【発明の詳細な説明】
小型エプスタイン・バーウイルスレプリコン
発明の背景
エプスタイン・バーウイルス(EBV)は、環状ウイルス性DNAの安定的なエピソー
ム増殖に特徴づけられる潜伏相を石するヒトヘルペスウイルスである。2つの非
連続性DNAエレメントは、潜伏相での複製、複製のシス作用オリジン、OriP、お
よびエプスタイン・バー核抗原1(EBNA1)に必要であり、複製に必要な唯一の
ウイルスにコードされるタンパク質である(Yates et al.,Proc.Natl.Acad.S
ci.USA 81:3806-10,1984;Yates et al.,Nature 313:812-5,1985)。ほとんどの
EBVベクターは、EBNA1およびOriPセグメントの大きさが大きいために、10kbま
たはそれ以上の単位と(挿入は除く)、非常に大きい。大半の既存のEBV EBNA1
およびOriPセグメントの大きさにより、改良発現ベクターの開発、ならびにレト
ロウイルスやアデノウイルスのようなほかの遺伝子輸送担体への埋め込みが可能
な小型の遺伝子発現および持続性カセットの作製が妨げられている。
発明の概要
概して、本発明は哺乳類細胞内でのエピソーム複製を助ける核酸配列を特徴と
する。この核酸配列は(a)切り取られたOriP配列と(b)プロモーターに機能
的に結合された、切り取られたEBNA1配列とを含み、且つ核酸配列の長さは3kb
未満、好ましくは2kb未満、そして最も好ましくは1.8kb未満である。
好ましい態様において、OriP配列は配列番号:1のおよそ1〜495残基を含
み、EBNA1配列は配列番号:1のおよそ627〜1718残基を含み、核酸配列は
ポリアデニル化コンセンサス配列(例えば、配列番号:2の配列とほぼ同じ)を
さらに含み、且つプロモーターはウイルス性のプロモーターである。
これに関連して、本発明はこのような核酸配列を含んでいるベクターおよび細
胞(例えば、哺乳類細胞、そして好ましくはヒト細胞)をも特徴とする。
詳細な説明
図をはじめに示す。
図1は短縮したEBV OriPおよびEBNA-1カセットの配列である。
図2は小型合成双方向型ポリアデニル化配列であり、小型EBVレプリコンとの
連
結に使うことができる配列である。
広範にわたる切り取りと変異誘発の結果、EBVエピソーム複製に必要なシスお
よびトランス作用機能を2kb未満の断片に挿入できることが判った。特に、小型
EBVレプリコンとして機能する1748塩基対の典型的な断片を図1に示す(配列
番号:1)。この断片は、ポリアデニル化コンセンサス配列を除き、EBNA-1タンパ
ク質を効率良く発現するために必要なすべての配列を含んでいる。この断片はBg
l2からBamH1までのセグメントであり、1から495残基間にOriPエレメント、
496から616残基間に単純ヘルペスウイルス1チミジンキナーゼ遺伝子由来
の修飾プロモーター、ならびに627から1718残基間に切り取って修飾され
たEBNA1遺伝子をコードする配列を含んでいる。この配列に基づいたプラスミド
ベクターは非齧歯類由来のトランスフェクトされた細胞の核内でエピソームとし
て複製する(Yates et al.,1985,前記)。
全体の配列の長さを最小にするために、上記断片を、適当なベクター中で双方
向ポリアデニル化配列の上流部位に挿入されるよう設計した。小型合成双方向ポ
リアデニル化配列の例を図2(配列番号:2)に示す。
小型EBVレプリコンは遺伝子治療ベクター、例えば、発現ベクターのような遺
伝子輸送担体としての使用が見出されている。
