Description
- Die vorliegende Erfindung bezieht sich auf die Verwendung einer Zusammensetzung, die ein Öl mit einem hohen Anteil an essentiellen Fettsäuren des ω3- und ω6-Typs, bevorzugter Weise Samenöl der schwarzen Johannisbeere, und mindestens eine Verbindung umfaßt, die ausgewählt ist aus β-Carotin, Lycopin, Tocopherol und seinen Derivaten, Tocotrienolen und deren Derivaten, Ascorbinsäure und Nikotinamid, als ein Nahrungsmittelzusatz, welcher dazu gedacht ist, schädliche Effekte von Xenobiotika auf die Haut, insbesondere auf das Immunsystem der Haut, zu verhüten und/oder zu bekämpfen.
- Die Haut ist ständig in Kontakt mit Xenobiotika, was Substanzen sind, die als fremd für die natürlichen metabolischen Stoffwechselwege der Zelle bezeichnet werden, wie z.B. medizinische Produkte, Pestizide, Schadstoffe, Tabak oder UV. Die Haut leidet unter täglichen Angriffen, wenn sie diesen Mitteln, insbesondere den UV-Strahlen, ausgesetzt wird, die unmittelbaren Schaden bei der Haut, wie z.B. Sonnenbrand, Photosensitivitäts- oder Immunosuppressions-Reaktionen, aber auch Langzeiteffekte, wie z.B. Lichtalterung oder Hauttumore, verursachen.
- Die meisten dieser nachteiligen Effekte hängen mit der Produktion von oxidierten freien Radikalen zusammen. Diese haben die Eigenschaft, bestimmte Hautbestandteile zu depolimerisieren, wie z.B. Collagen oder Elastin, oder sonst Membranlipide oder DNA zu degradieren, was zur Produktion von Endobiotika führt, die für die Haut schädlich sind, wie z.B. toxische Metabolite oder Entzündungsmediatoren, was letztendlich zum Verlust der Integrität der Zellmembranen führt. Die Wirkung von Xenobiotika und Endobiotika ist insbesondere bedeutsam in Bezug auf die Immunverteidigung der Haut. Insbesondere kann eine Depletion der Langerhans-Zellen zur Penetration von Pathogenen in den Körper führen, ohne daß das allgemeine Immunsystem gewarnt wird. Das führt z.B. zu einer verstärkten Neigung des Auftretens von Infektionen durch Pathogene.
- Die Verwendung von Verbindungen, die in der Lage sind, die Wirkung von Xenobiotika und Endobiotika, insbesondere von oxidierten freien Radikalen, zu inhibieren oder zu neutralisieren, könnte es ermöglichen, die hervorgerufene Hautschädigung zu reduzieren.
- Unter diesen Verbindungen haben Vitamine und essentielle Fettsäuren in einer Reihe von in vitro und in vivo Studien eine schützende Aktivität in Bezug auf schädliche Xenobiotika und Endobiotika gezeigt.
- Es wurde gezeigt, daß α-Tocopherol die Oxidation von poly-ungesättigten Fettsäuren verhindern kann, welche essentielle Komponenten von Zellmembranen und insbesondere sensitiv gegenüber der Schädigung sind, die durch freie Radikale induziert wird (
1 ). Diese Verbindung hat auch anti-inflammatorische Aktivität durch direkte Wirkung auf die enzymatischen Systeme der Arachidonsäure-Kaskade (2 ). Andere Verbindungen, die zur Tocotrienol-Familie gehören (α-, γ-Tocotrienole und deren Derivate) haben beachtenswerte Wirkung bei der Verhinderung von Zellschädigung gezeigt, die mit einem Aussetzen der Haut und der oberflächlichen Körperauswüchse gegenüber freien Radikalen und UV-Strahlen (3 ) und gegenüber Ozon (4 ) assoziiert sind. Zusätzlich konnte demonstriert werden, daß die Rolle von Tocopherolen und Tocotrienolen bedeutsam in Bezug auf die Aktivierung des Immunsystems ist (5 ). Ascorbinsäure kann ebenso die oxidierten freien Radikale (insbesondere Singulett-Sauerstoff) fangen, die in viele oxidative Prozesse involviert sind, welche Zellen schädigen (6 ). Zusätzlich zeigt α-Tocopherol in Kombination mit Ascorbinsäure Effekte auf die Steigerung der minimalen Erythemdosis (Minimum Erythemal Dose) (7 ). - β-Carotin ist ein Vitamin A-Vorläufer, welches als ein chemisches Screening-Mittel (mit einem Absorptionsmaximum im UVA und sichtbaren Bereich) agiert und gegen Lipid-Peroxidation schützt (
8 ,9 ). Es wird oftmals verwendet, um Photodermatosen zu verhindern (10 ). Im allgemeinen sind Carotenoide (β-Carotin, Lycopin, Lutein, Zeaxanthin und Astaxanthin) ebenso in die Modellierung des Immunsystems involviert (WO 98/44808). Lycopin ist außerdem ein freier Radikal-Fänger (11 ) und Nikotinamid wird oftmals auf die gleiche Weise wie ein β-Carotin gegen Photodermatosen eingesetzt (12 ). - Das Samenöl der schwarzen Johannisbeere, welches essentielle Fettsäuren des ω3- und ω6-Typs enthält, hat ein gewisses Maß an Wirksamkeit bei der Reduktion von bestimmten inflammatorischen Prozessen (
13 ) oder bei der Verstärkung der Immunantwort (14 ,15 ) gezeigt. Es wurde ebenso in der Herstellung von Medikament-Zusammensetzungen verwendet (16 ). - Im Zusammenhang mit der vorliegenden Erfindung wurde überraschenderweise gefunden, daß eine Kombination aus Samenöl der schwarzen Johannisbeere und der zuvor erwähnten Verbindungen in einem Nahrungsmittelzusatz einen vorteilhaften Effekt auf die Reduzierung der schädlichen Wirkung von Xenobiotika und Endobiotika des freien Radikal-Typs oder des Typs haben, der freie Radikale generiert, insbesondere durch Verstärkung der Immunabwehr der Haut. Nach wiederholter täglicher Anwendung gewährt eine solche Zusammensetzung Schutz für die Langerhans-Zellen.
- Beschreibung
- Daher bezieht sich die vorliegende Erfindung auf die Verwendung einer Zusammensetzung, umfassend ein Öl mit einem hohen Anteil an essentiellen Fettsäuren des Typs ω3 und ω6 und mindestens eine Verbindung, ausgewählt aus β-Carotin, Lycopin, Tocopherol und seiner Derivate, Tocotrienolen und deren Derivaten, Ascorbinsäure und ihrer Derivate, Nikotinamid und ein Spurenelement, als ein Nahrungsmittelzusatz, welcher dazu gedacht ist, die Immunabwehr der Haut und der oberflächlichen Körperauswüchse zu verstärken.
- Spezifischer gesagt, kann dieser Nahrungsmittelzusatz dazu gedacht sein, die schädlichen Effekte von Xenobiotika auf die Haut, insbesondere auf das Immunsystem der Haut, zu verhüten und/oder zu bekämpfen, insbesondere für die Reduktion der zytotoxischen Effekte von Xenobiotika auf die Langerhans-Zellen. In diesem Sinn ist eine Zusammensetzung gemäß der Erfindung nützlich für die Bekämpfung oder Verhütung von Photodermatosen, für die Verbesserung der Hauttoleranz gegenüber Sonnenlicht und für die Verhütung der Alterung der Haut und der oberflächlichen Körperauswüchse und deren Ungleichgewicht aufgrund von freien Radikalen.
- In einer vorteilhaften Ausführungsform umfaßt die Zusammensetzung Samenöl der schwarzen Johannisbeere, da dieses Öl reich an essentiellen Fettsäuren des ω3- und ω6-Typs ist, insbesondere α-Linolsäure, α-Linolensäure und γ-Linolensäure.
- Dieses Öl umfaßt:
- Im wesentlichen haben die ω3-Fettsäuren einen vorteilhaften Effekt auf Erythem, das durch UVB induziert wird, und sind nützlich in der Verhinderung von Photo-Karzinogenese. Die ω6-Fettsäuren sind insbesondere bedeutsam im Zusammenhang mit dieser Erfindung, da sie an der Keratinisation beteiligt sind.
- Natürlich kann auch ein Öl, das zum Samenöl der schwarzen Johannisbeere äquivalent in Bezug auf die oben erwähnte essentielle Fettsäure-Zusammensetzung ist, in dem beschriebenen Nahrungsmittelzusatz gemäß der Erfindung verwendet werden.
- In einem bevorzugten Aspekt der Erfindung umfaßt die Zusammensetzung Samenöl der schwarzen Johannisbeere, β-Carotin, Lycopin, Tocopherol oder seine Derivate, Tocotrienole oder deren Derivate, Ascorbinsäure oder deren Derivate und Nikotinamid.
- Die Zusammensetzung kann auch ein Spurenelement umfassen, bevorzugter Weise Selenium in jeder Form, insbesondere in Form von Selenium-enthaltenden Hefen.
- Selenium ist ein Co-Faktor, welcher die Aktivität von Gluthathionperoxidase (GPX) erhöht, Zellen gegen schädliche Effekte von organischen und anorganischen Peroxiden schützt und Membranen durch Einbau in die Disulfidbrücken stabilisiert, was zur Erniedrigung der Menge von Arachidonsäure, die während der inflammatorischen Antwort freigesetzt wird, führt. Daher ist diese Verbindung ebenso als eine Verbindung zum Schutz der Haut gegen Xenobiotika geeignet.
- Die Zusammensetzung umfaßt vorteilhafter Weise mindestens 25 Gew-%, bevorzugter Weise ungefähr 50 Gew.-%, des Samenöls von schwarzer Johannisbeere. Es kann ebenso umfassen:
- – mindestens 5 Gew.-%, bevorzugter Weise 15–18 Gew.-% Ascorbinsäure;
- – mindestens 4 Gew.-%, bevorzugter Weise ungefähr 8 Gew.-% Lycopin;
- – mindestens 5 Gew.-%, bevorzugter Weise ungefähr 10 Gew.-% Selenium;
- – mindestens 2 Gew.-%, bevorzugter Weise ungefähr 4 Gew.-% β-Carotin;
- – mindestens 1 Gew.-%, bevorzugter Weise ungefähr 3 Gew.-% Nikotinamid;
- – und/oder mindestens 1 Gew.-%, bevorzugter Weise ungefähr 2 Gew.-% Tocopherol oder seine Derivate, insbesondere Tocopherylacetat.
- Eine Zusammensetzung gemäß der Erfindung kann daher ungefähr umfassen, nach Gewicht:
- – 53% Samenöl der schwarzen Johannisbeere,
- – 4% β-Carotin,
- – 8% Lycopin,
- – 2,2% Tocopherylacteat,
- – 15–18% Ascorbinsäure, und
- – 3% Nikotinamid;
- – und optional ungefähr 10 Gew.-% Selenium.
- Unter den Hilfsstoffen, welche addiert werden, werden diejenigen bevorzugt, die ausgewählt sind aus gelbem Bienenwachs, Sojabohnenlecithin, Glycerol, flüssigem Paraffin und Gelatine, bevorzugter Weise von mariner Herkunft.
- Natürlich ist es möglich, zu der Zusammensetzung jede andere aktive Verbindung, die im Stand der Technik bekannt ist, hinzuzufügen. Erwähnt werden könnten z.B. Nukleinsäuren oder Nukleotide, Aminosäuren, insbesondere Lysin und Arginin, Peptide, Zucker, insbesondere biologisch aktive Zucker, Retinoide, Pflanzenextrakte, insbesondere aus grünem Tee oder Sojabohnen, jeder Extrakt von Pflanzen, der reich an Isoflavonen ist, DHEA und Melatonin.
- Die Zusammensetzung gemäß der Erfindung kann in Form von Gelkapseln, weichen Kapseln, Zucker-beschichteten und einfachen Tabletten, einschließlich verzögerter Formen, geeignet für orale Verabreichung sein. Daher bezieht sich die Erfindung auf eine Zusammensetzung, wie oben definiert.

4 : Wirksamkeit des oralen Zusatzes gemäß der Erfindung auf die Lipidperoxidation. Messung der Variation in der Menge an Squalenperoxiden. - Beispiel 2: Bevorzugte Zusammensetzung Nr. 2 der Erfindung
- Diese Zusammensetzung ist identisch zu der oben in Beispiel 1 beschriebenen, enthält aber zusätzlich Selenium 10%; d.h. 23,5 mg/Gelkapsel.
- Beispiel 3: Effekte der Zusammensetzung auf den Schutz gegen Xenobiotika und auf die Verstärkung der Immunantwort der Haut, demonstriert in einer Doppelblind-klinischen Studie gegenüber Placebo.
- 2 Gruppen von 20 gesunden Freiwilligen, einschließlich Individuen mit sensitiver Haut (Minimum 8) wurden aufgenommen, nachdem sie eine freie, erklärte und ausgedrückte Einwilligung (free, explained and express consent) unterschrieben hatten.
- Erste Gruppe (20 Freiwillige): 2 Kapseln, enthaltend das aktive Mittel, eingenommen oral pro Tag für 90 aufeinanderfolgende Tage (3 Monate).
- Zweite Gruppe (20 Freiwillige): 2 Kapseln des Placebos (ohne aktive Inhaltsstoffe), eingenommen oral pro Tag für 90 Tage (3 Monate).
- Messungen
- Die minimale Erythemdosis (MED) oder schwächste wahrnehmbare Rötung mit klar definierten Rändern wurde auf dem Rücken der Freiwilligen bestimmt, die an der Studie teilnahmen, gemäß der Empfehlungen von Colipa, vor, 1 Monat nach und 3 Monate nach dem Beginn der oralen Ergänzung.
- Die Kapazität, die diese Ergänzung auf die Verstärkung des Schutzes, der durch eine Sonnenschutzcreme (Sonnenschutzfaktor 16) gewährt wird, wurde weiterhin vor und nach den 3 Monaten der Behandlung evaluiert und mit der Wirksamkeit der Sonnenschutzcreme allein verglichen.
- Biopsien
- Oberflächliche Hautproben wurden vom unteren Rücken von zehn Freiwilligen jeder Gruppe genommen, unter der Verwendung eines zirkularen Bistoury (Stanzer), 4mm im Durchmesser, 24 Stunden nach Bestrahlung mit 1 MED.
- Die Biopsien wurden danach mit Bouin-Flüssigkeit fixiert, in Paraffin eingeschlossen, in Serien geschnitten, um durch Immununhistochemie entwickelt zu werden, um die Langerhans-Zellen zu zeigen (Verwendung von Mausklon 010 IgG1 monoklonaler Antikörper, von Immunotech Laboratory, welcher einen spezifischen Membranmarker der Langerhans-Zelle, CD1a, erkennt).
- Ergebnisse
- Alle Ergebnisse und statistischen Vergleiche sind in den
1 bis4 hierin danach angegeben. - Die untersuchte orale Ergänzung verstärkt signifikant die Widerstandsfähigkeit der Haut gegenüber Sonnenlicht, im Vergleich zu ihrem Placebo:
- – Zunahme
der minimalen Erythemdosis (
1 ), - – Zunahme
der Wirksamkeit eines topischen Sonnenschutz-Screening-Mittels (
2 ), - – Wirkung
auf das Immunsystem der Haut, durch Reduktion der Depletion von
Langerhans-Zellen, welche durch UV induziert wird (
3 ), - – Reinigen
(Scavenging) von freien Radikalen, induziert durch UV (Squalenhydroxyperoxide)
(
4 ). - Diese Kombination von Vitaminen (α-Tocopherol, Ascorbinsäure, β-Carotin, Lycopin und Nikotinamid) und von Samenöl der schwarzen Johannisbeere, welches essentielle Fettsäuren enthält, erscheint daher angebracht bei der Verhütung von nachteiligen Effekten zu sein, welche durch Xenobiotika induziert werden, insbesondere zur Verstärkung der Immunabwehr der Haut.

Claims (16)
- Verwendung einer Zusammensetzung, welche wenigstens 25 Gew.-% Samenöl der schwarzen Johannisbeere und wenigstens eine Verbindung, ausgewählt aus β-Carotin, Lycopin, Tocopherol, Tocotrienolen, Ascorbinsäure, Nicotinamid und einem Spurenelement, umfasst, für die Herstellung eines Nahrungsmittelzusatzes, welcher dazu gedacht ist, Lichtdermatosen zu bekämpfen und/oder zu verhüten, um die Widerstandsfähigkeit der Haut gegenüber Sonnenlicht zu verbessern und um die Alterung der Haut und der oberflächlichen Körperauswüchse oder -wucherungen und deren gestörtes Gleichgewicht aufgrund von freien Radikalen zu verhüten.
- Verwendung nach Anspruch 1, dadurch gekennzeichnet, dass die Zusammensetzung Samenöl der schwarzen Johannisbeere, β-Carotin, Lycopin, Tocopherol, Ascorbinsäure und Nicotinamid umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung auch ein Spurenelement umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung auch Selen umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung 50 Gew.-% Samenöl der schwarzen Johannisbeere umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 5 Gew.-% Ascorbinsäure umfasst.
- Verwendung nach Anspruch 6, dadurch gekennzeichnet, dass die Zusammensetzung 15% bis 18% Ascorbinsäure umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 4 Gew.-% Lycopin umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 5 Gew.-% Selen umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 2 Gew.-% β-Carotin umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 1 Gew.-% Nicotinamid umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung wenigstens 1 Gew.-% Tocopherol umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung bezogen auf das Gewicht umfasst: – 53% Samenöl der schwarzen Johannisbeere, – 4% β-Carotin, – 8% Lycopin, – 2,2% Tocopherylacetat, – 15 bis 18% Ascorbinsäure – und 3% Nicotinamid.
- Verwendung nach Anspruch 13, dadurch gekennzeichnet, dass die Zusammensetzung auch 10 Gew.-% Selen umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung auch wenigstens einen Träger, ausgewählt aus gelbem Bienenwachs, Sojabohnen-Lecithin, Glycerol, flüssigem Paraffin und Gelatine, umfasst.
- Verwendung nach einem der vorangegangenen Ansprüche, dadurch gekennzeichnet, dass die Zusammensetzung in Form von Gelkapseln, die für eine orale Verabreichung geeignet sind, vorliegt.
