreren der zuvor beschriebenen Komponente(n) direkt auf der Haut bzw. Wunde gebildet werden oder es folgt die Anwendung eines zuvor gefertigten Polyurethan-Hydrogels, typischerweise in Form einer flächi- gen Wundauflage.
Die erfindungsgemäßen Polyurethan-Hydrogele eignen sich weiterhin als Haftungsbarriere, welche das unerwünschte Zusammenwachsen von Organen verhindert („post surgical adhesion prevention, PSA"). Hierbei ist von besonderem Vorteil, dass das Polyurethan-Hydrogel aus einer oder mehreren zunächst flüssigen Komponente(n) hergestellt wird, und so insbesondere bei endoskopischen Eingriffen in den Körper dosiert werde kann. Nach der Applikation bildet sich das Polyurethan- Hydrogel durch die beschriebene Vernetzung.
Je nach Wahl der Polyol-Komponente des Polyurethans kann das daraus hergestellte Polyurethan- Hydrogel biostabil oder bioabbaubar sein. Insbesondere bei Verwendung als PSA, ist der Einsatz eines bioabbaubaren Polyurethan-Hydrogels bevorzugt.
Die Anwendung ist beim Menschen oder bei Tieren möglich.
Beispiele
Beispiel 1:
Man legte 93,75g eines Polyethers mit einem Ethylenoxidgehalt von 63% und einem Propyleno- xidgehalt von 37% gestartet auf TMP (3-funtionell), 0,0625g Methylen-bis-tert-butylkresol (BKF) und 0,0625g DBTL bei 6O0C vor, gab 10g Methacryloyloxyethylisocyanat hinzu, rührte 4h und ließ 4d bei Raumtemperatur stehen.
Dann wurden 10g dieses hydrophilen Urethanacrylats und 0,2 g Ammoniumperoxodisulfat in 40g Wasser sowie getrennt davon wiederum 10 g des hydrophilen Urethanacrylats und Ig Ascorbin- säure in 40 g Wasser, der lOμl einer 3 proz. Lösung von Eisen(II)chlorid hinzugefügt wurden, gelöst. Dann wurden die beiden Portionen unter kurzem Rühren vereint, wobei nach 5 min ein Hydrogel erhalten wurde.
Beispiel 2:
Man legte 200g HDI und Ig Benzoylchlorid vor, tropfte innerhalb von 2h bei 800C 400g eines Polyethers mit einem Ethylenoxidgehalt von 63% und einem Propylenoxidgehalt von 37% gestar- tet auf TMP (3-funtionell) hinzu und rührte Ih nach. Dann wurde durch Dünnschichtdestillation bei 13O0C und 0,1 Torr das überschüssige HDI abdestilliert. Man erhielt 420g eines Präpolymeren mit einem NCO-Gehalt von 2,75%. 153g dieses Präpolymeren, 0,1g BKF und 0,1g DBTL wurden bei 600C mit 21,4g Methacrylsäure-[3-(acryloyloxy)-2-hydroxypropylester] (Glycerinmonoacry- latmonomethacrylat) versetzt. Man ließ auf Raumtemperatur abkühlen und 4d stehen.
Dann wurden 10g dieses hydrophilen Urethanacrylats und 0,2 g Ammoniumperoxodisulfat in 40g Wasser sowie getrennt davon wiederum 10 g des hydrophilen Urethanacrylats und Ig Ascorbin- säure in 40 g Wasser, der lOμl einer 3 proz. Lösung von Eisen(IT)chlorid hinzugefügt wurden, gelöst. Dann wurden die beiden Portionen unter kurzem Rühren vereint, wobei nach 5 min ein Hydrogel erhalten wurde.
Beispiel 3:
Man versetzte 112g eines hexafunktionellen, auf Sorbit gestarteten Polyethylenoxids mit einer Molmasse von 6740 g/mol mit 0,1g BKF und 0,1g DBTL und fügte 15g MOI hinzu. Man ließ 7 Tage bei Raumtemperatur stehen.
Dann wurden 10g dieses hydrophilen Urethanacrylats und 0,2 g Ammoniumperoxodisulfat in 40g Wasser sowie getrennt davon wiederum 10 g des hydrophilen Urethanacrylats und Ig Ascorbin- säure in 40 g Wasser, der lOμl einer 3 proz. Lösung von Eisen(II)chlorid hinzugefügt wurden, gelöst. Dann wurden die beiden Portionen unter kurzem Rühren vereint, wobei nach 5 min ein Hydrogel erhalten wurde.
Beispiel 4:
Man legte 100g HDI und 2g Benzoylchlorid bei 800C vor, tropfte innerhalb von 2h 1000g eines tetrafunktionellen, auf Ethylendiamin gestarteten Polyethers mit einem Ethylenoxidgehalt von 75% und einem Propylenoxidgehalt von 25% und einer Molmasse von 4800g/mol hinzu. Man ließ noch
6h bei 800C nachrühren. Dann wurde der HDI-Überschuss durch Dünnschichtdestillation bei
1300C entfernt, wobei man ein Präpolymer mit einem NCO-Gehalt von 3,11% erhielt. 67,5g dieses
Präpolymeren wurde mit 0,1g BKF, 0,1g DBTL und 6,5g Hydroxyethylmethacrylat (HEMA) ver- setzt.
Dann wurden 10g dieses hydrophilen Urethanacrylats und 0,2 g Ammoniumperoxodisulfat in 40g Wasser sowie getrennt davon wiederum 10 g des hydrophilen Urethanacrylats und Ig Ascorbin- säure in 40 g Wasser, der lOμl einer 3 proz. Lösung von Eisen(II)chlorid hinzugefügt wurden, gelöst. Dann wurden die beiden Portionen unter kurzem Rühren vereint, wobei nach 5 min ein Hydrogel erhalten wurde.

Claims

Patentansprflche:
1. Verfahren zur Herstellung von Polyurethan-Hydrogelen, bei dem
A) hydrophile Polyurethane mit olefϊnisch ungesättigten Gruppen in Gegenwart von
B) Wasser und
C) einem Redox-System, umfassend ein wasserlösliches Oxidationsmittel und wasserlösliches
Reduktionsmittel, wobei das Oxidationsmittel vom Redox-Potential her in der Lage ist, mit dem wasserlöslichen Reduktionsmittel unter Radikalbildung zu reagieren,
radikalisch vernetzt werden.
2. Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, dass die hydrophilen Polyurethane auf hydroxyfunktionellen Polyalkylenoxiden basieren.
3. Verfahren gemäß Anspruch 2, dadurch gekennzeichnet, dass die hydroxyfunktionellen Polyal- kylenoxide einen Ethylenoxidgehalt von mindestens 60 % bezogen auf die enthaltenen Oxal- kyleneinheiten aufweisen.
4. Verfahren gemäß Anspruch 2 oder 3, dadurch gekennzeichnet, dass die hydroxyfunktionellen Polyalkylenoxide ein Molekulargewicht von Mn = 2.000 bis 20.000 g/mol und eine OH-
Funktionalität von 2 bis 6 aufweisen.
5. Verfahren gemäß einem der Ansprüche 2 bis 4, dadurch gekennzeichnet, dass die hydrophilen Polyurethane als olefmisch ungesättigte Gruppen Acrylat- oder Methacylat-Gruppen aufweisen.
6. Verfahren gemäß einem der Ansprüche 2 bis 5, dadurch gekennzeichnet, dass die hydrophilen Polyurethane auf aliphatischen oder cycloaliphatischen Diisocyanaten basieren.
7. Verfahren gemäß einem der Ansprüche 2 bis 6, dadurch gekennzeichnet, dass das Redox- System aus wenigstens einer Verbindung der Gruppe bestehend aus Ammonium-, Natriumoder Kaliumperoxodisulfat als Oxidationsmittel und Ascorbinsäure als Reduktionsmittel be- steht.
8. Verfahren gemäß einem der Ansprüche 2 bis 7, dadurch gekennzeichnet, dass neben dem Redox-System als Aktivatoren Eisensalze eingesetzt werden.
9. Verfahren gemäß einem der Ansprüche 2 bis 8, dadurch gekennzeichnet, dass in A) 1,0 Gewichtsteil des hydrophilen, ungesättigten Polyurethans, in B) 0,2 bis 19 Gewichtsteile Wasser sowie in C) als Redox-System bezogen auf die Summe der Mengen von A) und B) jeweils 0,05 bis 5 Gew.-% des wasserlöslichen Oxidations- bzw. Reduktionsmittels eingesetzt werden.
10. Verfahren gemäß einem der Ansprüche 2 bis 8, dadurch gekennzeichnet, dass ferner antibiotisch wirksame Stoffe zugesetzt werden.
11. Hydrogele erhältlich nach einem Verfahren gemäß einem der Ansprüche 1 bis 10.
12. Verwendung von Hydrogelen nach Anspruch 11 zur Herstellung von Auflagen für die Wundbehandlung.
