Claims (13)
1. Fremgangsmåde til ikke-destruktionsbestemmelse af køn og/eller udviklingstrin for et fjerkræsembryo i et æg, hvilken fremgangsmåde omfatter (a) detektering af mindst en første udviklingsmarkør for bestemmelsen af udviklingstrin valgt fra betain, aspartat, arginin, glutamat, glutamin og prolin og/eller mindst en første udviklingsmarkør for kønsbestemmelsen valgt fra glucose, cholin og/eller valin i et ægs amnionvæske på et tidspunkt fra starten af udrugningen af ægget indtil udklækningen; (b) måling af mængden af den mindst første detekterede udviklingsmarkørforbindelse til bestemmelse af udviklingstrinnet og/eller kønnet; og (c) sammenligning af mængden med en baseline etableret for hankøn og hunkøn, og/eller udviklingstrin for embryoet, for at bestemme, om embryoet er hankøn eller hunkøn og/eller embryoets udviklingstrin.
2. Fremgangsmåde ifølge krav 1, hvor (c) indbefatter en univariat eller multivariat analyse af de målte metabolitter og en bestemmelse af associationen af et fjerkræsembryo med en bestemt population, der omfatter et hovedkomponentanalyseprogram og/eller et partielt mindste kvadraters regressionsanalyseprogram.
3. Fremgangsmåde ifølge krav 1 eller krav 2, hvor mindst en anden markør detekteres i trin (a), og hvor den mindst første og anden markør analyseres og sammenlignes med baseline og med hinanden for at fastslå et udviklingsmarkørforhold.
4. Fremgangsmåde ifølge krav 3, hvor fjerkræsarten er Gallus gallus domesticus, og hvor en første eller yderligere udviklingsmarkør er glucose i absolut mængde for et hunkønsembryo i intervallet fra 30 pM/ml til 70 μΜ/ml og for et hankønsembryo på fra 1 μΜ/ml op til 30 pM/ml.
5. Fremgangsmåde ifølge krav 4, hvor en første eller yderligere udviklingsmarkør er cholin i en absolut mængde for et hunkønsembryo i intervallet fra 110 μΜ/ml til 130 μΜ/ml og for et hankønsembryo på fra 90 μΜ/ml op til men ikke inklusive 110 μΜ/ml.
6. Fremgangsmåde ifølge krav 4 eller 5, hvor en første eller yderligere udviklingsmarkør er valin i en absolut mængde for et hunkønsembryo i intervallet fra 110 μΜ/ml til 130 μΜ/ml og for et hankønsembryo på fra 90 μΜ/ml op til men ikke inklusive 110 μΜ/ml.
7. Fremgangsmåde ifølge krav 3 til 6, hvor mindst en første og en anden udviklingsmarkør detekteres og analyseres, og hvor de absolutte mængder og forholdet mellem den mindst første og den anden markør anvendes til kønsbestemmelse.
8. Fremgangsmåde ifølge krav 7, hvor analysen udføres med en fremgangsmåde med magnetisk billeddannelse, der indbefatter nukleære resonansfremgangsmåder; ved en spektral resonansfremgangsmåde, der indbefatter infrarød eller Raman-spektroskopi; og/eller en analytisk fremgangsmåde, såsom GC eller HPLC koblet med egnede detektorer, fluorescensspektroskopi og/eller et enzymbundet immunsorbent assay.
9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, der endvidere omfatter mindst én af; (i) bestemmelse af, om et embryo i et æg er hankøn eller hunkøn og separation af en flerhed af hankønsæg fra en flerhed af hunkønsæg for at danne en overvejende hankøns- eller overvejende hunkønsægselektion; (ii) bestemmelse af, om et embryo i et æg er hankøn eller hunkøn og separation af en flerhed af hankønsæg fra en flerhed af hunkønsæg for at danne en overvejende hankøns- eller overvejende hunkønsægselektion, og udsættelse af hunkøns- eller hankønsægselektioneme for en udrugnings- og udklækningsproces for at danne en overvejende hunkøns- eller hankønskyllingepopulation.
10. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 9, der omfatter (iii) bestemmelse af embryoets udviklingstrin og tiden indtil sandsynlig udklækning, og separation af æg i flerheder af æg med tilsvarende eller ens udviklingstrin.
11. Fremgangsmåde ifølge krav 10, der endvidere omfatter udsættelse af ægget for en udrugnings- og udklækningsproces i overensstemmelse med den forventede udklækningsdato for at danne en kyllingepopulation af overvejende samme udviklingstrin.
12. Fremgangsmåde ifølge krav 9 eller 10, der endvidere omfatter (vi) injektion af et virus eller virus-lignende materiale i hvert æg identificeret som indeholdende et levende embryo og hankøn eller hunkøn og/eller af et bestemt udviklingstrin, og (vii) isolering af den opnåede vaccine fra de udrugede æg.
13. Fremgangsmåde ifølge krav 10, hvor fjerkræsarten er Gallus gallus domesticus, og hvor udviklingsmarkøreme er valgt fra mindst én af: trimethylglycin, aspartat og/eller asparagin, glutamat og/eller glutamin og/eller prolin.
