Description

- cis-Dichloro-aminosäure-platin(II)-Komplexe
- Die Erfindung betrifft cis-Dichloro-aminosäure-platin(II-Komplexe der Formel (I) in der R1 und R2 unabhängig voneinander Wasserstoff oder den Rest einer biologisch annehmbaren Aminosäure und R3 4 und R unabhängig voneinander einen Alkylrest mit wenigstens 2 C-Atomen, der durch eine Hydroxylgruppe substituiert sein kann, bedeuten, ein Verfahren zu ihrer Herstellung und diese enthaltende Arzneimittel.
- Aus einer Veröffentlichung von B. Rosenberg et al., Nature 222, 385 (1969) ist bekannt, daß cis-Platin(II)-amin--Komplexe, z.B.
- cis- EPt(NH3)2Cl2] cytostatische Eigenschaften aufweisen. Nachteilig bei diesen Verbindungen ist, daß sie sich als sehr toxisch erwiesen haben.
- Aus der DE-OS 26 26 559 ist ferner bekannt, daß Verbindungen des Typs d.h. Peptidester-Komplexe von Platin (11) bei erheblich geringerer Toxizität als die der einfachen Aminkomplexe eine diesen vergleichbare antitumorale Wirkung aufweisen. Die cis-Struktur scheint fUr die Antitumorwirkung wesentlich zu sein.
- Es wurde nun gefunden, daß Verbindungen der voranstehenden Formel (I) bei geringer Toxizität eine besonders starke Hemmwirkung im Versuch hervorrufen Gemäß der oben angegebenen Formel (1) handelt es sich bei dem über das N-Atom mit dem Platin verbundenen organischen Rest um einen α-Aminosäureester, insbesondere natürlicher und essentieller Aminosäuren.
- Natürliche 0C-Aminosäuren sind beispielsweise Glycin und die L-Formen von Alanin, Serin, Valin Leucin, Isoleucin, Phenylalanin, Tyrosin, Threonin, Methionin, Lysin, Prolin, Histidin. Als Alkohole werden bevorzugt ein- oder mehrwertige Alkohole mit 2 bis 8 C-Atomen verwendet.
- Erfindungsgemäße Verbindungen sind dementsprechend cis-PtCl2 (Gly OEthyl)2 cis-PtCl2 (L-SerOEthyl)2 cis-PtCl2 (L-Ala OEthyl)2 cis-PtCl2 (L-Val OButyl)2 cis-PtCl2 (L-Leu On-Hexyl)2 cis-PtCl2 (L-Pro OPropOH)2, wobei die üblichen Abkürzungen für die Aminosäuren verwendet sind und PropOH den einwertigen Rest des Propylenglykols bedeutet.
- Die erfindungsgemäßen Verbindungen können hergestellt werden, indem man entweder zunächst einen cis-Platin-Komplex der Formel (II) in der R1 und R die oben angegebenen Bedeutungen hat, in üblicher Weise herstellt und den Komplex nachträglich verestert, z.B. durch Umsetzung mit Propylenoxid oder durch unmittelbare Umsetzung von z.B. Kaliumtetrachloroplatinat (II) mit einem Aminosäureester, wie in den nachfolgenden Herstellbeispielen beschrieben.
- Bei der üblichen Herstellweise fallen die cis-Verbindungen in sehr reiner Form in Ausbeuten von 80 bis 90 S an.
- Die erfindungsgemäßen Verbindungen zeichnen sich durch wertvolle pharmakologische Eigenschaften aus. Insbesondere sei eine cytostatische, antimikrobielle, antivirale und immunsuppressive Wirkung angegeben, wobei sie verglichen mit bekannten Verbindungen weniger toxisch sind. Sie können daher als Chemotherapeutika bei der Behandlung von Krebs, von bakteriellen und viralen Infektionen sowie als Immunsuppressiva Verwendung finden.
- Dementsprechend sind ein weiterer Gegenstand der vorliegenden Erfindung therapeutische Mittel, enthaltend neben üblichen Träger- und Verdünnungsmitteln eine Verbindung der Formel I als Wirkstoff, die je nach Zubereitung intravenös, intramuskulär, intraperitoneal, subkutan oder peroral verabreicht werden können. Auch äußerliche Applikation ist möglich.
- Herstellbeispiel a) nach Wallin (Ofver. Akad. Stockholm 492 21 (1894): Man suspendiert 2,0 mmol des Chelat-Komplexes in 50 ml Alkohol und erwärmt zum Sieden Dann leitet man langsam trockenen Chlorwasserstoff durch die Suspension, bis man eine gelbgrüne, schwach trübe Lösung erhält (etwa eine Stunde). Man erwärmt noch 30 Minuten und filtriert heiß. Beim langsamen Abkühlen des Filtrats kristallisieren gelbgrüne Nadeln aus. Diese werden auf einer Fritte gesammelt, zweimal mit 10 ml Ether gewaschen und unter vermindertem Druck getrocknet. Ausgehend vom Komplex des Alanins und Ethylalkohol erzielt man eine Ausbeute von etwa 80 % b) direkt: Man löst 2,5 mmol K2PtCl4 und 5,0 mmol des α-Aminosäureester-hydrochlorids in 4 ml Wasser. Innerhalb von 2 bis 3 Stunden tropft man eine Lösung von 5,0 mmol Kaliumhydroxid in 2 ml Wasser zu. Nach eintägigem Rühren sammelt man den gelbgrünen Niederschlag auf einer Fritte, wäscht mit einigen ml Wasser und 10 ml Ether und trocknet im Vakuum. Ausbeute 50 bis 80 %.

Claims (5)
- 9 Patentansprüche 1. Verbindungen der Formel in der R¹ und R2 unabhängig voneinander Wasserstoff oder den Rest einer biologisch annhembaren Aminosäure und R³ und R4 unabhängig voneinander einen Alkylrest mit wenigstens 2 C-Atomen bedeuten, der durch eine Hydroxylgruppe substituiert sein kann.
- 2. Verbindungen gemäß Anspruch 1, in denen die Aminosäurereste die L-Formen natürlich vorkommender Aminosäuren sind
- 3. Therapeutische Mittel, enthaltend neben üblichen Träger- und Verdünnungsmitteln eine Verbindung der Formel (I) als Wirkstoff.
- 4. Therapeutische Mittel nach Anspruch 3, dadurch gekennzeichnet, daß es eine Verbindung der Formel (I) in Form einer Lösung oder Suspension in Wasser enthält.
- 5. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I, dadurch gekennzeichnet, daß man entweder einen cis-Platin-Komplex der allgemeinen Formel (II) in an sich bekannter Weise verestert oder ein Salz der Tetrachlorplatin (II) säure unmittelbar mit einem Aminosäureester umsetzt.
