Abstract
Die Erfindung betrifft biologisch aktive Peptide mit verbesserter Wirksamkeit, die erfindungsgemäß dadurch erreicht wird, dass die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell durch eine chemische Gruppe substituiert ist, die mindestens ein temporäres oder ein permanentes Dipolmoment aufweist. Insbesondere sind dafür chemische Gruppen geeignet, die unpolar sind, z.B. bizyklische oder polyzyklische aromatische Kohlenwasserstoffe mit einem Dipolmoment. Die erfindungsgemäßen biologisch aktiven Peptide können antimikrobielle, insbesondere antibakterielle, antifungale oder antivirale, oder antitumorale Eigenschaften aufweisen, oder Hormone, Neurotransmitter oder Neuropeptide sein. Sie können zu einer der folgenden Substanzgruppen gehören: Actinomycine, Polymyxine, Bacitracin, Capreomycin, Colistin, Daptomycin, Dermcidin, Duramycin, Feglymycin, Fusafungin, Gramicidine wie Gramicidin S, Nisin, Lugdunin, Plectasin, Pristinamycine, Teixobactin, Tyrothricin. Die erfindungsgemäßen biologisch aktiven Peptide können zur Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und / oder Prophylaxe von Krankheiten, insbesondere von Infektionskrankheiten, verwendet werden.
Description
- Die vorliegende Erfindung betrifft biologisch aktive, insbesondere antimikrobielle Peptide mit verbesserter Wirksamkeit, sowie ihre Verwendung für die Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und / oder Prophylaxe von Krankheiten, primär von Infektionskrankheiten.
- Antimikrobielle Peptide sind das Wachstum von Mikroorganismen, bevorzugt Bakterien, hemmende oder Bakterien abtötende Peptide, an deren strukturellem Aufbau proteinogene und nicht-proteinogene Aminosäuren beteiligt sein können. Außerdem zählen zu den antimikrobiellen Peptiden auch in der Humanmedizin zugelassene, antibiotisch wirksame Glykopeptide, die neben dem Peptidanteil auch Kohlenhydratstrukturen aufweisen, und Depsipeptide, bei denen die Verknüpfung der einzelnen Aminosäurebausteine über eine Esterfunktion anstelle der Amidbindung erfolgt. Häufig handelt es sich dabei um Naturstoffe oder ihre Derivate. Inzwischen sind mehr als 200 antimikrobielle Peptide charakterisiert worden.
- Natürlich vorkommende antimikrobielle Peptide werden sowohl von Prokaryoten als auch von Eukaryoten produziert. Sie wurden in unterschiedlichen Organismen nachgewiesen und dienen z.B. zur Bekämpfung von Nahrungskonkurrenten, potentiellen Pathogenen oder zur Regulation der körpereigenen Mikrobenflora. So produziert der menschliche Organismus antimikrobielle Peptide, die in den Darm oder an die Hautoberfläche abgegeben werden.
- Viele antimikrobielle Peptide besitzen einen anderen Wirkmechanismus als herkömmliche Antibiotika der medizinischen Anwendung (1, 2). Die Substanzen der Gruppe der antimikrobiellen Peptide beeinträchtigen Funktionen der Zellmembran, indem sie entweder ihre osmotische Barriere-Funktion stören oder den Aufbau der Zellhülle bzw. die Biosynthese von lebenswichtigen Bestandteilen der Zellhülle verhindern. Dies geschieht über Porenbildung oder Einlagerung der Peptide in die Membran und die daraus resultierende Störung deren Integrität und / oder Transportfähigkeit. Die Zellmembran kann den elektrochemischen oder chemischen Gradienten nicht mehr aufrechterhalten und den Verlust von Ionen und das Einströmen von Wasser in die Zelle nicht kontrollieren, die als Folge anschwillt und lysiert.
- Die biologische Wirkung antimikrobieller Peptide kann gegen verschiedene Eukaryoten, Prokaryoten und auch Viren gerichtet sein. Inzwischen sind mehrere Peptide bekannt, die nicht nur gegen Bakterien wirksam sind, sondern auch gegen Viren, Pilze und sogar Tumorzellen.
- Zu allgemeinen Eigenschaften, Wirkmechanismen sowie der therapeutischen Bedeutung von antimikrobiellen Peptiden kann auf einen Review-Artikel (3) verwiesen werden.
- Bei vielen bekannten Wirkstoffen handelt es sich um Oligopeptide, die bis zu zehn Aminosäuren enthalten. Größere antimikrobielle Peptide sind ebenfalls beschrieben worden und werden als Polypeptide bezeichnet. Viele dieser Substanzen liegen in Form von Zyklopeptiden vor, deren Aminosäuresequenz kovalent zu mindestens einem Ring geschlossen ist.
- Auf der Basis von strukturell sehr unterschiedlichen antimikrobiellen Peptiden werden Forschungsreagenzien, Wertstoffe in Industrie und Landwirtschaft, Zusatzstoffe in der Lebensmittelherstellung oder auch pharmazeutisch anwendbare Wirkstoffe bis hin zu zugelassenen Arzneistoffen etabliert. Zu ihnen gehören beispielsweise Gramicidine wie Gramicidin S, Bacitracin, Tyrocidin, Capreomycin, Polymyxine wie Polymyxin B und Colistin. Weitere antimikrobielle Peptide befinden sich derzeit in der Entwicklung (1).
- Verglichen mit niedermolekularen antimikrobiellen Substanzen haben Peptide einige Vorteile, insbesondere ihre geringe Toxizität. Wenn antimikrobielle Peptide aus proteinogenen Aminosäuren aufgebaut sind, sind ihre Metaboliten meistens nicht toxisch, im Gegensatz zu den Metaboliten vieler synthetisch hergestellten organischer Substanzen. Umgekehrt ist die Molekülgröße antimikrobieller Peptide klein genug und daher in der Regel nicht mit problematischen Antworten des Immunsystems bei der Anwendung behaftet. Sie sind weniger immunogen, verglichen mit größeren Proteinen oder Antikörpern. Zusätzlich weisen Peptide geringere Halbwertszeiten auf, so dass die Gefahr einer Gewebsakkumulation von Peptiden sehr gering ist. Auf der anderen Seite können Peptide tiefer ins Gewebe eindringen. Hinzu kommen geringere Herstellungskosten, höhere Aktivität und hohe Stabilität z.B. bei Raumtemperatur (4).
- Kürzlich wurde ein neues antimikrobielles Peptid, Lugdunin, aus Staphylococcus lugdunensis isoliert und charakterisiert (4, 5). Es ist der erste bekannte Vertreter einer neuen Klasse von makrozyklischen Thiazolidin-Anibiotika, die als Fibupeptide (6) bezeichnet werden. Lugdunin inhibiert neben anderen Organismen das Wachstum des human-pathogenen multi-resistenten Erregers Staphylococcus aureus USA300. Wie bei anderen mikrobiellen Peptiden basiert der Wirkmechanismus von Lugdunin auf einer Interaktion mit der bakteriellen Membran (6).
- Die Aufgabe der vorliegenden Erfindung ist es, biologisch aktive, insbesondere antimikrobielle Peptide mit verbesserter Wirksamkeit sowie ihre Verwendung für die Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und / oder Prophylaxe von Krankheiten, insbesondere von Infektionskrankheiten bereitzustellen.
- Diese Aufgabe wird durch die Bereitstellung von biologisch aktiven Peptiden gelöst, bei denen die Seitenkette an mindestens einem α-Kohlenstoffatom (C-2-Kohlenstoffatom von 2-Aminocarbonsäuren) komplett oder teilweise durch eine chemische Gruppe substituiert ist, die mindestens ein temporäres oder ein permanentes Dipolmoment aufweist. Insbesondere ist die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder teilweise substituiert, unpolar.
- Das Ziel dieser erfindungsgemäßen Substitution ist es, eine durch das zusätzlich eingeführte Dipolmoment herbeigeführte Verstärkung der molekularen Wechselwirkung zwischen dem Peptid und der Membran zu erreichen. Diese Wechselwirkung steigert die Wirksamkeit der Peptide.
- Zur Steigerung der Wirksamkeit von biologisch aktiven Peptiden sind insbesondere Substitutionen mit chemischen Gruppen geeignet, deren Dipolmoment größer als 0,5 Debye ist.
- Besonders geeignet sind Substitutionen mit aromatischen Kohlenwasserstoffen, die ein Dipolmoment aufweisen.
- Insbesondere sind bizyklische oder polyzyklische aromatische Kohlenwasserstoffe mit einem Dipolmoment als Substituenten geeignet. Die aromatischen Kohlenstoffatome können nichtbenzoid oder benzoid sein.
- Ein besonders bevorzugtes Ausführungsbeispiel stellen biologisch aktive Peptide dar, bei denen die Seitenkette an mindestens einem α-Kohlenstoffatom durch einen Azulen-Rest oder einen 1-Carboxy-1-(6-(dialkylamino)naphthalen-2-yl)-Rest oder einen 1-Carboxy-1-(6-(dialkylamino)anthracen-2-yl)-Rest oder einen Pentafluorphenyl-alanyl-Rest (2-Amino-3-(perfluorphenyl)propanoyl-) oder einen 3-(Benzothiazol-2-yl)-L-alanyl-Rest substituiert ist (
1 ). - Als Beispiel enthält das aromatische Ringsystem von Azulen zehn Kohlenstoffatome in zyklisch planarer Anordnung und verfügt über zehn π-Elektronen mit einer nichtalternierenden Verteilung der Elektronendichte. Das Dipolmoment des Azulen-Moleküls beträgt etwa 1 Debye (6).
- Ferner können Substitutionen mit Azulen-, Naphthalin- oder Anthracen-Derivaten verwendet werden, um die Wirksamkeit von biologisch aktiven Peptiden erfindungsgemäß zu erhöhen. Diese können synthetischen oder auch natürlichen Ursprungs sein, wie beispielsweise Chamazulen, Matricin, Guajazulen, Lactaroviolin, 1,4-Dimethyl-7-isopropylazulen, Vetivazulen oder 4,8-Dimethyl-2-isopropylazulen. Letztere könnten aufgrund ihrer im Vergleich zur Azulen-Stammverbindung höheren chemischen Stabilität zusätzliche Vorteile in der Handhabung erfindungsgemäßer biologisch aktiver Peptide haben.
- Derivate der Azulen-, Naphthalin- oder Anthracen-Reste umfassen Halogenierungen (Fluor-, Chlor-, Brom-), Alkylierungen, Acetylierungen, Amidierungen, Substitutionen mit aromatischen Verbindungen oder andere organische Modifikationen.
- Biologisch aktive Peptide, deren Wirksamkeit durch die erfindungsgemäße Einführung eines zusätzlichen Dipolmoments gesteigert werden kann, können z.B. antimikrobielle Peptide (darunter antibakterielle Peptide, antifungale Peptide und antivirale Peptide), Hormone, Neurotransmitter oder Neuropeptide sein. Ihre Wirkung, die es erfindungsgemäß zu verbessern gilt, kann beispielswese eine antibakteriell, antivirale, antifungale, antitumorale oder eine sonstige physiologische Aktivität umfassen (8, 9).
- Als Beispiele für antimikrobielle Peptide, deren Wirksamkeit erfindungsgemäß erhöht werden kann, kommen folgende Substanzen und Substanzklassen in Frage: Actinomycine, Polymyxine, Bacitracin, Capreomycin, Colistin, Daptomycin, Dermcidin, Duramycin, Feglymycin, Fusafungin, Gramicidine wie Gramicidin S, Nisin, Lugdunin, Plectasin, Pristinamycine, Teixobactin, Tyrothricin.
- Besonders bevorzugt kann die Wirksamkeit von makrozyklischen Thiazolidin-Peptiden durch die o.g. Substitution verbessert werden.
- Ein Ausführungsbeispiel für ein solches makrozyklisches Thiazolidin-Peptid mit verbesserter Wirksamkeit ist 6-D-Ala(Azulen)-Lugdunin (
2 ). Zur Herstellung dieser Substanz wurde das Valin an Position 6 in Lugdunin durch Azulen-Alanin substituiert. Die erfindungsgemäße Substanz 6-D-Ala(Azulen)-Lugdunin zeigte eine 9-fach erhöhte Aktivität im Vergleich zum unsubstituierten Lugdunin (s. Ausführungsbeispiele). - In einem weiteren Ausführungsbeispiel zeigt die erfindungsgemäß substituierte Substanz D-Pentafluorphenylalanin-Gramicidin S eine 2-fach erhöhte Aktivität im Vergleich zur Ausgangssubstanz Gramicidin S.
- Die Erfindung umfasst ferner die Verwendung von erfindungsgemäßen biologisch aktiven Peptiden, insbesondere von antimikrobiellen Peptiden, für die Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und / oder Prophylaxe von Krankheiten, insbesondere von Infektionskrankheiten.

- Darstellung von 6-D-Ala(Azulen)-Lugdunin
- -6-D-Ala(Azufen)-Lugdunin wurde mit der Merrifield-Festphasenpeptidsynthese unter Anwendung der Fmoc-Schutzgruppenstrategie dargestellt. Die Aktivierung der Carboxylatfunktion erfolgte mittels HATU (O-(7-Azabenzotriazol-1-yl)-N,N,N',N'tetramethyluronium-hexafluorphosphat) und HOBt. Als Lösungsmittel für die Kopplung der Aminosäuren wurde MeCN verwendet. Die Kupplung von Fmoc-D-Ala(Az)-OH wurde aufgrund der schlechten Löslichkeit dieser Aminosäure in MeCN/DMF (50/50) durchgeführt. Zur Synthese der Peptidaldehyde wurde das kommerzielle Spezialharz H-Väl-H NovaSyn® TG resin (Novabiochem) verwendet.
- Darstellung von D-Pentafluorphehylalanin-Gramicidin S
- 6-D-Pentafluorphenylalanin-Gramicidin S wurde mit der Merrifield-Festphasenpeptidsynthese unter Anwendung der Fmoc-Schutzgruppenstrategie dargestellt. Die Kupplung der ersten Aminosäure Fmoc-D-Phe-OH erfolgte mit DIPEA in DCM/DMF (90/10) auf ein Tritylchlorid-Polystyrol-Harz (TCP-Harz). Die Aktivierung der Carboxylatfunktion erfolgte mittels HATU (O-(7-Azabenzotriazol-1-yl)-N,N,N',N'tetramethyluronium-hexafluorphosphat) und HOBt. Als Lösungsmittel für die Kupplung der Aminosäuren wurde DMF verwendet.
- Die Abspaltung des linearen Gramicidin S-Vorläufers erfolgte durch Inkubation in TFA/MilliQ-H2O/Thioanisol/Phenol. (95/2.5/2.5/Spatelspitze). Dieser wurde in DCM unter Zugabe von HOBt, PyBop und DIPEA makrozyklisiert. Nach der Makrozyklisierung wurden die Dde-Schutzgruppen am Ornithin mit 2 % Hydrazin in THF abgespalten.
- Test auf minimale Hemmstoffkonzentration (MHK):
- Der MHK-Test wurde wie bereits beschrieben durchgeführt (5), dabei wurden folgende Kontrollen verwendet:
Negativkontrolle: 100 % DMSO Positivkontrollen: Daptomycin (10 mg/mL in DMSO, MHK: 6,25 µg/mL) Vancomycin (10 mg/mL in MilliQ-H2O, MHK: 0,78 µg/mL) Lugdunin (10 mg/mL in DMSO, MHK: 3,13 µg/mL) Gramicidin S (10 mq/mL in DMSO, MHK: 6,25 µg/mL) - Die MHK-Werte jedes Peptids wurden als drei biologische Replikate in voneinander unabhängigen Assays bestimmt (Wachstum 21 h bei 37°C, 160 rpm):

Claims (13)
- Biologisch aktive Peptide mit verbesserter Wirksamkeit, dadurch gekennzeichnet, dass die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell durch eine chemische Gruppe substituiert ist, die mindestens ein temporäres oder ein permanentes Dipolmoment aufweist.
- Biologisch aktive Peptide nach
Anspruch 1 , dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, unpolar ist. - Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, mindestens einen aromatischen Kohlenwasserstoff mit einem Dipolmoment aufweist.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, mindestens einen bizyklischen oder polyzyklischen aromatischen Kohlenwasserstoff mit einem Dipolmoment aufweist.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, einen Azulen-Rest oder einen 1-Carboxy-1-(6-(dialkylamino)naphthalen-2-yl)-Rest oder einen 1-Carboxy-1-(6-(dialkylamino)anthracen-2-yl)-Rest oder einen Pentafluorphenyl-alanyl-Rest (2-Amino-3-(perfluorphenyl)propanoyl-) oder einen 3-(Benzothiazol-2-yl)-L-alanyl-Rest aufweist.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, ein Azulen-, ein Naphthalin- oder ein Anthracen-Derivat aufweist.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die chemische Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, ein Chamazulen, Matricin, Guajazulen, Lactaroviolin, 1,4-Dimethyl-7-isopropylazulen, Vetivazulen oder 4,8-Dimethyl-2-isopropylazulen oder ein Derivat mindestens einer dieser Substanzen aufweist.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass die Derivate der chemischen Gruppe, die die Seitenkette an mindestens einem α-Kohlenstoffatom komplett oder partiell substituiert, zusätzlich einen Halogen-, Alkyl-, Acetyl-, Amid-Rest, oder einen aromatischen Ring enthalten.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass sie antimikrobielle, insbesondere antibakterielle, antifungale oder antivirale, oder antitumorale Eigenschaften aufweisen, oder Hormone, Neurotransmitter oder Neuropeptide sind, oder eine sonstige physiologische Aktivität aufweisen.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass sie zu einer der folgenden Substanzgruppen gehören: Actinomycine, Polymyxine, Bacitracin, Capreomycin, Colistin, Daptomycin, Dermcidin, Duramycin, Feglymycin, Fusafungin, Gramicidine wie Gramicidin S, Nisin, Lugdunin, Plectasin, Pristinamycine, Teixobactin, Tyrothricin.
- Biologisch aktive Peptide nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass sie eine der Substanzen 6-D-Ala(Azulen)-Lugdunin oder D-Pentafluorphenylalanin-Gramicidin S umfassen.
- Verwendung von biologisch aktiven Peptiden nach einem der vorangehenden Ansprüche zur Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und / oder Prophylaxe von Krankheiten, insbesondere von Infektionskrankheiten.
- Pharmazeutische Zusammensetzung, die mindestens ein biologisch aktives Peptid nach einem der vorangehenden Ansprüche enthält.
