Den foreliggende opfindelse angår en fremgangsmåde til fremstilling af en ester ud fra en alkohol og fedtsyre (glycerid) ved enzym-katalyseret omdannelse.
Fremstillingen af fedtsyreestere ud fra fedtsyre/-6 glycerid-udgangsmateriale under anvendelse af en enzymkatalyseret reaktion er velkendt.
GB Patentskrift (GB-A) nr. 1.577.933 beskriver inter-forestring af et fedtsyreglycerid med et andet triglycerid indeholdende forskellige fedtsyregrupper eller en fedtsyre 10 under anvendelse af en lipaseenzymkatalysator og en mængde vand. I EP-A-0.195.311 beskrives omsætning af en fedtsyre og visse C^- til C-^^alkoholer i nærværelse af lipase.
Fremstillingen af en ester ud fra alkohol og en fedtsyre ved enzym-katalyseret reaktion er beskrevet i US 15 patentskrift (US-A) nr. 4.451.565, der bl.a. beskriver forestring af oliesyre med ethanol i nærværelse af lipase.
Det fremgår ikke klart af denne beskrivelse, hvordan reaktanterne bringes i kontakt med hinanden.
Det har nu vist sig, at den hastighed, med hvilken 20 den ønskede ester dannes, kan forøges betydeligt, hvis fedtsyre (glycerid) og en C^g-alkohol kontaktes med lipase, og alkoholen tilsættes med sådan en hastighed, at alkoholkoncentrationen i reaktionsblandingen holdes under et molært forhold på 0,5 mol alkohol pr. mol fedtsyre/acylgruppe.
25 Fortrinsvis holdes dette forhold under 0,25.
Fedtsyre (glycerid) -udgangsmaterialet er C2~C24“ -mættet eller umættet monocarboxylsyre med fortrinsvis en lige kæde eller et glycerid deraf, fortrinsvis et triglycerid. Sådanne triglycerider er meget tilgængelige som ani-30 malske eller vegetabilske olier og fedtstoffer. Sojabønneolie, kokosnøddeolie, smørolie osv. er meget egnede.
Alkoholbestanddelen er en C^-Cg-alkohol, fortrinsvis en monovalent alkohol, og C2~C^-primære alkoholer foretrækkes. Ethanol og isopropanol er meget egnet. Polyalkoholer, såsom 35 glycoler, kan også anvendes.
Lipasen anvendt ifølge opfindelsen er sædvanligvis af mikrobiel oprindelse, og der kan anvendes enzymmateriale opnået fra Mucor miehei, Rhizopus-arter, Aspergillus niger, 0 2 DK 171810 B1
Candida cylindracae, Pseudomonas-arter eller Arthrobacter-arter.
Enzymet kan anvendes som sådan, dvs. i nogen renset form, men fortrinsvis knyttes det til noget bæremateriale, 5 såsom diatoméjord, så det kan fraskilles efter omsætning og genanvendes. Enzymer kan immobiliseres ved forskellige metoder, f.eks. ved absorption på bæremateriale, binding til ionbytterharpikser, kovalent binding til bærematerialer, indeslutning i hydrofobe geler osv.
10 Til effektiv anvendelse af enzymet er det ønskeligt, at der er noget vand til stede under esterdannelsen. Vand har imidlertid også tendens til at hydrolysere esteren, således at kun meget små mængder er ønskelige, især mængder, der opløses i udgangsblandingen. Når der dannes reak-15 tionsvand, efterhånden som reaktionen forløber, som det er tilfældet, når fedtsyre omsættes med alkohol, er det ønskeligt, at overskydende vand fjernes under reaktionen, f.eks. ved neddypning af en kold overflade i hovedrummet af reaktionsbeholderen (hvorved vandet fryses ud) eller 20 alternativt ved destillation ved en relativ lav temperatur, ved hvilken lipasen stadig er aktiv, især under formindsket tryk. Alternativt kan vand absorberes, f.eks. af tilsat molekylsigtemateriale.
Den esterdannende reaktion udføres sædvanligvis i 25 et temperaturområde mellem 10 og 90°C, fortrinsvis mellem 30 og 70°C. Reaktionen varer normalt mindst flere timer.
Alkoholysen bør udføres kontinuerligt (omrørt tank) eller halvkontinuerligt (kaskade-metode) eller batchvis.
Opfindelsen illustreres ved hjælp af de følgende 30 eksempler.

Eksempel 1 10 mmol (2,82 g) oliesyre omrøres ved 40°C med 40 mg lipasekatalysator bestående af Mucor miehei-lipase båret af diatoméjordpartikler. Katalysatoren, leveret af Novo 5 Industri, tørres under vakuum ved 30°C før anvendelse. Katalysatoren indeholder 9100 lipaseenheder pr. gram (1 lipaseenhed frigør 1 pmol fedtsyre pr. minut ved en pH-værdi på 7,0 og ved 30°C fra en 5%'s emulsion af glycerol-tributyrat i en 0,6%'s gummi-arabicum-opløsning).
10 Til påbegyndelse af forestringsreaktionen sættes 2,0 mmol (116 pi) ethanol til blandingen af oliesyre og katalysator. Der tilsættes yderligere syv portioner på 1,15 mmol (67 pi) ethanol med timeintervaller, hvorved fås en samlet ethanoltilsætning på 10 mmol. Prøver tages 15 periodisk fra reaktionsblandingen til analyse af ethanol-indhold og ethyloleatdannelse ved gaschromatografi.
Ved et kontrolforsøg sættes alle 10 mmol ethanol til blandingen af oliesyre og katalysator ved reaktionens start. Igen tages prøver periodisk til analyse.
20 Resultaterne anført i tabel I viser klart, at ethyl- oleat dannes hurtigere i det forsøg, i hvilket ethanol tilsættes i små portioner for at sikre, at ethanolkoncen-trationen i reaktionsblandingen forbliver lav. I dette forsøg er esterdannelsen i alt væsentligt tilendebragt 25 efter 12 timer (det maksimale opnåelige udbytte af ester er ca. 7 mmol).

I et kontrolforsøg sættes 16,5 ml ethanol i én portion til blandingen af smørolie og katalysator ved reaktionens 25 start. Igen tages prøver periodisk til analyse.
Resultaterne vist i tabel II viser, at ethylestere dannes hurtigere i det forsøg, i hvilket ethanol pumpes gradvis ind i reaktionsblandingen. I dette forsøg er ethanol-indholdet af reaktionsblandingen lavt og når et maksimums-30 niveau på 0,14 mol efter 8 timer.

med 2,5 g af katalysatoren beskrevet i eksempel 1. Ethanol pumpes gradvis ind i reaktionsblandingen med en hastighed på 1,03 ml/time i 24 timer til opnåelse af en samlet ethanoltilsætning på 24,7 ml (0,424 mol). Prøver tages 20 periodisk til analyse af reaktionsprodukterne.
I et kontrolforsøg sættes 24,7 ml ethanol i én portion til blandingen af kokosnøddeolie og katalysator ved reaktionens start. Produktet, der dannes efter 24 timer, analyseres.
25 Resultaterne anført i tabel III viser igen, at ethyl- estere dannes hurtigere i det forsøg, i hvilket ethanol gradvis sættes til reaktionsblandingen. I dette forsøg når ethanolindholdet af reaktionsblandingen et maksimumsniveau på 0,22 mol efter 24 timers omsætning.

En immobiliseret lipasekatalysator fremstilles under anvendelse af Candida cyclindracae-lipase og "Duolite ES 568"-ionbytterharpiks (Rohm & Haas, USA). Lipase MY, fra 15 Meito Sangyo Co. Ltd., opløses i 0,1M kaliumphosphatpuffer med en pH-værdi på 6,0 til dannelse af en opløsning indeholdende 1400 lipaseenheder ml”'*·. 79 ml af denne opløsning omrøres med 8,0 g af den ovennævnte harpiks ved stuetemperatur i 4 timer. Der sker adsorption af lipasen på harpikspartikler-20 ne, og immobiliserede lipasekatalysatorpartikler opsamles ved filtrering og tørres under vakuum ved stuetemperatur. Mængden af lipase adsorberet på harpiksen er 6.000 lipaseenheder g”^.
6,38 mmol (1,80 g) oliesyre omrøres ved 40°C med 25 50 mg af den immobiliserede Candida cylindracae-lipase- katalysator. Til påbegyndelse af en forestringsreaktion sættes 0,4 mmol (30 pi) n-propanol til blandingen. Der tilsættes yderligere 23 portioner på 0,267 mmol (20 pi) med intervaller på 20 minutter til opnåelse af en samlet 30 n-propanol-tiIsætning på 6,54 mmol. Prøver tages periodisk fra reaktionsblandingen til analyse af n-propanol-indhold og n-propyloleat-dannelse.
I et kontrolforsøg sættes alle 6,54 nmol n-propanol til blandingen af oliesyre og katalysator ved reaktionens 35 start, og igen tages prøver periodisk til analyse.
Resultaterne anført i tabel IV viser klart, at n-propyloleat dannes hurtigere i det forsøg, i hvilket 0 7 DK 171810 B1 n-propanol tilsættes langsomt i små portioner for at sikre, at koncentrationen af n-propanol i reaktionsblandingen forbliver lav.

Claims (9)
1. Fremgangsmåde til fremstilling af en ester ud fra en alkohol og fedtsyre (glycerid) ved lipase-katalyseret omdannelse, kendetegnet ved, at en fedtsyre 5 (glycerid) og en C1_8-alkohol kontaktes med lipase, og at alkoholen tilsættes med sådan en hastighed, at alkoholkoncentrationen i reaktionsblandingen holdes under et molært forhold på 0,5 mol alkohol pr. mol fedtsyre/acylgruppe.
2. Fremgangsmåde ifølge krav 1, kendeteg- 10 net ved, at alkoholen tilsættes med en sådan hastighed, at det molære forhold mellem alkohol og fedtsyre/acylgruppe er under 0,25.
3. Fremgangsmåde ifølge krav 1 eller 2, kendetegnet ved, at alkoholen er en monovalent alkohol.
4. Fremgangsmåde ifølge ethvert af kravene 1-3, kendetegnet ved, at fedtsyren er en C2-C24“fedt-syre.
5. Fremgangsmåde ifølge ethvert af kravene 1-3, kendetegnet ved, at fedtsyretriglyceridet er 20 afledt af Ci0“C24-fedtsyrer.
6. Fremgangsmåde ifølge krav 4, kendetegnet ved, at reaktionsvand fjernes, medens forestringen forløber.
7. Fremgangsmåde ifølge krav 6, kendeteg- 25 net ved, at vand fjernes ved neddypning af en kold overflade i den gasformige fase af reaktionsbeholderen.
8. Fremgangsmåde ifølge ethvert af kravene 1-7, kendetegnet ved, at esteren fremstilles i nærværelse vand.
9. Fremgangsmåde ifølge krav 6, kendeteg net ved, at vand fjernes ved destillation.
