Description
Die Erfindung betrifft kationische Lipide der allgemeinen Formel I,
wobei n = 2,3,4,6,8 und m = 2,3,6,8 sein kann und R₁ = H, CH₃, CH₂CH₂OH; R₂ = H, CH₃,

Kationische Lipide der allgemeinen Formel I-X sind geeignete Reagenzien für den
liposomalen Gentransfer (Transfektion). Anwendungsgebiete derartiger
Transfektionsreagenzien liegen in der Medizin und in der Gentechnik.
Das Einbringen genetischen Materials in eukaryontische Zellen ist ein grundlegendes
Verfahren zum Studium biologischer Funktionen und von zunehmender Bedeutung für die
gentherapeutische Behandlung verschiedener Erkrankungen, von denen die Tumoren an erster
Stelle zu nennen sind.
Man unterscheidet hierbei biologische, physikalische und physikochemische Methoden für
den Transfer von DNA, RNA und Proteinen in die Zielzelle [Wagner J, Madry, H., Reszka, R

Physikalische Methoden wie Elektroporation und Mikroinjektion sind nur für den ex vivo und
in vitro Transfer geeignet. Die sogenannte "Jet"- Injektions-Methode läßt sich darüberhinaus
auch für den in vivo Gentransfer anwenden (Leber, Haut). Physikochemische Methoden wie
Kalziumphosphatpräzipitationstechnik (KPP)- und DEAE-Dextran-Transfektion, sind auf in
vifro und ex vivo Anwendungen begrenzt. Der zur Zeit in einer Reihe von klinischen Studien
(Phase I/II) erprobte retrovirale Gentransfer mit Hilfe virusproduzierender Zellen ist durch
eine längeranhaltende, aber relativ niedrige Genexpression in sich teilende Zellen
gekennzeichnet. Problematisch beim retroviralen Gentransfer ist vor allem die Entwicklung
einer spezifischen Immunantwort gegen die implantierten virusproduzierenden Helferzellen,
die Möglichkeit der Generierung replikationskompetenter Viren und die Gefahr einer
Aktivierung von zellulären Onkogenen bzw. Inaktivierung von Suppressorgenen als Resultat
der zufälligen Lokalisation der Geninsertion. Die notwendigen umfangreichen
zellbiologischen und medizinischen Vorarbeiten und die aufwendigen
Sicherheitsvorkehrungen lassen hohe Kosten bei der klinischen Anwendung des retroviralen
Gentransfers erwarten.
Vektoren auf der Basis von Adenoviren sind ebenfalls aussichtsreiche Gentransfervehikel.
Diese erreichen hohe Transfektionsraten auch in sich nicht teilendem Gewebe. Da die DNA
nicht in das Genom integriert wird, ist die Dauer der Fremdgenexpression begrenzt und einer
wiederholten Anwendung in vivo steht die starke spezifische Immunantwort des
Wirtsorganismus bei wiederholten Applikationen entgegen.
Der liposomal vermittelte Gentransfer hat dagegen in den letzten Jahren auch für in vivo
Anwendungen an Bedeutung gewonnen. Die Genkonstrukte lassen sich entweder in die
Liposomen verkapseln oder werden mit der Membran assoziiert. Liposomale Präparationen zeichnen sich durch eine einfache Handhabung, geringe Immunreaktivität und damit wiederholbare Anwendbarkeit bei niedrigem Sicherheitsrisiko für Anwender und "Empfänger" aus. Noch am Anfang steht die Anwendung von Immunoliposomen als Transportvehikel für genetisches Material.

Ein ebenfalls interessanter Ansatz ist die Verwendung fusogener Liposomen, die in ihrem

Seit einigen Jahren werden kationische Liposomen für den Transfer von DNA (Felgner PL,
Gadek TR, Holm M et al. (1987). Lipofection: a highly efficient, lipid-mediated DNA
transfection procedure. Proc Natl Acad Sci USA 84 : 7413-7417, Felgner JH, Kumar R, Sridhar
CN et al. (1994). Enhanced gene delivery and mechanism studies with a novel series of
cationic lipid formulations. J Biol Chem 269 : 2550-2561), Antisense-Oligomeren, Proteinen
und Ribozymen mit Erfolg eingesetzt. DNA wird dabei über elektrostatische
Wechselwirkungen mit der Membran der Liposomen assoziiert und über einen bisher noch
unvollständig verstandenen Mechanismus in die Zielzelle transferiert. Die Transferrate in
vitro ist dabei in Abhängigkeit von der Zellinie mit der Effizienz des retroviralen Gentransfer
vergleichbar. Zur Zeit befinden sich die ersten kationischen Liposomen/DNA/Komplexe
[DMRIE/DOPE und DOTMA/DOPE (Lipofectin, Gibco BRL USA) sowie DC-Chol/DOPE]
in einer klinischen Phase I/II Prüfung (Gao, X., Huang, L. (1995) Cationic liposome-mediated
gene transfer. Gene Therapy 2 : 710-722).
Kationische, amphiphile Moleküle, bestehend aus einem Lipidteil (Steroid- oder Diglycerid-
Gerüst) und einer positiv geladenen Kopfgruppe (Ammonium-Ionen) sind in der Lage,
entweder spontan oder nach Zusatz eines Hilfslipids (Dioleoylphosphatidylethanolamin
(DOPE)) Liposomen auszubilden. DNA wird dabei über elektrostatische Wechselwirkungen
mit der Membran der Liposomen assoziiert und über einen bisher noch unvollständig
verstandenen Mechanismus in die Zielzelle transferiert.
Für einen möglichst effizienten in vitro und in vivo Gentransfer sollten die zur Generierung
der Liposomen verwendeten kationischen Lipide die folgenden Kriterien erfüllen:
- 1. Sie sollten nicht toxisch und biologisch abbaubar sein und keine Immunreaktionen hervorrufen
- 2. Sie sollten möglichst effizient mit der DNA komplexieren, diese vor Abbaureaktionen schützen und hohe Transferraten aufweisen
- 3. Sie sollten Zellen rezeptorspezifisch transfizieren
- 4. Sie müssen leicht und in größeren Mengen synthetisiert werden können.
Sowohl die Syntheseverfahren als auch die Moleküle selbst der bislang in der Literatur
beschriebenen kationischen Amphiphile weisen eine Reihe von Nachteilen auf. Die Synthese
der kommerzialisierten Transfektionsreagenzien DOTMA (Lipofektin), DOGS und DOSPA
verläuft über viele Stufen mit zum Teil sehr geringen Ausbeuten und die Aufreinigung der
Produkte erfordert eine aufwendige säulenchromatographische Reinigung. Bei vielen
Verbindungen wie z. B. DOTMA, DMRIE und DOSPA sind die kationischen Kopfgruppen mit
langkettigen Fettalkoholen über Etherbindungen verknüpft, welche eine sehr schlecht
biologische Abbaubarkeit und somit eine hohe Toxizität des Moleküls zur Folge haben.
Der Erfindung liegt die Aufgabe zu Grunde, neuartige kationische Lipide zu finden, welche im
Vergleich mit den bislang bekannten Verbindungen höhere Gentransferraten und geringere
Toxizitäten aufweisen. Weiterhin liegt der Erfindung die Aufgabe zu Grunde, Verfahren zu
finden, welche die Synthese der kationischen Lipide in wenigen Reaktionsstufen und mit hohen
Ausbeuten ermöglicht.
Diese Aufgaben wurde gemäß den Ansprüchen I-IX gelöst. Mit den neuartigen kationischen
Lipiden der Formeln I-XI werden Verbindungen zur Verfügung gestellt, die den
Anforderungen der Aufgabenstellung entsprechen. So wird z. B. in Abbildung 2-6 an
unterschiedlichen Tumorzellinien gezeigt, daß die aus den Lipiden SP-Chol, O-Chol, Put-Chol
und DOSGA und dem Helferlipid Dioleoylphosphatidylethanolamin (DOPE) (in den
beschriebenen molaren Verhältnissen) hergestellten Liposomen im Vergleich zu DC-
Chol/DOPE Vesikel deutlich höher transfizieren in Abwesenheit von Serum. An der Ratten-
Kolonkarzinomlinie CC531 (Abb. 3) transfizieren SP-Chol und O-Chol/DOPE signifikant höher
als DC-Chol/DOPE bei einem 5%igem Serumanteil. Besonders überraschend sind die in Abb. 5
und 6 gezeigen hohen Transferraten, die unter Verwendung von DOSGA/DOPE-Liposonien
bei einem 5%igem Serumanteil an humanen und Rattenglioblastomzellen (N64, F98) gewonnen
wurden. Auch sind SP-Chol/DOPE-Liposomen in serumhaltigen Medium sehr gut geeignet,
humane Brusttumorzellen (MaTu) zu transfizieren (Abb. 1) Zuordnung der Lipide:

Die Herstellung der erfindungsgemäßen Verbindungen erfolgt gemäß den beigefügten
Schemata.
Schema I beschreibt das Verfahren zur Herstellung neuartiger kationischer Lipide mit 1,3-
Diglycerid-Grundgerüst an denen die kationische Kopfgruppe über einen Carboxylat-Spacer
angeknüpft ist.
Schema II beschreibt das Verfahren zur Synthese 1,3-Diglycerid-modifizierter Aminosäuren.
Schema III beschreibt das Verfahren zur Synthese 1,3-Diglycerid-modifizierter Guanidin-
Derivate.
Schema IV beschreibt das Verfahren zur Synthese neuartiger Cholesterol-Derivate mit
biogenen Aminen als kationischen Kopfgruppen.
Schema V beschreibt das Verfahren zur Synthese Aminosäure-modifizierter Cholesterol-
Derivate.
Schema VI beschreibt das Verfahren zur Synthese Glucosamin-modifizierter Cholesterol-
Derivate.
Schema VII beschreibt das Verfahren zur Synthese kationisch-modifizierter Oestradiol-
Derivate.
Schema VIII beschreibt das Verfahren zur Synthese kationisch-modifizierter Oestron-
Derivate.
Zusammenfassend kann festgestellt werden, das es nunmehr möglich ist, durch die
beschriebenen Verfahren unter kostengünstigen Bedingungen und unter Verwendung
preiswerter Chemikalien Transfektionsreagenzien zu synthetisieren, welche eine unerwartet
hohe Gentransferrate und eine gute biologische Abbaubarkeit aufweisen.
Die Erfindung soll nachfolgend durch Ausführungsbeispiele näher erläutert werden.
247 µl (1 mmol) 1,3-Dibenzyloxy-2-propanol wurden mit 7 ml einer 20%igen Lösung von
Phosgen in Toluol versetzt und 48 h bei RT gerührt. Nach dieser Zeit wurde überschüssiges
Phosgen mit Argon ausgetrieben und der Ansatz zu einer Lösung von 302 mg (0.6 mmol)
N¹,N¹⁰,N¹⁴-tris-BOC-spermin und 300 µl Triethylamin in 5 ml Methylenchlorid zugetropft
und für 4 h bei RT gerührt. Anschließende Säulenchromatographische Reinigung (Kieselgel
60) ergaben 375 mg (47%) 2-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-1,3-
dibenzyloxy-propanol (zähes Öl).
670 mg (0.84 mmol) 2-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-1,3-
dibenzyloxy-propanol wurden daraufhin in 25 ml Ethanol gelöst, mit 150 mg Pd/C (5%)
versetzt und 24 h hydriert. Nach Absaugen des Katalysators erhielt man 520 mg (100%) 2-
(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-1,3-propantriol (zähes Öl).
472 mg (0.76 mmol) 2-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-1,3-propantriol-
wurden mit 452 mg (1.6 mmol) Ölsäure, 334 mg (1.62 mmol) DCC und 20 mg DMAP in 30
ml Methylenchlorid gelöst und 24 h bei RT gerührt. Anschließende
Säulenchromatographische Reinigung (Kieselgel 60) ergaben 457 mg (52%) 2-(N⁵-
carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-1,3-dioleoyloxy-propa-n (zähes Öl).
200 mg (0.17 mmol) 2-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetrazzadecan))-1,3-
dioleoyloxy-propan wurden mit 3 ml TFA/CH₂Cl₂ (1 : 1) versetzt und 30 min bei RT gerührt.
Der Ansatz wurde mit 10 ml Dioxan verdünnt und im Hochvakuum vom Lösungsmittel
befreit. Man erhielt 202 mg (99%) 2-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-ammoniumtetraazadecan))-
1,3-dioleoyloxy-propan-tri-trifluoracetat I (zähes, farbloses Öl).
4 mmol (756 mg) Boc-L-alanin (M = 189.21) und 4.3 mmol (436 mg 600 µl) Triethylamin
(M = 101.19, d = 0.726), wurden in 10 ml abs. Tetrahydrofuran gelöst und mit Aceton/N₂ (fl.)
auf -10°C abgekühlt. Nun wurden 4 mmol (320 µl) Chlorameisensäuremethylester (M = 94.50,
d 1.228) zugegeben und 30 min bei 0°C gerührt. Nach Zugabe von 5 mmol (456 mg) 2-
Amino-1,3-propandiol (M = 91.11) wurde noch 1 h bei Raumtemperatur gerührt. Nach
Entfernen des Tetrahydrofurans am Rotationsverdampfer wurden 50 ml Ethylacetat
zugegeben. Die organische Phase wurde mit 5 ml ges. NaHCO₃-Lösung und 10 ml ges. NaCl-
Lösung gewaschen und über Na₂SO₄ getrocknet. Nach Einrotieren im Wasserstrahlvakuum
wurden letzte Lösungsmittelreste im Ölpumpenvakuum entfernt. Man erhielt 332 mg (32%)
2-BOC-L-Alanyl-(1,3-dihydroxy)propylamid (farbloses Öl).
2.28 mmol (598 mg) 2-BOC-L-Alanyl-(1,3-dihydroxy)propylamid, 5.70 mmol (1.610 g)
Ölsäure (M = 282.47), 5.70 mmol (1.176 g) Dicyclohexylcarbodiimid (M = 206.33) und 0.23
mmol (28 mg) Dimethylaminopyridin (M = 122.17) gelöst in 40 ml abs. CH₂Cl₂ wurden 12 h
bei RT gerührt. Anschließende Reinigung des Rohprodukts durch Säulenchromatographie
(Kieselgel 60) ergab 1.450 g (80%) 2-BOC-L-Alanyl-(1,3-dioleoyloxy)propylamid.
0.89 mmol (702 mg) 2-BOC-L-Alanyl-(1,3-dioleoyloxy)propylamid wurden mit 6 ml
TFA/CH₂Cl₂ (1 : 1) versetzt und 30 min bei RT geführt. Der Ansatz wurde mit 10 ml Dioxan
verdünnt und im Hochvakuum vom Lösungsmittel befreit. Man erhielt 712 mg (99%) 2-L-
Alanyl-(1,3-dioleoyloxy)propylamid-trifluoracetat II.
1.892 g (10 mmol) N-BOC-β-alanin wurden analog zu Beispiel 2 zu 2-BOC-β-Alanyl-1,3-
dioleoyloxypropylamid (15% über beide Stufen) umgesetzt. 500 mg (0.64 mmol) 2-BOC-P-
Alanyl-1,3-dioleoyloxypropylamid wurden mit 3 ml TFA/CH₂Cl₂ (1 : 1) versetzt und 30 min
bei RT gerührt. Der Ansatz wurde dann mit 50 ml Methylenchlorid verdünnt und mit zwei
mal 5 ml ges. NaHCO₃-Lösung extrahiert. Die über Natriumsulfat getrocknete organische
Phase wurde am Rotationsverdampfer vom Lösungsmittel befreit und der Rückstand mit 103
mg (0.7 mmol) 1-H-Pyrazol-1-carboxamidin-hydrochlorid und mit 120 µl (0.7 mmol) DIEA
in 20 ml DMF gelöst und über Nacht bei RT gerührt. Anschließende Reinigung des
Rohprodukts durch Säulenchromatographie (Kieselgel 60) ergab 480 mg (98%) 2-(Guanidino
β-alanyl)-1,3-dioleoyloxypropylamid-hydrochlorid III (zähes Öl).
610 mg (1.2 mmol) N¹,N¹⁰,N¹⁴-tris-BOC-spermin wurden in 10 ml Methylenchlorid gelöst
und mit 674 mg (1.5 mmol) Cholesterolchlorformiat und mit 210 µl (1.5 mmol) NEt₃ versetzt.
Der Ansatz wurde für 4 h bei RT gerührt und anschließend säulenchromatographisch
gereinigt. Man erhielt 570 mg (52%) 3β-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-
tetraazadecan))-cholesterol (farbloser Schaum). 275 mg (0.3 mmol) 3β-(N⁵-carbamoyl-
(N¹,N¹⁰,N¹⁴-tris-BOC-tetrs-decan))-cholesterol wurden mit 6 ml MeOH (10 mmol HCl
enthaltend) versetzt, 30 min bei RT gerührt und im Hochvakuum vom Lösungsmittel befreit.
Man erhielt 205 mg (94%) 3β-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-ammoniumtetraazadecan))-
cholesterol-tri-hydrochlorid V (farbloser Feststoff). Auf analoge Weise wurde ausgehend von
BOC-Putrescin 3β-(N-Ammonium-N′-carbamoyl-1,4-diaminobutan)-cholesterol-hydrochlorid-
VII (farbloser Feststoff) synthetisiert.
997 mg (3 mmol) Di-BOC-Ornithin wurden in 40 ml Methylenchiorid gelöst, mit 1.16 g (3
mmol) Cholesterol, 640 mg (3.1 mmol) DCC, 50 mg DMAP versetzt, und 24 h bei RT
gerührt. Es wurde vom ausgefallenen Dicyclohexylharnstoff abfiltriert im Vakuum eingeengt
und der Rückstand aus 5 ml Hexan umkristallisiert. Man erhielt 1.62 g (77%) 3β-di-BOC-
Ornithyl-cholesterol (farbloser Feststoff). 190 mg (0.27 mmol) 3β-di-BOC-Ornithyl-
cholesterol wurden mit 6 ml MeOH (enthielt 10 mmol HCl) versetzt und 60 min bei RT
gerührt. Nach Entfernen des Lösungsmittels im Hochvakuum erhielt man 146 mg (94%) 3β-
Ornithyl-cholesterol-dihydrochlorid VI (farbloser Feststoff).
279 mg (1 mmol) N-BOC-Glucosamin wurden in 8 ml Pyridin gelöst, mit 449 mg (1 mmol)
Cholesterylchlorformiat versetzt und für 24 h bei RT gerührt. Daraufhin wurde im
Hochvakuum Pyridin entfernt, in 50 ml Ether aufgenommen und mit 0.01 n HCl extrahiert.
Die organische Phase wurde vom Lösungsmittel befreit und der Rückstand
säulenchromatographisch (Kieselgel 60) gereinigt. Man erhielt 210 mg (30%) 6-(3β-
Carboxycholesteroyl)-N-BOC-glucosamin (farbloser Feststoff). 100 mg (0.15 mmol) 6-(3β-
Carboxycholesteroyl)-N-BOC-glucosamin wurden mit 3 ml ml MeOH (enthielt 5 mmol HCl)
versetzt und 60 min bei RT gerührt. Nach Entfernen des Lösungsmittels im Hochvakuum
erhielt man 95 mg (99%) 6-(3β-Carboxycholesteroyl)-glucosamin-hydrochlorid VIII
(farbloses, zähes Öl).
216 mg (0.8 mmol) Östron wurden in 10 ml Dioxan gelöst, mit 241 µl (2 mmol) Diphosgen
und 152 µl (1.2 mmol) Dimethylanilin versetzt und 12 h bei RT gerührt. Daraufhin wurde im
Hochvakuum eingeengt und der Rückstand in 15 ml Methylenchlorid gelöst. Die so erhaltene
Lösung wurde zu einer Lösung von 300 mg (0.6 mmol) N¹,N¹⁰,N¹⁴-tris-BOC-spermin und
277 µl (2 mmol) Triethylamin in 5 ml Methylenchlorid zugetropft und 4 h bei RT gerührt.
Anschließende säulenchromatographische Reinigung ergab 290 mg (61%) 3-(N⁵-carbamoyl-
(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-oestron (zähes Öl).
220 mg (0.28 mmol) 3-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-oestron wurden
daraufhin in 5 ml Dioxan gelöst und mit 37 mg (0.96 mmol) Natriumborhydrid versetzt. Nach
2 stündigem Rühren bei RT wurde der Ansatz mit 0.1 N HCl auf pH 6 gebracht und drei mal
mit je 50 ml Methylenchlorid extrahiert. Die organischen Phasen wurden über Natriumsulfat
getrocknet und am Rotationsverdampfer vom Lösungsmittel befreit. Man erhielt 219 mg
(96%) 3-(N⁵-carbamoyl-(N¹,N¹⁰,N¹⁴-tris-BOC-tetraazadecan))-oestradiol (farbloser Schaum).
Die Verbindung wird analog dem Beispiel 4 mit MeOH/ HCl zu 3-(N⁵-carbamoyl-
(N¹,N¹⁰,N¹⁴-ammoniumtetraazadecan))-oestradiol-tri-hydrochlorid IX umgesetzt.
541 mg (2 mmol) Östron wurden in 20 ml Dioxan gelöst, mit 603 µl (5 mmol) Diphosgen und
380 µl (3 mmol) Dimethylanilin versetzt und 12 h bei RT gerührt. Daraufhin wurde im
Hochvakuum eingeengt und der Rückstand in 15 ml Methylenchlorid gelöst. Diese Lösung
wurde mit 800 µl (7.3 mmol) N,N-Dimethylaminoethylendiamin versetzt, 4 h bei RT gerührt
und dann mit 100 ml Methylenchlorid verdünnt. Es wurde fünf mal mit je 20 ml 0.01 N HCl
extrahiert, die organische Phase über Natriumsulfat getrocknet und vom Lösungsmittel
befreit. Anschließende säulenchromatographische Reinigung (Kieselgel 60) ergab 573 mg
(68%) 3-(N,N-dimethylammoniumethylendiamin-N′-carbamoyl)-oestron-hydrochlo-rid X
(farbloser Feststoff).
Claims (9)
1. Kationische Lipide der allgemeinen Formel I,

2. Kationische Lipide der Formel II,
wobei R₁ einen aliphatischen, aromatischen oder heteroaliphatischen α-C-Atom-
Substituenten der α-Aminosäuren Glycin, Alanin, Valin, Leucin, Isoleucin, Phenylalanin,
Tyrosin, Prolin, Hydroxyprolin, Serin, Threonin, Cystein, Cystin, Methionin, Tryptophan,
Arginin, Lysin, Ornithin, Histidin, R₂ einen geradkettigen, gesättigten oder ungesättigten
aliphatischen Rest C₇-C₂₁, X = Cl, Br, I, CH₃COO, CF₃COO, Y = CH₂, O, NH und Z = CH₂,
O, NH bedeutet.
3. Kationische Lipide der allgemeinen Formel III,
wobei R₁ = H, CH₃, (CH₂)₃NH₂⁺X⁻(CH₂)₃NH₃⁺X⁻, R₃ = H, (CH₂)₃NH₃⁺X⁻, R₂ einen
geradkettigen, gesättigten oder ungesättigten aliphatischen Rest C₇-C₂₁ und X = Cl, Br, I,
CH₃COO, CF₃COO bedeutet.
4. Kationische Lipide der allgemeinen Formel IV,
wobei n = 1-4 sein kann und R einen geradkettigen, gesättigten oder ungesättigten
aliphatischen Rest C₇-C₂₁, Y = CH₂, O, NH, Z = CH₂, O, NH und X = Cl, Br, I, CH₃COO,
CF₃COO bedeutet.
5. Kationische Lipide der allgemeinen Formel V,
wobei n = 2, 3, 4, 6, 8 und m = 2, 3, 6, 8 sein kann und R₁ = H, CH₃, CH₂CH₂OH; R₂ = H, CH₃,
CH₂CH₂OH, (CH₂)₃N⁺(R₁)₃ und X = Cl, Br, I, CH₃COO, CF₃COO bedeutet.
6. Kationische Lipide der allgemeinen Formel VI,
wobei R₁ = H, CH₃, (CH₂)₃NH₂⁺X⁻(CH₂)₃NH₃⁺X⁻, (CH₂)₃NH₃⁺X⁻, R₂= H,
(CH₂)₃NH₃⁺X⁻ und X = Cl, Br, I, CH₃COO, CF₃COO bedeutet.
7. Kationische Lipide der allgemeinen Formel VII,
wobei m = 2-6 sein kann und Y eine Gruppe N(R)₃⁺X⁻ mit R = H, CH₃, (CH₂)₂OH oder eine
Gruppe NH-C(NH₂⁺X⁻)NH₂ mit X = Cl, Br, I, CH₃COO, CF₃COO bedeutet.
8. Kationische Lipide der allgemeinen Formel VIII,
wobei Y eine Gruppe N(R)₃⁺X⁻ mit R = H, CH₃, (CH₂)₂OH oder eine Gruppe NH-C(NH₂⁺X⁻
)NH₂ mit X = Cl, Br, I, CH₃COO, CF₃COO bedeutet.
9. Kationische Lipide der allgemeinen Formel IX und X,
wobei n = 2,3,4,6,8 und m = 2,3,6,8 sein kann und R₁ = H, CH₃, CH₂CH₂OH; R₂ = H, CH₃,
CH₂CH₂OH, (CH₂)₃N⁺(R₁)₃, R = H, CH₃, (CH₂)₂OH, Y eine Gruppe Carbonyl (= O
(Östron)) oder eine Gruppe OH (Östradiol), Z eine Gruppe N(R)₃⁺X⁻ mit R = H, CH₃,
(CH₂)₂OH oder eine Gruppe NH-C(NH₂⁺X⁻)NH₂ und X = Cl, Br, I, CH₃COO, CF₃COO
bedeutet.
