Claims (12)
1. Fremgangsmåde til bestemmelse af, om det er sandsynligt, at en patient med en blodcancer vil respondere på terapi med en HSP90-inhibitor, som omfatter: (a) etablering af kontakt mellem en prøve, der indeholder cancerceller og ikke-cancerceller, fra patienten og en cellepermeabel fluorescensmærket PU-H71, der fortrinsvis binder til en tumorspecifik form af HSP90, der er til stede i cancercellerne hos patienten; (b) måling af mængden af fluorescensmærket PU-H71, der er bundet til cancercellerne og ikke-cancercellerne i prøven; og (c) sammenligning af mængden af den fluorescensmærkede PU-H71, der er bundet til cancercellerne, med mængden af den fluorescensmærkede PU-H71, der er bundet til ikke-cancercellerne, hvor en større mængde af fluorescensmærket PU-H71, der er bundet til cancercellerne end til ikke-cancercellerne, indikerer, at det er sandsynligt, at tumoren vil respondere på en HSP90-inhibitor, fortrinsvis hvor ikke-cancercellerne er lymfocytter, og yderligere fortrinsvis hvor målingen udføres ved hjælp af flowcytometri.
2. Fremgangsmåde ifølge krav 1, hvor det gælder, at jo større forholdet er mellem mængden af fluorescensmærket PU-H71, der er bundet til tumoren eller tumorcellerne målt i trin (b) , sammenlignet med den fluorescensmærkede PU-H71, der er bundet til ikke-cancercellerne, jo større er sandsynligheden for respons på terapi med en HSP90-inhibitor.
3. Fremgangsmåde ifølge krav 1 eller 2, hvor blodcanceren er en leukæmi, fortrinsvis kronisk myeloid leukæmi.
4. Fremgangsmåde ifølge et hvilket som helst af kravene 1-3, hvor prøven omfatter levende celler.
5. Fremgangsmåde ifølge et hvilket som helst af kravene 1-4, hvor prøven omfatter en biologisk væske, fortrinsvis blod eller knoglemarv, mere fortrinsvis blod.
6. Fremgangsmåde ifølge krav 1, hvor prøven omfatter splittede tumorceller.
7. Fremgangsmåde ifølge krav 1, hvor prøven omfatter frosne celler.
8. Fremgangsmåde ifølge krav 1, hvor prøven omfatter fikserede og permeabiliserede celler.
9. Fremgangsmåde ifølge krav 1, hvor HSP90-inhibitoren, der skal administreres som terapi, er PU-H71.
10. Fremgangsmåde ifølge et hvilket som helst af kravene 1-9, hvor den fluorescensmærkede HSP90-inhibitor er PU-H71-FITC2.
11. Fremgangsmåde ifølge et hvilket som helst af kravene 1-9, hvor den fluorescensmærkede HSP90-inhibitor er PU-H71-NBD1.
12. Fremgangsmåde ifølge krav 1, hvor blodcanceren er en hæmatologisk malignitet.
