Abstract
Verbindungen der Formel (I), worin der Ring mit dem Inkrement (N-A)5-, 6-, oder 7-gliedrig sein kann und zusätzlich noch ein Sauerstoff- oder Schwefelatom oder eine C-C-Doppelbindung enthalten kann, mit Ausnahme des 1,4-Benzoxazepingerüstes, A eine Carbonyl-oder Sulfonyl Gruppe darstellt, Y CH2, CH2-CH2, CH2-CH2-CH2 oder CH2-CH ist, Z ein Stickstoffatom, Kohlenstoffatom oder CH darstellt, wobei die Bindung zwischen Y und Z auch eine Doppelbindung sein Kann, n die Zahl 2,3 oder 4 bedeutet, R<1> ein Wasserstoffatom, Halogen oder eine C1-C4-Alkyl-, Trifluormethyl, Hydroxy-, C1-C4-Alkoxy- oder Amino Gruppe sein kann, R<2> eine gegebenenfalls durch C1-C4-Alkyl, Trifluormethyl-, Trifluormethoxy, Hydroxy-, Amino-, Monomethylamino-, Dimethylamino, Cyano-, oder Nitrogruppen mono oder disubstituierte Phenyl-, Pyridyl- oder Pyrazinyl-Gruppe darstellt, die mit einem Benzolkern, der durch Halogenatome, C1-C4-Alkyl, Hydroxy-, Trifluormethyl, C1-C4-Alkoxy, Amino-, Cyano- oder Nitrogruppen mono- oder disubstituiert sein kann und gegebenenfalls 1 Stickstoffatom enthalten kann, oder mit einem 5- oder 6- gliedrigen Ring, der 1 bis 2 Sauerstoffatome enthalten kann, anelliert sein kann, sowie deren physiologisch verträgliche Salze, zeigen Affinität zum 5-HT1A-Rezeptor und eignen sich zur Behandlung der zerebralen Ischämie.

R9 für Wasserstoff, (Cχ-6) Alkyl verzweigt oder unverzweigt, CO-(Cι_4) -Alkyl, C02tBu, CO-Aryl und einen Phenylalkyl-8 Cι~C4-Rest, der seinerseits am Aromaten durch F, Cl, Br, I, C1-C4 Alkyl, C1-C4 Alkoxy, Trifluormethyl, Hydroxy, Amino, Cyano oder Nitro substituiert sein kann, steht.
Diese Verbindungen eignen sich aufgrund ihrer 5-HTιA Affinität für die Behandlung von zerebraler Ischämie, insbesondere von Schlaganfall .
Hierbei spielt der 5-HTιΑ Agonismus eine besondere Rolle, wie man aus den Arbeiten von SMITHKLINE BEECHAM (EP 345 948), BAYER/ TROPON (EP 749 970; De Vry et al., Drugs of the Future 1997, 22(4), S. 341-349) und SUNTORY (WO 96/24594, 99/03847) ersehen kann.
Es wurde nun gefunden, daß sich bicyclische Verbindungen der Formel I
worin
der Ring mit dem Inkrement (N-A) 5-, 6- oder 7-gliedrig sein kann und zusätzlich noch ein Sauerstoff- oder Schwefelatom oder eine C-C-Doppelbindung enthalten kann, mit Ausnahme des 1, 4-Benzoxazepingerüstes,
A eine Carbonyl- oder Sulfonyl Gruppe darstellt,
X ein Stickstoffatom bedeutet,
Y CH2, CH2-CH2, CH2-CH2-CH2 oder CH2-CH ist,
Z ein Stickstoffatom, Kohlenstoffatom oder CH darstellt, wobei' die Bindung zwischen Y und Z auch eine Doppelbindung sein kann,
n die Zahl 2, 3 oder 4 bedeutet,
R1 ein Wasserstoffatom, Halogen oder eine Cι-C4-Alkyl-, Trifluormethyl-, Hydroxy-, Cι-C4~Alkoxy- oder Amino Gruppe sein kann,
R2 eine gegebenenfalls durch Cχ-C4-Alkyl, Trifluormethyl-, Trifluormethoxy-, Hydroxy-, Amino-, Monomethylamino-, Dimethylamino-, Cyano- oder Nitrogruppen mono oder disubstituierte Phenyl-, Pyridyl- oder Pyrazinyl-Gruppe darstellt, die mit einem Benzolkern, der durch Halogenatome, Cι~C4-Alkyl, Hydroxy-, Trifluormethyl, Cι-C4-Alkoxy-, Amino-, Cyano- oder Nitrogruppen mono- oder disubstituiert sein kann und gegebenenfalls 1 Stickstoffatom enthalten kann, oder mit einem 5- oder 6-gliedrigen Ring, der 1 bis 2 Sauerstoffatome enthalten kann, anelliert sein kann,
sowie deren physiologisch verträgliche Salze,
zur Herstellung von Medikamenten zur Prophylaxe und Therapie von Neurodegeneration, Hirntrauma und zerebraler Ischämie, insbesondere Schlaganfall, bzw. den durch diese Krankheiten hervorgerufenen Folgeerkrankungen, eignen.
Eine erfindungsgemäδe Verwendung betrifft auch die Neuro- protektion.
Verbindungen der Formel I, die ein unten dargestelltes 1, 4-Benzoxazepingerüst besitzen, sind vom Patentanspruch ausgenommen.

Diese Verbindungen der Formel I lassen sich herstellen, indem man eine Verbindung der Formel II
in der R1, A und der Ring mit dem Inkrement (N-A) die oben angegebenen Bedeutungen haben, mit einem reaktiven Baustein der Formel III

Natriumhydrid oder dem Natriumsalz eines niederen Alkohols oder einem Alkalicarbonat, umsetzt und die so erhaltene Verbindung gegebenenfalls in das Säureadditionssalz einer physiologisch verträglichen Säure überführt.
Die Umsetzung erfolgt zweckmäßig in einem inerten organischen Lösungsmittel, insbesondere DMF oder einem niederen Alkohol, z.B. Methanol oder Ethanol, oder einem cyclischen gesättigten Ether, insbesondere Tetrahydrofuran oder Dioxan oder einem Kohlenwasserstoff, wie z.B. Toluol oder Xylol.
Die Umsetzung erfolgt in der Regel bei Temperaturen von 20 bis 190°C, insbesondere von 60 bis 90°C, und ist im allgemeinen innerhalb von 1 bis 10 Stunden beendet .
Oder man setzt eine Verbindung der Formel II
in der R1, A und der Ring mit dem Inkrement (N-A) die oben angegebenen Bedeutungen haben, mit einem reaktiven Baustein der Formel IV

worin W eine leaving group, wie z.B. Cl oder Br, darstellt, in Gegenwart einer Base, bevorzugt sind Alkalihydroxide, in einem inerten Lösungsmittel, vorzugsweise Halogenkohlenwasserstoffe, bevorzugt als zweiphasige Reaktion mit Wasser unter Zusatz eines Phasentransferkatalysators (Aralkyl- bzw. Alkylammoniumsalze) oder ohne Lösungsmittel unter Zusatz eines Aralkyl- bzw. Alkyl- ammoniumsalzes, bei Temperaturen zwischen 20 und 120°C zum Cyclisierungsprodukt V um.
Zuletzt setzt man das Halogenderivat der Formel V mit einem A in der allgemeinen Formel VI

worin X, Y, Z und R2 die oben angegebenen Bedeutungen haben, zum erfindungsgemäßen Endprodukt der Formel I um. Diese Umsetzung verläuft am besten in einem inerten organischen Lösungsmittel, vorzugsweise Toluol oder Xylol, in Gegenwart einer Base, wie z.B. Kaliumcarbonat oder Kaliumhydroxyd, bei Temperaturen zwischen 60 und 150°C.
Die erfindungsgemäßen Verbindungen der Formel I können entweder durch Umkristallisation aus den üblichen organischen Lösungsmitteln, bevorzugt aus einem niederen Alkohol, wie Ethanol, umkristallisiert oder durch Säulenchromatographie gereinigt werden.
Die freien bicyclischen Verbindungen der Formel I können in üblicher Weise in die Säureadditionssalze einer Lösung mit der stöchiometrischen Menge der entsprechenden Säure. Pharmazeutisch verträgliche Säuren sind beispielsweise Salzsäure, Phosphorsäure, Schwefelsäure, Methansulfonsäure, Amidosulfonsäure, Maleinsäure, Fumarsäure, Oxalsäure, Weinsäure oder Zitronensäure.
,Die als Ausgangsmaterialien eingesetzten Verbindungen der Formeln II, III, V und VI sind literaturbekannt oder lassen sich nach analogen Literaturvorschriften herstellen.
:Die Verbindungen der vorliegenden Erfindung besitzen eine über- .raschend hohe Affinität zum 5-HTιA-Rezeptor, wie BindungsStudien mit klonierten humanen 5-HTι-Rezeptoren zeigten.
Die folgende Testanordnung benutzte man zur Bestimmung der Rezeptorbindungs-Affinität :
'5-HTιA-Bindungsassay mit Membranen von 5-HTχA-Rezeptor iexpri ierenden HEK293-Zellen
jKultur von 5-HTιA-Rezeptor expri ierenden HEK293-Zellen 5-HTIA exprimierende HEK293-Zellen werden in RPMI/Glutamax-Mediurα (RPMI 1640, 25 M Hepes, 2 M Glutamax, 10 % FCS, 2 M Glutamin, Penicillin/Streptomycin (100 IU/ml each) , Geneticin G-418-Sulfate 400 mg/1, NaHC03 1,2 g/1) in Kulturflaschen (TripleFlasks T - 175) in einer 5 % CO2 Atmosphäre bei 37°C in kultiviert. Nach Erreichen der Konfluenz wird das Medium entnommen und die Flaschen mit 15 ml sterilen PBS (phosphate buffered saline) gefüllt. Die Zellen werden durch 10-minütige Inkubation (Brutschrank, 37°C) mit einer Trypsin-Lösung (0,05 % Trypsin, 0,0004 % EDTA, 0,02 % EGTA, 2,682 mM KCL, 1,47 mM KH2P0 , 6,46 M NaHP0 , 136,89 mM NaCl) gelöst. Das Ablösen der Zellen wird durch Klopfen auf den Flaschenboden gefördert. Nach Überführen in 50-ml-Röhrchen , (Greiner) werden die Zellen bei 250 x g bei Raumtemperatur zentrifugiert . Der Überstand wird verworfen und die Zellen in 10 ml Medium resuspendiert. Die Zellen werden erneut auf Kulturflaschen verteilt und weitere 5 bis 6 Tage bis zur Präparation der Membranen kultiviert.
Präparation der Membranen von 5-HTχ-Rezeptor-exprimierenden HEK293-Zellen
Die Überstände der Zellen werden abgenommen und die Kulturflaschen mit PBS gefüllt. Die Zellen werden daraufhin 10 Minuten mit einer Trypsin-Lösung (zur Zusammensetzung siehe oben) inkubiert. Das Ablösen der Zellen wird durch Klopfen auf den Flaschenboden gefördert. Die Zellsuspension wird entnommen und
die verbleibenden Zellen durch 2-maliges Waschen der Kulturflaschen mit PBS ebenfalls in PBS aufgenommen. Die gesammelte Zellsuspension wird auf 150-ml-Falcon-Röhrchen verteilt und 10 Minuten bei 250 x g bei 4°C zentrifugiert. Die Überstände werden verworfen und die Zellen im Pellet in PBS resuspendiert . 20 μl der Zeil-Suspension werden entnommen und die Zelldichte •bestimmt. Die Zellen werden erneut 10 Minuten bei 250 x g (4°C) zentrifugiert, der Überstand verworfen und die Zellen im Pellet in 50 mM Tris-HCl pH 7,4 (1 ml / 108 Zellen) mit Hilfe eines Ultra-Turrax (30 sec) homogenisiert. Das Homogenat wird auf 'Kryo-Röhrchen verteilt (1 ml / Kry-Röhrchen) und bis zur Verwendung im Bindungsassay in flüssigen Stickstoff gelagert.
5-HTιA-Bindungsassay
'Die eingefrorenen Membranen werden bei 37°C aufgetaut, bei 48000 * g (20 Minuten) zentrifugiert, und in Bindungspuffer (50 mM Tris- HCl pH 7,4, 5 mM CaCl2) resuspendiert. Ein Inkubationsansatz enthält Membranmaterial von 50 mg/Probe, 0,15 pmol (= 0,15 nM) 3H-8-OH-DPAT sowie die zu testenden Substanzen in insgesamt 1 ml Bindungspuffer. Die unspezifische Bindung wird in Gegenwart ,von 10~5 M 5-Carboxamidotryptamine bestimmt. Nach erfolgter 90-minütiger Inkubation bei 22°C wird gebundener und freier Ligand ,durch Filtration über CF/B-Filter und anschließendem Waschen mit '5 bis 9 ml eiskaltem Bindungspuffer voneinander getrennt. Die GF/B-Filter werden vor Verwendung mindestens 2 Stunden mit 0,3 % Polyethylenimin behandelt . Nach erfolgter Filtration werden die Filter mit 3 bis 4 ml Packard Ultima Gold XR versetzt und die Radioaktivität durch Flüssigkeits-Scintillationszählung im Packard Tricarb bestimmt.
Auswertung der Daten des 5-HTιA-Bindungsassays Die Verdrängungs-Kurven werden durch nichtlineare Regression mit Hilfe einer modifizierten Version des "Ligand"-Programmes von Munson & Rodbard (Anal. Biochem. , 107, 220 (1980)) ;analysiert. Der Wert für die theoretische unspezifische Bindung •wird als theoretische Radioligand-Bindung bei infinitisimal hohen Ligandenkonzentration geschätzt. Dabei werden die gemessenen Werte für die unspezifische Bindung als Datenpunkte der Verdrängungskurve behandelt, die Meßpunkte bei einer infinitisimal hohen Liganden-Konzentration entsprechen. Bei Testung von weniger als 4 Konzentrationen einer Substanz oder bei spezifischer Verdrängung des Radioliganden < 25 % (bei allen getesteten Konzentrationen) wird ein ICs0-Wert unter Verwendung der Hill-Gleichung geschätzt und der Ki-Wert nach der Gleichung von Cheng und Prusoff (Biochem. Pharmacol. 22, 3099 (1973)) berechnet .
Die folgenden Resultate (Ki-Werte) werden erhalten:
Die folgenden Beispiele dienen zur Erläuterung der Erfindung:
A Allgemeine Herstellung der Ausgangsmaterialien der Formel III, V und VI
a) 1. 1- [4- (2-Hydroxyethyl) -piperazin-1-yl] -isochinolin
17,8 g (109 mM) 1-Chlor-isochinolin in 100 ml Ethanol wurden mit 47,0 g (350 mM) 1- (2-Hydroxyethyl) - piperazin versetzt und 16 h am Rückfluß gekocht. Nach dem Abkühlen verteilte man die Reaktionsmischung zwischen Essigester und Wasser, stellte mit Ammoniumhydroxid auf pH = 9 und extrahierte die wäßrige Phase noch zweimal mit Essigester nach. Nach Trocknen und Einengen der organischen Phase isolierte man 26,2 g (94 %) Produkt als viskoses Öl.
2. 1- [4- (2-Chlorethyl) -ρiperazin-1-yl] isochinolin
26,2 g (102 mM) 1- [4- (2-Hydroxyethyl) -piperazin- 1-yl] -isochinolin in 200 ml DMF wurden mit 20,7 g (205 mM) Triethylamin versetzt. Anschließend tropfte man unter gutem Rühren bei Raumtemperatur 23,4 g (205 mM) Methansulfonylchlorid während 20 min hinzu und ließ das Reaktionsgemisch noch 2 h bei Raumtemperatur nachrühren.. Danach verteilte man den Ansatz zwischen Essigester und Wasser, stellte mit Ammoniumhydroxid auf pH = 9 und wusch die organische
Phase nochmals mit Wasser durch. Nach Trocknen und Einengen der organischen Phase isolierte man 26,0 g (93 %) Produkt als Öl, das langsam durchkristallisiert .
Das Produkt kann bei Bedarf durch Säulenchromatographie (Kieselgel, Laufmittel Essigester) noch weiter gereinigt werden, Schp 87 bis 89°C.
b) 1- (2-Chlorethyl) -4- (3-trifluormethyl-phenyl) -piperazin
100,0 g (434 mM) 1- (3-Trifluormethyl) -piperazin in 1 1 Toluol wurden mit 94,1 g (655 mM) l-Brom-2-chlor-ethan und 43,9 g (434 mM) Triethylamin versetzt und das Reaktionsgemisch 4,5 h am Rückfluß gekocht. Nach dem Abkühlen goß man den Ansatz auf Eis/Wasser, trennte die organische Phase ab und extrahierte die wäßrige Phase noch einmal mit Methyl-t-butyl-ether. Nach Trocknen und Einengen der organischen Phase isolierte man 133 g Rohprodukt, das über eine Kieselgelsäule (Laufmittel Heptan/Essigester 1/1) gereinigt wurde. Man isolierte 68,3. g (54 %) reines Produkt als Öl.
c) 2- (2-Chlorethyl) -2 , 3-dihydro-l, 2-benzisothiazol-l, 1- dioxid
Zu 144,1 g (852 mM) 2 , 3-Dihydro-l, 2-benzisothiazol- 1,1-dioxid und 244,2 g (1,7 M) l-Brom-2-chlor-ethan in 25 ml Methylenchlorid ließ man unter gutem Rühren eine Mischung aus 940 g 20 %iger Natronlauge und 13,7 g (42,6 mM) Tetrabutylammoniumbromid während 10 min zutropfen. Man ließ noch 5 h. nachrühren und gab anschließend 400 ml Toluol hinzu. Die organische Phase wurde noch zweimal mit Wasser nachgewaschen, die wäßrige Phase zweimal mit je 200 ml Toluol nachextrahiert. Die vereinigten organischen Phasen lieferten nach Trocknen und Einengen das Rohprodukt, das aus ϊsopropanol umkristallisiert wurde. Ausbeute 142 g (72 %) mit Schmp. 88 bis 90°C.
d) 2- (2-Chlorethyl) -1-isoindolinon
30,0 g (225 mM) Phthalimidin in 300 ml 1, 2-Dichlorethan wurden mit 21,6 g (338 mM) KOH Pulver (88 %) sowie mit 1,0 g (4,39 mM) Triethylbenzylammoniumchlorid versetzt und 3 h am Rückfluß gekocht. Nach dem Abkühlen setzte man 400 ml Wasser hinzu, ließ die Phasen bei pH = 7 trennen und extrahierte die wäßrige Phase noch einmal mit
Methylenchlorid nach. Die vereinigten organischen Phasen engte man nach dem Trocknen ein. Das Rohprodukt reinigte man durch Säulenchromatographie (Kieselgel, Laufmittel Methylenchlorid/Methanol 100/1). Man isolierte 13 , 3 g (30 %) Produkt mit Schmp. 77 bis 79°C.
e) 1- (4-Trifluormethyl-2-pyridinyl) -piperazin
28,0 g (154 mM) 2-Chlor-4-trifluormethyl-pyridin in 250 ml Ethanol wurden mit 93 , 0 g (1079 mM) Piperazin versetzt und das Reaktionsgemisch 5 h am Rückfluß gekocht. Nach dem Abkühlen engte man den Ansatz zur Trockene ein, verteilte den Rückstand zwischen Essigester und Wasser, wusch die organische Phase noch zweimal mit Wasser nach • und engte nach dem Trocknen ein. Man isolierte 34 g (95 %) Produkt als Öl.
f) 1- (3-Trifluormethyl-phenyl) -1, 4-diazepan
10,0 g (44,4 mM) l-Brom-3-trifluormethyl-benzol in 300 ml Xylol wurden mit 6,68 g (66,7 mM) 1,4-Diazepan (Homopiperazin) , 0,60 g (2,67 mM) Pd-II-acetat, 1,62 g (5,33 mM) Tri-o-tolylphosphin sowie 6,70 g (62,2 mM) Kalium-tert . -butanolat unter Stickstoff und unter gutem Rühren versetzt und 16 h am Rückfluß gekocht. Nach dem Abkühlen verdünnte man das Reaktionsgemisch mit Methylenchlorid, filtrierte und engte das Filtrat ein. Den Rückstand verteilte man zwischen Methyl-tert .-butyl- ether und Wasser und engte die organische Phase nach dem Trocknen ein. Das Rohprodukt reinigte man durch Säulen- chromtographie (Kieselgel, Laufmittel THF/Methanol/ Ammoniak 50/50/1). Man isolierte 3 , 64 g (34 %) Produkt als Öl.
g) 2- (3-Chlorethyl) -3, 4-dihydro-l (2H) -isochinolinon
2,0 g (13,6 mM) 3,4-Dihydro-l (2H) -isochinolinon in 80 ml Dimethylformamid wurden unter Stickstoff und unter gutem Rühren mit 0.49 g (16,3 mM) Natriumhydrid (80 %ig) versetzt und 30 min nachgerührt. Anschließend fügte man 3,2 g (20,4 M) l-Brom-3-chlorpropan hinzu und ließ das Reaktionsgemisch 1 h bei Raumtemperatur nachrühren. Nach dem Abkühlen verteilte man den Rückstand zwischen Essigester und Wasser und säuerte mit verdünnter Salzsäure an. Die organische Phase wurde noch einmal mit verdünnter Salzsäure gewaschen. Nach Trocknen und Einengen der organischen Phase isolierte man 2,1 g {66 %) Produkt, das genügend rein für die weiteren Umsetzungen war.
h) 1- (4-Ghlorbutyl) -3, 4-dihydro-2 (1H) -chinolinon
2,0 g (13,6 mM) 3,4-Dihydro-chinolinon-2 in 35 ml Dimethylformamid wurden unter Stickstoff und unter gutem Rühren mit 0.45 g (15, 0 mM) Natriumhydrid (80 %ig) versetzt und 30 min nacitgerührt. Anschließend fügte man 2,4 g (14,0 mM) 4-Brom-l-chlorbutan hinzu und ließ das Reaktionsgemisch 3 h bei Raumtemperatur nachrühren. Nach dem Abkühlen verteilte man den Rückstand zwischen Essigester und Wasser und säuerte mit verdünnter Salzsäure an. Die organische Phase wurde noch einmal mit verdünnter Salzsäure gewaschen. Nach Trocknen und Einengen der organischen Phase isolierte man 3,1 g (96 %) Produkt, das genügend rein für die weiteren Umsetzungen war.
i) tert-Butyl-4- (8-chinolinyl) -1-piperazin-carboxylat
Eine Lösung von 9,0 g 8-C lorchinolin (55,0 mmol) , 10,2 g tert-Butyl-1- piperazin-carboxylat (55,0 mmol), 0,66 g 2- (Di- (tert-butyl) -phosphino) - 1, 1 ' -biphenyl (2,2 mmol) und 8,23 g Natrium- ert-butoxid (85,6 mmol) in 300 ml wasserfreiem Toluol wurde mit 0,25 g Palladium(II) -acetat (1,1 mmol) versetzt und 10 h unter Stickstoff zum Rückfluss erhitzt. Das Reaktionsgemisch wurde abgekühltund vom Lösemittel befreit. Der erhaltene Rückstand wurde in Essigester aufgenommen und mit gesättigter wässriger Natriumchloridlösung extrahiert und über Natriumsulfat getrocknet. Nach dem Entfernen des Lösemittels erhielt man 17,6 g eines Rohproduktes, das durch Flash-Säulenchromatographie (Kieselgel; Heptan/Essigester, 3/1) gereinigt wurde. Als Hauptfraktion erhielt man 13,3 g (77 %) der Titelverbindung: 1H-NMR (CDCI3, 270 MHz) d = 1.5 (s, 9 H), 3.35 (t, 4 H) , 3.8 (t, 4 H) , 7.17 (m, 1 H) , 7.4 (m, 1 H) , 7.45 (m, 2 H), 8.15 (dd, 1H) , 8.9 (m, 1H) .
k) 8- (1-Piperazinyl) -chinolin
Eine Mischung aus 13,28 g tert-Butyl-4- (8-chinolinyl) - 1-piperazin-carboxylat (42,38 mmol), 13,0 g Trifluoressigsäure (169,5 mmol) und 9,2 ml Anisol (84,8 mmol) wurde 3 h unter Rühren auf 80°C erhitzt. Anschließend wurden die flüchtigen Bestandteile unter Vakuum abdestilliert, der erhaltene Rückstand in Dichlormethan aufgenommen, mit gesättigter wässriger Natriumhydrogencarbonatlösung gewaschen und über Natriumsulfat getrocknet. Nach dem Abziehen des Lösemittels erhielt man 7,16 g der leicht verunreinigten Titelverbindung, die ungereinigt weiter umgesetzt wurde: -""H-NMR (CDCI3, 400 MHz) d = 2.0 (br, 1H) , 3.25 (m, 4 H), 3.4 (m, 4 H), 7.15 (m, 1 H) , 7.4 (m, 1 H) , 7.45 (m, 2 H) , 8.1 (dd, 1 H) , 8.9 (m, 1H) .
Analog diesen Vorschriften stellte man auch die übrigen Vorprodukte der Formel III, V und VI her.
Herstellung der Endprodukte
Beispiel 1
2- [2- (4- (1-Isochinolinyl) -1-piperazinyl) -ethyl] -3 , 4-dihydro-
1(2H) -isochinolinon x 2 HC1 x 2 H20
1,0 g (6,8 mM) 3, 4-Dihydro-l (2H) -isochinolinon in 50 ml Dimethylformamid wurden unter Stickstoff und unter gutem Rühren mit 0,2 g (6,8 mM) Natriumhydrid (80 %ig) versetzt und 30 min nachgerührt. Anschließend fügte man 1,9 g (6,8 mM) 1- [4- (2-Chlorethyl) -piperazin-1-yl] -isochinolin hinzu und ließ das Reaktionsgemisch 2 h bei 100°C nachrühren. Nach dem Abkühlen engte man zur Trockene ein, verteilte den Rückstand
zwischen Methylenchlorid und Wasser und stellte mit verdünnter Natronlauge auf pH = 9. Die wäßrige Phase wurde noch einmal mit Methylenchlorid extrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 2,7 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Laufmittel Essigester/Methanol 10/1) gereinigt wurde. Man isolierte 0,9 g (34 %) Produkt, das in Ether gelöst und mit etherische Salzsäure in das Hydrochlorid mit Schmp. 118 bis 120°C übergeführt wurde.
Beispiel 2
2- [2- (4- (1-Isochinolinyl) -1-piperazinyl) -ethyl] -1 (2H) - isochinolinon x 2 HC1 x H20
1,45 g (10,0 mM) 1 (2H) -Isochinolinon (Isocarbostyril) in 50 ml Dimethylformamid wurden unter Stickstoff und unter gutem Rühren mit 0,35 g (11,7 mM) Natriumhydrid (80 %ig) versetzt und 30 min nachgerührt. Anschließend fügte man 2,75 g (10,0 mM) 1- [4- (2-Chlorethyl) -piperazin-1-yl] -isochinolin hinzu und ließ das Reaktionsgemisch 2 h bei 80°C nachrühren. Nach dem Abkühlen engte man zur Trockene ein, verteilte den Rückstand zwischen Essigester und Wasser und stellte mit verdünnter Natronlauge auf pH = 9. Die wäßrige Phase wurde noch einmal mit Essigester extrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 5,0 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Laufmittel Essigester/Ethanol 14/1) gereinigt wurde. Man isolierte 3,15 g (82 %) Produkt, das in Ether gelöst und mit etherischer Salzsäure in das Hydrochlorid mit Schmp. 146 bis 148°C übergeführt wurde.
Beispiel 3
2- [2- (4- (1-Isochinolinyl) -1-piperazinyl) -ethyl] -2, 3-dihydro-
1,2-benzisothiazol-l, 1-dioxid x 2 HCl
2,5 g (10,8 mM) 2- (2-Chlorethyl) -2, 3-dihydro-l,2-benz- isothiazol-1, 1-dioxid in 30 ml Xylol wurden mit 1,5 g (10,8 mM) fein pulverisiertem Kaliumcarbonat sowie mit 2,5 g (12,0 mM) l-(Piperazin-l-yl) -isochinolin versetzt und 24 h unter Rückfluß gekocht. Nach dem Abkühlen engte man den Reaktionsansatz zur Trockene ein und verteilte den Rückstand zwischen Methylenchlorid und Wasser bei pH = 10. Die wäßrige Phase wurde noch einmal mit Methylenchlorid nachextrahiert . Nach Trocknen und Einengen der organischen Phase isolierte man 5,6 g Rohprodukt, das durch Säulenchromatographie (Kieεelgel, Laufmittel Methylenchlorid/Methanol 20/1) gereinigt wurde. Man isolierte 2,4 g (55 %) Produkt, das
in Ether gelöst und mit etherischer Salzsäure in das Hydrochlorid mit Schmp. 158 bis 168°C übergeführt wurde.
Beispiel 4
2- [2- (4- (6-Methyl-2-pyridinyl)-l-piperazinyl) -ethyl] -1- isoindolinon
2,2 g (11,25 mM) 2- (2-Chlorethyl) -1-isoindolinon in 30 ml Xylol wurden mit 1,55 g (11,25 mM) fein pulverisiertem Kaliumcarbonat sowie mit 1,99 g (11,25 mM) 1- (2- (6-Methyl- pyridyl) -piperazin versetzt und 4 h unter Rückfluß gekocht. Nach dem Abkühlen engte man den Reaktionsansatz zur Trockene ein und verteilte den Rückstand zwischen Methylenchlorid und Wasser. Die wäßrige Phase wurde noch einmal mit Methylenchlorid nachextrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 4,5 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Laufmittel Methylenchlorid/ Methanol 30/1) gereinigt wurde. Man isolierte 2,2 g (58 %) Produkt mit Schmp. 130 bis 132°C.
Beispiel 5
1- [2- (4- (3-Trifluormethyl-phenyl) -1-piperazinyl) -ethyl] -1, 3- dihydro-2H-indol-2-on x 2 HC1
1,0 g (7,5 mM) Oxindol in 30 ml Toluol wurden mit 2,2 g (7,5 mM) l-(2-Chlorethyl) -4- (3-trifluormethyl-phenyl)- piperazin sowie mit 0,55 g (3,75 mM) fein pulverisiertem Kaliumcarbonat 12 h unter Rückfluß gekocht. Nach dem Abkühlen engte man den Reaktionsansatz zur Trockene ein und verteilte den Rückstand zwischen Methylenchlorid und Wasser. Die wäßrige Phase wurde noch einmal mit Methylenchlorid nachextrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 4,3 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Laufmittel Methylenchlorid/ Methanol 30/1) gereinigt wurde Man isolierte 1,9 g (65 %) Produkt, das in Ether gelöst und mit etherischer Salzsäure in das Hydrochlorid mit Schmp. 256 bis 258°C übergeführt wurde.
Beispiel 6
1- [2- (4- (l-Isochinolinyl)-l-piperazinyl) -ethyl] -3 ,4-dihydro-
2 (1H) -chinolinon
1,5 g (10,2 mM) 3, 4-Dihydrochinolinon-2 in 30 ml Dimethylformamid wurden mit 350 mg (11,7 mM) 80 %igem Natriumhydrid unter Stickstoff und unter gutem Rühren versetzt und 30 min nachgerührt. Anschließend fügte man 2,8 g (10,2 mM) 1- [4- (2-Chlorethyl)-piperazin-l-yl] -isochinolin hinzu und
ließ das Reaktionsgemisch 3 h bei 80°C nachrühren. Nach dem Abkühlen engte man zur Trockene ein, verteilte den Rückstand zwischen Essigester und Wasser und' stellte mit verdünnter Natronlauge auf pH = 9. Die wäßrige Phase wurde noch einmal mit Essigester extrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 3,7 g Rohprodukt, das in 20 ml Essigester ausgerührt, gekühlt und abgesaugt wurde. Die Kristalle wusch man mit wenig Essigester nach und ließ an der Luft trocknen. Man isolierte 2,4 g (61 %) Produkt mit Schmp. 133 bis 135°C.
Beispiel 7
1- [2- (4- (1-Isochinolinyl) -1-piperazinyl) -ethyl] -2 (1H) - chinolinon x 2 HC1 x H20
1,0 g (6,9 mM) 2-Hydroxychinolin in 25 ml Dimethylformamid wurden unter Stickstoff und unter gutem Rühren mit 0,25 g (8,3 mM) Natriumhydrid (80 %) versetzt und 1 h nachgerührt. Anschließend fügte man 2,0 g (7,0 mM) 1- [4- (2-Chlorethyl) - piperazin-1-yl] -isochinolin hinzu und ließ das Reaktionsgemisch 2 h bei 85°C nachrühren. Nach dem Abkühlen engte man zur Trockene ein, verteilte den Rückstand zwischen Essigester und Wasser und stellte mit verdünnter Natronlauge auf pH = 8. Die wäßrige Phase wurde noch einmal mit Essigester extrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 3,4 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Laufmittel Essigester/ Ethanol 14/1) gereinigt wurde. Man isolierte 2,0 g (75 %) Produkt, das in Ether/Essigester gelöst und mit etherischer Salzsäure in das Hydrochlorid mit Schmp. 257 bis 259°C übergeführt wurde . Beispiel 8
2- [2- (4- (1-Naphthyl) -1-piperazinyl) -ethyl] -2 , 3-dihydro-l, 2- benzisothiazol-1, 1-dioxid x HC1
3,0 g (12,96 mM) 2- (2-Chlorethyl) -2, 3-dihydro-l,2-benz- isothiazol-1, 1-dioxid in 30 ml Xylol wurden mit 1,79 g (12,96 mM) fein pulverisiertem Kaliumcarbonat sowie mit 2,73 g (12,96 mM) 1- (1-Naphthyl) -piperazin versetzt und 5 h unter Rückfluß gekocht . Nach dem Abkühlen engte man den Reaktionsansatz zur Trockene ein und verteilte den Rückstand zwischen Methylenchlorid und Wasser. Die wäßrige Phase wurde noch einmal mit Methylenchlorid nachextrahiert. Nach Trocknen und Einengen der organischen Phase isolierte man 7,2 g Rohprodukt, das durch Säulenchromatographie (Kieselgel, Lauf- mittel Methylenchlorid/Methanol 50/1) gereinigt wurde. Man isolierte 3,5 g (66 %) Produkt, das in Ether gelöst und mit
etherischer Salzsäure in das Hydrochlorid mit Schmp. 278 bis 280°C übergeführt wurde.
Analog der Beispiele 1 bis 8 wurden hergestellt :

Claims
1. Verbindungen der Formel I
worin
der Ring mit dem Inkrement (N-A) 5-, 6- oder 7-gliedrig sein kann und zusätzlich noch ein Sauerstoff- oder Schwefelatom oder eine C-C-Doppelbindung enthalten kann, mit Ausnahme des • 1 , 4-Benzoxazepingerüstes ,
A eine Carbonyl- oder Sulfonyl Gruppe darstellt,
X ein Stickstoffatom bedeutet,
Y CH2, CH2-CH2, CH2-CH2-CH2 oder CH2-CH ist,
Z ein Stickstoffatom, Kohlenstoffatom oder CH darstellt, wobei die Bindung zwischen Y und Z auch eine Doppelbindung sein kann,
n die Zahl 2 , 3 oder 4 bedeutet ,
R1 ein Wasserstoffatom, Halogen oder eine Cι-C4-Alkyl-, Trifluormethyl-, Hydroxy-, Cι-C4-Alkoxy- oder Amino Gruppe sein kann,
R2 eine gegebenenfalls durch Cχ-C4-Alkyl, Trifluormethyl-, Trifluormethoxy-, Hydroxy-, Amino-, Monomethylamino- , Dimethylamino-, Cyano- oder Nitrogruppen mono oder disubstituierte Phenyl-, Pyridyl- oder Pyrazinyl- Gruppe darstellt, die mit einem Benzolkern, der durch Halogenatome, Cχ-C4-Alkyl, Hydroxy-, Trifluormethyl, Cι-C4-Alkoxy-, Amino-, Cyano- oder Nitrogruppen mono- oder disubstituiert sein kann und gegebenenfalls 1 Stickstoffatom enthalten kann, oder mit einem 5- oder 6-gliedrigen Ring, der 1 bis 2 Sauerstoffato e enthalten kann, anelliert sein kann,
sowie deren physiologisch verträgliche Salze.
2. Verwendung von Verbindungen nach Anspruch 1 zur Herstellung von Medikamenten.
3. Verwendung nach Anspruch 2 zur Prophylaxe und Therapie von Neurodegeneration, Hirntrauma und zerebraler Ischämie, insbesondere Schlaganfall, bzw. den durch diese Krankheiten hervorgerufenen Folgeerkrankungen.
