Description
- Hintergrund der Erfindung
- Epstein-Barr-Virus (EBV) ist ein Human-Herpes-Virus mit einer latenten Phase, welche durch die stabile episomale Vermehrung einer zirkulären Form der viralen DNA gekennzeichnet ist. Für die Replikation der latenten Phase werden zwei diskontinuierliche DNA-Elemente benötigt, der cis-wirkende Replikationsursprung, OriP, und das Eppstein-Barr-Kern-Antigen 1 (EBNA1), das einzige viral codierte Protein, welches für die Replikation erforderlich ist (Yates et al., Proc. Natl. Acad. Sci. USA 81: 3806–10, 1984; Yates et al., Nature 313: 812–5, 1985). Die meisten EBV-Vektoren sind sehr groß, in der Größenordnung von 10 kb oder darüber (ohne Insert), aufgrund der beträchtlichen Größe der EBNA1- und OriP-Segmente. Die Größe der meisten existierenden EBV-EBNA1- und OriP-Segmente beeinträchtigen die Entwicklung von verbesserten Expressionsvektoren und die Ausbildung von kompakten Genexpressions- und -persistenzkassetten, welche in andere Genabgabevehikel, wie Retroviren oder Adenoviren, eingebettet werden können.

US 4939088 beschreibt ein kompaktes Epstein-Barr-Virus-Expressionssystem, welches die episomale Replikation unterstützt, wobei das Expressionssystem OriP und EBNA1-Nukleotide umfaßt. - Zusammenfassung der Erfindung
- Allgemein betrifft die Erfindung eine Nukleinsäuresequenz, welche die episomale Replikation in einer Säugetierzelle unterstützt. Diese Nukleinsäuresequenz umfaßt (a) eine deletierte OriP-Sequenz und (b) eine deletierte EBNA1-Sequenz, operabel an einen Promotor gebunden, und die Nukleinsäuresequenz besitzt eine Länge von weniger als 3 kb, vorzugsweise von weniger als 2 kb und am stärksten bevorzugt von weniger als 1,8 kb, wobei die OriP-Sequenz die Reste 1–495 der SEQ ID NR. 1 umfaßt; die EBNA1-Sequenz die Reste 627–1718 der SEQ ID NR. 1 umfaßt; die Nukleinsäuresequenz ferner vorzugsweise eine Polyadenylierungs-Konsensus-Sequenz (beispielsweise die Sequenz der SEQ ID NR. 2 umfaßt); und der Promotor ein viraler Promotor ist.
- In verwandten Aspekten betrifft die Verbindung auch Vektoren und Zellen (beispielsweise Säugetierzellen und vorzugsweise Humanzellen), welche derartige Nukleinsäuresequenzen enthalten.
- Detaillierte Beschreibung
- Zunächst werden die Zeichnungen beschrieben werden.

1 ist die Sequenz einer gekürzten EBV-OriP und EBNA1-Kassette. -
2 ist die Sequenz einer kompakten synthetischen bidirektionalen Polyadenylierungssequenz, welche in Verbindung mit einem kompakten EBV-Replikon verwendet werden kann. - Nach umfassender Deletion und Mutagenese hat es sich als möglich erwiesen, die cis- und trans-wirkenden Funktionen, die für die EBV-episomale Replikation erforderlich sind, in ein Fragment von weniger als 2 kb einzubetten. Insbesondere ist ein beispielhaftes Fragment von 1748 Basenpaaren, welches als ein kompaktes EBV-Replikon wirkt, in
1 , SEQ ID NR: 1, gezeigt. Dieses Fragment enthält alle für die effiziente Expression des EBNA1-Proteins erforderlichen Sequenzen, mit der Ausnahme einer Polyadenylierungs-Konsensus-Sequenz. Das Fragment ist ein Bg12 bis BamH1-Segment, welches das OriP-Element zwischen den Resten 1 und 495, einen modifizierten Promotor aus dem Herpes simplex-Virus 1-Thymidinkinase-Gen zwischen den Resten 496 bis 616 und die codierende Sequenz für ein deletiertes und modifiziertes EBNA1-Gen zwischen den Resten 627 und 1718 enthält. Die auf dieser Sequenz basierenden Plasmidvektoren replizieren als Episome im Kern von transfizierten Zellen von Nicht-Nagetier-Urspung (Yates et al., 1985, oben). - Um die Gesamtsequenzlänge zu minimieren, wurde das vorstehende Fragment ausgebildet, um stromaufwärts von einer bidirektionalen Polyadenylierungs-Sequenz in einem geeigneten Vektor insertiert zu werden. Ein Beispiel einer kompakten synthetischen bidirektionalen Polyadenylierungs-Sequenz ist in
2 , SEQ ID NR. 2, gezeigt. - Kompakte EBV-Replikone finden Verwendung in Gentherapievektoren, beispielsweise in Genabgabevehikeln, wie Expressionsvektoren.
Claims (8)
- Nukleinsäuresequenz, welche die episomale Replikation in einer Säugetierzelle unterstützt, welche Nukleinsäuresequenz (a) eine OriP-Sequenz und (b) eine EBNA1-Sequenz, operabel an einen Promoter gebunden, umfaßt, welche Nukleinsäuresequenz eine Länge von weniger als 3 kb besitzt, worin die OriP-Sequenz die Reste 1–495 der SEQ ID NR. 1 und die EBNA1-Sequenz die Reste 627–1718 der SEQ ID NR. 1 umfaßt.
- Nukleinsäuresequenz nach Anspruch 1, welche Nukleinsäure eine Länge von weniger als 2 kb besitzt.
- Nukleinsäuresequenz nach Anspruch 1, welche Nukleinsäure eine Länge von weniger als 1,8 kb besitzt.
- Nukleinsäuresequenz nach einem der Ansprüche 1 bis 3, welche Nukleinsäuresequenz ferner eine Polyadenylierungs-Konsensus-Sequenz umf aßt.
- Nukleinsäuresequenz nach Anspruch 4, wobei die genannte Polyadenylierungs-Konsenus-Sequenz die SEQ ID NR. 2 umfaßt.
- Nukleinsäuresequenz nach einem der Ansprüche 1 bis 5, wobei der genannte Promoter ein viraler Promoter ist.
- Vektor, welcher die Nukleinsäuresequenz nach einem der Ansprüche 1 bis 6 umf aßt, welcher Vektor zur Replikation als ein Episom im Kern einer transfizierten Zelle fähig ist.
- Zelle, umfassend die Nukleinsäuresequenz nach einem der Ansprüche 1 bis 6 und/oder den Vektor nach Anspruch 7.
