Abstract
Die vorliegende Erfindung betrifft Lebensmittel-, Nahrungsmittel- oder Arzneimittelzubereitungen, die konjugierte trans/cis Octadecatriensäure enthalten sowie Verfahren zu iher Herstellung. Erfindungsgemä beta e Zubereitungen enthalten vorzugsweise Calendulasäure. Die Erfindung betrifft zudem die Verwendung der Zubereitungen zur Redutkion der Nahrungsaufnahme bei Menschen und Tieren und zur besseren Nahrungsverwertung.

Beschreibung
Die vorliegende Erfindung betrifft Lebensmittel-, Nahrungsmittel- oder Arzneimittelzubereitungen, die konjugierte trans/cis Octa- decatriensäure enthalten sowie Verfahren zu ihrer Herstellung. Erfindungsgemäße Zubereitungen enthalten vorzugsweise Calendula- säure. Die Erfindung betrifft zudem die Verwendung der Zubereitungen zur Reduktion der Nahrungsaufnahme bei Menschen und Tieren, und und zur besseren Nahrungsverwertung bei Menschen und Tieren, zur Veränderung der KörperZusammensetzung, Verfahren zur Reduktion der Nahrungsmittelaufnahme und ein Kit, das die erfindungsgemäße Zubereitung enthält.
Fettsäuren und Triglyceride haben eine Vielzahl von Anwendungen in der Lebensmittelindustrie, der Tierernährung, der Kosmetik und im Pharmabereich. Je nachdem, ob es sich um freie gesättigte oder ungesättigte Fettsäuren oder um Triglyceride mit einem erhöhten Gehalt an gesättigten oder ungesättigten Fettsäuren handelt, sind sie für die unterschiedlichsten Anwendungen geeignet. So ist z.B. ein hoher Anteil an Lipiden mit ungesättigten Fettsäuren und speziell mit mehrfach ungesättigten Fettsäuren für die Ernährung von Tieren und Menschen wichtig, da diese z.B. einen positiven Einfluss auf den Triglyceridspiegel bzw. Cholesterinspiegel im Blut und damit das Risiko einer Herzerkrankung reduzieren. Ungesättigte Fettsäuren finden in verschiedenen diätischen Lebensmitteln oder Medikamenten Anwendung.
Besonders wertvolle und gesuchte ungesättigte Fettsäuren sind die sogenannten konjugierten ungesättigten Fettsäuren. Konjugierte mehrfach ungesättigte Fettsäuren sind im Vergleich zu anderen mehrfach ungesättigte Fettsäuren eher selten.
CLA ist ein Sammelbegriff für positioneile und strukturelle Isomere der Linolsäure, die sich durch ein konjugiertes Doppelbindungssystem am Kohlenstoffatom 8, 9, 10 oder 11 auszeichnen. Geometrische Isomere existieren für jedes dieser positionellen Isomere, also cis-cis, trans-cis, cis-trans, trans-trans . Vor allem C18:2 cis-9, trans-11 und C18:2 trans-10, cis-12 CLAs, die die biologisch aktivsten Isomere darstellen, sind von besonderem Interesse, da sie sich im Tierexperiment als krebsvorbeugend erwiesen haben, anti-arteriosklerotisch wirken und in Mensch und Tier den Körperfettanteil reduzieren. Kommerziell werden CLAs heute hauptsächlich als freie Fettsäure vertrieben.
Für den Menschen sind die wichtigsten natürlichen Quellen für CLA vor allem tierische Fette. So besitzen Fette von wiederkäuenden Tieren, wie Rindern (Chin, Journal of Food Composition and Analysis, 5, 1992: 185 - 197) und Schafen, sowie Molkereiprodukte sehr hohe CLA-Konzentrationen. Bei Rindern findet man 2,9 bis 8,9 mg/g Fett. Dagegen besitzen Pflanzenöle, Margarinen und Fette von nicht-wiederkäuenden Tieren CLA-Konzentrationen von nur 0,6 bis 0,9 mg/g Fett.

Für CLA sind eine Reihe positiver Effekte nachgewiesen worden. So reduziert die Verabreichung von konjugierter Linolsäure das Körperfett in Mensch und Tier bzw. erhöht den Futterumsatz pro Körpergewicht bei Tieren (WO 94/16690, WO 96/06605, WO 97/46230, WO 97/46118) . Durch Gabe von konjugierter Linolsäure lassen sich auch beispielsweise Allergien (WO 97/32008) , Diabetes (WO 99/29317) oder Krebs positiv (Banni, Carcinogenesis , Vol. 20, 1999: 1019-1024, Thompson, Cancer, Res . , Vol. 57, 1997: 5067-5072) beeinflussen. Mehrfach ungesättigte Fettsäuren werden auch Babynahrung zur "Erhöhung des Nährwertes" und als essentielle Bausteine, die Wachstum und Gehirnentwicklung gewährleisten, zugesetzt.
CLA hat wie oben beschrieben sehr weitreichende positive ernährungsphysiologische Wirkungen. CLA kommt jedoch natürlicherweise in signifikanten Mengen nur bei Wiederkäuern und deren Produkten, wie Milch, Käse etc. vor. Es besteht daher ein großer Bedarf an Alternativen des aus diesen tierischen Quellen stammenden CLA, insbesondere um eine ausgeglichene und gesunde Ernährung in weniger entwickelten Regionen zu gewährleisten, in denen die Versorgung mit diesen tierischen Fetten unzureichend oder eine synthetische Herstellung zu teuer ist. Aber auch in entwickelten Regionen hat der Verzehr an Fleisch- und Milchprodukten abge-
nommen, um den Anteil an als ungesund geltenden gesättigten Fettsäuren zu verringern. Dies bedeutet aber gleichzeitig auch eine Verringerung der Aufnahme an "gesunder" CLA.
Die Aufgabe der vorliegenden Erfindung ist somit, eine alternative Zubereitung zur Verfügung zu stellen, die ernährungsphysiologisch ähnlich vorteilhafte Wirkungen besitzt wie eine Zubereitung, die CLA enthält.
Die Lösung des Problems wird in den der vorliegenden Erfindung zugrunde liegenden Ausführungsformen beschrieben.
Die vorliegende Erfindung betrifft daher eine Lebensmittel-, Nahrungsergänzungsmittel-, Tierfuttermittel- oder Arzneimittelzubereitung, enthaltend konjugierte trans/cis Octadecatriensäure.
Der vorliegenden Erfindung liegt die überraschende Erkenntnis zugrunde, dass der Zusatz von konjugierter trans/cis Octadecatriensäure, insbesondere von Calendulasäure, zur Nahrung zu einer reduzierten Nahrungsaufnahme führt, ohne dass eine Verringerung des Körpergewichts beobachtet wird. D.h. die Futterverwertung ist bei Zugabe von konjugierter Octadecatriensäure zur Nahrung signifikant verbessert. Der Begriff "Octadecatriensäure" oder der Begriff "Calendulasäure" umfaßt hierin nicht nur die freie Säure, sondern auch die Salze, und Ester sowie weitere nicht-toxische Derivate. Vorzugsweise umfaßt der Begriff nicht-toxische Salze oder Glyceride, insbesondere Triglyceride, oder Ethyl oder Methylester, wie hierin unten beschrieben.
Überraschend wurde zudem gefunden, daß durch die Zugabe einer konjugierten trans/cis Octadecatriensäure, insbesondere von Calendulasäure, eine Veränderung der Körperzusammensetzung beobachtet werden konnte. So wurde gefunden, daß die Zugabe von Calendulasäure zu einer Reduktion des Körperfettanteils im tierischen Körper führt .
Die vorliegende Erfindung betrifft somitweiterhin ein Verfahren zur Herstellung einer Lebensmittel-, Nahrungsergänzungsmittel-, Tierfuttermittel- oder Arzneimittelzubereitung, umfassend die Zugabe einer konjugierten trans/cis Octadecatriensäure zur Zubereitung. Vorzugsweise hat die genannte trans/cis Octadecatriensäure eine tlO , cl2-Konfiguration.
In einer besonders bevorzugten Ausführungsform ist die genannte trans/cis Octadecatriensäure daher Calendulasäure. Calendulasäure ist eine C18 : 3-Fettsäure mit einer t8, tlO, cl2-Konfiguration.
Calendulasäure (C18:3; t8, t10, c12) ( I )
Sie ist somit eine konjugierte trans/cis Octadecatrien-Säure. Calendulasäure ist die verantwortliche Fettsäure für die beobachtete erfindungsgemäße Reduzierung der Nahrungsaufnahme und verbesserte Nahrungsverwertung bei Mäusen bei Zugabe von Calendulaöl zum Futtermittel, wie die Vergleichsversuche in den Beispielen mit Mais-Öl zeigen. Die chemische Herstellung von Calendulasäure ist in US 3,356,699 beschrieben. Calendulasäure kommt natürlich z.B. in Calendula officinalis vor (Earle et al . , Lipids, 1, 1964: 325-327, Takagi et al., Lipids, 17, 1981: 716-723, Ul'chenko et al . , Lipids of Calendula officinalis, Chemistry of Natural Compounds , 34, 1998: 272-274). Biochemische Untersuchungen zur Synthese von Calendulasäure sind in Crombie et al., J. Chem. Soc . Chem. Commun., 15, 1984: 953-955 und J. Chem. Soc. Perkin Trans., 1, 1985: 2425-2434 zu finden.
Andere konjugierte trans/cis Octadecatriensäuren können durch chemische Modifikation von Linolensäure haltiger Öle, z.B. aus Leinöl, Soja- oder Hanföl, entstehen.
Erfindungsgemäße Zubereitungen eignen sich hervorragend als Nahrungsmittel- oder Futterzusatz, z.B. in Diäten oder bei der Tiermast. So kann in Kombination oder auch alleine mit einer reduzierten Kalorienaufnahme Calendulasäure zur Unterstützung einer Diät, z.B. zur Reduzierung des Körpergewichts beim Menschen, eingesetzt werden, die vorteilhaft auch die Essgewohnheiten beeinflusst. Die verbesserte Verwertung der zugenommenen Nahrung führt zu einer Reduktion des Nahrungsmittelsverbrauchs, was besonders in wenig entwickelten Regionen mit Nahrungsmittel- knappheit oder in Extremsituationen (Krankheiten, Leistungssport) vorteilhaft sein kann. Desweiteren kann vorteilhafterweise eine Veränderung der Körperzusammensetzung, insbesondere eine Reduzierung des Körperfettgehaltes erreicht werden.
Erfindungsgemäße Zubereitungen können auch ökonomisch und ökologisch vorteilhaft in der Tierernährung, insbesondere für eine Reduzierung der Futtermenge oder zur Reduzierung des Fettgehalts bei Tieren eingesetzt werden. Gesellschaftlich besteht heute großes Interesse an dem Fettgehalt der Nahrung. Aufgrund der vermuteten Beziehung zwischen gesättigten Fettsäuren von Fleisch und dem Cholesteringehalt im Blut, wird auf eine fettarme Ernährung geachtet. Daher besteht großes Interesse an Verfahren zur Redu-
zierung des Fettgehaltes in Tieren, wie insbesondere durch die vorliegende Erfindung.
In einer Ausführungsform ist die konjugierte trans/cis Octadecatriensäure oder die Calendulasäure in der Zubereitung als Calendulaöl enthalten.
Unter dem Begriff "Calendulaöl" wird ein Fettsäuregemisch verstanden, das Calendulasäure enthält. Vorzugsweise ist der Anteil an Calendulasäure im Calendulaöl ungefähr 30 %, mehr bevorzugt ist ein Anteil von 50 %, noch mehr bevorzugt ist ein Anteil von 60 %, 70 %, 80 % oder 90 %. Am meisten bevorzugt ist ein Anteil 95 % oder mehr. Dazu kann z.B. der Anteil an Calendulasäure nach Verseifung der Fettsäuren und Überführung der Fettsäuren zu Methylestern gaschromatographisch bestimmt werden. Neben der Calendulasäure kann das Calendulaöl verschiedene andere gesättigte oder ungesättigte Fettsäuren z.B. Linolsäure, Palmitin- säure enthalten. Insbesondere kann je nach Herstellungsverfahren der Anteil der verschiedenen Fettsäuren in dem Calendulaöl schwanken. Jedes Fettsäuremuster, ist von der erfindungsgemäßen Zubereitung umfasst, insbesondere Fettsäuremuster, die bei der Herstellung von Öl aus pflanzlichem Material entstehen und das den genannten Calendulaöl-Anteil enthält. Bevorzugt ist, dass das Calendulaöl eine geringe Streuung an unterschiedlichen Fettsäuren enthält bzw. dass die Anzahl an verschiedenen Fettsäuren im Calendulaöl gering ist.
In einer weiteren Ausführungsform enthält die genannte Zubereitung weitere Additive.
Unter dem Begriff "Additive" werden weitere für die Ernährung oder Gesundheit vorteilhafte Zusätze verstanden, z.B. "Nährstoffe", "Diätzusatzstoffe" oder "Wirkstoffe". Die Zubereitung kann ein oder mehr Additive für die tierische oder menschliche Ernährung oder Behandlung enthalten und damit verdünnt oder gemischt werden. Additive können zusammen mit oder getrennt von dem Futtermittel, Nahrungsmittel, Nahrungsergänzungsmittel oder Arzneimittel verabreicht werden. Eine Lebensmittel-, Nahrungsergänzungsmittel-, Tierfutter- oder Arzneimittelzubereitung beinhaltet keine Additive oder keine Mengen von Additiven, die für die tierische oder menschliche Ernährung als schädlich angesehen werden können.
Unter "Nährstoffe" werden solche Zusätze verstanden, die für die Ernährung von Menschen oder Tieren vorteilhaft sind. Vorzugsweise enthält die erfindungsgemäße Zubereitung daher auch Vitamine, z.B. die Vitamine A, Bl, B2, B6, B12, C, D3 , und/oder E, Fol- säure, Nicotinsäure, Taurin, Carboxylsäuren, z.B. Besonders bevorzugt enthält das erfindungsgemäße Tierfutter unbelebte, lebende und/oder vermehrungsfähige Anteile an Pilzen der Gattung Mucor, Hefen der Gattung Saccharomy- ces und/oder Bakterien der Enterobakteriaceae, wie E. coli, Sal- monellen, wie Salmonella typhimurium, Proteus vulgaris, Pseudomo- naden, wie Pseudomonas matophila, Bacillaceae, wie Bacillus sub- tilis oder Bacillus cereus, coryneformer Bakterien, wie Coryne- bacterium glutamicum oder Brevibacterium breve und/oder Actinum mycetalis und/oder Mischungen davon. Ganz besonders bevorzugt sind Bakterien der Gattung Bacillus und hierbei der Spezies Bacillus subtilis. Ebenso sind genetisch veränderte und/oder transgene Organismen und/oder zur Herstellung der erfindungsgemäßen Zubereitung geeignete Produktionsstämme erfindungsgemäß umfasse Die vorangehende Aufzählung ist dabei nicht limitierend für die vorliegende Erfindung.
Werden konjugierte trans/cis Octadecatriensäuren, insbesondere Calendulasäure, einzeln oder in Kombination in Futtermittel verabreicht, so werden die aktiven Verbindungen als Reinsubstanz oder Stoffgemische oder flüssige oder feste Extrakte zusammen mit üblichen Futtermittelbestandteilen verabreicht. Beispiele üblicher Futtermittelbestandteile sind: Mais, Gerste, Weizen, Hafer, Roggen, Tritikale, Sorghum, Reis und Kleien, Grieskleien sowie Mehle dieser Getreidearten, Sojabohnen, Sojaprodukte wie Sojaextraktionsschrot, Raps, Rapsextraktionsschrot, Baumwollsaat und Extraktionsschrot, Sonnenblumen, Sonnenblumenextraktionsschrot, Leinsaat, Leinextraktionsschrot, Expeller von Ölsaaten, Ackerbohnen, Erbsen, Gluten, Gelatine, Tapioka, Hefen, Single Cell Protein, Fischmehl, Salze, Mineralstoffe, Spurenelemente, Vitamine, Aminosäuren, Öle/Fette und dergleichen. Vorteilhafte Zusammensetzungen werden z.B. in Jeroch, H. et al . Ernährung landwirtschaftlicher Nutztiere, UTB beschrieben.
Weitere geeignete Zusätze sind in Park, Lipids, 32, 1997, 853, beschrieben.
Die erfindungsgemäße Zubereitung kann als Pulver, Granulat, Pellet, Extrudate mit einem Überzug versehen ("coated") und/oder als Kombinationen davon vorliegen. Die Zubereitung des erfindungsgemäßen Tierfutters, beispielsweise durch umhüllende Verbindungen, dient z.B. zur Verbesserung der Produkteigenschaften, wie Staubverhalten, Fließeigenschaften, Wasseraufnahmefähigkeit und Lagerstabilität. Solche Zubereitungen sind im Stand der Technik weitreichend bekannt. So werden in der Tierernährung

fein oder Aminosäuren, wie z.B. vorzugsweise essentielle Aminosäuren oder wie z.B. Tyrosin, Valin, Leucin, Threonin, Trypto- phan, Methionin, Arginin, Histidin oder Glutamin. Je nach Zweck muß die Menge der eingesetzten konjugierten trans/cis Octadecatriensäure, z.B. Calendulasäure, angepaßt werden. Die Menge der eingesetzten Calendulasäure kann z.B. 0,01 % oder 0,1 % der bei der Ernährung zugesetzten Fettmenge darstellen. Ebenfalls bevorzugt sind 0,5 %, 1 %, 2 % oder 3 %, 5 % oder 10 % Calendulasäure. Dies kann auch für andere Fettsäuren gelten.
Weiterhin kann die erfindungsgemäße Zubereitung zur Reduktion der Nahrungsaufnahme Veränderung der KörperZusammensetzung, und zur Verbesserung der Nahrungsverwertung bei Menschen oder Tieren verwendet werden. Insbesondere kann die erfindungsgemäße Zubereitung zur Reduktion des Fettgehaltes im Verhältnis zum Körperproteingehalt eingesetzt werden.
Bei Tieren ist insbesondere bei der Mast eine verbesserte Nahrungsverwertung vorteilhaft. Bereits eine geringe Reduktion der Nahrungsaufnahme kann zu einer bedeutenden Verringerung der Nahrungsmittelkosten führen. Vorzugsweise kann der Futterverbrauch um 1 % oder 2 % gesenkt werden, noch mehr bevorzugt sind 3 %, noch mehr bevorzugt sind 5 %, 10 % oder mehr.
Zur Reduktion des Fettgehaltes wird der Anteil der zugeführten Fette durch die konjugierte trans/cis Octadecatriensäure ersetzt. Vorzugsweise beträgt der Anteil an konjugierter trans/cis Octadecatriensäure des zugeführten Fettes mindestens 0,1 bis 50 Gew.-%, vorzugsweise 0,5 bis 30 Gew.-%, mehr bevorzugt 1 bis 20 Gew.-%, bezogen auf eine Aufnahme an Energie, die 30 % aus Fett besteht, d.h. ca. 70 bis 90 g Fett/Tag [gemäß: Referenzwerte für die Nährstoffzufuhr, 2000, Deutsche Gesellschaft für Ernährung, Umschau Braus GmbH Verlagsgesellschaf ) .
In einer weiteren Ausführungsform betrifft die vorliegende Erfindung ein Verfahren zur Verfütterung der erfindungsgemäßen Zubereitung oder zur Ernährung, wobei konjugierte trans/cis Octadecatriensäure, insbesondere Calendulasäure, zum Fettträger der Nahrung hinzugegeben wird.
Dem Fachmann sind verschieden Herstellungsverfahren zur Herstellung der erfindungsgemäßen Zubereitung bekannt Als Ausgangsmaterial kann z.B. pflanzliches Material dienen. Calendulaöl kann beispielsweise aus dem Samen von Calendula officinalis (Ringelblume) gewonnenen werden. Tabelle 2 zeigt, dass das Fettsäuremuster verschiedener Calendulaöle aus Ringelblumensamen in
Abhängigkeit von ihrer Herstellungsart variiert. Eine solche Variation wird als von der erfindungsgemäßen Zubereitung umfasst angesehen. Calendulaöl kann aus pflanzlichen Samen durch Pressen von z.B. Samen mit einem hohen Schalenanteil oder von geschältem Samen gewonnen werden. Die Presslinge können auch mehrmals gepresst werden. Dabei können pflanzlicher Samen oder andere Pflanzenteile, wie z.B. Blätter, Knollen, Stängel, Blüten, Früchte etc. von jeder Pflanze verwendet werden, die Calendulasäure enthält. Auch ganze Pflanzen können verwendet werden. Bekannt ist, dass Calendula officialis einen hohen Calendula- säureanteil enthält und daher sich zur Herstellung von Calendulaöl besonders eignet. Calendulasäure und somit auch -öl kann aber auch synthetisch (US 3,356,694) hergestellt werden.
Calendulasäure kann auch durch selektive Einführung von Doppelbindungen durch Wittichreaktion oder Metathese hergestellt werden. Alternativ kann Calendulasäure aus Octadecatriensäuren- haltigen Ölen hergestellt werden. Diese Octadecatriensäuren können mit Propylenglykol bei 150°C konjugiert werden.
Die Herstellung von Calendulasäure oder deren Ester kann durch Isomerisierung an der Doppelbindung einer Octadecatriensäure in Gegenwart von NaHS0 oder KHS0 erfolgen. Diese Reaktion kann wie in DE 114 16 90 für dehydratisiertes Castor Öl (Rizinusöl) beschrieben durchgeführt werden.
Die Herstellung von Calendulasäure kann analog zum Herstellungsverfahren für CLA aus Ölen, die Octadecatriensäure enthalten, erfolgen: z.B. wird in einem Verfahren die Isomerisierung Linol- säure-haltiger Öle mit KOH in Propylenglykol bei 150°C durchgeführt. Es werden freie CLA-Säuren erhalten, die nur geringe Mengen ungewünschter Isomere enthält (EP-839897). In einem vorteilhaften Verfahren können Linolsäure-Alkylester mit kata- lytischen Mengen (0,3 bis 1 %) Base (Kaliumalkoholat) isomeri- siert werden, wobei CLA-Alkylester in hoher Reinheit erhalten werden (DE-1156788 und DE-1156789) . Konjugierte Fettsäuren und deren Ester können z.B. in Gegenwart von NaHS0 oder KHS0 an der Doppelbindung isomerisiert werden, s.a. DE-1141690.
Vorteilhaft ist ebenfalls, dass viel konjugierte trans/cis Octadecatriensäure, insbesondere Calendulasäure, im Gegensatz zu CLA in Pflanzen synthetisiert wird. Pflanzen lassen sich einfach und in großen Mengen anbauen und stellen daher eine preiswerte Ressource für die Produktion von z.B. Calendulaöl oder Calendulasäure dar. Insbesondere können Pflanzenarten, die für die Ernährung und Fütterung von großer Bedeutung sind, z.B. verschiedene Getreidearten, Kartoffeln, Reis, Mais usw. trans-

Besonders vorteilhaft ist es, wenn eine Aufarbeitung der in Pflanzen oder Mikroorganismen hergestellten erfindungsgemäßen konjugierten cis/trans Octadecatriensäure nicht erfolgen muß, z.B. durch Produktion in Pflanzen, die direkt zur Nahrungsaufnahme dienen, oder durch Mikroorganismen, die in Fermentations- prozeßen eingesetzt werden, wie z.B. bei der Herstellung von Käse, Joghurt, Bier, Folglich betrifft die vorliegende Erfindung auch solche Pflanzen. Insbesondere sind transgene Nutzpflanzen, wie z.B. Mais, Reis, Getreide, Kartoffeln oder transgene ölprodu- zierende Pflanzen, die Calendulasäure produzieren umfasst, z.B. Leinen, Sonnenblumen, Oliven oder Pflanzen, deren natürlicher Calendulagehalt durch klassische oder transgene Züchtung erhöht wurde .
In einer weiteren Ausführungsform betrifft die vorliegende Erfindung auch ein Kit, das die erfindungsgemäße Zubereitung enthält. Die Zubereitung kann in einem oder mehr Behältern abgepackt sein. Die Bestandteile der erfindungsgemäßen Zubereitung, insbesondere Calendulaöl oder Calendulasäure können getrennt oder zusammen in einem Behälter des Kits abgepackt sein. Das Kit kann für die Durchführung des erfindungsgemäßen Verfahrens eingesetzt werden und kann Anweisungen für die Durchführung enthalten.
Verschiedene Dokumente sind in dem vorliegenden Text dieser Beschreibung zitiert. Jedes der Dokumente (einschließlich von Anleitungen und Beschreibungen von Herstellern) gilt hiermit durch die Referenz als von der Beschreibung mit umfasst. Dies heißt jedoch nicht, dass jedes der genannten Dokumente tatsächlich Stand der Technik für die vorliegende Erfindung ist.
Die vorliegende Erfindung wird durch die folgenden Beispiele und Zeichnungen verdeutlicht, ohne dass diese in irgendeiner Weise als einschränkend gelten.
Figur 1 beschreibt die Veränderungen des Körpergewichts nach Calendulasäure-Behandlung. Figur 2 beschreibt die Nahrungsaufnahme von weiblichen und männlichen Mäusen nach Calendulasäure-Behandlung. Figur 3 beschreibt die Futterverwertung von weiblichen und männlichen Mäusen nach Calendulasäure-Behandlung. Figur 4 beschreibt die Körperfett und Protein-Zusammensetzung von weiblichen Mäusen nach Calendulasäure-Behandlung.
Beispiele
Tiere, Diäten und Analysen der Körperzusammensetzung
6 Wochen alte männliche und weibliche ICR Mäuse wurden von Charles River bezogen. 24 Tiere (12 w/m) wurden zufällig jeder Versuchsgruppe zugeteilt. Tiere wurden in Gruppen von 4 Tieren bei 25°C und 60 % relativer Luftfeuchtigkeit gehalten. Ein halbsynthetisches Futter wurde wie bei Park, J. Lipids 197997; 32, 8583-856, beschrieben, zubereitet, folgende Komponenten enthaltend:
Sucrose, Casein, Maisstärke, 5,5 % Maisöl, Mineralien- und Vitaminmix und Ethoxyquin (100 ppm) . In dem Versuchsfutter wurde das Maisöl durch eine entsprechende Menge von rohem Calendulaöl (BiRo, Sommerschenburg, Deutschland) ersetzt.

Das verwendete Calendulaöl enthielt ungefähr 62 % Calendulasäure (als Triglyzerid) . Die Fettsäurezusammensetzung des Calendulaöls ist in Tabelle 2 dargestellt.

Nach einer einwöchigen Adaptionsperiode mit Kontrollfutter wurde ein vierwöchiger Versuch durchgeführt. Körpergewicht und Futteraufnahme wurden wöchentlich gemessen. Am Ende der Studie wurden die Tiere getötet und Eingeweide und Organe wie Leber, Herz und Lunge entfernt. Die leeren Körper wurden in flüssigem Stickstoff

Ergebnisse:
Die Körpergewichtsentwicklung der beiden Gruppen war vergleichbar. Allerdings gab es deutliche Unterschiede in der Körperzusammensetzung der Tiere, vor allem was den Körperfettgehalt anbelangt .
Calendulasäure reduzierte den Fettgehalt in den Versuchstieren signifikant. Der Effekt war in männlichen Tieren stärker ausgeprägt als in weiblichen Tieren:

6. Zubereitung nach einem der Ansprüche 1 bis 4 oder Verfahren nach Anspruch 5, wobei die konjugierte trans/cis Octadecatriensäure Calendulasäure ist.
7. Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 oder Verfahren nach Anspruch 5 oder 6, wobei die konjugierte trans-cis Octadecatriensäure oder die Calendulasäure als Calendula-Öl enthalten ist.
8. Zubereitung nach einem der Ansprüche 1 bis 4, 6 oder 7 oder Verfahren nach einem der Ansprüche 5 bis 7 , wobei die genannte Zubereitung weitere Additive enthält.
9. Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 8 oder Verfahren nach einem der Ansprüche 5 bis 8, wobei die genannte Zubereitung weitere vielfach ungesättigte Fettsäuren (PUFAs) enthält.
10. Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 9 oder Verfahren nach einem der Ansprüche 5 bis 9, wobei die genannte Zubereitung konjugierte Linolsäure (CLA) enthält.
11. Zubereitung nach einem der Ansprüche 4 oder 6 bis 10 oder Verfahren nach einem der Ansprüche 5 bis 10, wobei die genannte Zubereitung ein Arzneimittel ist und einen pharmazeutisch verträglichen Träger umfasst.
12. Zubereitung nach einem der Ansprüche 3, oder 6 bis 10 oder Verfahren nach einem der Ansprüche 5 bis 10, wobei die Zubereitung für die Verwendung in der Tierernährung geeignet ist und einen oder mehr Zuschlagstoff (e) enthält.
13. Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 12 oder Verfahren nach einem der Ansprüche 5 bis 12, wobei die genannte Zubereitung weitere Wirkstoffe enthält.
14. Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 10, oder Verfahren nach einem der Ansprüche 5 bis 12, wobei die Zubereitung ein Diätmittel ist.
15. Verwendung der Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 14 zur Veränderung der Körperzusammensetzung.
16. Verwendung der Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 14, zur Reduktion der Nahrungsaufnahme oder zur Reduktion des Körperfettgehaltes bei Menschen oder Tieren.
17. Verwendung der Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 14 zur Verbesserung der Nahrungsverwertung bei Menschen oder Tieren.
18. Verwendung der Zubereitung nach einem der Ansprüche 4 oder 6 bis 14 zur Herstellung eines Arzneimittels zur Behandlung von Krebs, kardiovaskulären Erkrankungen, AIDS, Diabetes und/oder Allergien.
19. Verfahren zur Verfütterung der Calendulasäure, umfassend Zugabe von Calendulasäure zum Fettträger der Nahrung.
20. Verfahren zur Reduktion der Nahrungsmittelaufnahme bei Menschen oder Tieren, umfassend die Zugabe der Zubereitung nach einem der Ansprüche 1 bis 4 oder 6 bis 14 zu Lebensmitteln oder Tierfuttermitteln.
