Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von alpha -Dicarbonylverbindungen der Formel (I), worin R<1> und R<2> unabhängig voneinander C1-6-Alkyl bedeuten. Das Verfahren ist dadurch gekennzeichnet, dass ein alpha , beta -ungesättigtes Keton der Formel (II), worin R<1> und R<2> die oben angegebene Bedeutung haben und R<3> und R<4> unabhängig voneinander Wasserstoff, C1-6-Alkyl oder Di-C1-6-alkylamino bedeuten, mit Ozon umgesetzt und das gebildete Ozonid reduktiv aufgearbeitet wird.
Description
Verfahren zur Herstellung von Dicarbonyl-Nerbindungen
Die Erfindung betrifft ein Verfahren zur Herstellung von α-Dicarbonyl- Verbindungen (α-Diketonen), insbesondere zur Herstellung von 2,3-Pentandion.
2,3-Pentandion ist ein Bestandteil des Butteraromas und wird deshalb als synthetischer Aromastoff industriell hergestellt. Es eignet sich — wie auch andere α-Dicarbonylverbindungen — auch als Synthesebaustein für die Herstellung weiterer Feinchemikalien und Wirkstoffe.
2,3 -Pentandion kann beispielsweise durch Kondensation von Milchsäure (2-Hydroxy- propansäure) hergestellt werden (US-A-5 831 130). Die Kondensationsreaktion erfolgt bei Temperaturen von ca. 300 °C.
US-A-3 153 066 besclireibt die Ozonolyse von Ketonen in Gegenwart von Selendioxid zur Herstellung von α-Dicarbonylverbindungen.
Aufgabe der vorliegenden Erfindung war es, ein einfaches Verfahren zur Herstellung von α-Dicarbonylverbindungen bereitzustellen. ,
Es wurde gefunden, dass durch Umsetzung von α,ß-ungesättigten Ketonen mit Ozon α-Dicarbonylverbindungen erhalten werden können.
Die Erfindung betrifft somit ein Verfahren zur Herstellung von α-Dicarbonylverbindungen der Formel
1 9 worin R und R unabhängig voneinander Cι_6-Alkyl bedeuten. Das Verfahren ist dadurch
BESTÄTIGUΝGSKOPIE
gekennzeichnet, dass ein α,ß -ungesättigtes Keton der Formel
worin R1 und R2 die oben angegebene Bedeutung haben, und R3 und R4 unabhängig voneinander Wasserstoff, Cι_6-Alkyl oder Di-Cι_6-alkylamino bedeuten, mit Ozon umgesetzt und das Reaktionsgemisch anschliessend reduktiv aufgearbeitet wird.
Unter „Cι_6-Alkyl" sind hier und im folgenden alle linearen oder verzweigten Alkylgruppen mit 1-6 Kohlenstoffatomen zu verstehen, wie beispielsweise Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, sec-Butyl, tert-Butyl, Pentyl, Isopentyl, tert-Pentyl, Neopentyl, Hexyl oder Isohexyl.
Der Rest R ist vorzugsweise Methyl. Der Rest R ist vorzugsweise Ethyl.
Die Umsetzung mit Ozon erfolgt vorteilhaft in einem geeigneten Lösungsmittel bei Temperaturen im Bereich von -100 °C bis 30 °C. Geeignete Lösungsmittel sind beispielsweise: Alkohole, wie z. B. Methanol, Ethanol, Propanol, Isopropanol, Butanol, Pentanol oder Hexanol; organische Säuren, wie z. B. Essigsäure oder gebräuchliche inerte Lösungsmittel, wie z. B. Hexan, Dichlormethan, Essigsäureethylester. Es sind aber auch Gemische obiger Lösungsmittel einsetzbar. Besonders bevorzugt ist Methanol oder ein Gemisch von Methanol und Dichlormethan.
In das Reaktionsgemisch wird ein ozonhaltiger Gasstrom wie z.B. ein Luft-Ozon-Gemisch oder ein Sauerstoff-Ozon-Gemisch eingeleitet. Der Gasstrom enthält vorzugsweise 0,5 bis 10 Gew.% Ozon.
Die Ozonolyse ist beendet, sobald sich das Reaktionsgemisch durch überschüssiges Ozon blau färbt.
Nach beendeter Reaktion wird überschüssiges Ozon vorteilhaft durch Einleiten von Inertgas, beispielsweise durch Einleiten von Stickstoff, entfernt.
Das gebildete Ozonid wird reduktiv aufgearbeitet nach bekannten Methoden, beispielsweise durch Zusatz von Dimethylsufid, Schwefeldioxid, Trimethylphosphit, Thioharnstoff, Zinkstaub in Essigsäure, Natriumhydrogensulfit oder mittels katalytischer Hydrierung.
Die Ausgangsverbindungen der Formel II lassen sich nach bekannten Methoden herstellen.
Beispielsweise lässt sich in einer Mannich-Reaktion ein entsprechendes Keton mit Formaldehyd und Dialkylamin umsetzen. Die entstehende Mannich-Base wird danach erhitzt, wobei sich Dialkylamin abspaltet und das α,ß -ungesättigte Keton der Formel II gebildet wird.
α,ß-ungesättigte Ketone der Formel II können auch durch Aldoladdition aus den entsprechenden Ketonen durch Umsetzung mit Acetaldehyd bzw. Paraldehyd hergestellt werden.
Ebenso fülirt die Umsetzung von Ketonen mit „Gold's Reagenz" [3-(Dimethylamino)- 2-azaprop-2-en-l-yliden]dimethylammoniumchlorid zu α,ß -ungesättigten Ketonen.
Die folgenden Beispiele erläutern das Verfahren, ohne dass darin eine Einschränkung zu sehen ist.
Alle Ausbeuten wurden durch GC- Analyse der Reaktionsmischung ermittelt, soweit die Ausbeuten in Prozenten angegeben sind, handelt es sich um Molprozente.
Beispiel 1
Herstellung von Pentan-2,3-dion
(ausgehend von II, R1 = Methyl, R2= Ethyl, R3 = R4 = H)
Eine Lösung von 88 % 2-Methylpent-l-en-3-on (11,36 g; 0,102 mol) in Methanol (100 ml) wurde auf -78 °C gekühlt und Ozon wurde über 2 h mit einer Geschwindigkeit von 3,4 g/h eingeleitet. Anschliessend wurde überschüssiges Ozon mit Stickstoff ausgetrieben. Das Reaktionsgemisch wurde mit Dimethylsulfid (7,59 g; 0,122 mol) versetzt und langsam auf 25 °C erwärmt. Im Vakuum (240 mbar) wurde ein Teil des Reaktionsgemisches abdestilliert. Man erhielt als Rückstand 19,81 g Rohprodukt, welches 10 Gew.% Pentan- 2,3-dion enthielt. Das entspricht einer Ausbeute von 20% Pentan-2,3-dion im Rückstand. Das Destillat (66,93 g) enthielt 3 Gew% Pentan-2,3-dion, entsprechend einer Ausbeute von weiteren 20% Pentan-2,3-dion.
1H NMR (400 MHz, CDC13) δ = 2,78 (q, J= 8 Hz, 2H); 2,34 (s, 3H); l,09 (t, J= 8 Hz, 3H).
Beispiel 2 Herstellung von Pentan-2,3-dion ausgehend von II, R1 = Methyl, R2 = Ethyl, R3 = Methyl, R4 = H
Eine Lösung von 89 % 4-Methylhex-4-en-3-on (11,25 g; 0,089 mol) in Methanol (100 ml) wurde auf -78 °C gekühlt und Ozon wurde über 5 h mit einer Geschwindigkeit von 3,4 g/h eingeleitet. Überschüssiges Ozon wurde mit Stickstoff ausgetrieben. Das Reaktionsgemisch wurde mit Dimethylsulfid (6,64 g; 0,107 mol) versetzt und langsam auf 25 °C erwärmt. Im Vakuum (240 mbar) wurde ein Teil des Reaktionsgemisches abdestilliert. Man erhielt als Rückstand 20,58 g Rohprodukt, welches 10 Gew.% Pentan-2,3-dion enthielt, entsprechend einer Ausbeute von 23%. Das Destillat enthielt weitere 24% Pentan- 2,3-dion, entsprechend einer Gesamtausbeute von 47%.
Beispiel 3
Herstellung von Pentan-2,3-dion ausgehend von II, R1 = Methyl, R2 = Ethyl, R3 = Dimethylamino, R4 = H
Eine Lösung von 78 % l-(Dimethylamino)-2-methylpent-l-en-3-on (3,75 g; 0,021 mol) in Methanol (35 ml) wurde auf -78 °C gekühlt und Ozon wurde über 1 h mit einer Geschwindigkeit von 3,4 g/h eingeleitet. Überschüssiges Ozon wurde mit Stickstoff ausgetrieben. Das Reaktionsgemisch wurde mit Dimethylsulfid (1,51 g; 0,024 mol) versetzt und langsam auf 25 °C erwärmt. Die Ausbeute an Pentan-2,3-dion wurde im Reaktionsgemisch zu 19% bestimmt.
Beispiel 4
Herstellung von 2-Methylpent-l-en-3-on II, R1 = Methyl, R2 = Ethyl, R3 = R4 = H, Mannich-Reaktion
a- 1 ) 1 -(Dimethylamino)-2-methylpentan-3 -on
Eine Lösung von Diethylketon (200,00 g; 2,322 mol), Dimethylamin-Hydrochlorid
(94,68 g; 1,161 mol) und 36% Formalin (96,85 g; 1,161 mol) wurde mit 32% wässriger HCl auf pH 1 ,00 gestellt und 15 h unter Rückfluss gekocht. Nach Abkühlen auf 25 °C wurde das Reaktionsgemisch mit Diethylether (100 ml) versetzt und die Phasen wurden getrennt. Die wässrige Phase wurde unter Kühlung mit 30% NaOH (144 g) auf pH 12 gestellt, mit Wasser (60 g) verdünnt und mit Diethylether (3 <100 ml) extrahiert. Diese letztgenannten Diethylether-Phasen wurden vereinigt, über Natriumsulfat getrocknet und im Vakuum eingeengt. Als Rückstand erhielt man 144,00 g Produlct, welches 94,6 Gew% l-(Dimethylamino-2-methylpentan-3-on enthielt, entsprechend einer Ausbeute von 82%.

2,54-2,47 (m, 2H); 2,19 (s, 6H, -N(CH3)2); 2,17-2,11 (m, 1H); 1,07-1,01 (m, 6H, 2xCH3).
a-2) 2-Methylpent-l-en-3-on
In eine Lösung von l-(Dimethylamino)-2-methylpentan-3-on (143,08 g; 1,000 mol) in
Diethylether (1,0 1) wurde innerhalb von 2 h HCl Gas (47,4 g; 1,317 mol) bei 25 °C
eingeleitet. Der entstandene Feststoff wurde abfiltriert, mit Diethylether (2x500 ml) gewaschen und bei 30 °C im Vakuum (30 mbar) getrocknet. Man erhielt 169,72 g (100%) 1 -(Dimethy lamino)-2-methylpentan-3 -on-Hydrochlorid. l-(Dimethylamino)-2-methylpentan-3-on-Hydrochlorid (20,00 g; 0,112 mol) wurde auf 170-200 °C erhitzt und das entstandene 2-Methylpent-l-en-3-on wurde destilliert. Man erhielt 7,29 g Produlct, welches 88 Gew.% 2-Methylpent-l-en-3-on enthielt. Bei den restlichen 12 Gew.% handelte es sich um Diethylketon. Das entspricht einer Ausbeute von 58% 2-Methylpent-l-en-3-on.
Η NMR (400 MHz, CDC13) δ = 5,95 und 5,74 (2xs, 2*1H, C=CH2); 2,70 (q, J= 8 Hz, 2H, CH2CH3); 1,86 (s, 3H, CH3); 1,10 (t, J= 8 Hz, 3H, CH2CH3).
b) 2-Methylpent-l-en-3-on
Eine Lösung von Diethylketon (100,00 g, 1,1610 mol), Dimethylamin-Hydrochlorid (47,34 g; 0,5805 mol) und 36% Formalin (48,42 g; 0,5805 mol) wurde mit 1 M HCl
(1,25 g) auf pH 1,00 gestellt und 15 h unter Rückfluss gekocht. Nach Abkühlen auf 25 °C wurde das zweiphasige Reaktionsgemisch getrennt. Die wässrige Phase wurde mit Diethylether (2x50 ml) gewaschen und das Wasser (28,52 g) wurde unter Vakuum (120 mbar) bei einer Badtemperatur von 90 °C abdestilliert. Der Sumpf wurde mit Hydrochinon (4,50 g; 0,0499 mol) versetzt, auf 160-200 °C erhitzt und das entstandene 2-Methylpent-l-en-3-on wurde destilliert. Man erhielt 22,30 g eines Produkts, welches 84 Gew.% 2-Methylpent-l-en-3-on enthielt (entsprechend einer Ausbeute von 33% 2-Methylpent-l-en-3-on ) und 13,81 g eines Produkts mit 86 Gew.% 2-Methylpent-l-en- 3-on (entsprechend weiteren 21% Ausbeute).
Beispiel 5
Herstellung von 4-MethyIhex-4-en-3-on
II, R1 = Methyl, R2= Ethyl, R3 = Methyl, R4 = H, Aldoladdition
Paraldehyd (51,14 g; 0,387 mol) wurde über 5 h zu einer Lösung von Diethylketon (200,00 g; 2,322 mol) und 98% H2SO4 (17,08 g; 0,1742 mol) bei 55 °C zugetropft. Das Reaktionsgemisch wurde 16 h bei 55 °C gerührt, auf 30 °C abgekühlt, mit Wasser (24,0 g)
versetzt und mit 30% NaOH auf pH = 7 gestellt. Die Phasen wurden getrennt und die organische Phase wurde im Vakuum eingeengt. Man erhielt 107,42 g Produkt, welches 77 Gew% 4-Methylhex-4-en-3-on enthielt (entsprechend einer Ausbeute von 64%).
1H NMR (400 MHz, CDC13) δ = 6,73 (q, J= 8 Hz, IH); 2,66 (q, J= 8 Hz, 2H); 1,85 (d, J= 8 Hz, 3H); 1,78 (s, 3H); 1,09 (t, J= 8 Hz, 3H).
Beispiel 6 Herstellung von l-(Dimethylamino)-2-methyIpent-l-en-3-on
II, R1 = Methyl, R2 - Ethyl, R3 = Dimethylamino, R4 = H, Gold's Reagenz
a-1) Herstellung von [3-(Dimethylamino)-2-azaprop-2-en-l-yliden]dimethylammonium- chlorid Eine Lösung von Cyanurchlorid (70,0 g; 0,379 mol) und NN-Dimethylformamid
(183,14 g; 2,510 mol) in 1,4-Dioxan (380 ml) wurde unter Ν2 10 h bei 65 °C gerührt. Das Reaktionsgemisch wurde auf 20 °C abgekühlt. Der entstandene Feststoff wurde abfiltriert und bei 50 °C im Vakuum (30 mbar) über Phosphorpentoxid getrocknet. Man erhielt 149,29 g (80%) [3-(Dimethylamino)-2-azaprop-2-en-l-yliden]dimethylaιrιmoniumchlorid. 1H NMR (400 MHz, CDC13) δ = 9,60 (s, 2H); 3,43 (s, 3H); 3,25 (s, 3H);
IR (KBr) 3453(m), 340 l(m), 1613(s), 1502(w), 1418(m), 1345(s), 1126(m) cm-1.
a-2) 1 -(Dimethylamino)-2-methylpent- 1 -en-3-on
Diethylketon (29,71 g; 0,345 mol) und [3-(Dimethylamino)-2-azaprop-2-en-l-yliden]di- methylammoniumchlorid (37,64 g; 0,230 mol) wurden zu einer Lösung von 30% Natrium- methylat in Methanol (41,41 g; 0,230 mol) und Methanol (305 ml) bei 20 °C unter N2 hinzugegeben. Das Reaktionsgemisch wurde 18 h unter Rückfluss gekocht und danach im Vakuum eingeengt. Der Rückstand wurde in Dichlormethan (300 ml) gelöst und mit gesättigter Natriumbisulfit-Lösung (3 100 ml) gewaschen. Die organische Phase wurde über Na2SO4 getrocknet und im Vakuum eingeengt. Man erhielt 26,60 g Produkt, welches 42 Gew.% l-(Dimethylamino)-2-methylpent-l-en-3-on enthielt (entsprechend einer Ausbeute von 34%).

Claims
Patentansprüche
Verfahren zur Herstellung von α-Dicarbonylverbindungen der Formel
worin R1 und R2 unabhängig voneinander Cι_6-Alkyl bedeuten, dadurch gekennzeichnet, dass ein α,ß -ungesättigtes Keton der Formel
worin R1 und R2 die oben angegebene Bedeutung haben und R3 und R4 unabhängig voneinander Wasserstoff, Cj_6-Alkyl oder Di-Cι_6-alkylamino bedeuten, mit Ozon umgesetzt und das gebildete Ozonid reduktiv aufgearbeitet wird.
Verfahren nach Anspruch 1, worin R1 Methyl und R2 Ethyl bedeuten.
Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass ein Ozon-haltiger Gasstrom bei Temperaturen von -100 °C bis +30 °C in das Realctionsgemisch geleitet wird.
