Claims (9)
1. Fremgangsmåde til forøgelse af secemeringsudbyttet af et modent protein, hvilken fremgangsmåde omfatter: (i) at tilvejebringe et polynukleotid, der koder for et umodent protein, som omfatter en aminosyresekvens for en ledersekvens og for et modent ønsket protein; (ii) at modificere polynukleotidsekvensen for at indføre et aminosyresekvensmotiv i ledersekvensen og (iii) at muliggøre, at det umodne protein secerneres af en svampeværtscelle, for at generere et modent protein; hvor: (a) motivet omfatter -X1-X2-X3-X4-X5-, hvor Xi er phenylalanin, tryptophan eller tyrosin, X2 er isoleucin, leucin, valin, alanin eller methionin, X3 er leucin, valin, alanin eller methionin, X4 er serin eller threonin, og X5 er isoleucin, valin, alanin eller methionin; og (b) secemeringsudbyttet af det modne protein er forøget i forhold til secemeringsudbyttet af et modent protein, der stammer fra et umodent protein, som omfatter ledersekvensen uden motivet.
2. Fremgangsmåde ifølge krav 1, hvor ledersekvensen omfatter (a) HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1 eller (b) en variant deraf, som omfatter HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1 med den undtagelse, at der i en eller flere positioner, som ikke er de af Xi, X2, X3, X4 eller X5 definerede, er en aminosyreinsertion, -deletion eller -substitution, og hvor varianten uden for motivet har mindst syv eller mindst ni aminosyrer, som er identiske med dem i HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1.
3. Fremgangsmåde ifølge krav 1 eller 2, hvor motivet er -Phe-Ile-Val-Ser-Ile-,
4. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor Xi, X2, X3, X4 og X5 forefindes i positionerne henholdsvis -20, -19, -18, -17 og -16 i stedet for de naturligt forekommende aminosyrer i disse positioner, hvor nummereringen er sådan, at resten -1 er den C-terminale aminosyre i den native secemeringsprosekvens eller -præsekvens, og hvor Xi, X2, X3, X4 og X5 er aminosyrer som defineret i et hvilket som helst af kravene 1-3.
5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor ledersekvensen omfatter en secemeringsprosekvens, såsom: (a) en albuminsecemeringsprosekvens, fortrinsvis en human semmalbuminsecemeringsprosekvens eller en gær-MFa-1-secemeringsprosekvens; eller (b) en variant af en i (a) defineret secemeringsprosekvens, hvilken variant har det dibasiske par af aminosyrer fra den secemeringsprosekvens, hvorpå den er baseret, og kun konservative substitutioner i en eller flere andre positioner.
6. Fremgangsmåde ifølge krav 5, hvor ledersekvensen omfatter (a) HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1 eller (b) en variant deraf, som omfatter HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1 med den undtagelse, at der i en eller flere positioner, som ikke er de af Xi, X2, X3, X4 eller X5 definerede, er en aminosyreinsertion, -deletion eller -substitution, og hvor varianten uden for motivet har mindst syv eller mindst ni aminosyrer, som er identiske med dem i HSA- eller HSA MFa-l-secemeringspræsekvensen ifølge fig. 1; og ledersekvensen fusioneres i sin C-terminale ende til den N-terminale aminosyre i en secemeringsprosekvens ved hjælp af en peptidbinding, så der derved dannes en præ-prosekvens.
7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor sekvensen for det ønskede protein fusioneres i sin N-terminale ende til den C-terminale aminosyre i ledersekvensen.
8. Fremgangsmåde ifølge et hvilket som helst af kravene 1-7, hvor det modne ønskede protein er valgt blandt albumin, transferrin, lactoferrin, endostatin, angiostatin, collagener, immunoglobuliner, Fab’-fragmenter, F(ab’)2, ScAb, ScFv, interferoner, IL 10, IL11, IL2, interferon α-typer og -undertyper, interferon β-typer og -undertyper, interferon γ-typer og -undertyper, IL 1-receptorantagonist, EPO, TPO, prosaptid, cyanovirin-N, 5-helix, T20-peptid, T1249-peptid, HIV-gp41, HIV-gpl20, fibrinogen, urokinase, prourokinase, tPA (vævsplasminogenaktivator), hirudin, trombocytvækstfaktor, parathyroideahormon, proinsulin, insulin, insulinlignende vækstfaktor, calcitonin, væksthormon, transformerende vækstfaktor β, tumomekrosefaktor, G-CSF, GM-CSF, M-CSF, koagulationsfaktorer i såvel præ- og aktive former, herunder men ikke begrænset til plasminogen, fibrinogen, trombin, prætrombin, protrombin, von Willebrands faktor, al-antitrypsin, plasminogenaktivatorer, faktor VII, faktor VIII, faktor IX, faktor X og faktor XIII, nervevækstfaktor, LACI (lipoproteinassocieret koagulationshæmmer [lipoprotein associated coagulation inhibitor], som også er kendt som vævsfaktorreaktionsvejshæmmer [tissue factor pathway inhibitor] eller hæmmer af den ekstrinsiske reaktionsvej [extrinsic pathway inhibitor]), trombocyt-endotelcellevækstfaktor (PD-ECGF, platelet-derived endothelial cell growth factor), glucoseoxidase, serumcholinesterase, aprotinin, amyloid-precursor, inter-alfa-trypsinhæmmer, antitrombin III, apolipoproteintyper, protein C, protein S, en variant eller et fragment af en hvilken som helst af ovennævnte.
9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det modne ønskede protein er: (a) albumin (fortrinsvis humant albumin); (b) en variant af albumin, som udviser mindst 25% sekvensidentitet med det modne albuminpolypeptid, der kodes for af SEQ ID NO: 20; (c) et fragment af albumin, som omfatter mindst 5% af den komplette sekvens for det modne albuminpolypeptid, der kodes for af SEQ ID NO: 20; (d) et fusionsprotein, som omfatter en hvilken som helst af (a), (b) eller (c), (e) transferrin (fortrinsvis humant transferrin) eller (f) et fusionsprotein, som omfatter (e).
