Claims (15)
1. Mikroorganisme, som tilhørerslægten Escherichia, med forbedret L-aminosyre-produktivitet og saccharose-assimilerende evne, og som har aktiviteterne af saccha-rosepermease, saccharosehydrolase og saccharosetranskriptionsregulator og har en forøget mannokinaseaktivitet i sammenligning med en saccharose-ikke-assimilerende mikroorganisme, som tilhører slægten Escherichia.
2. Mikroorganisme ifølge krav 1, hvor den forøgede mannokinaseaktivitet opnås ved én eller flere fremgangsmåder, som er valgt fra gruppen bestående af en forøgelse af kopitallet af mannokinasegenet, promotorudskiftning, promotormutation og genmutation.
3. Mikroorganisme ifølge krav 2, hvor forøgelsen af kopitallet af mannokinasegenet opnås ved at indsætte et polynukleotid, som koder for mannokinase, i et kromosom eller indføre polynukleotidet i et vektorsystem.
4. Mikroorganisme ifølge krav 3, hvor vektoren indeholderen basesekvens som i SEQ ID NO:15.
5. Mikroorganisme ifølge krav 1, hvor aktiviteterne af saccharosepermease, saccharosehydrolase og saccharosetranskriptionsregulator bibringes ved transformation med en vektor, som indeholder basesekvenser, der koder for saccharosepermease, saccharosehydrolase og saccharosetranskriptionsregulator.
6. Mikroorganisme ifølge krav 5, hvor basesekvenserne er basesekvenser som i SEQ ID NO:4, 6 og 7.
7. Mikroorganisme ifølge krav 5, hvor saccharosetranskriptionsregulatoren kodes afen basesekvens som i SEQ ID NO:17.
8. Mikroorganisme ifølge krav 1, hvor en vektor, som indeholder alle basesekvenser, der koder for saccharosepermease, saccharosehydrolase, saccharosetranskriptions-regulator og mannokinase, anvendes til at udføre transformation.
9. Mikroorganisme ifølge krav 8, hvor vektoren kodes af en basesekvens som i SEQ ID NO:18.
10. Mikroorganisme ifølge krav 1, hvor den mikroorganisme, der tilhører slægten Escherichia, er E. col i.
11. Mikroorganisme ifølge krav 10, hvor E. coli er E. coli CA03-0308 med deponeringsnummer: KCCM10978P.
12. Mikroorganisme ifølge krav 1, hvor L-aminosyren er valgt fra gruppen bestående af L-threonin, O-succinyl-homoserin, O-acetyl-homoserin, L-methionin, L-lysin, L-homo-serin, L-isoleucin, L-valin og L-tryptophan.
13. Mikroorganisme ifølge krav 12, hvor L-aminosyren er valgt fra gruppen bestående af L-threonin, O-succinyl-homoserin, O-acetyl-homoserin og L-tryptophan.
14. Fremgangsmåde til fremstilling af en L-aminosyre, hvilken fremgangsmåde omfatter følgende trin: inokulering og dyrkning af en £sc/7en'c/7/'a-mikroorganisme ifølge et hvilket som helst af kravene 1 til 13, som har forbedret L-aminosyreproduktivitet og saccharose-assimilerende evne, i et dyrkningsmedium, som helt eller delvist indeholder saccharose som carbonkilde; og separering af L-aminosyren fra det i ovenstående trin opnåede dyrkningsmedium.
15. Fremgangsmåde ifølge krav 14, hvor L-aminosyren er valgt fra gruppen bestående af L-threonin, O-succinyl-homoserin, O-acetyl-homoserin, L-methionin, L-lysin, L-homo-serin, L-isoleucin, L-valin og L-tryptophan.
