in einem wässerigen oder wasserfreien Medium hergestellt.
Die Umsetzung der Verbindungen der allgemeinen Formel II mit den Verbindungen der Formel III oder IV wird
ebenfalls entweder in einem wasserfreien inerten Lösungsmittel oder in einem Zweiphasensystem aus einer wässerigen
Natriumhydroxid- oder Kaliuinhydroxidlösung und einem inerten, nicht mit Wasser mischbaren organischen Lösungsmittel
durchgeführt.

Als inerte Lösungsmittel eigenen sich für beide genannten alternativen Verfahrensweisen aromatische Kohlenwasserstoffe
wie Benzol, Toluol und Xylole oder chlorierte Lösungsmittel wie ζ. B. Dichloräthan und
Tetrachloräthan; besonders geeignet ist die Verwendung
von Toluol oder Chlorbenzol ±m Hinblick auf die Produktausbeute
.
Die Umsetzung im Zweiphasensystem läßt sich ferner unter Verwendung von Phasenübertragungskatalysatoren durchführen,
jedoch bietet diese Verfahrensweise keinen wesentlichen Vorteil.
Die Reaktionsgemische werden nach erfolgter Umsetzung nach für die angeführten Alkylierungsreaktionen üblichen
Verfahren,je nach der gewählten Durchführungsform der
Umsetzung, aufgearbeitet. Ein geeignetes Verfahren beruht z. B. auf der Methode von E.R. Andrews et al.,
J.Med.Chem. V7 (1974), 882. Das rohe Reaktionsprodukt wird durch Kristallisation aus einem geeigneten einheitlichen
Lösungsmittel oder einem Lösungsmittelgemisch gereinigt; das Produkt wird in Form der Base charakterisiert
oder durch Einwirkung eines 10 %igen Überschusses einer anorganischen oder organischen und insbesondere einer pharmazeutisch
geeigneten Säure im Medium eines niederen Alkohols, vorzugsweise von Methanol oder Äthanol, in das entsprechende
Säureadditionssalz übergeführt, das zuletzt
durch Kristallisation aus einem geeigneten Lösungsmittel gereinigt wird.
Als Säuren für die Herstellung der Additionssalze sind aus pharmazeutischer Sicht einige typische anorgani-

sehe Säuren wie z. B. Chlorwasserstoffsäure, Schwefelsäure
und Phosphorsäure sowie organische Carbonsäuren und Sulfonsäuren wie z. B. Maleinsäure, Weinsäure,
Methansulfonsäure und Äthansulfonsäure,2-Haphthalinsulfonsäure,
Fumarsäure und Citronsäure besonders geeignet.
Die für die oben angeführten Reaktionen erforderlichen
Ausgangssubstanzen, d. h. die Verbindungen der allgemeinen Formel V, sind bereits bekannt 'vgl CF. Koelsch,
J. Org.Chem. _26 (1961), 2590 und die CS-PS "en 200.094,
202.213 und 204.211) und präparativ leicht zugänglich.
Die Herstellung von Verbindungen der allgemeinen Formel I und ihren Additionssalzen mit anorganischen und
organischen Säuren ist in den folgenden Ausführungsbeispielen näher erläutert, die jedoch nicht einschränkend
sind. Die angegebenen Schmelzpunkte der Verbindungen wurden auf der Koflerbank bestimmt und sind unkorrigiert.
Beispiel 1
5- [2-(Dimethylamino)äthoxy]-7-oxo-VH-benzo fcjfluoren
5- [2-(Dimethylamino)äthoxy]-7-oxo-VH-benzo fcjfluoren
Eine durch Auflösen von 2,76 g (0.12 mol) Natrium in
45 ml Methanol hergestellte Natriummethylatlösung wird mit 150 ml Chlorbenzol und 14,8 g (0,06 mol) 5-Hydroxy-
-7-oxo-7H-benzo fcj f luoren versetzt; das Reaktionsgemisc-n
wird allmählich bis zur Siedetemperatur dps Chlorbenzols
unter gleichzeitigem Abdestillieren von MuLhanol erwärmt,
wonach eine Lösung von 13,0 g (0,12 mol) 2-(Dimethylamine)-äthylchlorid
in 30 ml Chlorbenzol zugegeben und das Reaktionsgemisch 5 h am Rückfluß zum Sieden erhitzt wird. Nach
dem Abkühlen und Zersetzung des Reaktionsgemischs mit Wasser wird die organische Schicht abgetrennt, mit einer
10 %igen wässerigen Kaliumhydroxidlösung extrahiert,
über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft. Der kristalline Rückstand wird aus einem
Tetrachlormethan-Hexan-Gemisch (1:1) umkristallisiert;
es werden 14,8 g (78,3 %) des obigen Produkts erhalten. F. 91-93 °C.
5- [2- (Dimethylamine) äthoxy] -7-oxo-7H-benzo [cjf luoren-
-hydrogenmaleinat
Eine nach Beispiel 1 hergestellte Lösung von 318 mg (1 mol) 5- J~2- (Dimethylamino) äthoxy] -7-oxo-7H-benzo (c] f luoren
in 5 ml Methanol wird mit 127 mg (1,1 mol) Maleinsäure versetzt. Nach Abkühlen auf -Γ·>
C scheidet sich aus der Lösung ein Feststoff aus, der abgesaugt und aus Äthanol
umkristallisiert wird, worauf das obige Maleinat erhalten wird. F. 119-124 °C.
In analoger Weise sind:
5- [2- (Dimethylamino) äthoxy] -7-oxo-7H-benzo[cJ f luoren-
-napsilat; F. 85-87 0C (Äthanol) und
5- [2- (Dimethylamino) äthoxy] -7-oxo-7H-benzo [cT] f luoren-hydrochlorid;
F. 268-269 °C (Äthanol) erhältlich.
3. 9-Diäthyl-5-[2-(Dimethylamino) äthoxy] -7-oxo-7H-benzo [c]-fluoren-hydrochlorid
Eine durch Auflösen von 0,92 g (0,04 mol) Natrium in 15 ml Methanol hergestellte Natriummethylatlösung wird

mit 50 ml Chlorbenzol und 6,04 g (0,02 mol) 3.9-Diäthyl-
-5-hydroxy-7-oxo-7H-benzo Jc]fluoren versetzt; die Reaktion wird wie in Beispiel 1 unter Verwendung von 4,3 g
2-(Dirnethylamino)äthylchlorid durchgeführt. Die als Produkt
angefallene Base (F. 88-90 °C) wird in Äthanol gelöst und mit einer äthanolischen Chlorwasserstoffsäurelösung
angesäuert; der nach dem Abkühlen ausgeschiedene kristalline Niederschlag wird abgesaugt und aus Äthanol
kristallisiert. Man erhält 6,2 g (76 %) des obigen Produkts. F. 241-243 °C.
In analoger Weise wird 3.9-Dimethyl-5- \l-(dimethylamino)äthoxy]-7-oxo-7H-benzo[cj
fluoren-hydrochlorid erhalten; F. 250-252 °C (Äthanol-Hexan).
5-£2-(Diäthylamino)äthoxy] -7-oxo-7H-benzo [c]fluoren-hydrochlorid
Ein Gemisch von 320 ml Toluol und 80 ml Wasser mit 13,2 g (0,24 mol) Kaliumhydroxid wird mit 9,84 g (0,04 mol)
5-Hydroxy-7-oxo-7H-benzo [c]fluoren und 14,9 g (0,088 mol)
2-(Diäthylamino)äthylchlorid-hydrochlorid versetzt. Das Reaktionsgemisch wird 12 h am Rückfluß erwärmt; nach dem
Abkühlen wird die Toluolschicht von der wässerigen Schicht abgetrennt und wie in den Beispielen 1 und 3 verarbeitet.
Durch Kristallisation aus Äthanol erhält man 13,6 g (89,5 %)
des obigen Produkts; F. 248-250 °C.

Die Verbindungen der allgemeinen Formel I mit R und Z wie oben weisen in biologischen Tests im Tierversuch eine
wachstumshemmende Wirkung auf implantierte Geschwülste und eine die Überlebenszeit verlängernde Wirkung bei den
Tieren mit Geschwülsten auf.
Die Untersuchungen wurden an 5-(3-Dimethylaminopropoxy)-
-7-oxo-7H-benzo(_c] fluoren-hydrochlorid (Verbindung A),
5-- [3- (Dimethylamine) -2. 2-dimethylpropoxy] -7-oxo-7H-benzo [c] fluoren-hydrochlorid
(Verbindung B) und 5- |2-(Dimethylamino) äthoxy! -7-oxo-7H-benzo [c~j fluoren-hydrochlorid
(Verbindung C) durchgeführt. Die erhaltenen Ergebnisse sind in den Tabellen 1-3 angeführt.

Die Hemmung der Adenin- und Valin-Inkorporation in
das Geschwulstzellensystem wird dabei als Maß für die Nucleinsäure- und Proteinsynthesehemmung in den entsprechenden
Zellen und damit als Maß für die tumorhemmende cytostatische Wirkung angesehen.
Die Ergebnisse sind in Tabelle 4 angegeben.
Aus den Ergebnissen ist ersichtlich, daß Verbindung C
zu einer signifikanten Verringerung der Inkorporation von 14C-Adenin und 14C-Valin führt und in
als 1Ofach wirksamer ist als Tiloron.
C-Adenin und C-Valin führt und in dieser Hinsicht mehr
Die tumorhemmende cytostatische Wirksamkeit von Verbindung C ist damit signifikant höher als die der strukturell
nahestehenden Vergleichsverbindung Tiloron.

Die Verbindung C weist ferner bei Mäusen immunsuppressive
Wirkungen auf die Antikörperbildung auf.
Die Untersuchungen wurden nach dem Verfahren von M. ^vorcovä, K. Rezäbek und M. Semonsky, Arzneimittelforschung
30 (I) 6 (1980) 978-9 80, durchgeführt.
Die Verbindung C wurde subcutan gespritzt. Die Verabreichung erfolgte 1 χ täglich entweder 5 Tage lang
(2 Tage vor der Immunisierung, am Tag der Immunisierung und 2 Tage danach) oder 11 Tage lang (2 Tage vor der
Immunisierung, am Tag der Immunisierung und 8 Tage danach). Es wurden 12 Tiere pro Gruppe herangezogen.
Die erhaltenen Ergebnisse sind in Tabelle 5 aufgeführt. Aus den Titerwerten (Mittelwert der höchsten Serumverdünnung,
die vollständige Agglutination bzw. vollständige Hämolyse hervorruft) geht hervor, daß die Verbindung C
die Serumantikörperbildung hemmt.

Aufgrund der obigen pharmakologischen Ergebnisse kann eine geschwulstheinmende und immunsuppressive Wirkung

Verfahren nach Anspruch 13, dadurch gekennzeichnet, daß die Umsetzung der Verbindung der Formel II mit der Verbindung der Formel III oder der Formel IV in einer Menge von1 bis 3 mol in einem Zweiphasensystem aus einer wässerigen Natriumhydroxid- oder Kaliumhydroxidlösung und einem inerten, mit Wasser nicht mischbaren organischen Lösungsmittel bei der Siedetemperatur des Reaktionsgemischs durchgeführt wird.
- 15. Verfahren nach Anspruch 14, dadurch gekennzeichnet, daß als inertes, mit Wasser nicht mischbares organisches Lösungsmittel Toluol verwendet wird.130081/0386§102517
- 16. Pharmazeutische Mittel,gekennzeichnet durchmindestens einen basischen Äther des 7-Oxo-7H-benzo j_cjfluorens der Formel I nach einem der Ansprüche 1-12 oder mindestens ein entsprechendes Additionssalz mit einer pharmazeutisch geeigneten anorganischen oder organischen Säure.130061/0386
