Description
- Die vorliegende Erfindung betrifft eine Kombination aus DL-Lysin-acetylsalicylat, Vitamin E und β-Carotin.
- Die verwendete Quelle für Vitamin E ist racemisches oder rechtsdrehendes, natürliches oder synthetisches α-Tocopherol in Form der Base oder eines Salzes, wie des Acetats, Palmitats oder Succinats.
- Die Acetylsalicylsäure ist ein bekanntes, gegen die Blutplättchenaggregation gerichtetes Mittel, das bei der sekundären Vorbeugung von Herz- und Gehirngefäßvorfällen verwendet wird, wie es beispielsweise in B. M. J. (1974), 436-440, J. A. M. A., Vol. 260, Nr. 14 (1988), 2088-2093 und Circulation, Vol. 87, Nr. 2 (1993), 659-675 beschrieben worden ist.
- Lysin-acetylsalicylat, eine in Wasser lösliche und stabile Form, verbessert die lokale Verträglichkeit der Acetylsalicylsäure im Verdauungstrakt beträchtlich.
- Vitamin E besitzt ebenfalls eine gegen die Blutplättchenaggregation gerichtete Wirkung. Darüber hinaus übt es eine gefäßerweiternde Wirkung auf die Arterienwandungen aus.
- Vitamin E und β-Carotin sind andererseits als Antioxidantien und Abfänger für freie Radikale bekannt.
- Es ist bekannt, daß freie Radikale Veränderungen von Nucleinsäuren, Proteinen und Zelllipiden verursachen und damit eine Rolle spielen bei der Cancerogenese, der Atherogenese und der Ischämie. Es ist bekannt, daß die Bildung von Atheromen durch oxidiertes LDL-Cholesterol begünstigt wird als Folge seiner inhibierenden Wirkung auf die Motilität der Gewebe-Makrophagen, seiner Einarbeitung in Monozyten in der Arterienwandung, seiner zytotoxischen Wirkung gegenüer endothelialen Zellen und seiner arteriellen gefäßverengenden Wirkung und aufgrund von freien Radikalen, die bei traumatischen Vorfällen des Endotheliums freigesetzt wer den.
- Vitamin E und β-Carotin wirken gegen die Oxidation von LDL-Cholesterol und inaktivieren die durch das Endothelium freigesetzten freien Radikale, verhindern oder vermindern die Bildung von Atheromen und verringern in dieser Weise die Risiken für kardio- und zerebrovaskuläre Erkrankungen.
- In Atherosclerosis, Vol. 82, Nr. 3 (1990), Seiten 247-252 ist eine klinische Untersuchung beschrieben, welche die Verwendung einer Kombination aus Aspirin und Vitamin E zur Inhibierung der Blutplättchenaggregation vorschlägt.
- Andererseits üben Vitamin E und β-Carotin als Antioxidantien und Abfänger von freien Radikalen, regulierenden Mitteln für die Zelldifferenzierung und Immunstimulantien, und Vitamin E aufgrund seiner blockierenden Wirkung auf die Umwandlung von Nitrosaminen in cancerogene Nitrosamide eine anti-neoplastische Wirkung aus.
- Die erfindungsgemäße Kombination ist daher für die primäre Vorbeugung von Herz- und Gehirngefäßerkrankungen, der sekundären Vorbeugung von Herz- und Gehirngefäßvorfällen, zur Vorbeugung der Restenose nach der Koronarangioplastie und zur Vorbeugung des Auftretens und/oder des Fortschreitens von atheromatösen Läsionen, für die Vorbeugung der Koronaratheromatose und der Verengung von Transplantaten nach Bypass-Operationen und zur Vorbeugung von Krebsen jeglicher Art bestimmt.
- Die erfindungsgemäße Kombination kann in Kombination mit geeigneten Trägermaterialien für die Herstellung von pharmazeutischen Zubereitungen verwendet werden, die auf oralem Wege verabreicht werden soll.
- Die Trägermaterialien werden aus den üblicherweise bei der Formulierung und der Herstellung von Arzneimitteln verwendeten Bestandteilen ausgewählt, wie Verdünnungsmitteln, Puffersubstanzen, Gleitmitteln, aromatisierenden Mitteln und Sü - Bungsmitteln.
- Die erfindungsgemäßen pharmazeutischen Zubereitungen können in Form von Tabletten, Gelkapseln, Kapseln oder Pulvern und insbesondere Pulvern für die Herstellung von trinkbaren Lösungen vorliegen.
- Sie können 54 bis 540 mg DL-Lysen-acetylsalicylat, das heißt ein Äquivalent von 30 bis 300 mg Acetylsalicylsäure, 10 bis 1000 IE Vitamin E und 5 bis 50 mg β-Carotin pro Einheitsdosis enthalten.
- Die erfindungsgemäßen Verbindungen in Form eines Pulvers werden durch aufeinanderfolgendes Vermischen der verschiedenen Bestandteile in trockenem Zustand in einem Pulvermischer hergestellt.
- Das Pulver kann anschließend unter Anwendung an sich bekannter Verfahrensweisen in Tabletten umgeformt oder in Gelkapseln, Kapseln oder Sachets für die Einheitsdosis abgefüllt werden.
- Das folgende Beispiel dient der weiteren Erläuterung der Erfindung:
- DL-Lysin-acetylsalicylat 320 mg
- DL-α-Tocopherol-acetat 250 IE
- β-Carotin 25 mg
- Glycin 35,5 mg
- Aromastoffe 30 mg
- für ein Sachet
- Die tägliche Dosierung beträgt 54 bis 540 mg DL-Lysin-acetylsalicylat, das heißt ein Äquivalent von 30 bis 300 mg Acetylsalicylsäure, 10 bis 1000 IE Vitamin E und 5 bis 50 mg β-Carotin.
Claims (5)
1. Pharmazeutische Zubereitung, dadurch gekennzeichnet, daß sie aus einer
Mischung aus D,L-Lysin-acetylsalicylat, racemischem oder rechtsdrehendem
α-Tocopherol in Form der Base oder eines Salzes und β-Carotin in Kombination mit
geeigneten Trägermaterialien gebildet ist.
2. Pharmazeutische Zubereitung nach Anspruch 1, dadurch gekennzeichnet,
daß a-Tocopherol in Form des Acetats, Palmitats oder Succinats eingesetzt wird.
3. Pharmazeutische Zubereitung nach einem der Ansprüche 1 oder 2, dadurch
gekennzeichnet, daß sie 54 bis 540 mg D,L-Lysin-acetylsalicylat, 10 bis 10000 IE
Vitamin E und 5 bis 50 mg β-Carotin pro Einheitsdosis enthält.
4. Pharmazeutische Zubereitung nach einem der Ansprüche 1 oder 3, dadurch
gekennzeichnet, daß sie in Form eines Pulvers, von Tabletten, Gelkapseln oder
Kapseln vorliegt.
5. Pharmazeutische Zubereitung nach Anspruch 4, dadurch gekennzeichnet,
daß sie in Form eines Pulvers für trinkbare Lösungen in einem Einheitsdosis-Sachet
vorliegt.
