Definitions
- the invention relates to a method for producing liposomes or proliposomes, a mixture suitable for their preparation is ground in an agitator ball mill.
- the liposomes Due to the ability of the liposomes to penetrate the skin, they can be used, for example, in cosmetics or in dermal therapy be used. You can use moisture or the most varied Active ingredients, among other things, loaded with vitamins. Thanks The amphiphilic properties of liposomes make these active ingredients far transported into the skin, and thus the release of these active ingredients is on guaranteed the desired position.
- liposomes and proliposomes are known. However, most of these methods are only suitable on a laboratory scale, for example the film method. Such methods are known to the person skilled in the art and for example in Liposome Technology; Gregoriadis G., (ed.), Vol. I-III (1993) .
- rotor-stator homogenization or high-pressure liquid homogenization are suitable for the production of liposomes on a larger scale.
- the high-pressure liquid homogenizers which include, for example, high-pressure liquid jet homogenizers (microfluidizers) or high-pressure liquid gap nozzle homogenizers, are very suitable for the production of liposomes, they have the disadvantage that they can only be used to process liquid materials.
- the procedure should enable liposomes or proliposomes with a regular structure, in which, if necessary added active ingredients are evenly distributed. It should also The method can also be readily used on an industrial scale.
- liposomes and / or proliposomes are present gently by grinding one to make liposomes and / or proliposome-suitable mixture in an agitator ball mill can be produced.
- An agitator ball mill is a special form of ball mill. The homogenization and downsizing is done in this mill caused by forces that move freely through the agitator Balls, preferably made of glass, and transferred from there to the regrind will.
- the method according to the invention is very gentle and can also be applied on an industrial scale. It is equally suitable for Production of proliposomes, for the production of liposomes Proliposomes as well as for the direct production of liposomes. Through the Choice of homogenization conditions (e.g. rotor peripheral speed, Grinding time, ball size, degree of filling, temperature) Size and lamellarity of the proliposomes and size of the liposomes be varied in a targeted manner.
- homogenization conditions e.g. rotor peripheral speed, Grinding time, ball size, degree of filling, temperature
- the inventive method can both unloaded as well as with one or more lipophilic and / or hydrophilic Proliposomes or liposomes loaded with substances are produced.
- lecithins are different Origin such as egg lecithin or soy lecithin used; such lecithins are commercially available.
- Polar organic solvents for example alcohols such as ethanol, isopropanol and the like be used.
- lipophilic or hydrophilic Substances such as lipophilic or hydrophilic vitamins, pharmaceuticals as well Flavors and fragrances in question.
- these are, for example fat-soluble vitamins and vitamin derivatives (A, K and E), their precursors ( ⁇ -carotene), carotenoids or fat-soluble pharmaceuticals such as diazepam, Amphotericin B, econazole, corticoids and the like.
- As a hydrophilic The water-soluble vitamins (B complex or vitamin C) or hydropable pharmaceuticals such as heparin, Cisplatin and the like, in question.
- fat-soluble flavors and Fragrance substances are citrus oils, peppermint oil and the like.
- hydrophilic Flavors and fragrances can, for example, vanilla extract, cocoa flavor or tobacco flavors.
- the added lipophilic or hydrophilic compounds in a very gentle way Liposomes or proliposomes incorporated. It will be a very good one Mixing and thus a homogeneous distribution of the added Connections guaranteed in the bilayer.
- the method according to the invention can be used Process in contrast to the conventional methods with very concentrated and viscous lecithin mixtures.
- proliposomes by the inventive Process typically takes place in that a mixture that has been prepared according to the recipe given above, in an agitator ball mill is ground.
- the agitator ball mill with between 50 and 90% of the volume commercially available beads from 0.2 to 6 mm, preferably glass beads from 0.3 to 1.5 mm in diameter.
- the rotor peripheral speed is preferably 5-15 m / s.
- Proliposomal formulations made by the procedure are characterized by a very regular, long lamellar Structure. They contain very high lecithin concentrations, and any added active ingredients are distributed very homogeneously.
- the proliposomes by the inventive method are manufactured because of their high concentration and their relatively high alcohol content very easy to preserve.
- proliposomes described above can be added after adding water in a known manner, such as by homogenization with a Liquid jet high pressure homogenizer, converted into liposomes will. It is noteworthy here that the liposomes according to the invention proliposomal preparations produced are generally smaller are as liposomes, which are produced directly.
- the liposomes can be derived from the proliposomes described above Addition of water also according to the invention by grinding the proliposomal preparation made in an agitator ball mill will.
- the proliposomal preparation is typically used for this purpose Water is added until the water content is between 35 and 95% and then ground in an agitator ball mill.
- the agitator ball mill is between 50 and 90% of the Volume with commercially available beads from 0.2 to 6 mm, preferably from 0.3 to 1.5 mm in diameter, loaded.
- the grinding speed as well as the grinding time can also in this case Structure of the product can be influenced.
- the rotor peripheral speed is 5-15 m / s. This way you can target large and very homogeneous liposomes (1 - 5 ⁇ m) are produced.
