Description
- Die Erfindung bezieht sich auf eine Mikropipette zur berührungslosen, strichförmigen Auftragung eines abgemessenen Volumens einer Suspension, die Zellen oder Partikel enthält, wobei in der Pipette ein Kolben flüssigkeitsdicht angeordnet und die Austrittsöffnung der Pipette durch den Kolben bündig abschließbar ist. Insbesondere betrifft die Erfindung eine Mikropipette, mit der eine bestimmte Menge einer Zell- oder Partikelsuspension in Form eines Strichs mit definierter Länge und Breite auf einem geeigneten Träger deponiert werden kann.
- Für medizinisch-diagnostische Zwecke, z.B. bei Blut- oder Zell- untersuchungen, werden routinemäßig Suspensionen hergestellt und präpariert. Eine Standardmethode ist die flächige Präparation von Zellsuspensionen auf Mikroskop-Objektträgern. Hierfür wird die Zellsuspension auf der gesamten Oberfläche des Objektträgers mit Hilfe eines zweiten Objektträgers verteilt bzw. in dünner Schicht aufgebracht. Dieses Verfahren hat den Nachteil, daß die Zellen und die Flüssigkeit am Hilfsobjektträger, mit dem die Suspension verteilt wird, haften bleiben. Somit geht ein Teil der Zellsuspension verloren. Aus diesem Grunde ist eine dem Volumen proportionale Zählung der Zellen nicht mehr möglich. Ferner ist die Zellverteilung flächig und zum Teil ungleichmäßig. Bei Anwendung starker mikroskopischer Vergrößerungen, bei der die zu untersuchende Fläche größer ist als das Gesichtsfeld des Mikroskops, ist eine automatische Analyse der Zellen durch auftretende Randzellen erschwert, wodurch eine automatische Auswertung des Prüfstoffes sehr aufwendig wird.
- Zur Entnahme und Deposition kleiner Flüssigkeitsvolumina sind Mikropipetten bereits beschreiben (E. Stahl, Dünnschichtchromatografie, Springer-Verlag 1967, 64-66), Mit solchen Pipetten kann jedoch eine strichförmige automatische Präparation von Zellsuspensionen mit definiertem Volumen und einer Strichbreite, die kleiner ist als der Durchmesser des Mikroskopgesichtsfeldes bei 250-facher Vergrößerung, nitch erfolgen. Auch die Verwendung einer pinselartigen Bürste, die in die Pipettenspitze gesteckt ist, gestattet nicht die strichförmige Präparation von Zellsuspensionen, da der Pinsel aufgrund seiner großen Kapillarkraft Zellen oder Partikel zurückhält, wodurch nur die Flüssigkeit geschrieben wird und ein Teil der Flüssigkeit im Pinsel verbleibt, dort eintrocknet oder auch in die nächste Probe verschleppt wird. Auch handbetriebene Dosier- und Pipettiergeräte sind zur Präparation von Zellsuspensionen nicht geeignet (DE-A1-2 541 642, US-A-3 606 086 und FR-A-927 862). Ein größeres Glaspipettensystem mit einer geschwungenen Glaszunge, die an der ebenfalls geschwungenen Mündung des Auslaufrohres angebracht ist, ist für den genannten Zweck nicht verwendbar (DE-C-41 515). Die Glaszunge dient der völligen Gleichheit der an ihre hängenbleibenden Tropfen. Würde man eine solche Pipette für Zellsuspensionen verwenden, wären Zellablagerungen nicht zu vermeiden.
- Es sind Vorrichtungen bekannt, die eine linienförmige Anbringung des Prüfstoffes auf einem Träger und eine automatische Auswertung des zu untersuchenden Materials ermöglichen (z.B. DE-A-19 00 808). Dabei wird die Suspension in Form eines Striches auf einem bandförmigen Träger, ohne diesen vollständig zu berühren, deponiert und die Breite des Striches ist dem Durchmesser des Gesichtsfeldes des Mikroskops angepaßt. Die Deposition erfolgt mit einer durch zwei Metallzungen variablen Abstands gebildeten Ausziehfeder, zwischen die das zur Deposition gelangende Suspensionsvolumen gegeben werden muß. Beim Ausschreiben muß der Präparateträger von den Metallzungenspitzen leicht berührt werden, um ein Ausfließen der Suspension zu ermöglichen. Hierdurch erfolgt eine ständige Abnutzung. Die volumengenaue, volumenproportionale Deponierung von Partikeln aus Suspensionen in Form eines Strichs mit definierter Breite und Länge bei vorgegebener Zeit und Schreibgeschwindigkeit ist mit einer derartigen Vorrichtung nicht möglich, da sowohl Flüssigkeit als auch Parikel an den Flächen der Metallzungen, die von der Suspension berührt werden, hängenbleiben und die Ausflußgeschwindigkeit der Suspension sowohl von ihrer Oberflächenspannung als auch den Benetzungseigenschaften des Präparateträgers abhängig ist. Ein weiterer wesentlicher Nachteil ist die erforderliche Reinigung des Systems vor jedem Probenwechsel, um eine Zellverschleppung auszuschließen.
- Auch der Ersatz der Metallzungenschreibvorrichtung durch eine Mikropipette, bei der in einem Kapillarrohr flüssigkeitsdicht ein Kolben so angeordnet ist, daß er mittels Miniatureelektromotor mit Untersetzungsgetriebe in axialer Richtung entsprechend dem aufzunehmenden und dann beim Schreiben wieder auszustoßenden Volumen in Richtung Kapillaraustrittsöffnung bewegt werden kann und nach beendetem Vorgang die Kapillare bündig abschließt, erfüllt die gestellten Forderungen insbesondere nicht für größere Suspensionsvolumina (z.B. > 1 pl) bei vorwählbarer Strichbreite.
- Die erzielbare Zellstrichbreite entspricht dem Außendurchmesser der Pipettenspitze und ist nur wenig breiter als der Kapillardurchmesser, wenn das Rohr an der Austrittsöffnung eine konisch spitze Form hat. Das Ausstoßen der Partikelsuspension erfolgt zwangsweise durch Bewegung des Kolbens und kann motorgetrieben mit stufenlos regelbarer Geschwindigkeit erfolgen. Aufgrund der Kolbendichtung wird die Kapillare rückstandslos geleert. Ein wesentlicher Vorteil einer solchen Pipette ist, daß durch die Zwangsausstoßung der Suspension aus der Pipette die Pipettenspitze den Träger nicht mehr berühren muß, wodurch eine Abnutzung der Spitze bzw. Beschädigung der Trägeroberfläche ausgeschlossen ist. Verschleppung von Material in die nächste Probe entfällt, Reinigung der Pipette kann in größeren Zeitabständen erfolgen. Mit einem solchen System kann mit einer Flüssigkeitsmenge von z.B. 0,15 ,ul bei Vewendung einer Kapillare mit einem Spitzendurchmesser von 0,3 mm und einem Kapillardurchmesser von 0,2 mm bei einem Kolbenhub von ca. 4,8 mm ein 0,3 mm breiter Strich berührungslos im Bereich von 5 bis 50 cm Länge beschrieben werden. Der Nachteil einer solchen Pipette ist jedoch, daß mit ihr nur Volumina im Bereich von 0,1 bis 0,5 yl verarbeitet werden können. Um z.B. ein Volumen von größer als 1,5 ,ul zu verarbeiten, müßte der Kolbenweg länger als 50 mm sein, wodurch große Instabilitäten entstehen, die diese Anordnungen unbrauchbar machen.
- Auf dem Gebiet der Medizintechnik, speziell bei der Krebsfrüherkennung, ist es erforderlich, daß größere Zellsuspensionsvolumina bis zu mehreren ,ul mit gleiche, vorstehend genannten GenauIgkeiten verarbeitet werden können. In einfacher Weise kann durch Vergrößerung des Kapillardurchmessers eine größere Menge einer Zellsuspensionsprobe. entnommen werden. Die geforderte Strichbreite von 0,3 mm oder kleiner kann jedoch nicht mehr erreicht werden, da diese direkt vom Durchmesser der Kapillarspitze abhängig ist und nicht kleiner als der Kapillardurchmesser sein kann.
- Der vorliegenden Erfindung liegt daher die Aufgabe zugrunde, die vorgenannten Nachteile zu beseitigen und ein Pipettensystem zu entwickeln, mit dem aus einer Zell- oder Partikelsuspension definierte vorbestimmte Mengen entnommen werden können und diese Mengen in Form eines geradlinigen oder spiralförmigen Striches auf einem geeigneten Träger, z.B. Kunststoffband oder Glasplatte, berührungslos deponiert werden können.
- Es hat sich nun geziegt, daß diese Aufgabe mit dem erfindungsgemäßen Pipettensystem in technisch fortschrittlicher Weise gelöst werden kann, bei dem die Austrittsöffnung der Pipette an einem Teil ihres Umfangs einen geraden Fortsatz aufweist, dessen Breite unabhängig von dem Pipettendurchmesser die Strichbreite bestimmt, und der senkrecht zur Stirnfläche des Kolbens verläuft.
- Der Fortsatz ist in der Richtung des Kolbenweges vorzugsweise mit einer schlitzförmigen Aussparung bzw. einer Einkerbung versehen, dessen Breite kleiner ist als der Durchmesser der suspendierten Zellen oder Partikel, um eine Ablagerung zu verhindern. Damit das gesamte Volumen der Zellsuspension wieder ausgestoßen werden kann, schließt die Einkerbung bzw. Aussparung am Ende des Kolbenweges an. Iry dieser Weise kann die Flüssigkeit mit Hilfe der Kapillarkraft, die in der Aussparung bzw. Einkerbung entsteht, bis an die Spitze des Fortsatzes fließen. Der gleiche Effekt kann auch durch hydrophile Beschichtung des Fortsatzes erreicht werden. Verbindungen, die zur Bildung hydrophiler Beschichtungen herangezogen werden könne, sind bekannt.
- Der Fortsatz kann an die Austrittsöffnung der Pipette anmontiert sein. Vorzugsweise ist die Austrittsöffnung der Pipette an einem Teil ihres Umfangs so geformt, daß sich ein Fortsatz bildet.
- Im folgenden wird die Erfindung anhand von lediglich einen Ausführungsweg darstellenden Zeichnungen näher erläutert. Es zeigen in schematischer Vereinfachung
- Figur 1 in perspektivischer Darstellung eine erfindungsgemäße Mikropipette und
- Figur 2 dieselbe Pipette aus der Seitenansicht.
- Aus den Figuren geht hervor, daß die Austrittsöffnung 1 der Pipette 2 einen Fortsatz 3 aufweist. Die Austrittsöffnung ist so geformt, daß sie durch den Kolben bündig abgeschlossen wird, wenn der Ausstoßvorgang beendet ist. Die Breite des Fortsatzes 3 bestimmt die Breite der Zellspur. Der Fortsatz 3 ist mit einer Einkerbung 4 versehen. Durch die zusätzlich in der Einkerbung entstehende Kapillarkraft kann die Flüssigkeit leicht bis an die Spitze fließen und einen Tropfen bilden. Zur Vermeidung von Zellablagerungen ist die Breite der Einkerbung kleiner als der Durchmesser der suspendierten Zellen bzw. Partikel.
- Mit dem erfindungsgemäßen Pipettensystem kann die Strichlänge so vorbestimmt werden, daß optimale Belegungsdichte wie auch Verteilung der Zellen oder Partikel im Strich gegeben ist. Durch die spezielle Formgebung der Pipettenspitze kann die Strichbreite unabhängig vom Innendurchmesser der Pipette den Erfordernissen, z.B. einer automatisierten mikroskopischen Auswertung, angepaßt werden. Wird die Strichbreite z.B. so eingestellt, daß im Gesichtsfeld eines Mikroskops immer die gesamte Strichbreite erfaßt wird, so können bei kontinuierlicher Bewegung des Striches durch das Gesichtsfeld alle Zellen bzw. Partikel nacheinander analysiert werden, ohne daß Probleme mit randständigen Zellen auftreten.
