Claims (14)
1. Saccharomyces-celle, der er i stand til at omdanne en eller flere pentose-sukkerarter og en eller flere hexose-sukkerarter til et fermenteringsprodukt, som omfatter en nukleotidsekvens, der koder for en xyloseisomerase, der bibringer evnen til at omdanne xylose ved hjælp af isomerisering eller reduktion, hvilken celle konstitutivt udtrykker et eller flere heterologe eller homologe polypeptider med aminosyresekvensen ifølge SEQ ID NO: 20 eller et variant polypeptid deraf, der har mindst 45 % identitet med SEQ ID NO: 20 og har glyoxylase I-aktivitet, hvor cellen omfatter en eller flere genmodifikationer, som resulterer i et fald i aldosereductaseaktivitet i cellen.
2. Celle ifølge krav 1, hvor det heterologe polypeptid har glyoxalase I-aktivitet.
3. Celle ifølge krav 1 eller 2, hvor cellen omfatter en eller flere genmodifikationer, som resulterer i: (a) en forøgelse af transporten af xylose i cellen; (b) en forøgelse af xylulosekinaseaktivitet; (c) en forøgelse af strømmen gennem pentosephosphatsystemet; (d) et fald i følsomheden for katabolitrepression; (e) en forøgelse af toleransen for ethanol, osmolaritet eller organiske syrer; eller (f) en reduceret produktion af biprodukter.
4. Celle ifølge krav 3, hvor den ene eller de flere genmodifikationer resulterer i overekspression af mindst ét gen, der koder for et enzym i den ikke-oxidative del af pentosephosphatsystemet.
5. Celle ifølge krav 4, hvor genet er et gen, der koder for en ribulose-5-phosphatisomerase, en ribulose-5- phosphatepimerase, en transketolase eller en transaldolase.
6. Celle ifølge et hvilket som helst af kravene 3 til 5, hvor den ene eller de flere genmodifikationer resulterer i overekspression af et gen, der koder for en xylulosekinase.
7. Celle ifølge et hvilket som helst af kravene 3 til 6, der har evnen til at anvende L-arabinose, hvor generne TALI, TKL1, RPE1 og RKI1 overudtrykkes.
8. Celle ifølge et hvilket som helst af kravene 1 til 7, hvor det kodende område af GRE3-genet er inaktiveret ved substitution af det kodende område med en nukleotidsekvens, der omfatter generne TALI, TKL1, RPE1 og RKI1.
9. Celle ifølge et hvilket som helst af kravene 1 til 8, hvor generne araA, araB og araD udtrykkes.
10. Celle ifølge et hvilket som helst af kravene 1 til 9, hvor et eller flere konstitutivt udtrykte eller konstitutivt overudtrykte gener er stabilt integreret i genomet i cellen.
11. Fremgangsmåde til frembringelse af et fermenteringsprodukt, hvilken fremgangsmåde omfatter fermentering af et medium, der indeholder en kilde af xylose og/eller arabinose, med en celle ifølge et hvilket som helst af kravene 1 til 10, således at cellen fermenterer xylose til fermenteringsproduktet.
12. Fremgangsmåde ifølge krav 11, hvor fermenteringsproduktet er valgt fra listen, der indbefatter ethanol, butanol, mælkesyre, 3-hydroxypropionsyre, acrylsyre, eddikesyre, ravsyre, citronsyre, æblesyre, fumarsyre, itaconsyre, en aminosyre, 1,3-propandiol, ethylen, glycerol, butanol, et β-lactam-antibiotikum og et cephalosporin.
13. Fremgangsmåde ifølge krav 12, hvor fermenteringsproduktet er ethanol.
14. Fremgangsmåde ifølge krav 12 eller 13, hvor fremgangsmåden er anaerob.
