Claims (9)
- TERMOSTABIL TRICHODERMA-CELLULASE E Fremgangsmåde til fremstilling af et termostabilt polypeptid af interesse i en modificeret Trichoderma-værtscelle, hvilken fremgangsmåde omfatter: en modifikation, der i alt væsentligt reducerer eller forhindrer produktion af en termostabil endoglucananse V (EGV) ved hjælp af værtscellen, en exogent nukleinsyremolekyle, der koder det termostabile polypeptid af interesse operativt bundet til en promoter, hvorved mindst ét endogent polypeptid og polypeptidet af interesse hvert udtrykkes in vivo af Trichoderma-væitscellen, hvilken fremgangsmåde omfatter: inkubation af den modificerede værtscelle i et medium, der er egnet for den modificerede værtscelle til at producere det termostabile polypeptid af interesse og det endogene polypeptid, hvorved hvert af det endogene polypeptid og det termostabile polypeptid af interesse udtrykkes af den modificerede værtscelle; og udsættelse af det termostabile polypeptid af interesse og det endogene polypeptid for en forhøjet temperatur, hvor den forhøjede temperatur er tilstrækkelig til i alt væsentligt at inaktivere det endogene polypeptid, men utilstrækkelig til at inaktivere det termostabile polypeptid af interesse og EGV, hvis den er til stede; hvorved det termostabile polypeptid af interesse produceres i en aktiv eller funktionel form i alt væsentligt i fravær af aktivitet fra det endogene polypeptid.
- 2. Fremgangsmåde ifølge krav 1, der endvidere omfatter isolering af en proteinblanding fra den modificerede Trichoderma-væitscelle før trinnet med udsættelse af polypeptidet af interesse og det endogene polypeptid for den forhøjede temperatur.
- 3. Fremgangsmåde ifølge krav 1 eller krav 2, hvor den forhøjede temperatur er en temperatur på ca. 90 °C eller højere.
- 4. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 3, hvor modifikationen omfatter én/ét eller flere deletioner eller brud i nukleotider, der er involveret i ekspression af EGV.
- 5. Fremgangsmåde ifølge krav 3, hvor bruddet eller deletionen omfatter deletion eller brud af det gen, der koder, eller kodningsområdet for eller promoteren eller regulatoriske elementer til ekspression af EGV.
- 6. Fremgangsmåde ifølge krav 5, hvor deletionen eller bruddet omfatter deletion eller brud af egl5.
- 7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor Trichoderma er T. reesei.
- 8. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor polypeptidet af interesse er hydrophobin II.
- 9. Fremgangsmåde ifølge krav 8, hvor hydrophobin II stammer fra en Trichoderma spp., eventuelt hvor Trichoderma spp. er Trichoderma reesei.
