Die Bildung einiger spezieller Oxo-hexahydropyrimidinderivate durch thermische Disproportionierung
von Propandiurethanen ist bekannt (»Liebigs Annalen der Chemie«, Bd. 232, 1886, S. 222 bis 228;
»Journal of the American Chemical Society«, Bd. 79, 1957, S. 3786 bis 3788).
Es wurde nun gefunden, daß man 2-Oxo-hexahydropyrimidine
neben 1,3-Diaminopropanderivaten erhält, wenn man Additionsprodukte, die aus Methylendiurethanender
allgemeinen Formel R — CH(NHCO2R^2,
worin R ein Wasserstoffatom oder eine Alkyl-, Halogenalkyl-,
Vinyl-, Phenylvinyl-, Cyclohexyl-, Benzyl-, Formyl-, Pyridyl-, Furyl-, Thienyl-, N-Alkylindolylgruppe
oder eine gegebenenfalls durch eine Alkyl-, Alkoxy-, Dialkylamino-, Nitro- oder Sulfonsäuregruppe
oder 1 oder 2 Halogenatome substituierte Arylgruppe und R1 eine Alkyl- oder Benzylgruppe bedeutet,
und kationisch polymerisierbaren Monoolefinen in Gegenwart von sauren Katalysatoren hergestellt
worden sind, bei 50 bis 2500C unter sauren oder neutralen Bedingungen oder in Gegenwart von
einer unter 2 Äquivalenten liegenden Menge Base verseift.
Die Herstellung der als Ausgangsverbindungen Verwendung findenden Additionsprodukte ist Gegenstand
eines eigenen älteren Vorschlags (deutsches Patent 1 158 492, belgisches Patent 608 904). Als
kationisch polymerisierbare Olefine werden dabei vorzugsweise verwendet: Isobutylen, Vinylcyclohexan,
Styrole, Vinyläther und Vinylpyridine.
Zur Durchführung des erfindungsgemäßen Verfahrens werden die linearen Additionsprodukte der vorgenannten
Additionsreaktion eingesetzt.
Die Verseifung der N,N'-Bis-carbalkoxy-l,3-diamino-propane erfolgt gegebenenfalls unter erhöhtem
Druck.
Als Basen kommen z. B. Alkali- oder Erdalkalimetalloxyde, -hydroxyde oder Carbonate, wie Lithium-,
Natrium-, Kaliumhydroxyd, Calciumoxyd oder -hydroxyd, Magnesiumoxyd, Bariumhydroxyd, Natrium-
oder Kaliumcarbonat oder Ammoniumhydroxyd oder -carbonat, in Frage.
Bei der sauren Verseifung werden gegebenenfalls nur katalytische Mengen, jedoch auch höhere Mengen
einer beliebigen Säure verwendet. Beispiele für Säuren, die sich für diese Verseifungsreaktion eignen, sind
Chlorwasserstoff, Bromwasserstoff, Schwefel-, Phosphor- oder Borsäure, sowohl stark saure als auch
schwach saure (Sulfonsäure- oder Carbonsäure-) Gruppen enthaltende Ionenaustauscher, Kohlendioxyd,
ferner organische Säuren, wie Essigsäure, Trichloressig säure, p-Toluolsulfonsäure, Benzolsulfonsäure.

Es ist darüber hinaus ohne weiteres auch möglich, die Verseifung in neutralem Medium, also z. B. mit
Wasser bei erhöhten Temperaturen, d. h. bei 100 bis 25O0C, unter Druck zu bewirken.
Das Reaktionsmedium kann sowohl wäßriger, alkoholischer als auch wäßrig-alkoholischer Natur
sein. Als alkoholische Reaktionsmedien seien Methanol, Äthanol, Glykol, Glykolmonobutyläther (Cellosolve)
bzw. deren wäßrige Gemische genannt.
Als Reaktionszeiten kommen z. B. Zeiträume von 1 bis 20 Stunden in Frage.
Die Isolierung der 2-Oxo-hexahydro-pyrimidine kann nach Durchführung der Verseifung auf Grund
der geringen Löslichkeit in z. B. Wasser oder Alkoholen durch Abfiltrieren usw. erfolgen. Die gemäß vorliegendem
Verfahren erhältlichen 2-Oxo-hexahydropyrimidine können als Zwischenprodukte Verwendung
finden, insbesondere zur Herstellung pharmazeutischer Pyrimidinderivate.
Die nachfolgenden Beispielsteile b) erläutern das erfindungsgemäße Verfahren. Die angegebenen Teile
sind Gewichtsteile, sofern nicht anders vermerkt.
a) 100 Teile Paraformaldehyd, 450 Teile Carbamidsäuremethylester,
10 Teile konzentrierte Schwefelsäure und 1000 Teile Benzol werden so lange unter
Rückfluß zum Sieden erhitzt, bis die hierbei mit Hilfe einer Wasserfalle bewirkte azeotrope Wasserabspaltung
beendet ist. Anschließend werden noch 50 Teile Schwefelsäure zugesetzt und bei 900C in 4 Stunden
312 Teile Styrol eingetropft. Man erhitzt 15 Stunden auf 90 bis 1000C, wäscht den sauren Katalysator mit
verdünntem Alkali aus und erhält beim Einengen bis 1600C bei einem Druck von 12 Torr insgesamt
689 Teile des entsprechenden Diurethans mit 9,8 °/0 N
(berechnet für 1: 1-Addukt: 10,5 °/0 N).

b) 200 Teile des nach Teil a) erhaltenen Additionsproduktes werden in 300 Teilen Äthanol gelöst,
100 Teile Wasser werden zugesetzt, in einem Autoklav 60 at Kohlendioxyd aufgepreßt; dann wird die Lösung
4 Stunden auf 23O0C erhitzt. Nach dem Abkühlen werden etwa 70 Teile des 4-Phenyl-2-oxo-hexahydropyrimidins
abfiltriert, das nach Umkristallisieren aus Äthanol bei 237 bis 2380C schmilzt.

Das Filtrat liefert nach Reinigung über das Hydrochlorid noch 40 Teile des l-Phenyl-l,3-diaminopropans.

Die Verseifung kann auch unter Verwendung von 1 % Phosphorsäure als Katalysator in der oben beschriebenen
Weise durchgeführt werden und führt praktisch zum gleichen Ergebnis.
a) 178 Teile Carbamidsäureäthylester, 175 Teile 2,6-Dichlorbenzaldehyd, 400 Teile Benzol und 3 Teile
Borfluorid-Ätherat werden analog Beispiel 1, a) in das Alkyliden-diurethan übergeführt, und dieses wird
nach Zugabe von 20 Teilen Borfluorid-Ätherat an 104 Teilen Styrol addiert. Bei analoger Aufarbeitung
wird das rohe Bisurethan mit einer Ausbeute von 380 Teilen erhalten.

b) 200 Teile des nach Teil a) erhaltenen Additionsproduktes werden mit 300 Teilen Äthanol, 100 Teilen
Wasser und 2 Teilen Phosphorsäure 2 Stunden auf 2300C erhitzt und aus dem Reaktionsprodukt wird
ein festes Material abfiltriert. Dieses wird aus Äthanol umkristallisiert und liefert das 4-Phenyl-6-(2',6'-dichlorphenyl)-2-oxo-hexahydro-pyrimidin
in einer Ausbeute von 25 Teilen mit einem Schmelzpunkt von 278 bis 2800C.

Der saure Katalysator wird durch Filtration entfernt und das rohe Urethan durch Destillation bis 1500C
bei einem Druck von 12 Torr vom Lösungsmittel und von flüchtigen Anteilen befreit. Es wird in einer
Ausbeute von 620 Teilen ein Additionsprodukt mit 5,9 % N und 10,85 % Cl erhalten, während ein reines
1: 1-Addukt theoretisch 6,9% N und 8,8% Cl enthielte,
b) 293 Teile des nach a) hergestellten Adduktes

ίο werden in 300 Teilen Äthanol gelöst und zusammen mit
80 Teilen Kaliumhydroxyd in 300 Teilen Wasser 15 Stunden unter Rückfluß erhitzt. Nach Einengen
der alkoholischen Phase und Zugabe von Wasser fällt in einer Ausbeute von 50 % der Theorie das 4-Phenyl-
1S 6-(4'-chlorphenyl)-2-oxo-hexahydro-pyrimidin an, das
nach Umkristallisieren aus Äthanol bei 249 0C schmilzt.

a) 240 Teile p-Tolylaldehyd und 300 Teile Carbamidsäuremethylester
in 400 Teilen Benzol werden in der im Beispiel 2, a) erläuterten Weise zunächst mit
2 Teilen Borfluorid-Ätherat in das Alkyliden-bisurethan
übergeführt und dann nach Zugabe weiterer 40 Teile Borfluorid-Ätherat bei 90 bis 100° C an 208
Teile Styrol addiert. Nach analoger Aufarbeitung erhält man das Anlagerungsprodukt in einer Ausbeute
von 550 Teilen.

b) 488 Teile des nach a) hergestellten Adduktes werden in einem Autoklav in 500 Teilen Methanol mit
100 Teilen Wasser versetzt, und 60 at Kohlendioxyd werden aufgepreßt. Dann wird 12 Stunden auf 240° C
geheizt. Nach Einengen des Lösungsmittels und Zugabe von Wasser werden 60% der theoretischen
Menge an 4-Phenyl-6-p-tolyl-2-oxo-hexahyd]ro-pyrimidins
erhalten, das nach Umkristallisieren aus Äthanol bei 234 bis 235°C schmilzt.

Das Filtrat des cyclischen Harnstoffes wird eingeengt, wobei noch 117 Teile eines nicht kristallisierenden
Materials erhalten werden, das laut Ultrarotspektrum einen linearen Aminoharnstoff darstellt.
a) 280 Teile p-Chlorbenzaldehyd, 356 Teile Carbamidsäureäthylester,
150 Teile einer mit 2% Divinylbenzol vernetzten Polystyrolsulfonsäure und 800 Teile
Benzol werden so lange unter Rückfluß zum Sieden erhitzt, bis die hierbei mit Hilfe einer Wasserfalle
bewirkte azeotrope Wasserabspaltung beendet ist; dann werden bei 80° C 208 Teile Styrol eingetropft.
Das Gemisch wird 20 Stunden auf 80 bis 1200C erhitzt, wobei eine entsprechende Benzolmenge abdestilliert
wird, um diese Temperatur zu erreichen. Aus dem Filtrat können nach Reinigung über das
Salz noch 20% an l-Phenyl-S-p-tolyl-l^-diaminopropan
isoliert werden.
Claims (2)
1. Verfahren zur Herstellung von 2-Oxo-hexahydro-pyrimidinen
neben 1,3-Diaminopropanderivaten, dadurch gekennzeichnet, daß
Additionsprodukte, die aus Methylendiurethanen der allgemeinen Formel R — CH(NHCO2R1).;,
worin R ein Wasserstoffatom oder eine Alkyl-, Halogenalkyl-, Vinyl-, Phenylvinyl-, Cyclohexyl-,
Benzyl-, Formyl-, Pyridyl-, Furyl-, Thienyl-, N-Alkylindolylgruppe oder eine gegebenenfalls
durch eine Alkyl-, Alkoxy-, Dialkylamino-, Nitro- oder Sulfonsäuregruppe oder 1 oder 2 Halogen-
atome substituierte Arylgruppe und R1 eine Alkyl-
oder Benzylgruppe bedeutet, und kationisch polymerisierbaren Monoolefinen in Gegenwart von
sauren Katalysatoren hergestellt worden sind, bei 50 bis 2500C unter sauren oder neutralen Bedingungen
oder in Gegenwart von einer unter 2 Äquivalenten liegenden Menge Base verseift werden.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß als Additionsprodukt ein solches
verwendet wird, das aus den oben bezeichneten Methylendiurethanen und Styrol erhalten worden
ist.
