Abstract
Die vorliegende Erfindung betrifft ein Verfahren zur molekularen Charakterisierung von Altershaar in vitro sowie ein Haarbehandlungsmittel, enthaltend mindestens eine Verbindung, die die Expression mindestens eines der in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimierten Gene positiv oder negativ zu beeinflussen vermag.

Verfahren zur molekularen Charakterisierung von Altershaar
Beschreibung
Die vorliegende Erfindung betrifft ein Verfahren zur molekularen Charakterisierung von Altershaar sowie ein Haarbehandlungsmittel, enthaltend mindestens eine Verbindung, die die Expression mindestens eines der in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimierten Gene positiv oder negativ zu beeinflussen vermag.
Bisher sind die Veränderungen im gealterten Haar nur unzureichend untersucht. Bekannt sind die Reduktion der Haardichte und des Haardurchmessers sowie die Abnahme der Zellteilungsrate der Haarfollikelzellen. Die vorhandenen Daten basieren allerdings größtenteils auf empirischen Beobachtungen. Die Untersuchung der molekularen und zellphysiologischen Gründe für diese phänotypischen Veränderungen während des Alterungsprozesses sowie die Analyse von alters- und geschlechtsspezifischen Unterschieden fehlen weitestgehend. So ist der Fachwelt zwar das Expressionsprofil von gealterten Rattenschnurrhaaren bekannt, jedoch haben Untersuchungen gezeigt, dass die Übertragung der Ergebnisse auf den Menschen nur sehr eingeschränkt möglich ist. Grund hierfür ist, dass sich der Haarzyklus in Nagern deutlich von dem des Menschen unterscheidet. Der Haarzyklus in Nagern läuft synchronisiert und in Wellen ab. D.h. eine große Anzahl von beieinanderliegenden Haarfollikeln befindet sich im selben Stadium. Beim Menschen ist der Haarzyklus der einzelnen Haarfollikel eines Areals unsynchron isiert. Auch die Verteilung der Haare in den einzelnen Zyklusstadien unterscheidet sich bei Nager und Mensch. Während sich beim Menschen ca. 80 % der Haare immer im Anagen (Wachstumsphase) befinden, ist der überwiegende Anteil der Nagerhaare in der Telogenphase. Eine umfassende Charakterisierung von Altershaar ist bislang aus dem Stand der Technik nicht bekannt.
Bei den klassischen in-vivo-Studien werden nur einzelne, makroskopisch erkennbare Parameter untersucht (Haardichte, Haardurchmesser, Wachstumsrate, Anagen/Telogen Ratio). Effekte und altersbedingte Veränderungen auf zellulärer Ebene, die über den Parameter „Haarwachstum" hinausgehen, wie z.B. Haarstruktur, Metabolismus oder Pigmentierung, werden dabei nicht erfasst.
Das geringe Verständnis der molekularen Mechanismen, die bei der Follikelalterung bedeutsam sind, führt zu einer unzureichenden Anzahl von Targets, die für eine kosmetische oder pharmakologische Einflussnahme auf den Haarfollikel zur Verfügung stehen. Dies führt dazu, dass es nur wenige Produkte gibt, die das Problem der Haaralterung adressieren, wobei hier in den meisten Fällen
lediglich die Haarfaser im Fokus steht. So sind lediglich zwei Anmeldungen bekannt, in denen die Verwendung von Taurin und seinen Derivaten (FR 2841129) sowie von retinolhaltigen Formulierungen (EP 972511 ) beschrieben wird.
Haare besitzen neben ihrer eigentlichen physiologischen Aufgabe, wie Wärmeisolierung und Lichtschutz, eine nicht zu unterschätzende psychosoziale Funktion. Sie dienen unter anderem als Mittel der zwischenmenschlichen Kommunikation und stellen ein Zeichen der eigenen Individualität dar. Veränderungen, auch altersbedingte, im Haarwachstum können zu einer massiven Beeinträchtigung des Selbstbewusstseins der betroffenen Person führen.
Durch geeignete Wirkstoffformulierungen die Jugendlichkeit der Haare zu erhalten oder sie verjüngen zu können, ist eine Herausforderung für die kosmetische Forschung.
Es besteht daher ein Bedarf an der Identifikation möglichst vieler, vorzugsweise aller, für die altersrelevanten Veränderungen im menschlichen Haarfollikel wichtigen Gene.
Aufgabe der vorliegenden Erfindung ist es, einen möglichst großen Teil der für die altersrelevanten Veränderungen im menschlichen Haarfollikel bedeutsamen Gene zu identifizieren.
Diese Aufgabe wird erfindungsgemäß gelöst durch ein Verfahren zur molekularen Charakterisierung von menschlichem Altershaar, das dadurch gekennzeichnet ist, dass man a) ein erstes Gemisch von in Haarfollikeln älterer Menschen exprimierten, d. h. transkribierten und gegebenenfalls auch translatierten genetisch codierten Faktoren, also von Proteinen, mRNA- Molekülen oder Fragmenten von Proteinen oder mRNA-Molekülen aus Haarfollikeln älterer Menschen, gewinnt, b) ein zweites Gemisch von in Haarfollikeln junger Menschen exprimierten, d. h. transkribierten und gegebenenfalls auch translatierten genetisch codierten Faktoren, also von Proteinen, mRNA- Molekülen oder Fragmenten von Proteinen oder mRNA-Molekülen aus Haarfollikeln junger Menschen gewinnt und c) die in a) und b) gewonnenen Gemische einer differentiellen Analyse der Genexpression unterwirft, und dadurch die Gene identifiziert, die in Haarfollikeln älterer bzw. junger Menschen unterschiedlich stark (differentiell) exprimiert werden.
Einzelnen gezupften humanen Haarfollikeln haftet ausreichend biologisches Material an, um relevante Parameter, die gegebenenfalls der Follikelalterung unterliegen, wie z.B. Haarwachstum, - metabolismus, -struktur sowie -pigmentierung auf zellulärer Ebene zu erfassen. Das anhaftende biologische Material besteht aus Zellen der äußeren Bindegewebsscheide (CTS-Fibroblasten),
Keratinozyten der äußeren und inneren Wurzelscheide (ORS- und IRS-Keratinozyten), Matrixmelanozyten sowie Matrixkeratinozyten. Die ebenfalls im Haarfollikel angesiedelte dermale Papille verbleibt beim Zupfen in der Kopfhaut, so dass an dieser Stelle ein neues Haar gebildet werden kann.
Makroskopische, altersbedingte Veränderungen im Längenwachstum, in der Haarstruktur, der Haardichte sowie der Färbung des Haares sind unmittelbare Folgen einer Modulation von Genen, Proteinen und Stoffwechselprodukten in den Haarfollikelzellen. Durch die Analyse und Quantifizierung der auftretenden zellulären Effekte können somit Rückschlüsse auf die zu erwartenden makroskopischen Veränderungen des Haares, wie z.B. Längenwachstum, gezogen werden, bevor sich diese im sichtbaren Haarschaft manifestieren.
Darüber hinaus können durch moderne molekularbiologische und proteinbiochemische Verfahren verschiedenste Endpunkte bestimmt werden, die über die klassischen Parameter wie Längen- und Dickenwachstum hinausgehen. So kann beispielsweise die Expression von speziellen Haarkeratinen, die die Haarstruktur beeinflussen, mittels quantitativer PCR auf Transkriptionsebene untersucht werden. Auch der Einfluss einer Testsubstanz auf Marker, die relevant für die Pigmentierung sind (z.B. gp100, TRP1 ) sowie auf haarzyklussteuernde Wachstumsfaktoren (z.B. HGF, KGF) und ihre Rezeptoren (z.B. c-met) lassen sich mit der vorgeschlagenen Methode direkt in vivo bestimmen.
Mittels verschiedener proteinanalytischer Methoden wie beispielsweise Westernblot oder Proteinarray können die haarrelevanten Marker auch auf Proteinebene bestimmt und alterabhängige Effekte auf das Proteinexpressionsmuster untersucht werden. Durch die molekulare Analyse von älteren Haarfollikeln gegenüber jungen Haarfollikeln kann eine umfassende Charakterisierung der altersbedingten Veränderungen im Haarfollikel durchgeführt werden und es können so neue Targets und Signalwege identifiziert werden, die die Follikelalterung entscheidend beeinflussen.
Durch das erfindungsgemäße Verfahren wird es vorteilhafterweise möglich, den komplexen Prozess der Haaralterung und die kausalen Zusammenhänge der Veränderungen am gealterten Haar zu begreifen. Nur mit diesem Wissen können neue Konzepte für kosmetische Haar-Produkte entwickelt werden, die ihre Wirkung auf das breite Spektrum der Genexpression im gealterten Haarfollikel ausüben. Die im Rahmen des erfindungsgemäßen Verfahrens durchgeführte differentielle Analyse zeigt erstmals in umfassender Weise, welche Gene in Altershaar exprimiert werden, und welche Gene dort anders exprimiert werden als in jungem Haar.
Erst durch die systematische Identifizierung der im Alter modifizierten zellulären Prozesse wird die gezielte Entwicklung biologisch aktiver Haarpflegeprodukte, die genau auf die Bedürfnisse des älteren Haarfollikels abgestimmt sind und so den Zeichen der Follikelalterung entgegenwirken, ermöglicht.
Vorteilhafterweise ermöglicht das erfindungsgemäße Verfahren somit erstmals eine umfassende systematische Identifikation und Clusterung (Zusammenfassung in Pathways) der altersrelevanten Veränderungen im Haarfollikel.
Vorzugsweise erhält man die in den Schritten a) und b) zu gewinnenden Gemische aus Haarfollikeln behaarter Kopfhaut.
Es ist erfindungsgemäß bevorzugt, dass man die Untersuchung in Schritt c) mittels einer Methode durchführt, die ausgewählt ist unter i. Ein- oder zweidimensionaler Gelelektrophorese ii. Affinitätschromatographie iii. Protein-Protein-Komplexierung in Lösung iv. ELISA (Enzyme-linked Immunosorbent Assay) v. enzymgekoppelte Stoffwechselassays vi. Massenspektrometrie, insbesondere Matrix Assistierter Laser Desorptions
Ionisation (MALDI), vii. Einsatz von Proteinchips, Northern Blots, viii. Real Time Quantitative Polymerasekettenreaktion (RT-PCR), ix. Multiplex-PCR, x. RNase-Schutzexperimente, xi. Dot-Blots, xii. CDNA-Sequenzierung, xiii. Klon-Hybridisierung, xiv. Differential Display, xv. Subtraktive Hybridisierung, xvi. cDNA-Fragment-Fingerprinting, xvii. Durchflusszytometrieanalysen, xviii. Serielle Analyse der Genexpression (SAGE) und insbesondere xix. Einsatz von Nukleinsäurechips, • oder mittels geeigneter Kombinationen dieser Methoden.
Im Rahmen der vorliegenden Erfindung sind unter „älteren Menschen" insbesondere solche in einem Alter von über 50 Jahren und unter „jungen Menschen" insbesondere solche in einem Alter von unter 25 Jahren zu verstehen.
Zur Erfassung der differentiellen Genexpression älterer bzw. junger Haarfollikel können die dem Fachmann bekannten Methoden eingesetzt werden, beispielsweise die Technik der „Seriellen Analyse der Genexpression" (SAGE™). Diese Technik erlaubt gleichzeitig die Identifikation und Quantifizierung aller in älteren bzw. jungen Haarfollikeln exprimierten Gene. Die Analyse der Genexpression ist auch mit der Quantifizierung spezifischer mRNA-Moleküle möglich (z.B. Northern-Blot, RNase- Schutzexperimente). Außerdem können die Techniken MPSS (Massive Parallel Signiture Sequencing) oder Techniken, die auf Differential display beruhen, erfindungsgemäß eingesetzt werden.
Diese erfindungsgemäß einsetzbaren Methoden sind in den Übersichtsartikeln von Akhilesh Pandey und Matthias Mann: „Proteomics to study genes and genomes", Nature, Volume 405, Number 6788, 837 - 846 (2000), und „Genombs, gene expression and DNA arrays", Nature, Volume 405, Number 6788, 827 - 836 (2000), und den dort angegebenen Referenzen beschrieben, worauf hiermit in vollem Umfang Bezug genommen wird.
Zur Erfassung der differentiellen Genexpression älterer bzw. junger Haarfollikel kann erfindungsgemäß auch die Analyse mittels Ganzgenom-Microarray eingesetzt werden. Insbesondere wurde für die vorliegenden Untersuchungen der Whole Human Genome Microarray der Firma Agilent eingesetzt. Dieser umfasst ca. 34.000 Gene.
Die 2D-Gelelektrophorese wird beispielsweise in L. D. Adams, Two-dimensional Gel Electrophoresis using the Isodalt System oder in L. D. Adams & S. R. Es können jedoch erfindungsgemäß auch andere dem Fachmann bekannte Methoden zur Erfassung der differentiellen Genexpression älterer bzw. junger Haarfollikel eingesetzt werden.
Die folgenden Beispiele erläutern die Erfindung, ohne sie jedoch darauf einzuschränken.
Beispiele:
Beispiel 1 : Haarkeratine
Ein altersabhängiges Phänomen ist das Dünnerwerden und die gesteigerte Fragilität der Haare. Dabei ist die Festigkeit der Haare und die Haarstruktur im Wesentlichen von der Zusammensetzung spezieller haarspezifischer Strukturproteine abhängig, den Haarkeratinen. Durch die altersbedingte Veränderung der Zusammensetzung dieser spezifischen Proteine wird auf biologischer Ebene Einfluss auf die Haarstruktur genommen.
Die Expression verschiedener Haarkeratine in gezupften Haarfollikel von Probanden unterschiedlicher Altersgruppen (<25 und > 50 Jahre) kann mit Hilfe eines quantitativen Real-Time-PCR-Verfahrens untersucht werden. Zur Durchführung der PCR wird zunächst mit Hilfe des RNeasy Mini Kits der Fa. Qiagen die RNA aus 15 Haarfollikeln isoliert und mittels reverser Transkription in cDNA umgeschrieben. Bei der anschließenden PCR Reaktion, die mit Hilfe genspezifischer Primer für die jeweiligen Haarkeratine durchgeführt wird und die der Amplifikation der gesuchten Genabschnitte dient, wird die Bildung der PCR-Produkte online über ein Fluoreszenzsignal detektiert. Das Fluoreszenzsignal ist dabei proportional zur Menge des gebildeten PCR-Produktes. Je stärker die Expression eines bestimmten Gens ist, desto größer ist die Menge an gebildetem PCR-Produkt und um so höher ist das Fluoreszenzsignal.
Sowohl die weibliche als auch die männliche (jeweils junge und alte) Probandengruppe wurde in jeweils 3 Pools unterteilt, wobei die Expressionsänderung im Alter durch Normierung auf die entsprechende junge Kontrollgruppe berechnet wurde.
Gezeigt sind in Tabelle 1 die durchschnittlichen Expressionen der Probandenpools. Die jeweils dritte Spalte dokumentiert die Unterschiede zwischen jungen und älteren Probanden.

Die Ergebnisse verdeutlichen, dass insbesondere die Keratine hHa3-l und hHa4 bei beiden Geschlechtern eine starke Abnahme der Genexpression im Laufe des Alterungsprozesses zeigen.
Beispiel 2: Apoptose
Unter Apoptose versteht man den programmierten Zelltod. Er manifestiert sich zunächst in morphologischen Veränderungen, wie dem Schrumpfen der Zellen und Ausbildung von Membraneinstülpungen; weiterhin zeigt sich eine Fragmentierung der chromosomalen DNA und es kommt letztlich zum Absterben und Abbau der betroffenen Zellen. Im Haarfollikel ist insbesondere die Regressionsphase (Katagen) von apoptotischen Prozessen in den Keratinozyten geprägt. Hier leitet die Apoptose den Übergang von der Wachstumsphase in die Regressionsphase ein. Darüber hinaus sind jedoch mit voranschreitendem Alter auch Melanozyten von Apoptose betroffen, was zum Ergrauen des entsprechenden Follikels führt.
Zentrale Regulatoren der Apoptose sind die zur Familie der Cysteinproteasen gehörende Caspasen.
Die Untersuchungen der Genexpression in Haarfollikeln unterschiedlicher Altersgruppen (<25 und > 50 Jahre) und unterschiedlichen Geschlechts mit Hilfe eines cDNA-Microarrays und quantitativer PCR zeigen ein im Alter gesteigertes Maß an proapoptotischen Signalmolekülen wie das der Caspasen 2, 7 und 9.
Hierbei ist insbesondere die weibliche Probandengruppe betroffen. Die hochregulierten Caspasen kann man in zwei Gruppen einteilen, zum einen in Initiator- und zum anderen in Effektor-Caspasen. Zu den Initiatoren des Zelltods zählen die Caspasen 2 und 9, die hier am stärksten reguliert sind. Eine
Aktivierung der Caspase 2 führt zu DNA-Schädigungen während Caspase 9 als kritisches Element für die intrazelluläre Vervielfältigung apoptotischer Signale anzusehen ist.
Ausgelöst durch die Signaltransduktion geschädigter Mitochondrien aktiviert sie die Caspase 7, die als Effektor-Caspase Substrate spaltet, die wiederum die morphologischen Veränderungen des Apoptoseprozesses auslösen. Darüber hinaus ist der Tumor Nekrosefaktor TNFSF11 bei den Frauen signifikant hochreguliert. Seine Bindung an den Membranrezeptor bewirkt über ASK1 eine Stimulierung von JNK (ebenfalls hochreguliert) und nimmt damit Einfluss auf die Mitochondrien- Schädigung. Relevante Marker hierfür sind die Gene Bax und Bcl-2. Sie wirken beide an der Mitochondrienmembran, haben aber unterschiedliche Effekte. Bax ist ein apoptoseförderndes und BcI- 2 ein apoptosehemmendes Signalmolekül.
Die Tabelle 2 zeigt die Ergebnisse der Untersuchung differenzieller Genexpression in jungen und älteren Follikeln von apoptose-assoziierten Genen (w50 weiblich>50 Jahre, w25 weiblich<25Jahre, m50 männlich >50 Jahre, m25 männlich <25 Jahre)

Beispiel 3: MAPK
Mitogenaktivierte Proteinkinasen (MAPK) gehören zur Familie der Serin/Threonin Proteinkinasen und sind an einer Vielzahl von Zellfunktionen wie Proliferation, Differenzierung und Zelltod beteiligt. Ihre Signalkaskaden sind hierarchisch in drei Stufen eingeteilt. Die MAPK-Aktivierung erfolgt durch MAPK- Kinasen (MAPKK), die ihrerseits wiederum von MAPKK-Kinasen (MAPKKK) aktiviert werden. Auslöser für diese Kettenreaktion sind äußere Stimuli wie Mitogene, Wachstumsfaktoren oder inflammatorische Cytokine
In Abbildung 1 ist die Signalkaskade mitogenaktivierter Protein kinasen (MAPK) dargestellt. Von einem äußeren Reiz ausgehend (z.B. Wachstumsfaktor), erfolgt die Aktivierung der MAPK über MAPK- Kinasen (MAPKK) und MAPKK-Kinasen (MAPKKK). Je nach Signalweg werden unterschiedliche biologische Antworten wie Wachstum, Differenzierung oder Zelltod ausgelöst. Hellgrau: keine Genregulation bei beiden Geschlechtern. Dunkelgrau: erhöhte Genexpression bei Frauen über 50 Jahren.
Bei der Probandinnengruppe über 50 Jahre wurde in gezupften Follikeln eine gesteigerte Genexpression von verschiedenen mitogenaktivierten Proteinkinasen nachgewiesen. Die Expression von MAPK 8 (=JNK1 ) ist bei den Frauen um den Faktor 3,5 und die von MEKK4 um den Faktor 1 ,5 im Vergleich zu jungen Follikeln erhöht. MAPK 14 (=p38) und MAPK 11 (=p38ß) weisen ebenfalls eine tendenzielle Zunahme auf. Die bei den Frauen angesprochenen Gene sind charakteristisch für die mit Nummer zwei und drei bezeichneten Signalwege, wohingegen Signalweg eins (über Raf-Proteine, MEK1/2 und ERK1/2) keine Regulationen aufweist.
Die wichtige Rolle der MAP-Kinasen liegt in ihrer Fähigkeit, Matrixmetalloproteinasen (MMP) zu aktivieren. MMPs sind Enzyme, deren Hauptaufgabe die proteolytische Degradation von dermalen Strukturproteinen (z.B. Kollagen) ist. Für die Haare ist dies von erheblicher Bedeutung, da Kollagen ein Matrixprotein der dermalen Papille und der Dermal Sheath ist. Kollagen unterliegt weiterhin Veränderungen während des Haarzykluses, sodass ihm möglicherweise auch eine Kontrollfunktion diesbezüglich zuzuschreiben ist.
Neben den MAP-Kinasen ist auch der epidermale Wachstumsfaktor EGF für die MMP-Aktivierung verantwortlich. Seine Expression ist bei der älteren Probandinnengruppe ebenfalls erhöht.

Interleukine sind von den Zellen des Immunsystems gebildete Glycoproteine, die als chemische Botenstoffe zwischen den Zellen fungieren. Basierend auf einer Ligand-Rezeptor-Wechselwirkung vermögen sie Signale ins Zellinnere zu vermitteln und bestimmte Antworten der Zielzellen herbeizuführen. Nach ihren Wirkungen unterscheidet man entzündungsauslösende (proinflammatorische) oder entzündungsabschwächende (antiinflammatorische) Interleukine. Interleukin 6 ist ein klassischer Vertreter der proinflammatorischen Interleukine. Es ist Vermittler der so genannten Akutphaseantwort und kann als Entzündungsmarker eingestuft werden. Dieser Reaktion entgegen wirkt die Ausschüttung von IL14, welches die Proliferation von B-Zellen induziert. Ihre Immunglobulinsekretion wird unterdrückt und sie entwickeln sich bevorzugt zu B-Gedächtniszellen.
Von besonderem Interesse bei entzündlichen Vorgängen der Haare könnte das erst kürzlich entdeckte Interleukin22 sein: Es konnte gezeigt werden, dass es ausschließlich auf Gewebezellen einwirkt und hier die unspezifische Abwehrkraft fördert. Sein Rezeptor (IL22R) kommt besonders auf solchen Zellen vor, die in ständigem Kontakt zur Außenwelt stehen und wird ebenfalls vermehrt bei Entzündungsprozessen gebildet. Die übermäßige Produktion von IL22 regt Hautzellen an, körpereigene Abwehrstoffe, so genannte Defensine zu produzieren. Überraschenderweise werden die Defensine in den gealterten Follikeln der weiblichen Probandengruppe heraufreguliert. Die Ergebnisse deuten darauf zwar darauf hin, dass die Haarfollikelalterung (zumindest bei Frauen), analog zur Hautalterung, als chronischer Entzündungsprozess verstanden werden kann. Jedoch ist bewerkenswert, wie die Abwehrleistung gegenüber eindringenden Keimen mit zunehmendem Alter aufrechterhalten bzw. noch verstärkt wird. Die Vorstellung, dass das in den Alterungsprozess einbezogene Immunsystem mit einer abnehmenden Abwehrfähigkeit pathologischer Prozesse assoziiert ist, trifft demnach auf das komplexe Gebilde des Haarfollikels nicht zu.
Beispiel 5: Cytokeratine
Die differenzielle Genexpression in gezupften Haarfollikeln unterschiedlicher Altersgruppen (<25 und > 50 Jahre) und unterschiedlichen Geschlechts wurde mit Hilfe eines cDNA-Microarrays und quantitativer PCR analysiert.

Cytokeratine stellen eine große Familie von Proteinen dar, deren Hauptaufgabe der Aufbau und Erhalt der Zellform ist. Sie bilden ein filamentöses Netzwerk im Cytoplasma und tragen damit zur Stabilität der Zelle bzw. des gesamten Gewebeverbandes bei. Cytokeratin 1 (CK1 ) ist bei beiden Geschlechtern im Alter stark herunterreguliert. Bei dem das Cytokeratin 19 (CK19) kodierende Gen ist diese Regulation ebenfalls zu verzeichnen, hier jedoch nur bei den älteren Frauen. Die Expression von CK19 bleibt bei den Männern im Alter konstant. Die verringerte Expression des Cytokeratin 1 , welches charakteristisch für differenzierende Epithelzellen ist, könnte auf eine Störung in der Differenzierung des epithelialen Gewebes des Haarfollikels im Alter hindeuten. Da CK19 als Stammzellmarker
diskutiert wird könnte seine Unterexpression auf eine gestörte Neubildung der Haarfollikelzellen hinweisen, was eine Verlangsamung des Haarzyklus zur Folge hätte.
Beispiel 6: Proliferation
Ein Beispiel für die kompensatorischen Fähigkeiten des Haarfollikels, um sich vor altersbedingte Veränderungen zu schützen zeigen verschieden proliferations-assoziierte Marker. Diese Marker wurden mit Hilfe eines cDNA Microarrays und Western-Blot untersucht. Zum zeigt der cDNA Microarray eine deutliche Abnahme von Cyclin E bei beiden älteren Probandengruppen. Cycline sind die zentralen Elemente bei der Kontrolle der Zellteilung. Sie regulieren Cyclinabhängige Kinasen (Cdk), eine weitere Gruppe von Proteinen, die durch Phosphorilierung die enzymatischen Aktivitäten ihrer Substrate ein- oder ausschalten können. So werden, je nach Vorhandensein unterschiedlicher Cycline, verschiedene Prozesse des Zellzyklus eingeleitet. Cyclin E reguliert dabei den Übergang von der Ruhephase G1 in die Synthese-Phase. Da ein hohes Level an Cyclin E den Übergang von der G1- Phase in die Synthesephase beschleunigt, könnte im Umkehrschluss eine stark verminderte Expression dieses Gens auf ein vermehrtes Verbleiben der Follikelzellen in der Ruhephase hinweisen. Damit würden mit zunehmendem Alter weniger neue Zellen produziert. Dem entgegen steht die im Alter insbesondere bei der männlichen Probandengruppe gesteigerte Expression von HGF (Hepatocyte Growth Factor), einem Wachstumsfaktor, der die Proliferation der Matrixkeratinozyten im Haarfollikel stimuliert. Dem entsprechend unverändert ist die Expression von PCNA, einem Protein, dass ausschließlich in proliferierenden Zellen lokalisiert ist. Dies zeigt, dass trotz einer verringerten Cytokeratin 14- und Cyclin E- Expression der Haarfollikel eine Strategie entwickelt hat, diesen Aspekt des Alterungsprozesses zu kompensieren, um weiterhin seine regenerativen Eigenschaften innerhalb des Haarzyklus zu bewahren.

Die S100 Proteine stellen eine weitere Gruppe von inflammationsassoziierten Proteinen dar. Sie werden verstärkt bei Entzündungsprozessen gebildet, was den Zusammenhang von Alterung und chronischen Inflammationen unterstützt. Es ist bekannt, dass die proinflammatorischen Moleküle S100A7, S100A8 und S100A9 durch IL22 hochreguliert werden. Dieses passt zu der oben beschriebenen Regulation des IL22-Rezeptors, sofern man von der Überexpression des IL22- Rezeptors auf vermehrt vorhandenes IL22 schließen kann. Im Alterungsprozess spielen die S100 Proteine noch eine weitere Rolle. S100A8 und S100A9 können ein Heterodimer formen, welches als Ligand an den „Altersrezeptor" RAGE (Receptor of Advanced Glycation End-Products) binden kann. Glykierungsendprodukte (AGE) sind nicht mehr abbaubare langkettige Verbindungen, die aus der Reaktion von Eiweißen mit Zuckermolekülen (Maillard-Reaktion) resultieren. Sie ist eine dauerhafte Modifikation und bewirkt, dass Proteine ihre normalen Funktionen im Körper verlieren. Während des normalen Alterungsprozesses wird eine verstärkte AGE-Bildung beobachtet, was wiederum einen Anstieg ihrer Rezeptoren (RAGE) auf nahezu allen Körperzellen zur Folge hat (Bierhaus und Nawroth, 2002). Kommt es zu einer Bindung von AGE an den Rezeptor, so erfolgt eine RAGE-vermittelte
Aktivierung von MAP-Kinasen und es kommt zur Ausschüttung von proinflammatorischen Cytokinen. Neben den klassischen RAGE-Liganden wurde diese Wirkung auch bei dem Heterodimer aus S100A8/S100A9 als Bindungsmolekül beobachtet.
Beispiel 8: cDNA Microarray Daten
Mit Hilfe eine cDNA Microarrays wurden >850 haarrelevante Marker untersucht und die differentielle Genexpression in gezupften Haarfollikeln unterschiedlicher Altersgruppen (<25 und > 50 Jahre) und unterschiedlichen Geschlechts untersucht.

Beispiel 11 : Ganzgenomchip
Um die altersbedingten Veränderungen im Haarfollikel umfassend zu charakterisieren, sowie zusammenhängende Pathways und Parallelen bzw. Alterungsschutzmechanismen zu identifizieren wurde die Genexpression mit Hilfe eines Ganzgenomchips untersucht.
Die so erhobenen Daten wurden mittels Pathway-Analyse weiterprozessiert. Dabei wurden die differentiell exprimierten Gene zunächst geclustert und dann hinsichtlich ihrer upstream- und downstream-lnteraktionapartner mit Hilfe einer speziellen Software (Interaction Explorer Software PathwayAssist) analysiert. Diese Analyse umfasst die automatische Suche nach Stichwörtern in Abstract- und Volltextdatenbanken. Die so identifizierten Interaktionen zwischen bestimmten Gengruppen werden anschliessend noch hinsichtlich ihrer Relevanz überprüft. Diese Pathway-Analyse ergab ein Set aus zusammenhängenden Regulationen.

Signalwege die neben kit auch Pax3 und WNT11 umfassen, sind insbesondere für die Pigmentierung beschrieben. Überraschenderweise konnte durch die Ganzgenomanalyse auch seine Bedeutung für Ergrauungs-unabhängige Phänomene nachgewiesen werden, da für die Analyse nur voll pigmentierte Follikel von unterdurchschnittlich ergrauten Probanden miteinbezogen wurden.
Die Ganzgenomanalyse ergab weiterhin, dass nicht nur wie in Beispiel 1 beschrieben die Expression von Haarkeratinen altersabhängig ist, sondern auch die Expression einer Reihe von keratin-
assoziierten Proteinen (s. Tabelle 13). Dies zeigt, das insbesondere die Keratinsynthese von der Follikelalterung beeinflusst wird.

Auch wenn die Bedeutung einzelner Faktoren für die Regulation des Haarzyklus bereits diskutiert wird, ist dieses Netzwerk ist in seiner Gesamtheit für die altersabhängigen Veränderungen im Haarfollikel noch nicht beschrieben worden.
Ein weiterer Gegenstand der vorliegenden Erfindung ist ein Haarbehandlungsmittel, enthaltend mindestens einen Wirkstoff, der die Expression mindestens eines der in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimierten Gene positiv oder negativ zu beeinflussen vermag.
Erfindungsgemäß bevorzugte Gene sind die in den Tabellen 1 bis 15 als in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimiert aufgelisteten Gene. Besonders bevorzugte Gene sind die als hochrelevant und/oder hochsignifikant in Gruppe 1 der Tabelle 8 aufgelisteten Gene.
Die Quotienten in den Tabellen 1 bis 15 geben die Stärke der differentiellen Expression an, d. h., um welchen Faktor das jeweilige Gen in jungen Haarfollikeln stärker exprimiert wird, als in alten Haarfollikeln, oder umgekehrt.
Unter ihrer UniGene-Accession-Number sind die jeweiligen Gene bzw. Genprodukte in der Datenbank des National Center for Biotechnology Information (NCBI) offenbart. Diese Datenbank ist im Internet unter folgender Adresse zugänglich: http://www.ncbi.nlm.nih.gov/.

Das erfindungsgemäße Mittel kann zusätzlich Proteinhydrolysate umfassen, vorzugsweise kationisierte Proteinhydrolysate, wobei das zugrunde liegende Proteinhydrolysat vom Tier, beispielsweise aus Collagen, Milch oder Keratin, von der Pflanze, beispielsweise aus Weizen, Mais, Reis, Kartoffeln, Soja oder Mandeln, von marinen Lebensformen, beispielsweise aus Fischcollagen oder Algen, oder biotechnologisch gewonnenen Proteinhydrolysaten, stammen kann. Die den kationischen Derivaten zugrunde liegenden Proteinhydrolysate können aus den entsprechenden Proteinen durch eine chemische, insbesondere alkalische oder saure Hydrolyse, durch eine enzymatische Hydrolyse und/oder einer Kombination aus beiden Hydrolysearten gewonnen werden. Die Hydrolyse von Proteinen ergibt in der Regel ein Proteinhydrolysat mit einer Molekulargewichtsverteilung von etwa 100 Dalton bis hin zu mehreren tausend Dalton. Bevorzugt sind solche kationischen Proteinhydrolysate, deren zugrunde liegender Proteinanteil ein Molekulargewicht von 100 bis zu 25000 Dalton, bevorzugt 250 bis 5000 Dalton aufweist. Weiterhin sind unter kationischen Proteinhydrolysaten quaternierte Aminosäuren und deren Gemische zu verstehen. Die Quaternisierung der Proteinhydrolysate oder der Aminosäuren wird häufig mittels quarternären Ammoniumsalzen wie beispielsweise N,N-Dimethyl-N- (n-Alkyl)-N-(2-hydroxy-3-chloro-n-propyl)-ammoniumhalogeniden durchgeführt. Weiterhin können die kationischen Proteinhydrolysate auch noch weiter derivatisiert sein. Obwohl das erfindungsgemäße Mittel prinzipiell auf dem Haar verbleiben kann, wird das Mittel vorzugsweise nach einer Einwirkzeit von 10 Sekunden bis 60 Stunden ausgespült. Dieses Ausspülen kann mit reinem Wasser oder einem marktüblichen Shampoo erfolgen. Einwirkzeiten von 1 bis 15 Minuten haben sich in den meisten Fällen als ausreichend erwiesen.
Unabhängig von dem genauen Ablauf der Behandlung hat es sich als vorteilhaft erwiesen, das erfindungsgemäße Mittel bei einer Temperatur von 20 bis 55 0C, insbesondere von 35 bis 400C, anzuwenden.
Hinsichtlich der Art, gemäß das erfindungsgemäße Mittel auf das Haar, aufgebracht wird, bestehen keine prinzipiellen Einschränkungen.
Erfindungsgemäß von besonderem Interesse sind Haartonics, insbesondere als leave on Formulierung. Diese werden vorzugsweise bei Raumtemperatur angewendet, der alkoholische Gehalt liegt bevorzugtermaßen im Bereich von etwa 30 % bis etwa 35 % und der pH-Wert sollte etwa bei pH 7 liegen.
Als Konfektionierung der das erfindungsgemäße Mittel enthaltenden Zubereitungen sind beispielsweise Cremes, Lotionen, Lösungen, Wässer, Emulsionen wie W/O-, O/W-, PIT-Emulsionen (Emulsionen nach der Lehre der Phaseninversion, PIT genannt), Mikroemulsionen und multiple Emulsionen, Gele, Sprays, Aerosole und Schaumaerosole geeignet. Diese werden in der Regel auf wäßriger oder wäßrig-alkoholischer Basis formuliert. Als alkoholische Komponente kommen dabei niedere Alkanole sowie Polyole wie Propylenglykol und Glycerin zum Einsatz. Ethanol und Isopropanol sind bevorzugte Alkohole. Wasser und Alkohol können in der wäßrig alkoholischen Basis in einem Gewichtsverhältnis von 1 : 10 bis 10 : 1 vorliegen. Wasser sowie wäßrig-alkoholische Mischungen, die
bis zu 50 Gew.-%, insbesondere bis zu 25 Gew.-%, Alkohol, bezogen auf das Gemisch Alkohol/Wasser, enthalten, können erfindungsgemäß bevorzugte Grundlagen sein. Der pH-Wert dieser Zubereitungen kann prinzipiell bei Werten von 2 - 11 liegen. Er liegt bevorzugt zwischen 2 und 7, wobei Werte von 3 bis 5 besonders bevorzugt sind. Zur Einstellung dieses pH-Wertes kann praktisch jede für kosmetische Zwecke verwendbare Säure oder Base verwendet werden. Üblicherweise werden als Säuren Genußsäuren verwendet. Unter Genußsäuren werden solche Säuren verstanden, die im Rahmen der üblichen Nahrungsaufnahme aufgenommen werden und positive Auswirkungen auf den menschlichen Organismus haben. Genußsäuren sind beispielsweise Essigsäure, Milchsäure, Weinsäure, Zitronensäure, Äpfelsäure, Ascorbinsäure und Gluconsäure. Im Rahmen der Erfindung ist die Verwendung von Zitronensäure und Milchsäure besonders bevorzugt. Bevorzugte Basen sind Ammoniak, Alkalihydroxide, Triethanolamin sowie N,N,N',N'-Tetrakis-(2-hydroxypropyl)-ethylendiamin.
Das Mittel kann als Einkammersystem oder als Zweikammersystem konfektioniert werden.
Neben der erfindungsgemäß zwingend erforderlichen Verbindung, die die Expression mindestens eines der in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimierten Gene positiv oder negativ zu beeinflussen vermag, kann das Mittel prinzipiell alle weiteren, dem Fachmann für solche kosmetischen Mittel bekannten Komponenten enthalten.
Weitere Wirk-, Hilfs- und Zusatzstoffe sind beispielsweise

Das erfindungsgemäße Mittel kann außerdem Tenside enthalten. Bei diesen kann es sich sowohl um anionische, ampholytische, zwitterionische oder nichtionogene Tenside als auch um kationische Tenside handeln.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung wird, beispielsweise in einem Shampoo, eine Kombination aus anionischen und nichtionischen Tensiden oder eine Kombination aus anionischen und amphoteren Tensiden eingesetzt. In einem Haartonic kann der Fachmann jedoch auch weitgehend oder vollständig auf den Einsatz von Tensiden verzichten.
Es hat sich in Einzelfällen als vorteilhaft erwiesen, die Tenside aus amphoteren oder nichtionischen Tensiden auszuwählen.
Als anionische Tenside eignen sich in erfindungsgemäßen Mitteln alle für die Verwendung am menschlichen Körper geeigneten anionischen oberflächenaktiven Stoffe. Diese sind gekennzeichnet durch eine wasserlöslich machende, anionische Gruppe wie z. B. eine Carboxylat-, Sulfat-, Sulfonat- oder Phosphat-Gruppe und eine lipophile Alkylgruppe mit etwa 10 bis 22 C-Atomen. Zusätzlich können im Molekül Glykol- oder Polyglykolether-Gruppen, Ester-, Ether- und Amidgruppen sowie Hydroxylgruppen enthalten sein.
Nichtionogene Tenside enthalten als hydrophile Gruppe z. B. eine Polyolgruppe, eine Po- lyalkylenglykolethergruppe oder eine Kombination aus Polyol- und Polyglykolethergruppe. Solche Verbindungen sind beispielsweise
Anlagerungsprodukte von 2 bis 30 Mol Ethylenoxid und/oder 0 bis 5 Mol Propylenoxid an lineare
Fettalkohole mit 8 bis 22 C-Atomen, an Fettsäuren mit 12 bis 22 C-Atomen und an Alkylphenole mit 8 bis 15 C-Atomen in der Alkylgruppe,
Ci2-C22-Fettsäuremono- und -diester von Anlagerungsprodukten von 1 bis 30 Mol Ethylenoxid an
Glycerin,
C8-C22-Al kylmono- und -oligoglycoside und deren ethoxylierte Analoga sowie
Anlagerungsprodukte von 5 bis 60 Mol Ethylenoxid an Rizinusöl und gehärtetes Rizinusöl.
Bevorzugte nichtionische Tenside sind Alkylpolyglykoside der allgemeinen Formel R1O-(Z)x. Diese Verbindungen sind durch die folgenden Parameter gekennzeichnet.
Der Alkylrest R1 enthält 6 bis 22 Kohlenstoffatome und kann sowohl linear als auch verzweigt sein. Bevorzugt sind primäre lineare und in 2-Stellung methylverzweigte aliphatische Reste. Solche Alkylreste sind beispielsweise 1-Octyl, 1-Decyl, 1-Lauryl, 1-Myristyl, 1 -Cetyl und 1-Stearyl. Besonders bevorzugt sind 1-Octyl, 1-Decyl, 1-Lauryl, 1-Myristyl. Bei Verwendung sogenannter "Oxo-Alkohole" als Ausgangsstoffe überwiegen Verbindungen mit einer ungeraden Anzahl von Kohlenstoffatomen in der Al ky I kette.
Die erfindungsgemäß verwendbaren Alkylpolyglykoside können beispielsweise nur einen bestimmten Alkylrest R1 enthalten. Üblicherweise werden diese Verbindungen aber ausgehend von natürlichen Fetten und Ölen oder Mineralölen hergestellt. In diesem Fall liegen als Alkylreste R Mischungen entsprechend den Ausgangsverbindungen bzw. entsprechend der jeweiligen Aufarbeitung dieser Verbindungen vor.
Besonders bevorzugt sind solche Alkylpolyglykoside, bei denen R1 im wesentlichen aus C8- und CiO-Alkylgruppen, im wesentlichen aus C12- und C14-Alkylgruppen, im wesentlichen aus C8- bis C16-Alkylgruppen oder im wesentlichen aus C12- bis C16-Alkylgruppen besteht.
Als Zuckerbaustein Z können beliebige Mono- oder Oligosaccharide eingesetzt werden. Üblicherweise werden Zucker mit 5 bzw. 6 Kohlenstoffatomen sowie die entsprechenden Oligosaccharide eingesetzt. Solche Zucker sind beispielsweise Glucose, Fructose, Galactose, Arabinose, Ribose, Xylose, Lyxose,
Allose, Altrose, Mannose, Gulose, Idose, Talose und Sucrose. Bevorzugte Zuckerbausteine sind Glucose, Fructose, Galactose, Arabinose und Sucrose; Glucose ist besonders bevorzugt.
Die erfindungsgemäß verwendbaren Alkylpolyglykoside enthalten im Schnitt 1 ,1 bis 5 Zuckereinheiten. Alkylpolyglykoside mit x-Werten von 1 ,1 bis 1 ,6 sind bevorzugt. Ganz besonders bevorzugt sind Alkylglykoside, bei denen x 1 ,1 bis 1 ,4 beträgt.
Die Alkylglykoside können neben ihrer Tensidwirkung auch dazu dienen, die Fixierung von Duftkomponenten auf dem Haar zu verbessern. Der Fachmann wird also für den Fall, dass eine über die Dauer der Haarbehandlung hinausgehende Wirkung des Parfümöles auf dem Haar gewünscht wird, bevorzugt zu dieser Substanzklasse als weiterem Inhaltsstoff der erfindungsgemäßen Zubereitungen zurückgreifen.
Auch die alkoxylierten Homologen der genannten Alkylpolyglykoside können erfindungsgemäß eingesetzt werden. Diese Homologen können durchschnittlich bis zu 10 Ethylenoxid- und/oder Propylenoxideinheiten pro Alkylglykosideinheit enthalten.
Weiterhin können, insbesondere als Co-Tenside, zwitterionische Tenside verwendet werden. Als zwitterionische Tenside werden solche oberflächenaktive Verbindungen bezeichnet, die im Molekül mindestens eine quartäre Ammoniumgruppe und mindestens eine -COO( )- oder -SO3 ( )-Gruppe tragen. Besonders geeignete zwitterionische Tenside sind die sogenannten Betaine wie die N-Alkyl-N,N-dime- thylammonium-glycinate, beispielsweise das Kokosalkyl-dimethylammonium-glycinat, N-Acyl- aminopropyl-N,N-dimethylammoniumglycinate, beispielsweise das Kokosacylaminopropyl-dime- thylammoniumglycinat, und 2-Alkyl-3-carboxylmethyl-3-hydroxyethyl-imidazoline mit jeweils 8 bis 18 C- Atomen in der Alkyl- oder Acylgruppe sowie das Kokosacylaminoethylhydroxyethylcarboxymethylgly- cinat. Ein bevorzugtes zwitterionisches Tensid ist das unter der INCI-Bezeichnung Cocamidopropyl Betaine bekannte Fettsäureamid-Derivat.
Ebenfalls insbesondere als Co-Tenside geeignet sind ampholytische Tenside. Unter ampholytischen Tensiden werden solche oberflächenaktiven Verbindungen verstanden, die außer einer C8-C18-Al kyl- oder Acylgruppe im Molekül mindestens eine freie Aminogruppe und mindestens eine -COOH- oder - SO3H-Gruppe enthalten und zur Ausbildung innerer Salze befähigt sind. Beispiele für geeignete ampholytische Tenside sind N-Alkylglycine, N-Alkylpropionsäuren, N-Alkylaminobuttersäuren, N- Alkyliminodipropionsäuren, N-Hydroxyethyl-N-alkylamidopropylglycine, N-Alkyltaurine, N- Alkylsarcosine, 2-Alkylaminopropionsäuren und Alkylaminoessigsäuren mit jeweils etwa 8 bis 18 C- Atomen in der Alkylgruppe. Besonders bevorzugte ampholytische Tenside sind das N-Kokosalkyl- aminopropionat, das Kokosacylaminoethylaminopropionat und das C12.18-Acylsarcosin.
Erfindungsgemäß werden als kationische Tenside insbesondere solche vom Typ der quartären Ammoniumverbindungen, der Esterquats und der Amidoamine eingesetzt.
Bevorzugte quaternäre Ammoniumverbindungen sind Ammoniumhalogenide, insbesondere Chloride und Bromide, wie Alkyltrimethylammoniumchloride, Dialkyldimethylammoniumchloride und Trialkyl- methylammoniumchloride, z. B. Cetyltrimethylammoniumchlorid, Stearyltrimethylammoniumchlorid, Di- stearyldimethylammoniumchlorid, Lauryldimethylammoniumchlorid, Lauryldimethylbenzylammonium- chlorid und Tricetylmethylammoniumchlorid, sowie die unter den INCI-Bezeichnungen Quaternium-27 und Quaternium-83 bekannten Imidazolium-Verbindungen. Die langen Alkylketten der oben genannten Tenside weisen bevorzugt 10 bis 18 Kohlenstoffatome auf.
Bei Esterquats handelt es sich um bekannte Stoffe, die sowohl mindestens eine Esterfunktion als auch mindestens eine quartäre Ammoniumgruppe als Strukturelement enthalten. Bevorzugte Esterquats sind quaternierte Estersalze von Fettsäuren mit Triethanolamin, quaternierte Estersalze von Fettsäuren mit Diethanolalkylaminen und quaternierten Estersalze von Fettsäuren mit 1 ,2- Dihydroxypropyldialkylaminen. Solche Produkte werden beispielsweise unter den Warenzeichen Stepantex®, Dehyquart® und Armocare® vertrieben. Die Produkte Armocare® VGH-70, ein N,N-Bis(2- Palmitoyloxyethyl)dimethylammoniumchlorid, sowie Dehyquart® F-75 und Dehyquart® AU-35 sind Beispiele für solche Esterquats.
Die Alkylamidoamine werden üblicherweise durch Amidierung natürlicher oder synthetischer Fettsäuren und Fettsäureschnitte mit Dialkylaminoaminen hergestellt. Eine erfindungsgemäß besonders geeignete Verbindung aus dieser Substanzgruppe stellt das unter der Bezeichnung Tegoamid® S 18 im Handel erhältliche Stearamidopropyl-dimethylamin dar.
Bei den als Tensid eingesetzten Verbindungen mit Alkylgruppen kann es sich jeweils um einheitliche Substanzen handeln. Es ist jedoch in der Regel bevorzugt, bei der Herstellung dieser Stoffe von nativen pflanzlichen oder tierischen Rohstoffen auszugehen, so dass man Substanzgemische mit unterschiedlichen, vom jeweiligen Rohstoff abhängigen Alkylkettenlängen erhält.
Bei den Tensiden, die Anlagerungsprodukte von Ethylen- und/oder Propylenoxid an Fettalkohole oder Derivate dieser Anlagerungsprodukte darstellen, können sowohl Produkte mit einer "normalen" Homologenverteilung als auch solche mit einer eingeengten Homologenverteilung verwendet werden. Unter "normaler" Homologenverteilung werden dabei Mischungen von Homologen verstanden, die man bei der Umsetzung von Fettalkohol und Alkylenoxid unter Verwendung von Alkalimetallen, Al- kalimetallhydroxiden oder Alkalimetallalkoholaten als Katalysatoren erhält. Eingeengte
Homologenverteilungen werden dagegen erhalten, wenn beispielsweise Hydrotalcite, Erdalkalimetallsalze von Ethercarbonsäuren, Erdalkalimetalloxide, -hydroxide oder -alkoholate als Katalysatoren verwendet werden. Die Verwendung von Produkten mit eingeengter Homologenverteilung kann bevorzugt sein.
Bezüglich weiterer fakultativer Komponenten sowie die eingesetzten Mengen dieser Komponenten wird ausdrücklich auf die dem Fachmann bekannten einschlägigen Handbücher, z. B. Kh. Schrader, Grundlagen und Rezepturen der Kosmetika, 2. Auflage, Hüthig Buch Verlag, Heidelberg, 1989, verwiesen.

Claims
1. Verfahren zur molekularen Charakterisierung von menschlichem Altershaar, dadurch gekennzeichnet, dass man a. ein erstes Gemisch von in Haarfollikeln älterer Menschen exprimierten, d. h. transkribierten und gegebenenfalls auch translatierten genetisch codierten Faktoren, also von Proteinen, mRNA-Molekülen oder Fragmenten von Proteinen oder mRNA-Molekülen aus Haarfollikeln älterer Menschen, gewinnt, b. ein zweites Gemisch von in Haarfollikeln junger Menschen exprimierten, d. h. transkribierten und gegebenenfalls auch translatierten genetisch codierten Faktoren, also von Proteinen, mRNA-Molekülen oder Fragmenten von Proteinen oder mRNA-Molekülen aus Haarfollikeln junger Menschen gewinnt und c. die in a) und b) gewonnenen Gemische einer differentiellen Analyse der Genexpression unterwirft, und dadurch die Gene identifiziert, die in Haarfollikeln älterer oder junger Menschen unterschiedlich stark exprimiert werden.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass man die Untersuchung in Schritt c) mittels einer Methode durchführt, die ausgewählt ist unter a) Ein- oder zweidimensionaler Gelelektrophorese b) Affinitätschromatographie c) Protein-Protein-Komplexierung in Lösung d) ELISA (Enzyme-linked Immunosorbent Assay) e) enzymgekoppelte Stoffwechselassays f) Massenspektrometrie, insbesondere Matrix Assistierter Laser Desorptions Ionisation (MALDI), g) Einsatz von Proteinchips, Northern Blots, h) Real Time Quantitative Polymerasekettenreaktion (RT-PCR), i) Multiplex-PCR, j) RNase-Schutzexperimente, k) Dot-Blots,
I) CDNA-Sequenzierung, m) Klon-Hybridisierung, n) Differential Display, o) Subtraktive Hybridisierung, p) cDNA-Fragment-Fingerprinting, q) Durchflusszytometrieanalysen, r) Serielle Analyse der Genexpression (SAGE) und insbesondere s) Einsatz von Nukleinsäurechips, • oder mittels geeigneter Kombinationen dieser Methoden.
3. Haarbehandlungsmittel, enthaltend mindestens einen Wirkstoff, der die Expression mindestens eines der in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimierten Gene positiv oder negativ zu beeinflussen vermag.
4. Haarbehandlungsmittel nach Anspruch 3, dadurch gekennzeichnet, dass die zu beeinflussenden Gene ausgewählt sind unter den in den Tabellen 1 bis 15 als in älteren Haarfollikeln und in jungen Haarfollikeln differentiell exprimiert aufgelisteten Genen.
5. Haarbehandlungsmittel nach Anspruch 3 oder 4, dadurch gekennzeichnet, dass die zu beeinflussenden Gene ausgewählt sind unter den als hochrelevant und/oder hochsignifikant in Gruppe 1 der Tabelle 8 aufgelisteten Genen.
