Description
- Die vorliegende Erfindung betrifft Pyrido[2,3- d]pyrimidinderivate, und insbesondere Derivate von N-[2- (5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-yl)alkyl]benzoyl- L-glutaminsäure, die antineoplastische Mittel sind, und ihre Herstellung und Verwendung.
- Die Folsäure-Antimetaboliten Aminopterin und Amethopterin (auch als 10-Methylaminopterin oder Methotrexat bekannt) sind antineoplastische Mittel. Diese Verbindungen inhibieren enzymatische Umwandlungen, in denen Stoffwechselderivate von Folsäure involviert sind. Amethopterin inhibiert z.B. Dihydrofolatreductase, ein Enzym, das zur Regenerierung von Tetrahydrofolat aus Dihydrofolat notwendig ist, das während der Umwandlung von 2-Deoxyuridylat in Thymidylat durch das Enzym Thymidylatsynthetase gebildet wird.
- Es wurden andere Derivate von Folsäure und Aminopterin synthetisiert und als Antimetaboliten getestet. Unter diesen sind verschiedene "Deaza"-Verbindungen, in denen eine Methylen- oder Methyliden-Gruppe eine Stellung im Molekül einnimmt, die normalerweise durch eine Imino- oder Nitrilgruppe besetzt ist. Diese Derivate besitzen verschiedene Grade an antimetabolischer Aktivität. 10-Deaza-aminopterin ist hochaktiv (Sirotak et al., Cancer Treat. Rep., 1978, 62, 1047), während 10-Deazafolsäure keine signifikante Aktivität zeigt (Struck et al., J. Med. Chem., 1971, 14, 693). 5- Deazafolsäure ist nur schwach cytotoxisch, während 5- Deazaaminopterin eine Aktivität besitzt, die der von Amethopterin ähnlich ist (Taylor et al., J. Org. Chem, 1983, 48, 4852). 5,6,7,8-Tetrahydro-5,10-dideazaaminopterin ist ebenfalls aktiv (US-Patent 4,684,653). 8,10-Dideazafolsäure ist als Dihydrofolatreductase-Inhibitor nur geringfügig wirksam (De Graw et al., "Chemistry and Biology of Pteridines", Elsevier, 1979, 229), während 5,8,10-Trideazafolsäure gegen Mäuse-L1210-Leukämie auch nur eine geringfügige Aktivität besitzt (Oatis et al., J. Med. Chem., 1977, 20, 1393). 8,10- Dideazaaminopterin wird als aktiv beschrieben (US-Patent Nr. 4,460,591) und 5,8,10-Trideazaaminopterin zeigt eine Aktivität gegenüber Mäuse-L1210-Leukämie (Yan et al., J. Heterocycl. Cheia., 1979, 16, 541).
- Die europäische Patentveröffentlichung Nr. 0248573A2 (the Trustees of Princeton University) beschreibt, daß beide diastereoisomeren Formen von N-(4-[2-(2-amino-4-hydroxy- 5,6,7,8-tetrahydropyrido[2,3-d]-pyrimidin-6- yl)ethyl]benzoyl]-L-glutaminsäure antineoplastische Mittel sind und ein Verfahren zur Trennung dieser diastereoisomeren Formen durch Behandlung ihrer Mischungen mit einer chiralen Säure.
- Gemäß einem Aspekt der vorliegenden Erfindung wird eine Verbindung bereitgestellt, ausgewählt aus:
- (i) Tetrahydropyrido[2,3-d]pyrimidin der Formel:
- oder
- worin R¹ Hydroxy oder Amino ist;
- R² Wasserstoff, Methyl oder Ethyl ist; und
- R³, R&sup4;, R&sup5;, R&sup6; und R&sup7; so ausgewählt sind, daß
- (a) R³
- ist, und jeder der Reste R&sup4;, R&sup5;, R&sup6; und R&sup7; unabhängig voneinander Wasserstoff, Chlor oder Fluor ist; oder
- (b) R&sup4;
- ist, und ein Rest von R³, R&sup5;, R&sup6; und R&sup7; Chlor oder Fluor ist; und
- die verbleibenden Reste R³, R&sup5;, R&sup6; und R&sup7; Wasserstoff, Chlor oder Fluor sind;
- (ii) den pharmazeutisch annehmbaren Alkalimetall-, Erdalkalimetall-, nichttoxischen Metall-, Ammonium- und substituierten Ammoniumsalzen davon; und
- (iii) diastereoisomeren Mischungen des Tetrahydropyrido[2,3-d]pyrimidins oder von dessen Salzen.
- Die erfindungsgemäßen Verbindungen sind Derivate des Pyrido[2,3-d]pyrimidin-heterocyclischen Ringes, der wie folgt numeriert wird:
- Die Verbindungen der Formeln IA und IB existieren in einem tautomeren Gleichgewicht mit den entsprechenden 4-Oxo- und 4-Imino-Verbindungen:
- Aus Zweckmäßigkeitsgründen werden die 3,4-Dehydro-4- hydroxy--
- und 3,4-Dehydro-4-aminoformen ausgewählt und in der Beschreibung die entsprechende Nomenklatur verwendet, wobei es aber selbstverständlich ist, daß in jedem Fall auch die tautomeren 4(3H)-Oxo- und Imino-Formen umfaßt werden.
- In einem Aspekt der vorliegenden Erfindung kann R¹ Hydroxy sein, und R² Wasserstoff. Die erfindungsgemäße Verbindung kann die Formel besitzen:
- worin R&sup4;, R&sup5;, R&sup6; und R&sup7; Wasserstoff, Chlor und Fluor sein kann, und die verbleibenden Reste R&sup4;, R&sup5;, R&sup6; und R&sup7; jeweils Wasserstoff sein können.
- Die erfindungsgemäße Verbindung kann die Formel besitzen:
- worin R³, R&sup5;, R&sup6; und R&sup7; Chlor oder Fluor sein können, und die verbleibenden Reste R³, R&sup5;, R&sup6; und R&sup7; Wasserstoff sein können.
- In einem weiteren erfindungsgemäßen Aspekt können die Verbindungen hergestellt werden, indem man eine Verbindung der Formel:
- worin X Brom oder Jod ist;
- R3'
- ist, einer der Reste R4', R&sup5;, R&sup6; und R&sup7; Wasserstoff, Chlor oder Fluor ist; oder
- R4'
- ist, und einer der Reste R3', R&sup5; und R&sup6; Chlor oder Fluor ist;
- ein zweiter der verbleibenden Reste R3', R&sup5; und R&sup6; Wasserstoff, Chlor oder Fluor ist; und
- der verbleibende Rest von R3', R&sup5; und R&sup6; Wasserstoff ist; und
- jeder der Reste R&sup8; und R&sup9; eine Carbonsäureschutzgruppe ist, mit einer Verbindung der Formel:
- worin R¹&sup0; Wasserstoff oder eine Aminoschutzgruppe ist;
- Z unabhängig von R² Wasserstoff ist; und
- R² unabhängig von Z Wasserstoff, Methyl oder Ethyl ist; oder
- Z und R² zusammen eine Kohlenstoff-Kohlenstoff-Bindung bedeuten;
- in Gegenwart eines Palladiumkomplexes reagieren läßt, um eine Verbindung der Formel:
- zu ergeben, worin R¹, R², R3', R4', R&sup5;, R&sup6;, R&sup7;, R¹&sup0; und Z die hier angegebene Bedeutung besitzen.
- In einem anderen erfindungsgemäßen Aspekt wird eine Verbindung bereitgestellt, ausgewählt aus
- (i) einem Tetrahydropyrido[2,3-d]pyrimidin der Formel:
- worin R¹ Hydroxy oder Amino ist;
- R² Wasserstoff, Methyl oder Ethyl ist; und
- R3', R4', R&sup5;, R&sup6; und R&sup7; so ausgewählt sind, daß
- (a) R3'
- ist, und jeder der Reste R4', R&sup5;, R&sup6; und R&sup7; unabhängig voneinander Wasserstoff, Chlor oder Fluor ist; oder
- (b) R4'
- ist, ein Rest R3', R&sup5;, R&sup6; und R&sup7; Chlor oder Fluor ist; und
- die verbleibenden Reste R3', R&sup5;, R&sup6; und R&sup7; Wasserstoff, Chlor oder Fluor sind;
- jeder der Reste R&sup8; und R&sup9; eine Carbonsäureschutzgruppe ist; und
- R¹&sup0; Wasserstoff oder eine Aminoschutzgruppe ist; und
- (ii) diastereoisomeren Mischungen dieser Tetrahydropyrido[2,3-d]pyrimidine.
- Die Palladiumkomplexe sind solche, wie sie früher bei der Umsetzung von Arylhalogeniden und Allylalkoholen verwendet wurden, wie dies z.B. beschrieben wird von Melpoler et al., J. Org., Chem., 41, Nr. 2, 1976, 265; Chalk et al., J. Org., Chem., 41, Nr. 7, 1976, 1206; Arai et al., J. Insbesondere brauchbar sind die Palladium/-trisubstitutierten Phosphin-Komplexe von Sakamoto, Synthesis, 1983, 312; z.B. ein trisubstituiertes Phosphin wie ein Triarylphosphin, z.B. Triphenylphosphin, oder ein Trialkylphosphin; ein Palladiumsalz, wie z.B. Palladiumacetat oder ein Palladiumhalogenid, wie Palladiumchlorid; und ein Kupfer(II)-halogenid, wie z.B. Kupfer(II)-jodid.
- Vorzugsweise wird die Umsetzung in Gegenwart von mindestens einem molaren Äquivalent eines sekundären oder tertiären Amins durchgeführt, das als Säureakzeptor dient, wie z.B. Triethylamin oder Diethylamin, und unter einer inerten Atmosphäre, gegebenenfalls in Gegenwart eines inerten polaren Lösungsmittels, wie z.B. Acetonitril, Dinethylformamid, N-Methylpyrrolidon und dergleichen. Besonders bevorzugt ist die Verwendung von Acetonitril, das nicht nur für die Reaktionsteilnehmer als Lösungsmittel dient, sondern auch für das aus dem Säureakzeptor und der gebildeten Säure ausgebildete Salz. Im allgemeinen sind schwach erhöhte Temperaturen, wie z.B. von ca. 75ºC bis 125ºC, und vorzugsweise bei oder unter 100ºC, vorteilhaft.
- Die hier erwähnten Amino- und Carbonsäureschutzgruppen sind solche, wie sie üblicherweise verwendet werden, und wie sie z.B. von Greene in "Protective Groups in Organic Synthesis", John Wiley & Sons, Inc., 1981, und McOmie in "Protective Groups in Organic Chemistry", Plenum Press, 1983, beschrieben werden. Insbesonders bevorzugte R¹&sup0;-Schutzgruppen sind Alkanoylgruppen, wie z.B. Acetyl, Propionyl, Pivaloyl und dergleichen.
- Die katalytische Hydrierung einer Verbindung der Formel IV ergibt das entsprechende 2-Amino (oder 2-geschütztes Amino)-6-substituierte-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin der Formel:
- Die Verbindungen der Formel V werden dann einer Hydrolyse unterworfen, um die Schutzgruppen R&sup8;, R&sup9; und R¹&sup0; zu entfernen. Dies wird bei Normaltemperaturen unter Verwendung von wäßriger Säure oder Base, wie z.B. einem wäßrigen Alkalimetallhydroxid, gegebenenfalls in Gegenwart eines mit Wasser mischbaren organischen Lösungsmittels, wie z.B. Methanol, Ethanol, Tetrahydrofuran, Dimethylformamid und dergleichen, oder einer Säure, wie z.B. Trifluoressigsäure, durchgeführt. Wenn eine Base verwendet wird, wird die kationische Gruppe des Salzes freigesetzt und das Produkt als dikationisches Glutamatsalz gebildet, das leicht durch pH-Einstellung ausgefällt werden kann, wie durch Ansäuerung mit z.B. Essigsäure. Die resultierenden Produkte sind im allgemeinen hochschmelzende Krystalline oder microkrystalline Feststoffe.
- Die absolute Konfiguration am chiralen Kohlenstoffatom in der Glutaminsäurekette in R³ oder R&sup4; in den Formeln IA und IB ist (S) oder L, was die gleiche absolute Konfiguration ist wie die am korrespondierenden alpha-Kohlenstoffatom im L- Alanin. Zusätzlich stellt das Kohlenstoffatom in 6-Stellung des 5,6,7,8-Tetrahydropyrido[2,3-di]pyrimidin-Rings ein chirales Zentrum dar, was die beiden (R, S)- und (S, S)Diastereomeren, die durch die Formeln IA und IB dargestellt werden, ergibt. Die Mischung der Diastereomeren kann therapeutisch verwendet werden, wobei beide als Substrate für relevante Folatenzyme dienen. Die Diastereomeren können auch so getrennt werden, daß sie in einer Form vorliegen, die im wesentlichen frei ist von der anderen, d.h. in einer Form mit einer optischen Reinheit von > 95 %.
- Die Diastereomeren können mechanisch getrennt werden, wie z.B. durch Chromatographie, oder eine Mischung der Diastereomeren kann mit einer chiralen Säure, die damit ein Salz bildet, behandelt werden. Die resultierenden diastereoisomeren Salze werden dann über eine oder mehrere fraktionierte Kristallisationen getrennt, und danach die freie Base der kationischen Gruppe von mindestens einem der getrennten Salze durch Behandlung mit einer Base und Entfernung der Schutzgruppen freigesetzt. Die Freisetzung des Kations aus dem Salz kann durch eine getrennte Stufe vor oder nach der Entfernung der Schutzgruppen R&sup8;, R&sup9; und R¹&sup0; durchgeführt werden, oder gleichzeitig mit der Entfernung solcher Gruppen unter basischer Hydrolyse.
- Geeignete chirale Säuren umfassen die individuellen Enantiomeren von 10-Camphersulfonsäure, Camphersäure, alpha- Bromcamphersäure, Menthoxyessigsäure, Weinsäure, Diacetylweinsäure, Äpfelsäure, Pyrrolidin-5-carbonsäure und dergleichen.
- Die Ausgangsmaterialien der Formel III können hergestellt werden aus dem entsprechenden 6-Halogenpyrido[2,3- d]pyrimidin auf die in der gleichzeitig anhängigen US-Anmeldung Serial No. 922,511 beschriebene Weise. Die hydrierten Ausgangsmaterialien der Formel II können durch Kupplung der geeigneten Halogenbenzoesäure und eines geschützten Derivats von L-Glutaminsäure auf die in der PCT-Anmeldung WO 86/05181 beschriebenen Weise hergestellt werden. Die Kupplungsreaktion verwendet übliche Kondensationsverfahren zur Ausbildung von Peptidbindungen, wie z.B. die Aktivierung der Carbonsäuregruppe durch Bildung eines gemischten Anhydrids, Behandlung mit DCC oder die Verwendung von Diphenylchlorphosphonat.
- Erfindungsgemäße repräsentative Verbindungen umfassen:
- A. N-(3-[2-(2-Amino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3- d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutaminsäure.

- Die resultierenden Verbindungen der Formeln IA und IB einschließlich ihrer Mischungen und ihrer pharmazeutisch annehmbaren Alkalimetall-, Erdalkalimetall-, nichttoxischen Metall-, Ammonium- und substituierten Ammoniumsalze, wie z.B. des Natrium-, Kalium-, Lithium-, Calcium-, Magnesium-, Aluminium-, Zink-, Ammonium-, Trimethylammonium-, Triethylammonium-, Triethanolammonium-, Pyridinium-, substituiertes Pyridinium-Salzes und dergleichen, wirken auf eine oder mehrere Enzyme, die Folsäure und insbesondere Stoffwechselderivate von Folsäure als Substrat verwenden. Die Verbindungen können allein oder in Kombination verwendet werden zur Behandlung von Neoplasmen einschließlich Choriokarzinom, Leukämie, Adenokarzinom der weiblichen Brust, von Hautkrebsen des Kopfes und des Nackens, squamösem oder kleinzelligen Lungenkrebs und verschiedenen Lymphsarkomen. Die Verbindungen können auch zur Behandlung von Pilzerkrankungen, Psoriasis und anderen Autoimmunerscheinungen, wie z.B. rheumatoider Arthritis, die auf Methotrexat ansprechen, verwendet werden.
- Die Verbindungen können entweder oral oder vorzugsweise parenteral, allein oder in Kombination mit anderen antineoplastischen Mitteln, Steroiden usw. einem Säugetier, das unter einem Neoplasma leidet und eine Behandlung benötigt, verabreicht werden. Parenterale Verabreichungswege umfassen eine intramuskuläre, intrathecale, intravenöse oder intraarterielle Verabreichung.
- Gemäß einem anderen erfindungsgemäßen Aspekt wird eine pharmazeutische Zusammensetzung zur Bekämpfung von neoplastischem Wachstum in Säugern bereitgestellt, die eine erfindungsgemäße Verbindung in einer Menge, die bei Verabreichung an den Säuger in einer Einzel- oder Mehrfachdosis zur Bekämpfung dieses Wachstums wirksam ist, in Kombination mit einem pharmazeutisch annehmbaren Träger enthält.
- Das Dosierungsschema kann sich nach dem besonderen Neoplasma, dem Zustand des Patienten und seinem Ansprechen richten, aber im allgemeinen beträgt die Dosis von ca. 10 bis ca. 100 mg/Tag während 5-10 Tagen, oder eine einzelne tägliche Dosis von 250-500 mg, die periodisch, z.B. alle 14 Tage, wiederholt wird. Orale Dosierungsformen können Tabletten und Kapseln, die 1-10 mg des Arzneimittels pro Dosiseinheit enthalten, umfassen. Isotonische Kochsalzlösungen, die 20-100 mg/ml enthalten, können für eine parenterale Verabreichung verwendet werden.
- Repräsentative IC&sub5;&sub0;-Werte, die in einer Ganzzellenmenschlichen Leukämiezellinie (CCRF-CEM) bestimmt wurden, sind die folgenden: Verbindung
- Mäusen, die mit C3H-Brustadenokarzinom implantiert waren, wurden intraperitoneal fünf Tage lang mit der Verbindung H behandelt. Bei einer Dosis von 30 mg/kg wurde eine 72%ige Inhibierung beobachtet. Bei einer Dosis von 100mg/kg wurde ein 80%ige Inhibierung beobachtet. Bei diesen Dosierungen wurde keine Toxizität festgestellt.
- Die folgenden Inhibitionswerte wurden mit der Verbindung H in Mäusen mit 6C3HED-Lymphsarkom beobachtet: 6C3HED-LYMPHSARKOM DOSIS (MK/KG) % INHIBIERUNG TOXISCH/TOTAL
- Die nachfolgende Beschreibung zur Duchführung der vorliegenden Erfindung ist nur eine beispielhafte.
- (In den NMR-Daten bedeutet "s" Singlett, "d" Doublett, und "t" Triplett, "q" Quartett, und "m" Multiplett.)
- Eine Mischung aus 0,016 g (0,093 mmol) Palladiumchlorid und 0,0486 g (0,185 mmol) Triphenylphosphin in 5 ml Acetonitril wird unter Stickstoff bei Raumtemperatur 30 Minuten lang gerührt. Zu dieser Mischung werden zusätzliche 10 ml Acetonitril und 0,80 g (1,89 mmol) Dimethyl N-(2-fluor-5-jodbenzoyl)-L-glutamat, 0,50 g (1,85 mmol) 2-Pivaloylamino-4- hydroxy-6-ethynylpyrido[2,3-d]pyrimidin, 0,0088 g (0,046 mmol) Kupferjodid und 0,54 ml (3,89 mmol) Triethylamin zugegeben. Die Mischung wird dann unter Rückfluß 3 Stunden lang erhitzt. Nach Abkühlen auf Raumtemperatur wird das Lösungsmittel im Vakuum entfernt und der Rückstand einer Flash-Chromatographie an Silica-Gel unter Verwendung von 1:19/Methanol:Chloroform als Eluens unterworfen und ergibt 0,92 g (88 %) Dimethyl N-(2-fluor-5-[2-(2-pivaloylamino-4- hydroxypyrido[2,3-d]pyrimidin-6-yl)ethynyl]benzoyl)-L-glutamat als dunkelgelben Feststoff, Schmelzpunkt 189-191ºC (Zersetzung); Rf = 0,48 (1:19/Methanol:Chloroform), Mass (FD) = 565; IR (KBr, cm&supmin;¹) = 1146, 1228, 1263, 1440, 1488, 1549, 1596, 1607, 1622, 1673. 1742; UV (EtOH) λmax = 319 (epsilon = 30600), 259 (epsilon = 11200), 212 (epsilon = 42100); ¹NMR (300 MHz, CDCl&sub3;) delta 1,38 (s, 9H), 2,19 (m, 1H), 2,3-2,6 (m, 3H), 3,72 (s, 3H), 3,85 (s, 3H), 4,95 (m, 1H), 7,21 (dd, 1H, J = 8,5, 11,2 Hz), 7,45 (m, 1H), 7,68 (m, 1H), 8,26 (dd, 1H, J = 2,0, 7,2 Hz), 8,37 (s, 1H), 8,63 (d, 1H, J = 2,2 Hz), 9,0 (d, 1H, J = 2,0 Hz).
- Eine Probe von 0,115 g (0,202 mmol) Dimethyl N-(2-fluor- 5-[2-(2-pivaloylamino-4-hydroxypyrido[2,3-d]pyrimidin-6- yl)ethynyl]benzoyl)-L-glutamat und 0,045 g Platinoxid werden in 5 ml Eisessig gelöst. Diese Mischung wird dann unter atmosphärischem Wasserstoff bei Raumtemperatur 5 Stunden lang hydriert. Der Katalysator wird durch Filtration entfernt, und das Filtrat im Vakuum einkonzentriert, und der rohe Feststoff einer Flash-Chromatographie an Silica-Gel unter Verwendung von 1:19/Methanol:Chloroform als Eluens unterworfen, und ergibt 0,76 g (66 %) Dimethyl N-(2-fluor-5-[2-(2-pivaloylamino- 4-hydroxy-5,6,7,8-tetrahydropyrido-[2,3-d]pyrimidin-6- yl)ethyl]benzoyl)-L-glutamat als grauweißen Feststoff. Schmelzpunkt 165-170ºC (Zersetzung); Rf = 0,33 (1:19/Methanol:Chloroform), Masse (FD) = 573; IR (KBr, cm&supmin;¹) = 1161, 1205, 1223, 1232, 1439, 1460, 1478, 1492, 1527, 1536, 1556, 1573, 1619, 1645, 1743, 3392, 3395; UV (EtOH) λmax = 271 (epsilon = 10200), 234 (epsilon = 42900).
- Analyse berechnet für C&sub2;&sub8;H&sub3;&sub6;N&sub5;O&sub7;F: C, 58,63; H, 6,33; N, 12,21.

- Eine Probe von 0,024 g (0,042 mmol) Dimethyl N-(2-fluor- 5-[2-(2-pivaloylamino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3- d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutamat wird in 0,5 ml 1N Natriumhydroxid gelöst und bei Raumtemperatur 5 Tage lang gerührt. Die schwach gelbe Lösung wird dann mit Eisessig angesäuert und das Lösungsmittel im Vakuum entfernt. Der Rückstand wird mit Wasser verrieben und der Feststoff durch Saugfiltration gesammelt, mit einer kleinen Menge Wasser gewaschen und im Vakuum bei 100ºC getrocknet, und ergibt 0,04 g (73 %) N-(2-Fluor-5-[2-(2-amino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutaminsäure als weißen Feststoff. Schmelzpunkt 252-262ºC (Zersetzung); Rf = 0,04 (1:19/Methanol:Chloroform), Masse (FAB) = 462; IR (KBr, cm&supmin;¹) = 1230, 1248, 1263, 1307, 1349, 1379, 1398, 1462, 1442, 1544, 1616, 1650, 1705, 2928; UV (EtOH) λmax = 278 (epsilon = 13300), 221 (epsilon = 29800); ¹NMR (300 MHz, Me&sub2;SO-d6) delta 1,57 (brs, 3H), 1,70-2,10 (m, 4H), 2,34 (t, 2H J = 5,0 Hz), 2,60-2,85 (m, 3H), 3,20 (m, 1H), 4,37 (m, 1H), 5,92 (s, 1H), 6,26 (s, 1H), 7,19 (t, 1H, J = 9,2 Hz), (brs 1H), 7,45 (d, 1H, J = 6,0 Hz), 8,43 (d, 1H, J = 6,3 Hz), 9,70 (brs, 1H).
- Durch Substitution einer im wesentlichen molaren äquivalenten Menge von Dimethyl N-(3-jodbenzoyl)-L-glutamat für Dimethyl N-(2-fluor-5-jodbenzyol)-L-glutamat im Verfahren des Beispiels 1 wird Dimethyl N-(3-[2-(2-pivaloylamino-4-hydroxypyrido[2,3-d]pyrimidin-6-yl)ethynyl]benzoyl)-L-glutamat, Schmelzpunkt 161-166ºC.
- Analyse berechnet für C&sub2;&sub8;H&sub2;&sub9;N&sub5;O&sub7;: C, 61,42; H, 5,38; N, 12,77.
- Gefunden: C, 61,34; H, 5,23; N, 12,67.
- Bei Hydrierung dieses Materials auf im wesentlichen die gleiche wie in Beispiel 2 beschriebene Weise wird Dimethyl N- (3-[2-(2-pivaloylamino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutamat, Schmelzpunkt 141-148ºC, erhalten.
- Analyse berechnet für C&sub2;&sub8;H&sub3;&sub7;N&sub5;O&sub7;: C, 60,53; H, 6,71; N, 12,60-
- Gefunden: C, 60,26; H, 6,74; N, 12,37.
- Die wie in Beispiel 3 beschriebene Hydrolyse ergibt dann N-(3-[2-(2-amino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3- d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutaminsäure, Schmelzpunkt 258-266ºC; [a] D25 = + 11,823.
- Eine Mischung aus 0,44 g (2,56 mmol) Palladiumchlorid und 1,34 g (5,12 mmol) Triphenylphosphin in 200 ml Acetonitril wird unter Stickstoff bei Raumtemperatur 30 Minuten lang gerührt. Zu dieser Mischung werden zusätzlich 150 ml Acetonitril, 22,09 g (52,2 mmol) Dimethyl N-(2-fluor-4-jodbenzoyl)- L-glutamat, 13,830 g (51,2 mmol) 2-Pivaloylamino-4-hydroxy-6- ethynylpyrido[2,3-d]pyrimidin, 0,0244 g (1,28 mmol) Kupferjodid und 15 ml (107,5 mmol) Triethylamin zugegeben. Die Reaktionsmischung wird dann unter Rückfluß 3 Stunden lang erhitzt und dann auf Raumtemperatur abgekühlt, und das Lösungsmittel dann im Vakuum entfernt und der rohe Feststoff an Silica-Gel mit 1:19/Methanol: Methylenchlorid chromatographiert und ergibt 18,1 g (62,5 %) Dimethyl N-(2-fluor-4-[2-(2- pivaloylainmo-4-hydroxypyrido[2,3-d]pyrimidin-6- yl)ethynyl]benzoyl)-L-glutamat als weißen Feststoff, Schmelzpunkt 204-207ºC (Zersetzung); Rf = 0,60 (1:19/Methanol:Chloroform), Masse (FD) = 565; IR (KBr, cm&supmin;¹) = 813, 1151, 1201, 1270, 1375, 1444, 1481, 1490, 1527, 1548, 1598, 1623, 1649, 1670, 1741, 1753; UV (EtOH) λmax = 325 (epsilon = 41100), 266 (epsilon = 14600), 210 (epsilon = 35700).

- Sechs Gramm (0,0106 mmol) Dimethyl N-(2-fluor-4-[2-(2- pivaloylamino-4-hydroxypyrido[2,3-d]pyrimidin-6- yl)ethynyl]benzoyl)-L-glutamat und 21 g Platinoxid werden in 480 ml Eisessig gelöst und bei 60 psi 2 Stunden lang hydriert. Der Katalysator wird durch Filtration entfernt, das Filtrat im Vakuum einkonzentriert und der Rückstand an Silica-Gel chromatographiert und ergibt 4,57 g (75 %) Dimethyl N-(2-fluor-4-[2-(2-pivaloylamino-4-hydroxy-5,6,7,8- tetrahydropyrido[2,3-d]pyrimidin-6-yl)ethy]benzoyl)-L-glutamat als weißen Feststoff, Schmelzpunkt 183-191ºC (Zersetzung); Rf = 0,21 (1:19/Methanol:Chloroform), Masse (FD) = 573; IR (KBr, cm&supmin;¹) = 1460, 1572, 1577, 1624, 1645, 1738; UV (EtOH) λmax = 270 (epsilon = 11100), 235 (epsilon = 49500).
- Analyse berechnet für C&sub2;&sub8;H&sub3;&sub6;N&sub5;O&sub7;F: C, 58,63; H, 6,33; N, 12,21.

- Eine Lösung von 0,50 g (0,872 mmol) Dimethyl N-(2-fluor- 4-[2-(2-pivaloylamino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3- d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutamat in 7,5 ml 1N Natriumhydroxid wird bei Raumtemperatur 66 Stunden lang gerührt. Die Lösung wird mit Eisessig angesäuert, das Lösungsmittel im Vakuum entfernt und der Feststoff mit 10 ml kaltem Wasser verrieben. Der Feststoff wird durch Filtration gesammelt, mit kaltem Wasser, Ether und 10 ml Ethanol gewaschen und ergibt 0,376 g (93 %) N-(2-Fluor-4-[2-(2-amino-4-hydroxy- 5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-yl)ethyl]benzoyl)- L-glutaminsäure als blaßgelben Feststoff, Schmelzpunkt 199- 209ºC, nach Trocknen im Vakuum bei 100ºC; Rf = 0,04 (1:1/Methanol:Chloroforin), Masse (FAB) = 462; IR (KBr, cm&supmin;¹) = 1315, 1389, 1401, 1495, 1542, 1545, 1555, 1565, 1584, 1627, 1647, 1718, 1721, 1724, 3290, 3306, 3310; UV (EtOH) λmax = 278 (epsilon = 13700), 223 (epsilon = 30900); ¹HNMR (300 MHz, Me&sub2;SO-d6) delta 1,57 (brs, 3H), 1,75-1,95 (m, 2H), 2,07 (m, 1H), 2,34 (t, 2H J = 7,3 Hz), 2,6-2,9 (m, 3H), 3,20 (m, 2H), 4,39 (m, 1H), 5,92 (s, 2H), 6,26 (s, 1H), 7,15 (dd, 2H, J = 6,5, 10,1 Hz), 8,40 (d, 1H, J = 7,0 Hz), 9,68 (brs, 1H).
- Durch Substitution einer im wesentlichen molaren äquivalenten Menge von Dimethyl N-(2-chlor-4-brombenzoyl)-L- glutaminsäure für Dimethyl N-(2-fluor-4-jodbenzoyl)-L-glutamat des Beispiels 5 wird Dimethyl N-(2-chlor-4-[2-(2-pivaloylamino-4-hydroxypyrido[2,3-d]pyrimidin-6- yl)ethynyl]benzoyl)-L-glutamat, Schmelzpunkt 150-156ºC (Zersetzung) erhalten; Rf = 0,35; Masse (FD) = 581; IR (KBr, cm&supmin;¹) = 810, 1147, 1210, 1223, 1245, 1375, 1440, 1446, 1491, 1548, 1593, 1624, 1680, 1741; UV (EtOH) λmax = 323 (epsilon = 33200), 264 (epsilon = 12400); ¹HNMR (300 MHz, CDCl3) delta 1,39 (s, 9H), 2,33 (m, 1H), 2,39-2,60 (m, 3H), 3,72 (s, 3H, 4,94 (m, 1H), 7,25 (d, 1H, J = 10 Hz), 7,53 (dd, 1H, J = 1,0, 8,0 Hz), 7,63 (s, 1H), 7,72 (d, 1H, J = 7,94), 8,45 (s, 1H), 8,64 (d, 1H, J = 2,2 Hz), 8,89 (d, 1H, J = 2,0 Hz).
- Nach Hydrierung des Produktes wie in Beispiel 6 beschrieben wird Dimethyl N-(2-chlor-4-[2-(2-pivaloylamino-4- hydroxy-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6- yl)ethyl]benzoyl)-L-glutamat vom Schmelzpunkt 165-170ºC (Zersetzung) erhalten; Rf = 0,33 (1:19/Methanol:Chloroform), Masse (FD) = 573; IR (KBr, cm&supmin;¹) = 1161, 1205, 1223, 1232, 1439, 1460, 1478, 1492, 1527, 1536, 1556, 1573, 1619, 1645, 1743, 3392, 3395; UV (EtOH) λmax = 271 (epsilon = 10200), 234 (epsilon = 42900).
- Analyse berechnet für C&sub2;&sub8;H&sub3;&sub6;N&sub5;O&sub7;F: C, 58,63; H, 6,33; N, 12,21.
- Gefunden: C, 58,59; H, 6,38; N, 12,14.
- ¹NMR (300 NHz, CDCl&sub3;) delta 1,32 (s, 9H), 1,73 (m, 2H), 1,89 (m, 1H), 2,20 (m, 2H), 2,30-2,60 (m, 3H), 2,65-2,90 (m, 3H), 3,03 (m, 1H), 3,39 (m, 1H), 3,70 (s, 3H), 3,83 (s, 3H), 4,66 (s, 1H), 4,91 (m, 1H), 7,09 (dd, 1H, J = 8,4, 11,7 Hz), 7,36 (m, 2H), 7,75 (s, 1H), 7,91 (dd, 1H, J = 2,2, 7,4 Hz).
- Hydrolyse der letztgenannten Verbindung auf im wesentlichen die im Beispiel 1 beschriebene Weise ergibt dann N-(2- Chlor-4-[2-(2-amino-4-hydroxy-5,6,7,8-tetrahydropyrido[2,3- d]pyrimidin-6-yl)ethyl]benzoyl)-L-glutaminsäure, Schmelzpunkt 204-211ºC (Zersetzung); Rf = 0,04 (1:19/Methanol:Chloroform), Masse (FAB) = 478; IR (KBr, cm&supmin;¹) = 1263, 1311, 1351, 1357, 1396, 1455, 1486, 1555, 1609, 1650, 1723, 2925, 2929; UV (EtOH) λmax = 278 (epsilon = 8000), 219 (epsilon = 20400); ¹HNMR (300 MHz, Me&sub2;SO-d6) delta 1,56 (brs, 3H), 1,85 (m, 2H), 2,05 (m, 1H), 2,36 (t, 2H J = 72, Hz), 2,66-2,85 (m, 3H), 3,17 (m, 2H), 4,37 (m, 1H), 5,92 (s, 2H), 6,26 (s, 1H), 7,20- 7,40 (m, 3H), 8,63 (d, 1H, J = 7,8 Hz), 9,70 (brs 1H).
- Die in den vorstehenden Beispielen verwendeten Glutamatderivate können wie folgt hergestellt werden:
- Zu 20 ml Benzol werden 2,0 g (0,00752 mol) 2-Fluor-4- jodbenzosäüre zugegeben, und danach 2,3 ml (320 mmol) Thionylchlorid und einige Tropfen Dimethylformamid. Die Mischung wird 3,5 Stunden lang am Rückfluß erhitzt. Das Lösungsmittel wird dann im Vakuum entfernt und der Rückstand trockengesaugt. Das resultierende Säurechlorid wird wieder in 10 ml Methylenchlorid gelöst und tropfenweise zu einer gekühlten Mischung von 1,67 g (7,89 mmol) L-Glutaminsäuredimethylesterhydrochlorid, 5 mg 4-Dimethylaminopyridin und 2,2 ml (15,8 mmol) Triethylamin in 30 ml Methylenchlorid tropfenweise zugegeben. Nach der Zugabe wird die Reaktionsmischung bei Raumtemperatur 18 Stunden lang gerührt. Die Reaktionsmischung wird dann mit zusätzlichem Methylenchlorid verdünnt, mit 1N Chlorwasserstoffsäure, Wasser und 5 % Natriunbicarbonat gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird im Vakuum entfernt und es werden 2,34 g (74 %) Dimethyl N-(2-fluor-4-jodbenzoyl)-L-glutamat als weißer Feststoff erhalten, Schmelzpunkt 94-96ºC; IR (KBr, cm&supmin;¹) = 1159, 1177, 1194, 1208, 1229, 1398, 1437, 1477, 1537, 1601, 1642, 1728, 1739, 1756, 3326.
- Analyse berechnet für C&sub1;&sub4;H&sub1;&sub5;NO&sub5;Fl: C, 39,74; H, 3,57; N, 3,31.
- Gefunden: C, 39,98; H, 3,61; N, 3,27.
- ¹HNMR (300 MHz, CDCl&sub3;) delta 2,15 (m, 1H), 2,33-2,49 (m, 1H), 3,66 (s, 3H), 3,79 (s, 3H), 4,86 (m, 1H), 7,55 (d, 1H, J = 11 Hz), 7,63 (d, 1H, J = 8,0 Hz), 7,76 (t, 1H, J = 8,0 Hz).
- Auf ähnliche Weise werden Dimethyl N-(2-fluor-5-jodbenzoyl)-L-glutamat, Schmelzpunkt 69-72ºC, und Dimethyl N-(2- chlor-4-brombenzoyl)-L-glutamat, Schmelzpunkt 77-80ºC, hergestellt.
- Die Halogenbenzoesäure-Ausgangsmaterialien können ihrerseits gemäß dem folgenden Verfahren hergestellt werden:
- Zu 8,1 g (51,3 mmol) Kaliumpermanganat in 150 ml Wasser werden 5,0 g (21,1 mmol) 2-Fluor-5-jodtoluol zugegeben. Die Reaktionsmischung wird 4 Stunden lang am Rückfluß erhitzt. Nach Abkühlung auf Raumtemperatur wird die Reaktionsmischung in 250 ml Methylenchlorid, Natriumbisulfat und 1N-Chlorwasserstoffsäure eingegossen. Die organische Schicht wird abgetrennt, mit Wasser gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird im Vakuum entfernt und der Feststoff mit Hexan verrieben, und ergibt 0,86 g 15 %) 2-Fluor-5- jodbenzoesäure als weißen Feststoff, Schmelzpunkt 161-163ºC.
- Auf ähnliche Weise wird aus 2-Fluor-4-jodtoluol 2-Fluor- 4-jodbenzoesäure vom Schmelzpunkt 213-215ºC (Zersetzung) hergestellt.
Claims (12)
1. Verbindung ausgewählt aus
(i) einem Tetrahydropyrido[2,3-d]pyrimidin der Formel:
worin R¹ Hydroxy oder Amino ist;
R² Wasserstoff, Methyl oder Ethyl ist; und R³, R&sup4;, R&sup5;,
R&sup6; und R&sup7; so ausgewählt sind, daß
ist und jeder der Reste R&sup4;, R&sup5;, R&sup6; und R&sup7; unabhängig
voneinander Wasserstoff, Chlor oder Fluor ist; oder
(b) R4
ist, einer der Reste R³, R&sup5;, R&sup6; und R&sup7; Chlor oder Fluor ist;
und die übrigen Reste R³, R&sup5;, R&sup6; und R&sup7; Wasserstoff, Chlor
oder Fluor sind;
(ii) den pharmazeutisch annehmbaren Alkalimetall-,
Erdalkalimetall-, nicht-toxischen Metall-, Ammonium- und
substituierten Ammonium-Salzen davon; und
(iii) diastereoisomeren Mischungen des Tetrahydropyrido[2,3-
d]pyrimidins oder der Salze.
2. Verbindung nach Anspruch 1, dadurch
gekennzeichnet, daß R¹ Hydroxy und R² Wasserstoff ist.
3. Verbindung nacch Anspruch 1 oder Anspruch 2,
dadurch gekennzeichnet, daß das Tetrahydropyrido[2,3-
d]pyrimidin die Formel besitzt
4. Verbindung nach Anspruch 3, dadurch
gekennzeichnet, daß einer der Reste R&sup4;, R&sup5;, R&sup6; und R&sup7;
Wasserstoff, Chlor oder Fluor ist, und die übrigen Reste R&sup4;,
R&sup5;, R&sup6; und R&sup7; jeweils Wasserstoff bedeuten.
5. Verbindung nach Anspruch 3, dadurch
gekennzeichnet, daß sie N-(3-[2-(2-Amino-4-hydroxy-5,6,7,8,-
tetrahydropyrido-[2,3-pyrimidin-6-yl)ethyl]benzoyl)-L-
glutaminsäure ist.
6. Verbindung nach Anspruch 3 oder Anspruch 4,
dadurch gekennzeichnet, daß sie N-(2-Fluor-5-[2-(2-amino-4-
hydroxy-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-
yl)ethyl]benzoyl)-L-glutaminsäure ist.
7. Verbindung nach Anspruch 1 oder Anspruch 2,
dadurch gekennzeichnet, daß das Tetrahydropyrido[2,3-
d]pyrimidin eine Verbindung der Formel ist:
8. Verbindung nach Anspruch 7, dadurch
gekennzeichnet, daß einer der Reste R³, R&sup5;, R&sup6; und R&sup7; Chlor
oder Fluor ist, und die übrigen Reste R³, R&sup5;, R&sup6; und R&sup7;
jeweils Wasserstoff bedeuten.
9. Verbindung nach Anspruch 7, dadurch
gekennzeichnet, daß sie N-(2-Chlor-4-[2-(2-amino-4-hydroxy-
5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-yl)ethyl]benzoyl-L-
glutaminsäure ist.
10. Verbindung nach Anspruch 7 oder Anspruch 8,
dadurch gekennzeichnet, daß sie N-(2-Fluor-4-[2-(2-amino-4-
hydroxy-5,6,7,8-tetrahydropyrido[2,3-d]pyrimidin-6-
yl)ethyl]benzoyl)-L-glutaminsäure ist.
11. Pharmazeutische Zusammensetzung zur Bekämpfung
von neoplastischem Wachstum in einem Säuger, dadurch
gekennzeichnet, daß sie eine Verbindung nach einem der
vorhergehenden Ansprüche in einer Menge besitzt, die bei
Verabreichung an den Säuger nach einem Einzel- oder
Mehrfachdosis-Plan zur Bekämpfung dieses Wachstums wirksam
ist, in Kombination mit einem pharmazeutisch annehmbaren
Träger.
12. Verbindung ausgewählt aus
(i) Tetrahydropyrido[2,3-d]pyrimidin der Formel:
worin R¹ Hydroxy oder Amino ist;
R² Wasserstoff, Methyl oder Ethyl ist; und
R3', R4', R&sup5;, R&sup6; und R&sup7; so ausgewählt sind, daß
(a) R3'
ist und jeder der Reste R4', R&sup5;, R&sup6; und R&sup7; unabhängig von
einander Wasserstoff, Chlor oder Fluor ist; oder
(b) R4'
ist, ein Rest R3', R&sup5;, R&sup6;, und R&sup7; Chlor oder Fluor ist; und
die verbleibenden Reste R3', R&sup5;, R&sup6;, R&sup7; Wasserstoff, Chlor
oder Fluor sind;
jeder der Reste R&sup8; und R&sup9; eine Carbonsäureschutzgruppe ist;
und
R10 Wasserstoff oder eine Aminoschutzgruppe ist; und
(ii) diastereoisomeren Mischungen dieser Tetrahydropyrido[2,3-
d]pyrimidine.
