Abstract
Die vorliegende Erfindung betrifft anorganische monolithische Formkörper, deren Oberfläche mit physi- oder chemisorbierten organischen Polymeren beschichtet ist, sowie Verfahren zur Herstellung derartiger Materialien. Die erfindungsgemässen Materialien eignen sich hervorragend als Sorbenzien für die Chromatographie, insbesondere für die Hochdruck flüssigkeitschromatographie von biologischen Materialien.
Description
Mit organischen Polymeren beschichtete anorganische monolithische
Formkörper
Die vorliegende Erfindung betrifft anorganische monolithische Formkörper, deren Oberfläche mit physi- oder chemisorbierten organischen Polymeren beschichtet ist, sowie Verfahren zur Herstellung derartiger Materialien. Die erfindungsgemäßen Materialien eignen sich hervorragend als Sorbenzien für die Chromatographie, insbesondere für die Hochdruckflüssigkeitschromatographie von biologischen Materialien.
Chromatographische Trägermaterialien bzw. Sorbenzien, die zur Auftrennung oder Reinigung von biologischen Materialien, wie Proteinen, Nukleinsäuren etc. eingesetzt werden, müssen neben guten Trenneigenschaften eine hohe Alkalistabilität aufweisen. Grund dafür sind insbesonde- re spezielle Reinigungs- und Sterilisationsverfahren, denen die Sorbenzien unterworfen werden.
Beispielsweise wird bei dem sogenannten clean in place Verfahren das Sorbens in Abhängigkeit von der Säulendimension über einen Zeitraum von 10 Minuten bis zu drei Stunden mit 1 M Natronlauge behandelt. Um eine
Verkeimung zu verhindern, werden die Trägermaterialien für eine Langzeitlagerung in 0,1 M Natronlauge aufbewahrt. Nicht alle Trägermaterialien sind unter derartigen Bedingungen stabil. Beispielsweise sind anorganische Trägermaterialien auf Basis von Siliziumdioxid unter den genannten Bedingungen oft nicht ausreichend stabil.
Aus diesem Grund werden in der Biochromatographie häufig organische Polymere, wie Dextran, Agarose, Cellulose, Polystyrol oder Methacrylat- ester, oder mit derartigen organischen Polymeren beschichtete anorgani- sehe partikuläre Materialien als Trägermaterialien eingesetzt.
Beispiele für Polymer-beschichtete anorganische Partikel finden sich z.B. in US 4,308,254 oder US 5,271 ,833. US 4,308,254 offenbart anorganische poröse Partikel, z.B. aus Silika, Aluminium-, Magnesium- oder Titan-Oxid, die mit Polysaccharid-Polymer beschichtet sind. US 5,271 ,833 offenbart anorganische Oxid-Partikel, die mit einem organischen Polymer umgeben sind.
Nachteil der rein organischen Polymere ist eine häufig ungünstige Porenstruktur und daraus resultierend eine zu geringe spezifische Oberfläche. Zudem quellen insbesondere wenig vernetzte Polymere in bestimmten Lösungsmitteln stark auf.
Bei Polymer-beschichteten Partikeln können diese Nachteile nur bis zu einem gewissen Grad durch den stabilen anorganischen Kern behoben werden. Insbesondere müssen zur Gewährleistung einer ausreichend hohen Trennleistung zumeist recht kleine Partikel eingesetzt werden, was zu einer starken Erhöhung des Säulenrückdrucks führt. Insbesondere bei hohem Druck lagern sich die Teilchen relativ dicht zusammen. Dabei wird die flexible Polymerschicht, vor allem durch den unflexiblen anorganischen Kern stark deformiert und teilweise sogar zerstört. Auch das führt natürlich zu einer erheblichen Verschlechterung der Trennleistung und der Standzeiten der Säule.
Einen weiteren Ansatz offenbart DE 199 29 073. Hier werden die Poren eines porösen anorganischen Stabs vollständig mit einer Polymerphase aus untereinander verbundenen kugelförmigen Teilchen gefüllt. Zwar lassen sich Formkörper, deren Poren vollständig mit Polymer gefüllt, relativ leicht herstellen, sie zeigen aber durch die vollständig mit Polymerkugeln gefüllten Poren nur eine mäßige Eignung für chromatographische Anwendungen. Zum einen haben die Polymerkugeln in den Kanälen des
Monolithen natürlich dieselben Eigenschaften wie Polymerkugeln, die direkt in eine Säule gepackt sind, und somit auch dieselben Nachteile. Zum
anderen ist die Gefahr groß, dass gerade kleinere Kanäle völlig verstopfen und so ungleichmäßige Druckverhältnisse im Monolithen entstehen, die wiederum zu einer Verschlechterung der Trenneigenschaften führen.
Es wurde nun gefunden, dass die oben genannten Nachteile durch den
Einsatz von Polymer-beschichteten anorganischen monolithischen Formkörpern behoben werden können. Die anorganischen porösen monolithischen Formkörper werden dazu gleichmäßig mit einer Beschichtung aus organischem Polymer überzogen. Durch die vorgegebene rigide Struktur des monolithischen Materials wird eine Verformung oder Zerstörung des
Sorbens auch bei hohem Druck verhindert. Zudem können durch geeignete Wahl der Porenstruktur des anorganischen Formkörpers und der Beschich- tungsverfahren Sorbenzien hergestellt werden, die selbst bei hohen Flussraten nur einen moderaten Säulenrückdruck zeigen.
Gegenstand der vorliegenden Erfindung sind daher poröse anorganische monolithische Formkörper, die mit mindestens einem organischen Polymer beschichtet sind.
In einer bevorzugten Ausführungsform handelt es sich bei den porösen anorganischen monolithischen Formkörpern um Materialien auf Basis von Si02.
In einer besonders bevorzugten Ausführungsform weisen die porösen anorganischen monolithischen Formkörper eine bimodale Porenstruktur mit Mesoporen mit einem Durchmesser zwischen 2 und 100 nm und Makroporen mit einem mittleren Durchmesser von über 0,1 μm auf.
In einer weiteren bevorzugten Ausführungsform handelt es sich bei den organischen Polymeren um Polystyrole oder Poly(meth)acrylate, sowie andere Poly(meth)acrylsäurederivate wie z.B. Poly(meth)acrylamidderivate.
In einer bevorzugten Ausführungsform ist das organische Polymer auf dem anorganischen Formkörper physisorbiert.
Gegenstand der vorliegenden Erfindung ist zudem ein Verfahren zur Her- Stellung von porösen anorganischen monolithischen Formkörpern, die mit mindestens einem organischen Polymer beschichtet sind, das folgende Verfahrensschritte aufweist: a) Bereitstellen eines porösen anorganischen monolithischen Formkörpers b) Tränken des porösen anorganischen monolithischen Materials aus Schritt a) mit einer Beschichtungslösung enthaltend zumindest organische Präpolymere oder Mono- und/oder Oligomere. c) Beschichtung des Formkörpers, wobei der Formkörper während der Beschichtung zumindest an den Längsseiten mit einem inerten Material dicht ummantelt ist oder in einem inerten Lösungsmittel gelagert wird d) Waschen und Trocknen des Formkörpers aus Schritt c) zur Entfernung von Reaktionsrückständen und Lösungsmittel
In einer bevorzugten Ausführungsform werden in Schritt c) die Präpolymere oder Monomere und/oder Oligomere aus der Beschichtungslösung auf den anorganischen Formkörper aufgefällt.
In einer besonders bevorzugten Ausführungsform erfolgt die Ausfällung durch Temperaturerniedrigung.
Gegenstand der vorliegenden Erfindung ist auch die Verwendung der erfindungsgemäßen Formkörper zur chromatographischen Trennung mindestens zweier Substanzen, insbesondere zur Trennung und/oder Aufreinigung von biologischen Materialien.
Formkörper im Sinne der vorliegenden Erfindung sind insbesondere Formkörper, die als Einzelstück für chromatographische Trennungen eingesetzt werden können und nicht wie Partikel in großer Zahl in eine
Säuie gefüllt werden. Insbesondere sind dies flache oder säulenförmige Formkörper. Flache Formkörper haben ihre größte Ausdehnung quer zur Durchflussrichtung des Elutionsmittels. Besonders bevorzugt werden säulenförmige Formkörper, die eine gleich lange oder längere Ausdehnung entlang der Achse haben, die von den Elutionsmitteln durchströmt wird.
Die Größe und Dimension der Formkörper entspricht den in der zum Einsatz in der Chromatographie üblichen Dimensionen. Flache Formkörper haben typischerweise eine Dicke zwischen 0,2 und 20 μm, säulenförmige Formkörper haben typischerweise einen Durchmesser zwischen 0,1 cm und 5 cm und eine Länge (längste Ausdehnung) zwischen 1 und 30 cm. Für präparative Trennungen können die angegebenen Dimensionen entsprechend der Dimensionen bekannter Säulendimensionen überschritten werden. Für miniaturisierte Anwendungen können die angegebenen Dimensionen bis in den Bereich von Kapillaren unterschritten werden.
Die porösen anorganischen monolithischen Formkörper bestehen typischerweise aus anorganischen Oxiden, wie Aluminiumoxid, Titandioxid oder bevorzugt Siliziumdioxid.
Von großer Bedeutung für die Eignung der erfindungsgemäßen beschichteten Formkörper als chromatographische Sorbenzien ist die Porenstruktur der anorganischen Formkörper. Sie beeinflusst zum einen, wie homogen die Polymer-Beschichtung auf den Formkörper aufgebracht werden kann. Zum anderen hat sie Einfluss auf die Trennleistung und den Säulenrückdruck der beschichteten Formkörper. Erfindungsgemäß geeignete Formkörper weisen zumindest Poren auf, die auch nach der Beschichtung noch ein Durchströmen des Formkörpers ermöglichen. Bevorzugt sind daher Materialien, die zumindest Makroporen aufweisen.
Als besonders vorteilhaft haben sich Materialien mit einer bimodalen Porenverteilung erwiesen, d.h. Formkörper, die sowohl Makro- als auch Meso- poren aufweisen. Durch die zusätzlichen Mesoporen gelingt es, bei gerin-
gern Flüssigkeitsgegendruck eine höhere Oberfläche zur Verfügung zu stellen.
Besonders bevorzugt werden erfindungsgemäß daher anorganische poröse monolithische Formkörper eingesetzt, die Makroporen mit einem mittleren
Durchmesser von über 0,1 μm aufweisen sowie Mesoporen in den Wänden der Makroporen, wobei die Mesoporen einen Durchmesser zwischen 2 und 100 nm haben. Derartige Materialien können beispielsweise nach einem Sol-Gel Prozess entsprechend WO 95/03256 und besonders bevorzugt entsprechend WO 98/29350 hergestellt werden.
Zur Beschichtung der Formkörper geeignete organische Polymere sind organische Materialien, die als Oligomer und/oder Polymer auf den Formkörper aufgebracht werden können oder organische Oligomere und/oder Monomere, die durch Polymerisation oder Polykondensation auf den
Formkörper aufgebracht werden. Die organischen Polymere können auf dem Formkörper chemi- oder physisorbiert sein.
Geeignete organische Polymere sind beispielsweise Polystyrole, Polymeth- acrylate, Melamine, Polysaccharide, Polysiloxane sowie deren Derivate oder Copolymere zweier oder mehrerer geeigneter Verbindungen, wie beispielsweise eine Beschichtung aus Tetraalkoxysilan und Methyltri- alkoxysilan. Geeignet sind auch Copolymere der zuvor genannten Stoffe mit Monomeren, die bereits für die Chromatographie geeignete Separa- tionseffektoren tragen, wie z.B. Copolymerisate aus Polystyrolen mit Verbindungen, die lonenaustauschergruppen tragen. Bevorzugt sind chemi- oder physisorbierte Polystyrole oder Polystyrol-Derivate, besonders bevorzugt sind physisorbierte Poly(meth) acrylate oder Poly(meth)acrylat- Derivate, sowie andere Poly(meth)acrylsäurederivate wie z.B. Poly(meth)acrylamidderivate. Dazu gehören insbesondere Poly(meth- acrylat), Poly(2-hydroxyethylmethacrylat), ein Copolymer aus 2-
Hydroxyethylmethacrylat und Ethylmethacrylat oder Poly(octadecyl- methacryiat).
Die Polymerbeschichtung kann erfindungsgemäß auf verschiedene Weise erfolgen: 1) durch Polymerisation bzw. Polykondensation von physisorbierten
Monomeren und/oder Oligomeren ohne Ausbildung von kovalenten
Bindungen zum anorganischen Formkörper
2) durch Polymerisation bzw. Polykondensation von physisorbierten Monomeren und/oder Oligomeren mit Ausbildung von kovalenten Bindungen zum anorganischen Formkörper
3) durch Immobiiisation (Physisorption) von Präpolymeren ohne Ausbildung von Bindungen zum anorganischen Formkörper
4) durch Chemisorption von Präpolymeren auf den anorganischen Formkörper. Eine Lösung, die für die erfindungsgemäße Beschichtung der Formkörper eingesetzt wird, enthält demnach entweder organische Präpolymere oder Monomere und/oder Oligomere. Zudem enthält sie typischerweise ein geeignetes Lösungsmittel und optionale weitere Bestandteile, wie z.B. Radikalstarter. Sie wird erfindungsgemäß als Beschichtungslösung bezeichnet.
Präpolymere bedeutet hierbei, dass bereits oligomerisierte und/oder poly- merisierte Verbindungen eingesetzt werden, die nach dem Einbringen in den Formkörper keiner weiteren Polymerisationsreaktion mehr unterzogen werden, d.h. nicht mehr weiter miteinander vernetzt werden. Je nach Art des Aufbringens werden sie an den Formkörper adsorbiert (Physisorption) oder kovalent gebunden (Chemisorption).
Monomere und/oder Oligomere dagegen sind zur Polymerisation oder Polykondensation geeignete Verbindungen, die nach dem Einbringen in den Formkörper durch Polymerisation oder Polykondensation weiter vernetzt oder polymerisiert werden. Oligomere sind dabei Verbindungen,
die vorab bereits durch Vernetzung oder Polymerisation von Monomeren erzeugt wurden.
Es wurde gefunden, dass Verfahren zur Beschichtung von Partikeln nur schlecht auf die Beschichtung von monolithischen Formkörpern übertragen werden können. Grund dafür ist insbesondere die schlechte Zugänglichkeit des Inneren eines Formkörpers. Um beschichtete Formkörper mit guten chromatographischen Trenneigenschaften zu erzeugen, muss die Beschichtung des Formkörpers bezüglich ihrer Dicke und ihrer chemischen Zusammensetzung über den gesamten Formkörper homogen sein. Da der Formkörper erfindungsgemäß nicht vollständig mit dem organischen Polymer ausgefüllt wird, muss, um ein Verstopfen der Poren des Formkörpers zu verhindern, die Verteilung der Beschichtung im Inneren des Formkörpers bestmöglich kontrolliert werden oder dem organischen Polymer ein Porogen zugesetzt werden, das Durchgangsporen bildet.
Verfahren zur Beschichtung von Partikeln beinhalten häufig das Aufbringen einer Polymerlösung oder einer Lösung aus Monomer und Radikalstarter. Anschließend wird das Lösungsmittel entfernt. Dieses Verfahren lässt sich bei der Beschichtung von Formkörpern nicht anwenden. Bei der Trocknung von monolithischen Formkörpern erfolgt der Verdampfungsprozess des Lösungsmittels über eine relativ kleine Oberfläche (der Monolithaußenoberfläche). Durch die Trocknung wird aus dem Inneren des Formkörpers immer wieder Polymerlösung nachgezogen (Löschblatteffekt; Kapillar- Wirkung), was eine Herstellung von homogenen Beschichtungen nahezu unmöglich macht.
Erfindungsgemäß werden beschichtete Formkörper erhalten, die verbesserte Eigenschaften, insbesondere verbesserte Stabilität bespielsweise gegen alkalische Lösungen, aufweisen. Für die Herstellung derartiger verbesserter beschichteter Formkörper sind Beschichtungsverfahren geeignet, bei denen die Beschichtungslösung in den Formkörper ein-
gebracht wird und die Chemi- oder Physisorption der Polymerbeschichtung erfolgt, ohne das Lösungsmittel der Beschichtungslösung zuvor zu entfernen. Erst nach erfolgter Chemi- oder Physisorption wird das Lösungsmittel entfernt.
Insbesondere sind Verfahren geeignet, bei denen eine Polymerisation oder Polykondensation von Monomeren und/oder Oligomeren in Anwesenheit des Lösungsmittels erfolgen kann oder bevorzugt Verfahren, bei denen Präpolymere oder Monomere und/oder Oligomere aus dem Lösungsmittel auf den Formkörper aufgefällt werden können.
In beiden Fällen ist weiterhin darauf zu achten, dass die Beschichtung des Formkörpers nicht zu einem übermäßig dicken Polymerüberzug auf der Außenwand des Formkörpers führt. Da die Formkörper zum Einsatz in der Chromatographie in der Regel mit einem Säulenmantel flüssigkeitsdicht umschlossen werden müssen und mit Anschlüssen für Elutionsmittelzu- und ablauf versehen werden müssen, würde eine dicke und gegebenenfalls unregelmässige Beschichtung der Außenwand eine Ummantelung stark erschweren. In der Regel müsste die Beschichtung der Außenwand zu- nächst entfernt werden. Das wiederum kann die Außenwand des Formkörpers beschädigen.
Daher wird die Beschichtung der Formkörper erfindungsgemäß bevorzugt bereits in einer Ummantelung durchgeführt, die zumindest die Längsseite des Formkörpers dicht umschließt. Die Ummantelung sollte inert gegenüber den bei der Beschichtung beteiligten Reagenzien und Lösungsmitteln sein. Die Beschichtungslösung kann dann bevorzugt, ähnlich wie bei einer Chromatographiesäule, über geeignete Anschlüsse in den ummantelten Formkörper gepumpt werden.
Genauso kann die Beschichtung erfolgen, in dem ein nicht ummantelter Formkörper für die Beschichtung in ein inertes Lösungsmittel eingebracht
wird. Dabei wird der Formkörper zunächst mit der Beschichtungslösung getränkt. Anschließend wird er zur Durchführung der Beschichtung möglichst vollständig in ein inertes Lösungsmittel getaucht. Inert bedeutet in diesem Fall, dass das Lösungsmittel nicht an der Polymerisation oder Polykondensation beteiligt ist.
Zur Durchführung des erfindungsgemäßen Verfahrens kann es gegebenenfalls notwendig sein, den Formkörper vor dem Befüllen mit der Beschichtungslösung chemisch zu modifizieren, um die Oberfläche des anorganischen Materials mit funktionellen Gruppen zu versehen, die zur Physi- oder Chemisorption der organischen Polymerbeschichtung notwendig sind. Geeignete Ver ahren sind dem Fachmann auf dem Gebiet der Sorbenzien bekannt. Zur Einführung der funktioneilen Gruppen eignen sich im Prinzip die Reaktionen, die auch zur Einführung von Separationseffek- toren in chromatographische Trägermaterialien verwendet werden. Für Formkörper auf Basis von Si02 ist dies insbesondere die Umsetzung mit geeignet funktionalisierten Silanen. Die Silane können beispielsweise polymerisationsfähige Gruppen tragen, die bei der Aufpolymerisation von Monomeren und/oder Oligomeren eine kovalente Anbindung an den Form- körper ermöglichen. Verfahren zur Einführung derartiger Funktionalitäten werden beispielsweise in WO 94/19687 offenbart.
Genauso kann es für eine homogene Beschichtung der Formkörper vorteilhaft sein, einen geeigneten Radikalstarter vor dem Tränken des Form- körpers mit der Beschichtungslösung homogen auf der Oberfläche des
Formkörpers zu verteilen, insbesondere durch kovalente Anbindung an den Formkörper.
Zur Beschichtung der anorganischen Formkörper mit dem organischen Polymer wird der Formkörper zunächst mit einer Beschichtungslösung getränkt. Dies kann z.B. durch Eintauchen des Formkörpers in eine ent-
sprechende Lösung erfolgen oder, im Falle eines ummantelten Formkörpers durch Durch- bzw. Aufpumpen der Beschichtungslösung.
Die Beschichtung des Formkörpers erfolgt dann je nach Art der Be- schichtungslösung. Enthält die Beschichtungslösung Präpolymere, werden diese unter geeigneten Bedingungen auf den Formkörper aufgefällt. Wird eine Chemisorption der Präpolymere angestrebt, kann diese beispielsweise durch Temperaturerhöhung, Temperaturerniedrigung, Bestrahlung oder Zugabe von Reagenzien erfolgen, die die chemische Reaktion zwischen Formkörper und Präpolymer starten.
Als besonders vorteilhaft zur Herstellung einer homogenen Beschichtung hat sich eine Verfahrensvariante erwiesen, bei der die Präpolymere aus der Beschichtungslösung durch Unterschreitung des Löslichkeitsproduktes aus- gefällt werden, bevorzugt durch Temperaturerniedrigung. Hierbei entsteht durch die starke Temperaturerniedrigung eine übersättigte Lösung, aus der die Präpolymere sich gleichmäßig auf dem Formkörper ablagern. Bei diesen Verfahrensvarianten kann die Geschwindigkeit der Ausfällung z.B. durch die Geschwindigkeit der Temperaturveränderung gesteuert werden.
Bei Präpolymeren, die in der Beschichtungslösung unlöslich sind und daher als Suspension aufgetragen werden, kann auch eine Erhöhung der Temperatur gegebenenfalls eine gleichmäßige Beschichtung des Formkörpers bewirken, insbesondere, wenn die Polymere durch die Temperatur- erhöhung schmelzen und sich dadurch auf der Oberfläche des Formkörpers ablagern.
Sollen die Präpolymere kovalent an den Formkörper gebunden werden, kann die Anbindung nach der Ausfällung durch geeignete Initiierung, wie Bestrahlung, Zugabe von Reaktionsstartern etc., gestartet werden.

Es ist zudem möglich, nacheinander mehrere, auch unterschiedliche, Polymerschichten auf den Formkörper aufzutragen.
Nach Beendigung der Beschichtung wird der Formkörper gründlich mit geeigneten Lösungsmitteln gespült, um Reaktionsrückstände, wie nicht abreagierte Monomere oder nicht adsorbiertes Polymer, zu entfernen.
Anschließend wird der beschichtete Formkörper, bevorzugt im Vakuum, getrocknet.
Die erfindungsgemäß beschichteten Formkörper können direkt für chromatographische Trennungen eingesetzt werden oder zuvor mit Separationseffektoren funktionalisiert werden. Dies sind beispielsweise ionische, hydrophobe, chelatisierende oder chirale Gruppen. Verfahren zur Einführung derartiger Funktionalitäten sind dem Fachmann auf dem Gebiet der chromatographischen Trägermaterialien bekannt und in einschlägigen Lehrbüchern, z.B. Handbuch der HPLC, Hrsg. K.K. Unger; GIT-Verlag (1989) und Porous Silica, K.K. Unger, Elsevier Scientific Publishing Company (1979).
Besonders geeignet sind die erfindungsgemäßen Formkörper für die Biochromatographie, da sie sich bei Verwendung basenstabiler organischer Polymere durch eine gute Basenstabilität auszeichnen und zugleich gute Trennleistungen und einen geringen Säulenrückdruck zeigen.
Weiterhin können die erfindungsgemäß beschichteten Formkörper als Festphase für die Immobilisierung von Katalysatoren, z.B. Biokatalysatoren wie Enzymen, eingesetzt werden. Auch können geeignet funktionalisierte Formkörper als Reaktionspartner für die Durchflusssynthese dienen.
Auch ohne weitere Ausführungen wird davon ausgegangen, dass ein Fachmann die obige Beschreibung im weitesten Umfang nutzen kann. Die

bevorzugten Ausführungsformen und Beispiele sind deswegen lediglich als beschreibende, keineswegs als in irgendeiner Weise limitierende Offenbarung aufzufassen.

Beispiele
Beispiel 1: Vergleich der Eigenschaften eines C18-funktionalisϊerten 5 Formkörpers mit einem erfindungsgemäßen polymerbeschichteten Formkörper
1.1 Herstellung von Monolithen mit Standard-C18-Modif izierung
Die übliche Methode zur Abschirmung einer Kieselgeloberfläche in der 0 Chromatographie ist die Oberflächenmodifizierung unter Reaktion mit einem hydrophoben Silan.
Dafür werden 6 gut getrocknete (100°C im Vakuum) poröse monolithische Silika-Formkörper (Herstellung entsprechend WO 98/29350; Länge 15 cm), die eine spezifische Oberfläche von 300 m2/g und ein Porenvolumen von £> 1ml/g aufwiesen, in ein Reaktionsgemisch von 100 ml Toluol und 15 ml Octadecyldimethylchlorsilan gestellt. Um die Luft aus dem Formkörper zu entfernen wird das Reaktionsgemisch evakuiert und mit Stickstoff wieder belüftet. Die Reaktion erfolgt durch Erhitzen für 24 Stunden auf 110°C. Nach der Reaktionszeit werden die Formkörper entnommen und durch 0 Extraktion mit Toluol von nicht reagiertem Silan befreit. Nach Trocknung bei 100°C im Vakuum wird eine Gewichtszunahme von 20% festgestellt.

Der Messung der Elementaranalyse ergibt einen Gehalt von 16,2% Kohlenstoff.
1.2 Herstellung von Formkörpern mit einer Polymerbeschichtung aus Styrol/Divinylbenzol
Die Beschichtung der Kieselgeloberfläche der Formkörper (gleiches Ausgangsmaterial wie unter 1.1) erfolgt durch Polymerisation einer Mischung aus Styrol und Divinylbenzol. Dazu wird zunächst eine Monomermischung aus 12 g Styrol und 6 g Divinylbenzol mit 0,5 g Azoisobutyronitril in 50 ml Toluol hergestellt.
6 gut getrocknete (100°C im Vakuum) Silika-Formkörper von 15 cm Länge, die eine spezifische Oberfläche von 300 m2/g und ein Porenvolumen von 1 ml/g aufweisen, werden in das Reaktionsgemisch gestellt. Um die Luft aus den Formkörpern zu entfernen, wird das Reaktionsgemisch evakuiert und mit Stickstoff wieder belüftet.
Zur Polymerisation werden die getränkten Stäbe dann in eine Reaktionsapparatur mit frischem, auf 80°C vorgeheiztem, Toluol gestellt. Der Grund dafür ist, dass nur die Beschichtungslösung im Inneren des Formkörpers zur Reaktion gebracht werden soll, damit nicht eine äußere Polymer- Schicht auf dem Formkörper und/oder Lösungspolymer die Reinigung erschweren und die spätere Beständigkeitsuntersuchung verfälschen. Nach 5 Stunden bei 80°C werden die Formkörper entnommen und durch Extraktion mit Toluol von nicht abreagiertem Monomer und nicht adsorbiertem Polymer befreit. Nach Trocknung bei 100°C im Vakuum wird eine Gewichtszunahme von 6% festgestellt.
Der Messung der Elementaranalyse ergibt einen Gehalt von 5,5% Kohlenstoff.
1.3 Umsetzung eines ummantelten Formkörpers, der eine poly- merisierbare Funktionalisierung trägt, mit Styrol /Divinylbenzol
Die Beschichtung der Kieselgeloberfläche erfolgt in diesem Fall durch eine in-situ Polymerisation von einer Mischung aus Styrol und Divinylbenzol. Um

das Polymer kovalent anbinden zu können, wurde der Formkörper zunächst mit einer Mischung aus Toluol und Methacryloxypropyltrimethoxy- silan umgesetzt.
Ein gut getrockneter, zuvor mit PEEK ummantelter, Formkörper von 10 cm
Länge (Herstellung entsprechend WO 98/29350), der eine spezifische Oberfläche von 300 m2/g und ein Porenvolumen von 1 ml/g aufweist, wird in einem HPLC Ofen auf 80°C temperiert. Eine Mischung aus 20 g Toluol und 2 g Methacryloxypropyltrimethoxysilan wird mit einem Fluss von 0,5 ml/Minute durch den Formkörper gepumpt und anschließend mit Toluol nachgewaschen.
Im Anschluss wird eine Monomermischung aus 6 g Styrol und 3 g Divinylbenzol mit 0,5 g Azoisobutyronitril in 50 ml Toluol hergestellt. Das zuvor entgaste Reaktionsgemisch wird mit einem Fluss von 0,5 ml/Minute durch den Formkörper, der in einem HPLC Ofen auf 80°C temperiert wurde, gepumpt. Zur Entfernung von nicht abreagiertem Monomer und nicht adsorbiertem Polymer wird im Anschluss der Formkörper noch mit 50 ml Toluol bei 80°C gespült. Nach Trocknung bei 100°C im Vakuum wird eine Gewichtszunahme von 10% festgestellt. Der Messung der Elementaranalyse ergibt einen Gehalt von 9,3% Kohlenstoff.
1.4 Test auf Stabilität der Polymerschicht A) Statische Testung Zur Testung der Abschirmung der Kieselgeloberfläche und Untersuchung auf Gleichmäßigkeit und Dichtigkeit der Polymerbeschichtung werden die Formkörper einer Natronlaugebehandlung ausgesetzt. Da Kieselgel eine hohe Löslichkeit in Natronlauge aufweist, muss die Oberflächenbelegung sich reduzieren, wenn das Trägermaterial angegriffen und aufgelöst wird.
Zur Testung werden die Formkörper in 100 ml einer Lösung aus 4 g NaOH in 1 I Wasser (0,1 N Natronlauge) gestellt und nach 1 h, 5h und 24h jeweils

Proben zur Untersuchung abgenommen. Die Muster werden durch intensives Extrahieren mit Wasser von löslichen Anteilen und restlicher Natronlauge befreit. Im Anschluss werden die Formkörper zunächst mit Methanol und dann mit Toluol extrahiert, um nicht mehr adsorbierte Polymeranteile zu entfernen.
Die erhaltenen Muster werden getrocknet und durch Elementaranalyse untersucht.
Als Vergleich wurde der Versuch auch mit einer 1 N Natronlauge-Lsg. durchgeführt.
B) Dynamische Testung
Als Ergänzung wird der PEEK ummantelte Formkörper mit einer Lösung aus 4 g NaOH in 1 I Wasser (0,1 N Natronlauge) 24 Stunden mit einem Fluss von 0,2 ml/Minuten durchpumpt und durch Nachwaschen mit Wasser, Methanol und Toluol gewaschen.
C) Ergebnis der Testung
Tabelle 1 zeigt den verbleibenden Kohlenstoffgehalt der Formkörper bzw. ihre Beschaffenheit nach unterschiedlich langer Behandlung mit Natron- lauge:
FK1: C18 funktionalisierter Formkörper (siehe 1.1)
FK2: mit Styrol/Divinylbenzol in Toluol beschichteter Formkörper (siehe 1.2)
FK3: mit Styrol/Divinylbenzol in Ummantelung beschichteter Formkörper
(siehe 1.3) - hier konnte nur ein Wert ermittelt werden, da der Formkörper für die Bestimmung des Kohlenstoffgehaltes aus der Ummantelung herausgebrochen werden musste.

Es konnte festgestellt werden, dass ein C-18 funktionalisierter Formkörper in Natronlauge deutlich an Masse und somit auch an Kohlenstoffmodifizierung verliert und teilweise zerstört oder sogar komplett aufgelöst wird. Dem gegenüber zeigen die polymerbeschichteten Muster eine gute Stabilität gegenüber der Natronlauge. Der Versuch zeigt, dass durch die Beschichtung eines anorganischen Monolithen mit einem Polymer ein Material mit neuen, verbesserten Eigenschaften bereitgestellt wird.
Durch die Stabilitätsprüfung in alkalischer Lösung kann die Verkapselung der Formkörper durch die Polymerbeschichtung gezeigt werden. Es ist aber durchaus für manche Anwendungen auch sinnvoll, Beschichtungen mit
Polymeren zu verwenden, die in Natronlauge instabil sind.
Dabei ist z.B. auch an die Herstellung von biokompatiblen Oberfläche mit hydrophilen Polymeren zu denken oder mit Polysacchariden.
Das Beispiel schränkt auch nicht die Herstellung von Polymeren auf anderen anorganischen Trägern ein.
1.5 BET Untersuchung
Die im beschichteten Formkörper FK2 wurden durch eine Messung der spezifischen Oberfläche und des Porenvolumens charakterisiert. Dabei

ergab sich eine Oberfläche von 280m2/g und ein Porenvolumen von
0,92ml/g.
Dieses Ergebnis zeigt, dass die Poren des Formkörpers nicht
„zupolymerisiert", d.h. verstopft, wurden, sondern sich eine gleichmäßige
Schicht auf der Oberfläche gebildet hat. Somit sind die Poren des
Formkörpers nicht vollständig mit Polymer gefüllt. In dem erfindungsgemäß beschichteten Monolithen verbleiben somit zugängliche Porenräume.
Beispiel 2: Herstellung von beschichteten Fremdkörpern durch Ausfällung von Präpolymeren 2.1 Synthese
Die folgenden Reaktionen wurden in einer Apparatur nach Abbildung 1 durchgeführt. Der Formkörper (1) ist mit einer druckstabilen Ummantelung (2) umgeben. Die Beschichtungslösung, sowie weitere Waschlösungen etc. werden in Behälter (3) vorgemischt und mittels einer Pumpe (4) über ein Druckreservoir (5) in einen Vorlagebehälter (6) aus druckfestem Stahl und von dort durch den Formkörper (1) in einen Abfallbehälter (7) gepumpt.
Zunächst wird der Formkörper (Chromolith®, Firma Merck KGaA, Darmstadt, Material Si02; Innendurchmesser ID = 4,6 mm; Länge L = 25 mm) mit dem verwendeten Lösungsmittel DMF konditioniert. Somit liegt ein vollständig mit Lösungsmittel getränkter Formkörper als Ausgangsmaterial vor.
Es wird eine Beschichtungslösung aus Poly(methacrylat)-Lösung (Konzentration c = 10 mgm."1) in Dimethylformamid (DMF; p.a.) hergestellt und in den Vorlagebehälter (6) gegeben. Mittels einer HPLC-Pumpe (Marke Bischoff), wird in dem teilweise mit Luft gefüllten Druckreservoir (5) Druck erzeugt und somit die Beschichtungslösung in den Formkörper gedrückt. Da Luft als Druckvermittler fungiert (Luftpolster), ist sichergestellt, dass die

Konzentration des Polymers bei diesem Verfahren konstant bleibt. Der hier aufzuwendende Druck beträgt - 0,4 bar bei einer Flussrate von 1 mimin"1. Das Volumen der nun im Formkörper befindlichen Lösung beträgt ca. V = 0,33 ml. Nach Durchfluss von 3 ml wird der Formkörper abgenommen, mittels Verschraubungen dicht verschlossen und anschließend für 40 min in das vorbereitete bei -55 °C temperierte Kältebad (Methanol/Trockeneis) gegeben. Danach wird das Lösungsmittel mittels einer Vakuumlinie bei 40 °C, 1 Torr für 12 h, entfernt.
In ähnlicher Weise kann die Beschichtung statt mit Poly(methacrylat)
[PEMA] z.B. auch mit Poly(2-hydroxyethylmethacrylat) [P2HEMA], einem Copolymer aus 2-Hydroxyethylmethacrylat und Ethylmethacrylat [P2HE.-E] oder Poly(octadecylmethacrylat) [POMA] erfolgen.
Zur Druckerzeugung kann anstelle einer HPLC-Pumpe auch ein Behälter mit komprimiertem Gas, z.B. eine Stickstoffbombe, genommen werden.
2.2 Kohlenstoffgehalt und Morphologie
Der über Elementaranalyse bestimmte Kohlenstoffgehalt der beschichteten Formkörper ist in Tabelle 2 aufgeführt.

Rasterelektronenmikroskopische Aufnahmen (REM) zeigen, dass bezüglich der Morphologie kein Unterschied zwischen den beschichteten und den unbeschichteten Formkörpern besteht. Die Makroporenstruktur der Formkörper bleibt erhalten. Abbildung 2 zeigt den Vergleich einer REM- Aufnahme eines unbeschichteten Formkörpers (Chromolith® der Firma Merck KGaA) (Figur 2 A) und eines mit einem Copolymer aus 2-Hydroxy-

ethylmethacrylat und Ethylmethacrylat beschichteten Formkörpers (Figur 2 B).
2.3 Chromatographisches Verhalten Vor den chromatographischen Tests wird der nun mit Poly(methacrylat) beschichtete Formkörper noch 1 h mit der mobilen Phase bei 1 mimin'1 konditioniert.
Für die chromatographischen Untersuchungen wurden drei ausgewählte 0 Proteine: Lysozym, Cytochrom C, Myoglobin (LCM-Testgemisch) als Testmischung verwendet.
Die Proteine wurden in Wasser ohne Triflüoressigsäure (TFA)-Zusatz gelöst. Als mobile Phase wurde ein organisch-wässriger Eluent mit TFA- Zusatz im Gradienten-Modus verwendet:
Fluss: 1 ml/min; Detektion: 215 nm, 400 nm; Raumtemperatur Injektionsvolumen: 5 /I von 95/5 % (v/v) A/B in 5 min zu 5/95 % (v/v) A/B; mit 0 A: Wasser + 0.1 % TFA,
B: Acetonitril + 0,09 % TFA
Der Gegendruck betrug für Poly(methacrylat)-beschichtete Formkörper ca. _ 20 bar, für den unbeschichteten Formkörper (Chromolith®) ca. 30 bar.
Die erhaltenen Chromatogramme zeigen, dass sich die Elutionsprofile der Proteinmischung wie erwartet in Abhängigkeit von der Hydrophobie der Beschichtung ändern. Die Trennleistung und der geringe Säulenrückdruck 0 belegen die gute Eignung der erfindungsgemäßen Materialien als Sorbens für die Chromatographie,

Beispiel 3: Herstellung von beschichteten Fremdkörpern durch Chemisorption
In einer Vorrichtung entsprechend Beispiel 2.1 wird an einen mono- lithischen Formkörper (Chromolith™ Si, Firma Merck KGaA, Darmstadt,
Material Si02; Innendurchmesser ID = 4,6 mm; Länge L = 25 mm) ein Azoderivat als Initiator kovalent angebunden und anschließend Monomere aufpolymerisiert.
3.1. Kopplung des p-(Chlormethyl)phenyltrimethoxysilan an die Kieselgel-Oberfläche
Es wird eine Mischung aus 3,95 g (16,04 mmol) p-(Chlormethyl)phenyl- trimethoxysilan und 25 ml trockenem Tetrahydrofuran (THF; p.a.) hergestellt. Der Monolith wurde mit trockenem Tetrahydrofuran (THF) konditioniert. Danach wurden die obige Mischung mit Stickstoff (über
H2S04 getrocknet) behandelt und in den Vorlagebehälter gegeben. Mittels einer HPLC-Pumpe (Marke Bischoff), wurde in dem mit Luft gefüllten Druckreservoir Druck erzeugt und somit die Lösung in denMonolithen gedrückt. Anschließend wurde der Monolith abgenommen, mittels Verschraubungen dicht verschlossen und senkrecht für 35 h bei
Raumtemperatur stehen gelassen. Danach wurde der behandelte Monolith mit THF und anschließend mit Methanol gewaschen.
Die Elementaranalyse ergab einen Kohlenstoffgehalt %C: ~ 2,87 %.
3.2. Kopplung des 4,4'-Azobis-(4-cyan-pentansäure) (ACPA) an den mit Silan derivatisierten Monolithen
Der nach 3.1 mit Silan derivatisierte Monolith wurde mit trockenem Toloul konditioniert. Danach wurde eine Lösung aus 3,63 g (10,66 mmol) 4,4'-Azo- bis-(4-cyan-pentansäure) (ACPA), 5,40 g α-Picolin und 15 ml trockenem Toluol mit Stickstoff (über H2S04 getrocknet) behandelt und in den Vorlagebehälter gegeben. Mittels einer HPLC-Pumpe (Marke Bischoff), wurde in der mit Luft gefüllten Druckreservoir Druck erzeugt und somit die Lösung in

den mit Silan derivatisierten Monolithen gedrückt. Anschließend wurde der Monolith abgenommen, mittels Verschraubungen dicht verschlossen und für 11 h bei 50 °C senkrecht in einen Ofen gestellt, in dem der Monolith guten Wärmekontakt hatte. Nach beendeter Reaktion wird der Monolith mit Toluol und anschließend mit Methanol gewaschen.
Die Elementaranalyse ergab einen Kohlenstoffgehalt %C: ~ 4,64
3. Thermische Polymerisation mit Ethylmethacrylat
Der mit Azo-Initiator derivatisierte Monolith wurde mit trockenem Toluol konditioniert. Danach wurde eine Lösung aus 1 ,86 g (15 mmol)
Ethylmethacrylate (EMA), 0,30 g (1,5 mmol) Ethylenglycoldimethacrylat (EDMA) und 15 ml trockenem Toluol mit Stickstoff (über H2S0 getrocknet) behandelt und in den Vorlagebehälter gegeben. Mittels einer HPLC-Pumpe (Marke Bischoff), wurde in der mit Luft gefüllten Druckreservoir Druck erzeugt und somit die Lösung in den mit Azo-Initiator gekoppelten
Monolithen gedrückt. Anschließend wurde der Monolith abgenommen, mittels Verschraubungen dicht verschlossen und für 4 h bei 90 °C senkrecht in einen Ofen gestellt, in dem der Monolith guten Wärmekontakt hatte. Nach beendeter Reaktion wird der Monolith mit Toluol und anschließend mit Methanol gewaschen.
Die Elementaranalyse ergab einen Kohlenstoffgehalt %C: - 12,16
Durch Veränderung der Konzentrationen der eingesetzten Reaktanden lässt sich der Kohlenstoffgehalt des Produktes beeinflussen.
Claims
1. Formkörper bestehend aus einem porösen anorganischen monolithischen Formkörper, der mit mindestens einem organischen Polymer beschichtet ist.
2. Formkörper nach Anspruch 1 , dadurch gekennzeichnet, dass der poröse anorganische monolithische Formkörper aus Si02 besteht.
3. Formkörper nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass der poröse anorganische monolithische Formkörper eine bimodale Porenstruktur mit Mesoporen mit einem Durchmesser zwischen 2 und 100 nm und Makroporen mit einem mittleren Durchmesser von über 0,1 μm aufweist.
4. Formkörper nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass es sich bei dem organischen Polymer um Polystyrol und/oder Polymethacrylat handelt.
5. Formkörper nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass das organische Polymer auf dem anorganischen Formkörper physisorbiert ist.
6. Verfahren zur Herstellung von porösen anorganischen monolithischen Formkörpern, die mit mindestens einem organischen Polymer beschichtet sind, durch a) Bereitstellen eines porösen anorganischen monolithischen Formkörpers b) Tränken des porösen anorganischen monolithischen Formkörpers aus Schritt a) mit einer Beschichtungslösung enthaltend zumindest organische Präpolymere oder organische Mono- und/oder Oligomere. - 25 -
c) Beschichtung des Formkörpers, wobei der Formkörper während der Beschichtung zumindest an den Längsseiten mit einem inerten Material dicht ummantelt ist oder in einem inerten Lösungsmittel gelagert wird d) Waschen und Trocknen des Formkörpers aus Schritt c) zur Entfernung von Reaktionsrückständen und Lösungsmittel
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass die Schritt c) die Präpolymere oder Monomere und/oder Oligomere aus der Beschichtungslösung auf den anorganischen Formkörper aufgefällt werden.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass das Auffällen durch Temperaturerniedrigung erfolgt.
9. Verwendung eines Formkörpers nach einem oder mehreren der Ansprüche 1 bis 5 zur chromatographischen Trennung mindestens zweier Substanzen.
