Som ovenfor nævnt, omhandler opfindelsen også salte af forbindelserne med formlen I, som er syrer, og således danner salte med baser. Som eksempler på salte, som er dannet med farmaceutisk acceptable, ugiftige baser, kan nævnes alkalimetalsalte og jordalkalimetalsalte, såsom lithium, natrium, kalium, mag-5 nesium og kalciumsalte, såvel som salte med ammoniak og passende ugiftige aminer, såsom lavere alkylaminer, fe triethylamin, lavere alkanolaminer, fe diethanolamin eller triethanolamin, procain, cykloalkylaminer, fe dicy-klohexylamin, benzylaminer, fe N-methylbenzylamin, N-ethylbenzylamin, N-benzyl-P-fenethylamin, Ν,Ν’-dibenzylethylendiamin eller dibenzylamin og 10 heterocykliske aminer, fe morfolin, N-ethylpiperidin og lignende.
Normale knogler er levende væv, som undergår en konstant resoiption og genaflejring af kalcium, hvis nettoeffekt er opretholdelsen af en konstant mineralbalance. Denne dobbelte proces kaldes almindeligvis "knogfeomsætnin-gen". I normalt voksende knogler overstiger mineralaflejringen mineral-15 resorptionen, medens knogleresoiptionen ved visse patologiske tilstande overstiger knogleaflejringen, hvilket fe resulterer i hyperkalcæmi, for eksempel på grund af maligne tilstande eller primær hyperparatyroidisme, eller i osteoporose. Ved andre patologiske tilstande kan kalciumaflejringen finde sted i uønskede mængder og på uønskede steder, som fe kan føre til osteoarthritis, rheumatoid 20 arthritis, nyre- og blærestene, atherosclerose, og Paget’s sygdom, som er en kombination af en abnorm høj knogleresorption, efterfulgt af en abnorm kalciumaflejring.
De fleste hidtil tilgængelige terapeutiske stoffer til behandling af osteoporose, fe østrogener og kalcitonin, virker ved at reducere knogleresorptionen 25 hos den osteoporotiske patient. Da benbrud er et alvorligt problem ved osteoporose, burde det ideelle terapeutiske stof være i stand til at forøge knoglemassen til et niveau, som overstiger brudgrænsen.
Forsøg med rotter har vist, at forbindelserne ifølge den foreliggende opfindelse, ligesom fe kendte bisfosfonater (fe l-hydroxyethyliden-l,l-bisfos-30 fonsyre (etidronate) og 3-amino-l-hydroxypropyliden-l,l-bisfosfonsyre (APD) DK 169678 B1 3 reducerer knogleresorptionen, hvilket ses af deres hæmning af urinudskillelsen af hydroxyprolin, men i modsætning til disse kendte stoffer, som også hæmmer knoglevævets alkalinfosfatase, stimulerer de nye forbindelser ifølge opfindelsen overraskende knoglevævets alkalinfosfatase, hvilket tyder på en stimulation af 5 de knogledannende celler - osteoblasteme - og en væsentlig forøgelse af knoglemassen ses faktisk under behandlingen med de foreliggende forbindelser.
Fra tysk offentliggørelsesskrift nr. 3.232.998 kendes nogle thiazindioxid-difosfonsyrederivater, der bl.a. anvendes som sekvestrerende midler til beskyttelse mod korrosion og belægningsdannelser, herunder modvirkning af tandsten. 10 Forbindelsernes effekt i knogleomsætningen er ikke beskrevet i trykskriftet, der heller ikke omtaler de tilsvarende sulfoxider og sulfider. Vi har sammenlignet forbindelsen ifølge eksempel 1 i nævnte offentliggørelsesskrift, 1,4-thiazin-dioxid-N-methandifosfonsyre (C) i tabellen nedenfor, med det tilsvarende sul-foxid (B) og sulfid (A) ifølge den foreliggende opfindelse på følgende måde: 15 Hunrotter (140-160 g) anbragtes i stofskiftestudie-bure. Test-forbindel serne opløstes i 2% NaHCOg pufferopløsning og blev indgivet subkutant dagligt i en mængde på 16 pmol/kg i 7 dage til grupper på 3 rotter hver. En kontrolgruppe blev doseret med pufferopløsningen alene. Urinen opsamledes dagligt, og hydroxyprolin-indholdet heri (som er en indikator for knogleresorptionen) 20 bestemtes ved Prockop og Udenfriends metode (Anal. Biochem. 1, 228-29, 1960). Inhiberingen af knogleresorptionen udtryktes som %-vis ændring i hydro-xyprolinudskillelsen fra dag 3 til dag 7, sammenlignet med kontrolgruppen.
Forøgelsen i knoglemassen blev bestemt som %-vis ændring i vægten af tibia metaphysea, sammenlignet med kontrolgruppen.

Tabellen viser, at forbindelserne A og B ifølge den foreliggende opfin-15 delse er klart mere aktive end forbindelsen ifølge offentliggørelsesskriftet. Sidstnævnte må betegnes som i praksis inaktiv, og testen viser samtidig, at introduktionen af en methylen-difosfonat-enhed i et molekyle ikke nødvendigvis fører til forbindelser, der er effektive overfor osteoporose og/eller beslægtede sygdomme.

Spaltning af estrene med formlen IV ved hydrolyse, fe med kogende saltsyre, eller ved hydrogenolyse, fører til forbindelser med formlen I (n = 0).
Estrene kan også spaltes ved en alternativ metode, som er beskrevet i Jour. f. prakt. Chemie 320. 344 (1978). Behandling med bromtrimethylsilan ved 15 stuetemperatur eller let forhøjet temperatur fører til en tetratrimethylsilylester, som let spaltes med vand eller alkohol, hvorved man far den frie syre med formlen I.
Alternativt kan forbindelserne med formlen I (n = 0) fremstilles ved at omsætte forbindelserne med formlen II med fosforsyre og fosfortriklorid, efter-20 fulgt af hydrolyse af reaktionsblandingen.
Forbindelserne med formlen I, hvori n = 1 kan fremstilles ved oxidation af forbindelserne med formlen I eller IV (n = 0) med de velkendte reagenser til fremstilling af sulfoxider ud fra sulfider, fe 3-klorperbenzoesyre eller hydrogen-peroxid, fortrinsvis efterfulgt af esterspaltning.
25 Let hydrolyserbare estre af forbindelserne med formlen I kan fremstilles ved at omsætte et salt, fe et sølvsalt eller et kvatemært ammoniumsalt, af en forbindelse med formlen I med et reaktivt halogenid svarende til den ønskede ester.
Tetra-estre kan spaltes ved omsætning med et jodid, fe natriumjodid, 30 hvorved der dannes di-estre af forbindelserne med formlen I.

Forbindelserne ifølge den foreliggende opfindelse er som ovenfor nævnt tænkt anvendt i farmaceutiske præparater, som er anvendelige i behandlingen af osteoporose, rheumatoid arthritis og andre arthritisformer, atherosclerose, hyper-kalcæmi på grund af maligne tilstande eller primær hypeiparathyroidisme, 5 Paget’s sygdom og andre tilstande med en abnormal kalciumbalance.
Forbindelserne ifølge den foreliggende opfindelse kan også anvendes i tandpasta for at hindre kalkaflejring i form af tandsten eller som beskyttelse mod kalciumresorption på grund af syreopløsning.
Den mængde af en forbindelse med formlen I (herefter kaldet den 10 aktive bestanddel), som er nødvendig for den terapeutiske virkning vil naturligvis både afhænge af den pågældende forbindelse, indgivelsesvejen og patienten under behandling. En egnet dosis af en forbindelse med formlen I til en patient, som lider af fe en hyperkalcæmi, som allerede defineret, er 0,001 til 25 mg/kg legemsvægt, idet den mest foretrukne dosis er fra 0,002 til 10 mg/kg af patien-15 tens legemsvægt, fe fra 0,005 til 5 mg/kg; indgivet én eller flere gange dagligt.
I tilfælde af profylaktisk behandling af fe postmenopausal osteoporose er den passende dosis af en forbindelse med formlen I 0,001 til 10 mg/kg legemsvægt, idet den mest foretrukne dosis er fra 0,002 til 5 mg/kg af patients legemsvægt.
20 Selvom det er muligt at indgive den aktive bestanddel alene som den rene forbindelse, foretrækker man at give den i form af et farmaceutisk præparat. Den aktive bestanddel udgør passende fra 0,1 til 99,9 vægt% af præparatet. Dosisenheder af et præparat indeholder passende mellem 0,1 mg og 1 g af den aktive bestanddel. Til topisk indgift udgør den aktive bestanddel 1 til 2 vægt% 25 af præparatet, men den aktive bestanddel kan udgøre op til 10 vægt%.
Ved udtrykket "dosisenhed" forstås en enhed, dvs. en enkelt dosis, som kan indgives til en patient, og som let kan håndteres og pakkes, idet den forbliver en fysisk stabil enhedsdosis, som indeholder enten den aktive bestanddel som farmaceutiske fortyndingsmidler eller bærere.

Præparater ifølge den foreliggende opfindelse, som er egnet til oral indgift, kan være i form af adskilte enheder, såsom kapsler, breve, tabletter eller sugetabletter, som hver indeholder en forudbestemt mængde af den aktive bestanddel; i form af et pulver; i form af en opløsning eller suspension i en 20 vandig eller ikke-vandig væske; eller i form af en olie-i-vand eller vand-i-olie emulsion. Den aktive bestanddel kan også indgives i form af en bolus, letværge eller pasta.
Præparater til rektal indgift kan være i form af et suppositorium, som indeholder den aktive bestanddel i et bæremiddel som fe kakaosmør, eller i 25 form af et lavement.
Egnede præparater til parenteral indgift kan passende omfatte en steril vandig præparation af den aktive bestanddel, som fortrinsvis er isotonisk med modtagerens blod.

Foruden de førnævnte bestanddele kan præparaterne ifølge opfindelsen indeholde en eller flere tilsætningsstoffer, såsom fortyndingsmidler, buffere, 5 smagsstoffer, bindemidler, overfladeaktive stoffer, fortykkelsesmidler, smøremidler, konserveringsmidler, fe methylhydroxybenzoat (herunder antioxidanter), emulgeringsmidler og lignende.
Præparaterne kan desuden indeholde andre terapeutisk aktive forbindelser, som sædvanligvis anvendes i behandlingen af de ovennævnte patologiske 10 tilstande, fe vitamin D2 og Dg og hydroxylerede derivater, fe la-hydroxy vitamin Dg, la-hydroxy-vitamin D2, la,25-dihydroxy vitamin Dg, la,25-dihy-droxy vitamin D2, calcitonin (humant, svine- eller lakse-), mitramycin, natriumfluorid, østrogener og ikke-steroide anti-inflammatoriske mediciner, fe acetylsalicylsyre, indomethacin, naprosyn og timegadin.

Præparation 1 5.5-Dimethvl-3-thiomorfolinon 35,5 g Dimethylaziridin tilsattes dråbevis under omrøring ved 100°C til 25 53 g methylmercaptoacetat i løbet af 1,5 timer. Efter yderligere 2 timer ved 100°C afkøledes blandingen, der krystalliserede fra ethylacetat.

Præparation 2 3.3-Dimethvl-thiomorfolin 50 g 5,5-Dimethyl-3-thiomorfolinon tilsattes portionsvis til en omrørt suspension af 25 g lithiumaluminiumhydrid i 1000 ml tetrahydrofuran. Efter 20 5 timers kogning under tilbagesvaling destrueredes uforbrugt lithiumaluminiumhydrid ved tilsætning af natriumsulfat-dekahydrat. Blandingen filtreredes, og filtratet inddampedes i vakuum. Destillation i vakuum gav titelforbindelsen med et kogepunkt på 71-72°C/15 mmhg.
10 Præparation 3 2.5.5- Trimethvl-3-thiomorfolinon
Man fik denne forbindelse fra 2,2-dimethylaziridin og methyl-2-mercap-topropionat som beskrevet i Præparation 1. Smeltepunkt: 137 - 138°C.
15 Præparation 4 2.5.5- Trimethvlthiomorfolin
Denne forbindelse fremstilledes som beskrevet i Præparation 2 ved at erstatte 2,5,5-dimethyl-3-thiomorfolinon med 5,5-dimethyl-3-thiomorfolinon. Kogepunkt: 76-77°C/15 mmhg.

Præparation 5 2-fl-ButvlV3-thiomorfblinon 56 g Kaliumhydroxyd sattes til en suspension af 56,5 g cysteamin, hydroklorid i 300 ml absolut ethanol, efterfulgt af dråbevis tilsætning af 111,5 g 25 ethyl-2-bromhexanoat. Da den eksoterme reaktion ophørte, kogtes blandingen under tilbagesvaling i 3 timer og filtreredes. Inddampning af filtratet i vakuum gav titelforbindelsen som en viskøs olie, som anvendtes i næste trin uden rensning.

Destillation i vakuum gav N-formylthiomorfolin med et kogepunkt på 140°C/15 mmhg. Når det rene produkt holdtes ved stuetemperatur, dannede det lavt-smeltende krystaller.

Blandingen holdtes i stuetemperatur i 48 timer, skylledes med kvælstof, fortyndedes med ether og vaskedes med vand. Den organiske fase DK 169678 B1 13 tørredes, og opløsningsmidlet inddampedes i vakuum. Destillation af remanensen gav titelforbindelsen med et kogepunkt på 145°C/15 mmhg.
Præparation 10 5 N-Formvl-3-ethvl-2-methvlthiomorfolin 68,2 g Cysteamin, hydroklorid (0,6 mol) sattes til en iskold omrørt opløsning af 27,6 g natrium (1,2 mol) i 1000 ml ethanol. 99 g a-Bromdiethyl-keton (0,6 mol) tilsattes derefter langsomt i en nitrogenatmosfære, medens temperaturen holdtes under 25°C.
10 Efter omrøring i 2 timer filtreredes blandingen, og filtratet inddampedes i vakuum, hvorved man fik en olie, som blev anbragt i 300 ml vand og ekstraheret med 2 x 500 ml ether. Den organiske fase fraskiltes, tørredes og inddam-pedes, hvorved man fik en olie, som destilleredes i vakuum. Destillatet med kp. 70°C/1 mmhg opvarmedes til 140°C under omrøring, og 100 ml myresyre 15 tilsattes med en hastighed, som holdt temperaturen på 135-140°C uden opvarmning udefra. Efter at være anbragt natten over ved stuetemperatur behandledes blandingen langsomt med 200 ml 40% kaliumhydroxyd og ekstraheredes med 2 x 500 ml ether. Den organiske fase tørredes og inddampedes til ca. 500 ml. 80 ml Kloral tilsattes, og blandingen kogtes under tilbagesvaling i 1 time, afkøledes 20 og inddampedes, hvorved man fik en olie, som destilleredes i vakuum og gav titelforbindelsen med et smeltepunkt på 102-104°C/1 mmhg.

Præparation 15 r+V3-tert-Butvlthiomorfolin 20 159 g 3-tert-Butylthiomorfolin (racemisk form beskrevet i Præparation 14) sattes til en varm opløsning af 150 g (+)-vinsyre i 100 ml vand. Blandingen 20 stod natten over og filtreredes derefter. Det krystallinske salt, som viste [a]j) = +20° (c=l, H2O) omkrystalliseredes gentagne gange af vand indtil [a]j^ = +32° (o=l, H2O). Den frie base frigjordes med overskud af 30% kaiiumhydro-25 xyd og opløstes i ether. Opløsningen tørredes og inddampedes, hvorved man fik titelforbindelsen som en olie med [a]j~^ = +36,1° (c=l, EtOH).

Præparation 18 20 Ved at følge fremgangsmåderne beskrevet i Eksempel 15, 16 og 17 kan andre substituerede thiomorfoliner opløses i enantiomereme og derefter for-myleres til dannelse af· fe (+VN-F ormvl-3 -isobutvlthiomorfolin f-VN-Formvl-3-isobutvlthiomorfolin 25
Præparation 19 N-F ormvl-tetrahvdro-1.4-thiazepin-3-on
Til en opløsning af 23 g natrium i 1 1 absolut ethanol sattes 60,1 g ethylthioglycolat og 109,5 g 3-brompropylamin, hydrobromid. Efter kogning 30 under tilbagesvaling i 12 timer, afkøledes og filtreredes blandingen. Filtratet DK 169678 B1 18 inddampedes i vakuum, og remanensen krystalliserede fra ethanol, hvorved man fik tetrahydro-l,4-thiazepin-3-on med et smeltepunkt på 145-148°C. 60 g af dette produkt sattes i portioner til en omrørt suspension af 25 g lithiumalumini-umhydrid i 2,5 1 tetrahydrofuran. Blandingen holdtes ved 45-50°C i 2 timer 5 efterfulgt af kogning under tilbagesvaling i yderligere to timer. Efter afkøling tilsattes dråbevis under omrøring en blanding af 62 ml vand og 250 ml tetrahydrofuran. Filtrering og inddampning af filtratet gav en remanens, som opløstes i 250 ml ether, tørredes over magnesiumsulfat, filtreredes og behandledes dråbevis med 40 ml kloral. Efter kogning under tilbagesvaling i 1 time inddampedes 10 opløsningsmidlet, og remanensen destilleredes i vakuum, hvorved man fik titelforbindelsen med et kogepunkt på 158-159°C/18 mmHg.
Præparation 20 N-F ormvl-2-methvltetrahvdro-1,4-thiazepin-3-on 15 Denne forbindelse fremstilledes ved at følge fremgangsmåden ifølge
Præparation 19, men erstatte ethylthioglycolat med ethyl-2-mercaptopropionat.
2-Methyltetrahydro-l,4-thiazepin-2-on med et smeltepunkt på 192-193°C isole-redes som et mellemprodukt. Titelforbindelsen var en farveløs olie med et kogepunkt på 104-105°C/1 mmHg.

Eksempel 1 (4-thiomorfolinvlmethvlenVbisfosfonsvre 16,6 ml Oxalylklorid tilsattes dråbevis ved 0°C til en omrørt opløsning af 26,2 g N-formylthiomorfolin i 200 ml methylenklorid. Blandingen omrørtes 25 ved stuetemperatur, indtil gasudviklingen ophørte (ca. 5 timer senere). 66 ml Triethylfosfit tilsattes derefter i løbet af 1,5 timer ved stuetemperatur. Uforbrugt triethylfosfit fjernedes i vakuum, og remanensen kogtes under tilbagesvaling med 150 ml 20% saltsyre i 3 timer. Eksempel 3 Γ1-Γ4-ThiomorfolinvlVethvlidenl-bisfosfonsvre. bisbenzvl-25 amin salt
En blanding af 14,5 g N-acetylthiomorfolin og 24,6 g fosforsyre opvarmedes til 100°C i 3 timer. 41,1 g fosfortriklorid tilsattes dråbevis under omrøring, og efter kogning under tilbagesvaling i 3 timer tilsattes langsomt 80 ml vand, og blandingen holdtes på 100°C i 3 timer. Filtrering og inddampning af 30 filtratet gav en olie, som opløstes i ethanol. Titelforbindelsen udfældedes ved DK 169678 B1 23 tilsætning af ether. Det rå produkt omdannedes til et bisbenzylamin salt med et smeltepunkt på 223-224°C.
NMR (D2O, HDO = 4,66 ppm som reference): δ = 1,51 (t, J=13 Hz, 3H); 2,95 (t, 4H);_3,85 (m, 4H); 4,07 (s, 4H) og 7,36 (s, 10H) ppm.

Eksempel 4
Tetrabenzvl-f4-thiomorfolmvlmethvlenVbisfosfonatsulfoxid 0,85 ml Oxalylklorid tilsattes dråbevis ved -10°C til en omrørt opløsning af 1,31 g N-formyl-thiomorfolin i 10 ml tetrahydrofuran, og blandingen 10 omrørtes ved -10°C i 1 time.
En opløsning af natriumdibenzylfosfit i 20 ml tetrahydrofuran (fremstillet ud fra 8,4 g dibenzylfosfit og 1,08 g 55% NaH) tilsattes, og efter omrøring i 1 time inddampedes opløsningsmidlet, og den tilbageblevne olie udiystedes mellem vand og methylenklorid. Den organiske fase tørredes og inddampedes, 15 hvorved man fik en olie, som rensedes ved flashkromatografi på silikagel (ether:acetone 90:10 som elueringsmiddel), hvorved man fik 1,27 g tetraben-zyl-(4-thiomorfolinylmethylen)-bisfosfonat, som blev opløst i 15 ml alkoholfri kloroform. Denne opløsning afkøledes i is og behandledes dråbevis med en opløsning af 0,38 g 90% 3-klorperbenzoesyre i 5 ml alkoholfri kloroform.
20 Isbadet fjernedes, og efter omrøring i yderligere 90 minutter, afdampedes opløsningsmidlet i vakuum, og remanensen rensedes ved kromatografi på silikagel, hvorved man fik titelforbindelsen som en farveløs olie.
NMR (CDCI3, TMS = 0,0 ppm som reference): 8 = 2,62 (bt, 4H); 3,0 (bm, 2H); 3,36 (t, J=25 Hz, IH), 3,60 (bm, 2H); 5,04 (m, 8H) og 7,30 (t, 20H) ppm. 25
Eksempel 5 f4-ThiomorfolinvlmethvlenVbisfosfonsvresulfoxid
Til en opløsning af 1 g tetrabenzyl-(4-thiomorfolinylmethylen)-bisfosfo-natsulfoxid i 15 ml ethylacetat sattes 15 ml vand og 1 g 10% palladium på kul.
30 Den resulterende blanding omrystedes kraftigt i en hydrogenatmosfære, indtil DK 169678 B1 24 hydrogenforbruget ophørte. Katalysatoren fjernedes ved filtrering, og den vandige fase fraskiltes og fiysetørredes, hvorved man fik titelforbindelsen som et amorft pulver.
NMR (NaOD, HDO = 4,66 ppm som reference): δ = 2,75 (t, J=22,6 Hz, IH); 5 2,7-3,25 (m, 6H) og 3,3-3,7 (m, 2H) ppm.

Man gentog fremgangsmåden ifølge Eksempel 1, idet N-formylthiomor-folin erstattedes af de passende N-formylderivater beskrevet i Præparation 13.

Eksempel 7 25 f3-Isobutvl-4-thiomorfolinvlmethvlenVbisfosfonsvre
En blanding af 18,7 g N-formyl-3-isobutylthiomorfolin og 16,4 g fosforsyre opvarmedes til 100°C i 2 timer. 30 ml Fosfortriklorid tilsattes dråbevis under omrøring, og efter kogning under tilbagesvaling i 3 timer tilsattes langsomt 180 ml vand. Blandingen blev kogt under tilbagesvaling i 1 time og filtre- DK 169678 B1 25 ret. Inddampning af filtratet gav en remanens, som behandledes med ethanol, hvorved man fik en krystallinsk forbindelse, som isoleredes ved filtrering.
Smp.: 237-238°C (under sønderdeling).
5 Eksempel 8
Fremgangsmåden ifølge Eksempel 7 blev gentaget, idet N-formyl-3-iso-butylthiomorfolin blev erstattet af de passende N-formylderivater beskrevet i Præparation 13, 17 og 18. 5 monohvdrat 11,1 ml 30% vandig brintoverilte sattes under omrøring til en opløsning af 27,7 g (4-thiomorfolinylmethylen)-bisfosfonsyre i 40 ml 5 M NaOH. Da den eksoterme reaktion var ophørt, inddampedes opløsningsmidlet, og remanensen behandledes med ethanol og filtreredes, hvorved man fik titelforbindelsen som 10 farveløse krystaller. NMR-spektret var identisk med det i Eksempel 5 beskrevne.

Eksempel 12
Tetra-pivalovloxvmethvl-f4-thiomorfolinvlmethvlenVbis-10 fosfonat
En opløsning af 2,8 g (4-thiomorfolinylmethylen)-bisfosfonsyre i 60 ml vand behandledes med fire ækvivalenter vandig tetrabutylammoniumhydroxid og frysetørredes. Overskydende vand fjernedes azeotropisk med toluen. Det resulterende salt suspenderedes i 120 ml tetrahydrofuran, og 9,4 ml jodmethyl-15 pivalat tilsattes under omrøring. Efter omrøring i 1 time ved stuetemperatur filtreredes blandingen, og filtratet inddampedes i vakuum. Remanensen blev optaget i ethylacetat og vasket med vandig natriumbikarbonat, tørret og inddam-pet i vakuum. Kromatografi på silikagel (elueringsmiddel: petroleumether -ethylacetat 7:3) gav den rene titelforbindelse, som krystalliserede efter henstand. 20 Smp.: 86-88°C.
NMR (CDCI3): δ = 1,24 (s, 36H); 2,65 (m, 4H); 3,25 (m, 4H); 3,55 (t, J=26 Hz, IH) og 5,70 (m, 8H) ppm.
Eksempel 13 25 Tetraacetoxvmethvl-(4-thiomorfolinvlmethvlenl-bisfosfonat
Ved at følge den i Eksempel 12 beskrevne fremgangsmåde, idet man erstattede jodmethylpivalat med jodmethylacetat, fik man titelforbindelsen som en farveløs olie.
NMR (CDCI3): δ = 2,14 (s, 6H); 2,16 (s, 6H); 2,65 (m, 4H); 3,26 (m, 4H); 30 3,53 (t, J=26 Hz, IH) og 5,70 (m, 8H) ppm.

Eksempel 14
Di-pivalovloxvmethvl-i4-thiomorfolinvlmethvlen)-bisfosfonat dinatriumsalt
En opløsning af 0,37 g tetra-pivaloyloxymethyl-(4-thiomorfolinylmethy-len)-bisfosfonat og 0,22 g natriumjodid i 5 ml acetone kogtes under tilbages-5 valing i 90 minutter, afkøledes og filtreredes, hvorved man fik titelforbindelsen som farveløse krystaller.
NMR (D20): δ = 1,16 (s, 18H); 2,65 (m, 4H); 3,05 (t, IH); 3,18 (m, 4H) og 5,52 (m, 4H) ppm.

Di-acetoxvmethvl-f4-thiomorfoiinvlmethvlen)bisfosfonat dinatrium salt
Denne forbindelse fremstilledes ud fra tetra-acetoxymethyl-(4-thiomor-folinylmethylen)-bisfosfonat ved at følge fremgangsmåden ifølge Eksempel 14.

Claims (8)
1. Bisfpsfonsyrederivater med den almene formel I 5 p R* R3 p O OH Rsy-^ VV-oh (0)1 a7C^p^oh i IX) m 0XOH 10 7 88 R,ri« Ru hvor Rj-Rj i er ens eller forskellige og betegner hydrogen, en lige eller forgrenet alifatisk eller alicyklisk Cj-CjQ-hydrokarbongruppe, en aiyl eller en aryl--Cj-C^-alkylgruppe; eller R2 og R4 sammen danner en mættet alifatisk 5-, 6-15 eller 7-leddet ring, som eventuelt er substitueret med en eller flere Cj-C^-alkyl-grupper; n er 0 eller 1, og m er 0, 1 eller 2; samt salte og estere deraf, hvor den esterdannende remanens er alkanoyloxymethyl- med tre til seks karbonatomer, 1 -(alkanoyloxy)ethyl- med fire til syv karbonatomer, 1 -methyl-1 -(alkanoyloxy)-ethyl- med fem til otte karbonatomer, (alkoxycarbonyloxy)methyl- med tre til 20 seks karbonatomer, 1 -(alkoxycarbonyloxy)ethyl- med fire til syv karbonatomer, 1 -methyl-1 (alkoxykarbonyloxy)ethyl- med fem til otte karbonatomer, 3-ftalidyl-, 4-krotonolactonyl-, y-butyrolacton-4-yl-, (2-oxo-1,3-dioxolen-4-yl)methyl-, (5-methyl-2-oxo-1,3-dioxolen-4-yl)methyl-, (5-fenyl-2-oxo~ 1,3-dioxolen-4-yl)-, dialkylaminoalkyl-, acetonyl- eller methoxymethyl. 25

Salt ifølge krav 1, kendetegnet ved, at det er udvalgt fra gruppen 30 bestående af alkalimetalsalte, jordalkalimetalsalte, salte med ammoniak og salte DK 169678 B1 31 med egnede ugiftige aminer, såsom lavere alkylaminer, lavere alkanolaminer, procain, cykloalkylaminer, benzylaminer og heterocykliske aminer.
4. Forbindelse samt salte og estere deraf ifølge krav 1, kendetegnet ved, 5 at den er udvalgt fra gruppen bestående af (4-thiomorfolinylmethylen)-bisfosfonsyre, (3-isobutyl-4-thiomorfolinylmethylen)-bisfosfonsyre i racemisk form eller i form 10 af de enkelte enantiomerer, (3-tert-butyl-4-thiomorfolinylmethylen)-bisfosfonsyre i racemisk form eller i form af de enkelte enantiomerer, og 15 (2-methyl-4-thiomorfolinylmethylen)-bisfosfonsyre i racemisk form eller i form af de enkelte enantiomerer.

Fremgangsmåde til fremstilling af en forbindelse med formel I ifølge krav 1, 20 kendetegnet ved, at en forbindelse med formel II, som defineret i krav 5, opvarmes med en blanding af fosforsyre og fosfortriklorid eller fosforoxy-klorid, hydrolyseres til dannelse af den ønskede forbindelse med formel I (n=0), som om ønsket oxyderes til dannelse af den ønskede forbindelse med formel I (n=l), idet slutproduktet med formel I udvindes som sådant eller om ønsket som 25 et salt eller en ester som defineret i krav 1.
7. Farmaceutisk præparat, kendetegnet ved, at det indeholder en effektiv mængde af en eller flere af forbindelserne ifølge krav 1, sammen med farmaceutisk acceptable, ugiftige bærer- og/eller hjælpestoffer. 30 DK 169678 B1 33
8.
