Abstract

Description
엡스타인-바 바이러스(Epstein-Barr virus ; EBV)는 원형 바이러스성 DNA의 안정적인 에피솜(episomal) 증식을 특징으로 하는 잠복기를 갖는 인간 헤르페스 바이러스(herpes virus)이다. 잠복기 복제를 위해 두 개의 불연속적인 DNA 요소가 필요한데, 시스(cis)-활성 복제 개시점, OriP, 복제에 필요한 유일하게 바이러스에 의해 코드화되는 단백질인 엡스타인-바 핵 항원 1(Epstein-Barr Nuclear Antigen 1 ; EBNA1)이다(Yates et al., Proc. Natl. Acad. Sci. 대폭적인 결실 및 돌연변이 유발에 의해, EBV 에피솜 복제를 위해 필요한 시스- 및 트랜스-작용성 기능을 2kb 미만의 프래그먼트에 끼워넣는 것이 가능하다는 사실을 밝혀내었다. 특히, 컴팩트한 EBV 레플리콘으로 작용하는 1748 염기 쌍의 예시적 프래그먼트를 도 1a 및 도 1b에 도시한다(서열 1). 이 프래그먼트는 폴리아데닐화 공통 서열을 제외한 효과적 EBNA-1 단백질의 발현을 위해 필요한 모든 서열을 포함한다. 상기 프래그먼트는 잔기 1과 485 사이에 OriP 요소를 포함하고, 잔기 496과 616 사이에 헤르페스 단순 바이러스 1 티미딘 키나아제 유전자로부터의 변형 프로모터를 포함하며, 잔기 627과 1718 사이에 결실되고 변형된 EBNA1 유전자에 대한 코드화 서열을 포함하는 Bgl2에서 BamH1 세그먼트이다. 이 서열에 기초한 플라스미드 벡터는 설치류가 아닌 생물로부터 유래된 트랜스팩션된 세포의 핵내에서 에피솜으로 복제한다(Yates et al., 1985, supra).

컴팩트한 EBV 레플리콘은 유전자 치료 벡터로 사용되는데, 예를 들어, 발현 벡터와 같은 유전자 전달 비히클로 사용된다.

Claims (10)
- 포유류 세포 내에서 에피솜 복제를 유지하는 핵산 서열에 있어서, 상기 핵산 서열은 프로모터(promotor)에 작용적으로 연결된 (a)대략적으로 서열 1의 잔기 1∼495를 포함하는 OriP 서열 및 (b)대략적으로 서열 1의 잔기 627∼1718을 포함하는 EBNA1 서열을 포함하고, 상기 핵산 서열은 3kb 미만의 길이를 갖는 핵산 서열.
- 제 1항에 있어서, 상기 핵산 서열은 2kb 미만의 길이를 갖는 핵산 서열.
