Abstract
Die Erfindung betrifft 2-Mercapto-4,5-diarylimidazolderivate der Formel I DOLLAR F1 worin die Reste R·1·, R·2·, R·3· und R·4· die in der Beschreibung angegebene Bedeutung besitzen. Die erfindungsgemäßen Verbindungen besitzen eine immunmodulierende und Cyclooxygenase-inhibierende Wirkung und sind daher geeignet zur Behandlung von Erkrankungen, die mit einer Störung des Immunsystems im Zusammenhang stehen.
Description
Die vorliegende Erfindung betrifft 2-Mercapto-4,5-diaryl-
imidazolderivate mit immunmodulierender und Cyclooxygenase
inhibierender Wirkung, pharmazeutische Mittel, welche die Ver
bindung enthalten, und ihre Verwendung in der Pharmazie.
Die Wirkung herkömmlicher nicht-steroidaler Antiphlogistika,
wie Acetylsalicylsäure, beruht im Wesentlichen auf der Hemmung
der Cyclooxygenase, einem Enzym der Arachidonsäure-Kaskade,
das auch als Prostaglandin-G/H-Synthase bekannt ist. Inzwi
schen wurden zwei unterschiedliche Formen der Cyclooxygenase
gefunden, die als COX-1 und COX-2 bezeichnet werden. Trotz
zahlreicher Untersuchungen ist die biochemische Wirkungsweise
der beiden Enzyme noch nicht vollständig geklärt. Verschiedene
Studien haben jedoch gezeigt, dass sie eine wesentliche Rolle
bei zahlreichen Erkrankungen und entzündlichen Prozessen
spielen.
Pharmakologisch wirksame Imidazolverbindungen, welche die
Cyclooxygenase-1 und -2 (Cox-1 und Cox-2) hemmen, sind bereits
bekannt. In J Med. Chem. 1996, 39, 3927-37 werden beispiels
weise Imidazolderivate mit 5-Lipoxygenase- und Cyclooxygenase
inhibierender Wirkung beschrieben, wobei 2-(4-Methylsulfinyl
phenyl)-4-(4-fluorphenyl-5-(pyrid-4-yl)-imidazol auch eine
Cytokin-inhibierende Wirkung besitzt.
Die WO 95/00501 beschreibt ferner Phenylheterocyclen, die eine
Cyclooxygenaseinhibierende Wirkung besitzen, darunter auch
4,5-Diaryl-substituierte Imidazole, zur Behandlung von Erkran
kungen, die mit der Cyclooxygenase in Zusammenhang stehen.
Weiter bekannt sind pharmazeutisch wirksame Imidazolderivate,
die 4,5-Di(hetero)arylimidazol-Elemente enthalten und in 2-
Position eine Substitution aufweisen. Das US-Patent 4,585,771
offenbart beispielsweise 4,5-Diphenylimidazolderivate, die an
der 2-Position mit einem Pyrrolyl-, Indolyl-, Imidazolyl- oder
Thiazolylrest substituiert sind und eine antiinflammatorische
und antiallergische Aktivität besitzen. Ferner beschreiben die
US-Patente 4,528,298 und 4,402,960 4,5-Di(hetero)arylimidazol
derivate, die in 2-Position über eine Thio-, Sulfinyl- oder
Sulfonylgruppe mit einem Phenyl-, Pyridyl-, N-Oxypyridyl-,
Pyrimidyl-, Thiazolyl- oder Thienylrest substituiert sind, und
eine antiinflammatorische und antiallergische Aktivität be
sitzen.
DE 198 42 833 betrifft 4-Heteroarly-5-phenylimidazolderivate,
die an der 2- Position mit einer Phenylalkylthiogruppe
substituiert sind. Diese Verbindungen wirken als
Entzündungshemmstoffe und Hemmstoffe der Cytokinfreisetzung.
Die WO 99/03837, WO 93/14081 und DE 198 42 833 beschreiben 2-
substituierte Imidazole, die die Synthese einer Anzahl
inflammatorischer Cytokine hemmen. Diese Verbindungen haben
wahlweise einen weiteren Substituenten am Stickstoffatom in
der 1-Position.
Es ist bekannt, dass die herkömmlichen nicht-steroidalen Anti
phlogistika eine Reihe unerwünschter Nebenwirkungen aufweisen,
insbesondere gastrointestinale Nebenwirkungen, Nephrotoxizität
und allergische Reaktionen. Ferner wurde gefunden, dass die
bekannten Verbindungen nicht stabil und schwer zu verarbeiten
sind oder eine geringe Wirksamkeit aufweisen.
Trotz der zahlreichen bekannten Verbindungen besteht daher
weiterhin ein Bedarf nach Verbindungen mit antiphlogistischer,
antipyretischer und antialgetischer Wirkung. Insbesondere
durch die Bedeutung der COX-Enzyme für eine Reihe weiterer
pathologischer Prozesse, z. B. Coloncarcinom, überschießende
Angiogenese, excitatorische neuronale Prozesse, Wundheilung
etc., kommen der Entwicklung von COX-Inhibitoren neue und
zusätzliche Bedeutungen zu.
Die Aufgabe der Erfindung ist die Bereitstellung solcher Ver
bindungen.
Überraschenderweise wurde nun gefunden, dass bestimmte 2-Mer
capto-4,5-diarylimidazolderivate stabile, leicht zu verarbeitende
Verbindungen darstellen, die eine hohe Cyclooxygenase
inhibierende Wirkung mit variabler COX-1/COX-2-Selektivität
aufweisen.
Gegenstand der vorliegenden Erfindung sind daher die
2-Mercapto-4,5-diarylimidazolderivate der Formel I
worin

Soweit die erfindungsgemäßen Verbindungen Asymetriezentren
aufweisen, sind Racemate sowie optische Isomere (Enantiomere,
Diastereomere) umfasst.
Der Ausdruck "Alkyl" (auch in Verbindungen mit anderen Grup
pen, wie Aminoalkyl, Alkylsulfonyl etc.) umfasst geradkettige
und verzweigte Alkylgruppen mit vorzugsweise 1 bis 6 und
insbesondere 1 bis 4 C-Atomen, wie Methyl, Ethyl, n- und i-
Propyl, n-, i- und t-Butyl, sec-Butyl, n-Pentyl und n-Hexyl.
Wenn Alkyl mit Halogen substituiert ist, sind vorzugsweise 1,
2 oder 3 Halogensubstituenten vorhanden. Bevorzugte Beispiele
für halogensubstituiertes Alkyl sind Chlormethyl, CHF2 und
insbesondere CF3.
Der Ausdruck "Alkylen" steht vorzugsweise für eine
geradkettige oder verzweigte Alkylengruppe mit 1 bis 6,
vorzugsweise 1 bis 4 C-Atomen, wie Methylen, Ethylen,
Ethyliden, 1,2- oder 1,3-Propylen, 1,4-Butylen oder 1,6-
Hexylen.
Der Ausdruck "Alkenyl" steht für eine geradkettige oder
verzweigte Alkenylgruppe mit 2 bis 6 und insbesondere 2 bis 4
C-Atomen, wie -CH=CH2, -CH=CH-CH3 oder -CH2-CH=CH2.
Der Ausdruck "Halogen" steht für ein Fluor-, Chlor-, Brom-
oder Iodatom, insbesondere ein Fluor- oder Chloratom.
Nicht-aromatische heterocyclische Reste sind vorzugsweise
Piperidinyl, Pyranyl, Morpholinyl oder Pyrrolidinyl.
Bevorzugte aromatische heterocyclische Reste sind Pyridyl,
insbesondere 2-, 3- oder 4-Pyridyl, Furyl, insbesondere 2-
Furyl, Thienyl, insbesondere 2-Thienyl, Pyrimidinyl, Pyrrolyl,
Imidazolyl, Oxazolyl, Isoxazolyl oder Thiazolyl. Bevorzugt
sind Furyl und Thienyl.
Wenn R7 für einen mit einem Benzolring kondensierten Rest
steht, handelt es sich vorzugsweise um Chinolyl, insbesondere
2-Chinolyl, oder Benzimidazol, insbesondere 2-Benzimidazol.
Bevorzugt sind Verbindungen der Formel I, wobei einer der
Reste R3 und R4 für eine OH- oder C1-C6-Alkoxygruppe,
vorzugsweise eine Methoxygruppe steht. Weiter bevorzugt stehen
beide Reste R3 und R4 für eine OH- oder C1-C6-Alkoxygruppe,
vorzugsweise eine Methoxygruppe. Besonders bevorzugt stehen
einer oder beide Reste R3 und R4 für eine C1-C6-Alkoxygruppe,
insbesondere für OCH3.
Weiterhin bevorzugt sind Verbindungen der Formel I, wobei R2
für Wasserstoff oder Methyl steht, vorzugsweise Wasserstoff.
R1 steht vorzugsweise für CONR5R6, insbesondere für CON (C2H5)2;
C1-C6-Alkylen-R7, wobei R7 für den erwähnten aromatischen
heterocyclischen Rest oder für COOR8 steht, insbesondere CH2-
Pyridyl oder CH2COOH; oder C1-C6-Alkylen-NR10-CO-R11, wobei R11
für C1-C6-Alkyl oder Phenyl steht, insbesondere CH2CH2NHCO-
isopropyl.
Die physiologisch verträglichen Salze können im vorliegenden
Fall Säureadditions- oder Basenadditionssalze sein. Für Säure
additionssalze werden anorganische Säuren wie Salzsäure,
Schwefelsäure oder Phosphorsäure eingesetzt oder organische
Säuren wie Weinsäure, Citronensäure, Maleinsäure, Fumarsäure,
Äpfelsäure, Mandelsäure, Ascorbinsäure, Gluconsäure und der
gleichen.
Die Herstellung der erfindungsgemäßen Verbindungen erfolgt in
einem zweistufigen Verfahren. Zunächst erfolgt die Synthese
eines 4,5-Diaryl-1H-imidazol-2-thiols. Dieses wird dann im
zweiten Schritt so umgesetzt, dass die gewünschten Substituen
ten eingeführt werden. Die Substituenten werden vorzugsweise
in den Positionen 1 und 2 am Stickstoff bzw. Schwefelatom
eingeführt.
Die Herstellung der erfindungsgemäßen Verbindungen wird
beispielhaft an Verbindungen erläutert, worin R3 und R4 beide
für eine Methoxygruppe stehen. Verbindungen mit anderen Resten
R3 und R4 können in entsprechender Weise hergestellt werden.
Durch Umsetzen der entsprechenden Benzoine (Verbindungen 1),
die durch Benzoinkondensation, z. B. gemäß Houben-Weyl,
Methoden der organischen Chemie, Band 7/2a, Thieme-Verlag,
Stuttgart, 1973, 653-671, erhältlich sind, mit Thioharnstoff
werden nach dem Verfahren aus Schema I symmetrisch
substituierte 4,5-Diaryl-1H-imidazol-2-thiole (Verbindungen 2)
synthetisiert. Das Verfahren ist in Liebigs Ann. Chem. 1895,
284, 24-29 beschrieben.
Unsymmetrisch substituierte 4,5-Diaryl-1H-imidazol-2-thiole
werden hingegen vorzugsweise nach dem Verfahren aus Schema II
synthetisiert. Dabei wird ein Aldehyd in Gegenwart einer Säure
mit 1,3-Propandithiol umgesetzt, wobei der elektrophile
Kohlenstoff der Carbonylgruppe durch Umwandlung zum cyclischen
Dithioacetal umgepolt wird. Die Methylgruppe zwischen den
beiden Schwefelatomen lässt sich mit einer starken Base wie n-
Butyllithium deprotonieren, was die Addition an einen anderen
Aldehyd ermöglicht, siehe J. Org. Chem. 1966, 31, 4303-4304.
Die Thioacetalfunktion wird dann durch Zugabe eines
Quecksilbersalzes in die entsprechende Carbonylverbindung
überführt, siehe Liebigs Ann. Chem. 1981, 10-19. Die so
erhaltenen Benzoine werden dann nach der bereits beschriebenen
Kondensation mit Thioharstoff umgesetzt, wobei die
unsymmetrisch substituierten 4,5-Diaryl-1H-imidazol-2-thiole
der Formel 2 in Schema 2 entstehen.
Die Substitutionen der 4,5-Diaryl-1H-imidazol-2-thiole können
durch übliche Verfahren erfolgen. Dazu gehören Reaktionen zur
Substitution des Schwefels in Position 2 des Imidazolrings
durch nukleophile Substitution zur Einführung einer Gruppe mit
einer Alkyl-, Amin-, Arylketon-, Essigsäureethylester-,
Carbonyl- und substituierter Carbonylfunktion.
Die nukleophile Substitution der Thiole erfolgt durch
Umsetzung mit der einzuführenden Seitenkette, die eine
geeignete Abgangsgruppe aufweist, z. B. ein Halogenatom,
insbesondere ein Brom- oder Iodatom, die Methansulfonyl- oder
Toluolsulfonylgruppe. Die Umsetzung erfolgt üblicherweise in
Gegenwart einer Base, wie Natrium- oder Kaliumcarbonat oder es
wird das Natrium- oder Kaliumsalz des Thiols eingesetzt, das
durch Umsetzung des Thiols mit einer Base, wie einem Natrium-
oder Kaliumalkoholat, erzeugt wird. Die Substitution wird im
allgemeinen in einem polaren organischen Lösungsmittel, wie
Methanol, Ethanol, Dimethylformamid etc. und bei einer
Temperatur im Bereich von Raumtemperatur bis zum Siedepunkt
des Reaktionsgemisches durchgeführt.
Die Einführung einer Seitenkette mit einer Carbonsäureamid-
oder Sulfonamidgruppierung erfolgt zweckmäßigerweise gemäß dem
nachfolgenden Schema 3:
Die Verbindung T15 wird durch nucleophile Substitution wie
oben beschrieben hergestellt. Die Umsetzung von T15 erfolgt
dann in einem polaren, aprotischen organischen Lösugsmittel,
wie Tetrahydrofuran (THF), Dioxan etc., bei einer Temperatur
im Bereich von -60°C bis etwa Umgebungstemperatur.
Die Herstellung der Verbindungen der Formel I mit einer
Harnstoffgruppe in der Seitenkette erfolgt in analoger Weise.
Die Oxidation des Schwefels in 2-Position des Imidazols zur
entsprechenden Sulfinyl- oder Sulfonylverbindung erfolgt nach
üblichen Verfahren mit einem geeigneten Oxidationsmittel, z. B.
m-Chlorperbenzoesäure, Wasserstoffperoxid, Benzoylperoxid etc.
Die erfindungsgemäßen Verbindungen sind potente selektive In
hibitoren der Cyclooxygenase. Die Cyclooxygenase ist ein Enzym
der Arachidonsäure-Kaskade, bei der aus der C20-Carbonsäure
Arachidonsäure Prostaglandine, beispielsweise die Prostaglan
dine G2 und H2, und Thromboxane gebildet werden.
Die erfindungsgemäßen Verbindungen eignen sich durch ihre die
Cyclooxygenase-hemmende Wirkung als antiphlogistische, anti
pyretische und analgetische Wirkstoffe zur Behandlung von Er
krankungen, die im Zusammenhang mit einer Störung des Immun
systems stehen. Sie eignen sich beispielsweise zur Hemmung
vorzeitig einsetzender Wehen, zur Behandlung von Krebs, bspw.
Kolonkarzinom, und Morbus Alzheimer. Weitere Verwendungsmög
lichkeiten bieten sich bei der Behandlung von Autoimmunerkran
kungen, rheumatischer Arthritis, Gicht, septischem Schock, Os
teoporosis, neuropathischem Schmerz, Alopezie, Psoriasis,
akuter Pankreatitis, Abstoßungsreaktionen bei allogenen Trans
plantaten, allergisch bedingter Lungenentzündung, Arterio
sklerose, Multipler Sklerose, Kachexie und Inflammatory-Bowel-
Disease (IBD), adenomatöse Polyposis (Gardner-Syndrom), Colon-
Carcinom, zur Inhibierung der Angiogenese im Zusammenhang mit
Tumorerkrankungen. Darüber hinaus sind die Verbindungen
brauchbar zur topischen Behandlung von Entzündungen
unterschiedlicher Genesis (Kontaktekzem, Erytheme, wie UV-
Erythem).
Die erfindungsgemäßen Verbindungen können entweder als einzel
ne therapeutische Wirkstoffe oder als Mischungen mit anderen
therapeutischen Wirkstoffen verabreicht werden. Die Verbindun
gen können systemisch oder als topische Wirkstoffe verabreicht
werden. Die Verbindungen können alleine verabreicht werden, im
Allgemeinen werden sie jedoch in Form pharmazeutischer Mittel
dosiert und verabreicht, d. h. als Mischungen der Wirkstoffe
mit geeigneten pharmazeutischen Trägern oder Verdünnungs
mitteln. Zur systemischen Wirkung können die Verbindungen oder
Mittel oral oder parenteral verabreicht werden, vorzugsweise
werden sie in oralen Dosierungsformen gegeben.
Die Art des pharmazeutischen Mittels oder Trägers bzw. des
Verdünnungsmittels hängt von der gewünschten Verabreichungs
form ab. Orale Mittel können bspw. als Tabletten oder Kapseln
vorliegen und können übliche Exzipienzien enthalten wie Binde
mittel (z. B. Sirup, Akazia, Gelatine, Sorbit, Tragant oder
Polyvinylpyrrolidon), Füllstoffe (z. B. Lactose, Zucker, Mais
stärke, Calciumphosphat, Sorbit oder Glycin), Gleitmittel
(z. B. Magnesiumstearat, Talcum, Polyethylenglycol oder Silici
umdioxid), desintegrierende Mittel (z. B. Stärke) oder Netz
mittel (z. B. Natriumlaurylsulfat). Flüssige Oralpräparate
können in Form wässriger oder öliger Suspensionen, Lösungen,
Emulsionen, Sirupe, Elixiere oder Sprays und dergleichen sein.
Sie können auch als Trockenpulver vorliegen, das zur Rekonsti
tution mit Wasser oder einem anderen geeigneten Träger aufbe
reitet wird. Derartige flüssige Präparate können übliche Addi
tive, beispielsweise Suspendiermittel, Geschmacksstoffe, Ver
dünnungsmittel oder Emulgatoren enthalten. Für die parenterale
Verabreichung kann man Lösungen oder Suspensionen mit üblichen
pharmazeutischen Trägern einsetzen.
Die erfindungsgemäßen Verbindungen oder Mittel können an Säu
ger (Mensch oder Tier) in einer Dosis von etwa 0,5 bis 100 mg
pro kg Körpergewicht pro Tag verabreicht werden. Sie können in
einer Einzeldosis oder in mehreren Dosen gegeben werden. Das
Wirkungsspektrum der Verbindungen als Inhibitoren der Cyclo
oxygenasen (COX-1 und COX-2) wurde anhand nachstehender Test
systeme untersucht.
Die erfindungsgemäßen Verbindungen können auch als pharmazeu
tische Zubereitungen zur topischen Verabreichung vorliegen,
beispielsweise in Form von Emulsionen, Pasten, Salben, Gelen,
Crems, Lotionen, Puder oder Sprays. Diese pharmazeutischen
Zubereitungen können neben dem oder den Wirkstoffen übliche
Trägerstoffe enthalten. Zubereitungen in Form von Salben,
Pasten, Crems oder Gelen können z. B. tierische und/oder
pflanzliche Fette, Wachse, Paraffine, Stärke, Tragant, Cellu
losederivate, Polyethylenglykole, Silikone, Bentonite, Kiesel
säure, Talkum und Zinkoxid oder Gemische davon enthalten.
Puder und Sprays können neben den Wirkstoffen z. B. Milchzucker,
Talkum, Kieselsäure, Aluminiumhydroxid, Calciumsilikat
und Polyamidpulver enthalten. Emulsionen und Lotionen können
übliche Lösungsmittel, Verdünner und Emulgatoren enthalten.
Die Hemmung des Cyclooxygenase-Weges erfolgt durch fluori
metrische Quantifizierung von Malondialdehyd (MDA). Dieser
kondensiert im sauren Medium mit der doppelten molaren Menge
Thiobarbitursäure zu einem roten Pigmentfarbstoff, der eine
Fluoreszenz bei λExcitation: 533 nm, λEmission: 550 nm aufweist.
Eine aus Schweineblut gewonnene Thrombozytensuspension wird 10 min
bei 37°C mit der Testsubstanz vorinkubiert. Die Konzentra
tion der Suspension sollte 8 × 108 Zellen/ml betragen. Die
Arachidonsäurekaskade wird anschließend durch Zugabe des Anti
biotikums Calcium-Ionophor A23187 (5 mol/l) aktiviert. Nach
weiteren 10 min bei 37°C wird die Reaktion mit einer Trichlor
essigsäurelösung gestoppt. Die Proben werden zentrifugiert und
der Überstand mit Thiobarbitursäure 30 min bei 70°C inkubiert.
Nach dem Abkühlen (30 min bei Raumtemperatur) werden die
Proben fluorimetrisch bei 533/550 nm gemessen und daraus an
hand einer Eichkurve die IC50-Werte der Testverbindungen
ermittelt.
Das Verfahren erfolgt an LPS-stimulierten humanen Monocyten,
die durch Ficoll-Extraktion gewonnen werden. Als Messgröße
dient das gebildete PGE2, das mittels ELISA bestimmt wird.
Die Ergebnisse der in-vitro-Tests für die COX-1-Hemmung sind
in nachstehenden Tabellen 1 bis 17 gezeigt.
Erfindungsgemäß besonders geeignete Verbindungen sind die
Verbindungen T2, T11, T51, T70, T71 und T80 mit IC50-Werten im
molaren Bereich von 10-9. Diese Verbindungen zeigen eine
besonders hohe Wirksamkeit zur Hemmung der Cyclooxygenase-1.
Die Ergebnisse der in-vitro-Tests für die COX-2-Hemmung sind
in nachstehender Tabelle 18 gezeigt.
Es wurde gefunden, dass die erfindungsgemäße Verbindung T29
beide Isoenzyme, COX-1 und COX-2, mit etwa gleich hoher Potenz
hemmt. Eine mittelstarke Hemmung für beide Enzyme wurde bei
der Verbindung T109 beobachtet.
Die erfindungsgemäßen Verbindungen und die Verfahren zu ihrer
Herstellung werden nun an nachstehenden Beispielen näher be
schrieben, welche die Erfindung in keiner Weise beschränken.
In den Beispielen werden zunächst die Synthesen der mit Z
benannten Zwischenverbindungen Z1 bis Z22 beschrieben aus
denen die nachfolgenden erfindungsgemäßen Verbindungen T1 bis
T111 gewonnen wurden.
42,2 g 4-Chlorbenzaldehyd und 5 g KCN wurden in 300 ml einer
1 : 1 Mischung von Ethanol und Wasser 6 Stunden unter Rückfluss
erhitzt. Nach dem Abkühlen wurde das Ethanol abgezogen, der
Rückstand im Eisbad gekühlt und die überstehende wässrige
Phase abdekantiert. Das Produkt wurde aus Ethanol/Petrolether
umkristallisiert und getrocknet. Es wurden 22 g der im Titel
genannten Verbindung Z1 gewonnen.
50 g 4-Fluorbenzaldehyd wurden nach dem gleichen Verfahren wie
in Beispiel 1 umgesetzt. Es wurden 29 g gewonnen.
50 g 4-Fluorbenzaldehyd wurden nach dem gleichen Verfahren wie
in Beispiel 1 umgesetzt. Die Ausbeute betrug 28 g.
26,5 g (0,25 mol) Benzaldehyd und 27,1 g (0,25 mol) Propandi
thiol wurden in Chloroform gelöst und im Eisbad gekühlt. Dann
wurde HCl-Gas etwa 5 min bis zur Sättigung eingeleitet und die
Lösung bei RT (Raumtemperatur) stehen gelassen. Die
Reaktionsmischung wurde zweimal mit 100 ml Wasser, dreimal mit
100 ml 10%-iger KOH-Lösung und nochmals zweimal mit 100 ml
Wasser gewaschen. Die organischen Phasen wurden mit Na2SO4
getrocknet, im Rotationsverdampfer eingedampft und aus
Methanol umkristallisiert. Die Ausbeute betrug 34,9 g.
34 g (0,24 mol) 4-Chlorbenzaldehyd wurden mit der gleichen
molaren Menge Propandithiol nach dem Verfahren wie in Beispiel
4 beschrieben umgesetzt. Die Ausbeute betrug 48,64 g.
33,0 g (0,169 mol) der Verbindung Z4 aus Beispiel 4 wurden in
trockenem THF gelöst und auf -60°C abgekühlt. Dann wurden
unter Stickstoff 112 ml (0,178 mol) n-Butyllithium (BuLi)
zugegeben und das Reaktionsgemisch 15-30 min in der Kälte
gerührt. Es wurden 24,0 g (0,169 mol) 4-Chlorbenzaldehyd
zugegeben und 1 Stunde bei RT gerührt. Das THF
(Tetrahydrofuran) wurde dann abgezogen, der Rückstand mit
Wasser versetzt und viermal mit CH2Cl2 extrahiert. Die
organische Phase wurde je zweimal mit Wasser, 7%-iger KOH und
nochmals mit Wasser gewaschen, mit Na2SO4 getrocknet und im
Rotationsverdampfer eingedampft. Die Ausbeute betrug 48,5 g.
47 g (0,204 mol) der Verbindung Z5 aus Beispiel 5 und die
gleiche molare Menge 4-Methoxybenzaldehyd wurden zusammen mit
112 ml (0,178 mol) BuLi wie in Beispiel 6 umgesetzt. Die
Ausbeute der im Titel benannten Verbindung betrug 64,2 g.
46,6 g (0,138 mol) der Verbindung Z6 wurde mit 80,0 g (0,295 mol)
HgCl2 und 40,0 g (0,185 mol) HgO in 600 ml 90%-igem Metha
nol 5 Stunden unter Rückfluss erhitzt. Die Feststoffe wurden
abfiltriert und mit CH2Cl2 sowie Diethylether gewaschen. Das
Filtrat wurde eingedampft, der Rückstand mit Wasser versetzt
und viermal mit Diethylether extrahiert. Die vereinigten
organischen Phasen wurden nacheinander mit Wasser, 5%-iger
NH4Cl-Lösung und wieder mit Wasser gewaschen, über Na2SO4 ge
trocknet, im Rotationsverdampfer eingedampft und aus Petrol
ether umkristallisiert. Die Ausbeute betrug 24,8 g.
64,2 g (0,175 mol) der Verbindung Z7 wurden wie in Beispiel 8
umgesetzt. Die Ausbeute betrug 35,89 g.
21,2 g (0,10 mol) Benzoin wurden mit 10,8 g (0,15 mol) Thio
harnstoff unter Erwärmung in DMF (Dimethylformamid) gelöst und
15 Std. unter Rückfluss erhitzt. Nach dem Abkühlen wurde das
DMF abgezogen und der Rückstand mit Ethanol versetzt. Der
dabei entstandene Niederschlag wurde abgesaugt, mit kaltem
Ethanol nachgewaschen, getrocknet und ohne weitere Reinigung
weiterverwendet. Die Ausbeute betrug 18,4 g.
27,23 g (0,1 mol) p-Anisoin und 10,8 g (0,15 mol) Thioharn
stoff wurden wie in Beispiel 10 umgesetzt. Die Ausbeute betrug
20 g.
22 g (78,5 mol) der Verbindung Z1 aus Beispiel 1 und 12 g (157 mol)
Thioharnstoff wurden wie in Beispiel 10 umgesetzt. Die
Ausbeute betrug 15,7 g.
29 g (0,12 mol) der Verbindung Z2 aus Beispiel 2 und 18 g
(0,24 mol) Thioharnstoff wurden wie in Beispiel 10 umgesetzt.
Die Ausbeute betrug 17,9 g.
28 g (0,08 mol) der Verbindung Z3 aus Beispiel 3 und 12 g
(0,16 mol) Thioharnstoff wurden wie in Beispiel 10 umgesetzt.
Die Ausbeute betrug 7,4 g.
13,6 g (50 mmol) p-Anisoin und 6,8 g (75 mmol) N-Methylthio
harnstoff wurden wie in Beispiel 10 umgesetzt. Die Ausbeute
betrug 7,4 g.
24,8 g (0,10 mol) der Verbindung Z8 aus Beispiel 8 und 10,8 g
(0,15 mol) Thioharnstoff wurden wie in Beispiel 10 umgesetzt.
Die Ausbeute betrug 11,3 g.
35,9 g (0,13 mol) der Verbindung Z9 aus Beispiel 9 und 15,2 g
(0,20 mol) Thioharnstoff wurden wie in Beispiel 10 umgesetzt.
Die Ausbeute betrug 17 g.
3,4 g (30 mol) 2-Chloressigsäurechlorid wurden in wasserfreiem
Dioxan gelöst und eine Lösung mit 3,8 g (30 mol) 4-Chloranilin
zugetropft. Nach einer Stunde wurde der Ansatz in Eiswasser
gegossen, mit verdünnter Salzsäure angesäuert, der Überstand
abgesaugt und der Rückstand mit Wasser neutral gewaschen. Das
Reaktionsprodukt wurde aus Ethanol umkristallisiert. Die Aus
beute betrug 3,2 g.
11,6 g (100 mmol) 2-Chlorethylamin-HCl wurden in Dichlormethan
suspendiert. Es wurden 13,8 g (175 mmol) Pyridin zugegeben, 10 min
gerührt und dann 15,8 g (75 mmol) 4-Chlorbenzolsulfonyl
chlorid, gelöst in Dichlormethan, zugetropft. Das Reaktions
gemisch wurde 8-10 Stunden unter Rückfluss erhitzt, anschlie
ßend mit Wasser versetzt und mit HCl auf einen pH-Wert von 1-2
eingestellt. Die organische Phase wurde dreimal mit 3%iger HCl
gewaschen und die wässrige Phase 3-4-mal mit Dichlormethan
ausgeschüttelt. Die vereinigten organischen Phasen wurden mit
Na2SO4 getrocknet, im eingedampft und aus Ethanol umkristalli
siert. Die Ausbeute betrug 11 g.
15,4 g (75 mmol) 2-Bromethylamin-HBr, 11,8 g (150 mmol) Pyri
din und 10,6 g (50 mmol) 4-Chlorbenzolsulfonylchlorid wurden
wie in Beispiel 19 umgesetzt. Die Ausbeute betrug 6,5 g.
Zu einer Vorlage von 1,0 g (42 mmol) Na0 wurde trockenes
Methanol langsam zugetropft, so dass die Lösung mäßig siedete.
5,0 g (16 mmol) der Verbindung Z11 aus Beispiel 11 wurden
zugegeben und das Reaktionsgemisch 10 min bei RT gerührt. Dann
wurden 3,5 g (16 mmol) 3-Brompropylamin-HBr, gelöst in
Methanol, zugesetzt und das Gemisch 2 Std. unter Rückfluss
erhitzt. Nach dem Abkühlen wurde das Produkt aufgearbeitet,
indem das Methanol abgezogen, der Rückstand mit Dichlormethan
versetzt und mit 1N NaOH und Wasser gewaschen wurde. Die
organische Phase wurde über Na2SO4 getrocknet und eingedampft.
Die Ausbeute betrug 3,5 g.
387 mg (16,8 mmol) Na0, 2,5 g (7,89 mmol) der Verbindung Z17
aus Beispiel 17 und 1,3 g (7,89 mmol) 2-Chlormethylpyridin-HCl
wurden wie in Beispiel 21 umgesetzt. Die Aufarbeitung erfolgte
mittels Säulenchromatographie über Kieselgel, wobei mit Ethyl
acetat eluiert wurde. Die Ausbeute betrug 1,77 g.
230 mg (10 mmol) Na0, 2, 5 g (8 mmol) der Verbindung Z11 aus
Beispiel 11 und 748 g (8 mmol) 2-Chloracetamid wurden wie in
Beispiel 21 umgesetzt. Die Aufarbeitung erfolgte mittels
Säulenchromatographie über Kieselgel, wobei mit Ethylacetat
eluiert wurde. Es wurden 0,8 g der im Titel benannten
Verbindung gewonnen.

Kälte gerührt und dann wurden 0,4 g (3,2 mmol) N,N-Dimethyl
carbamoylchlorid, die in THF gelöst waren, zugetropft. Das
Reaktionsgemisch wurde über Nacht bei RT gerührt. Die Auf
arbeitung erfolgte mittels Säulenchromatographie über Kiesel
gel, wobei mit Ethylacetat eluiert wurde. Es wurden 740 mg der
im Titel genannten Verbindung gewonnen.

in trockenem THF suspendiert, auf -50°C abgekühlt und 1,0 ml
(1,6 mmol) BuLi unter Stickstoff zugespritzt. Nach 5 min
Rühren wurden 190 mg (1,4 mmol) Benzoylchlorid, die in
trockenem THF gelöst waren, zugetropft. Das Reaktionsgemisch
wurde über Nacht gerührt. Das Produkt wurde über eine Kiesel
gelsäule gereinigt, wobei mit Ethylacetat eluiert wurde. Es
wurden 234 mg der im Titel benannten Verbindung gewonnen.

1,3 g (3,3 mmol) der Verbindung T69 aus Beispiel 30 wurden in
absolutem THF gelöst und 120 mg (5 mmol) NaH unter Rühren in
kleinen Portionen zugegeben. Das Reaktionsgemisch wurde 10 min
bei RT gerührt. Anschließend wurden 0,5 g (3,3 mmol) Brom
essigsäureethylester, gelöst in trockenem DMF, zugetropft. Das
Reaktionsgemisch wurde dann 2 Std. bei RT gerührt. Zur Auf
arbeitung wurde das DMF abgedampft und der Rückstand mittels
Säulenchromatographie über Kieselgel, wobei mit Ethylacetat
eluiert wurde. Es wurden 1,37 g der im Titel genannten Verbin
dung gewonnen.
Die nachstehenden Verbindungen wurden in analoger Weise wie in
Beispiel 122 beschrieben aus den genannten Ausgangsstoffen
hergestellt:

(1,6 mmol) Bromessigsäureethylester; 72 mg (3 mmol) NaH

1,37 g (2,8 mmol) der Verbindung Z23 aus Beispiel 23 wurden in
Ethanol gelöst, mit 5 ml (10 mmol) 2 N NaOH versetzt und 2 Std.
unter Rückfluss gekocht. Nach dem Abkühlen wurde das
Ethanol abgezogen, der Rückstand mit Wasser versetzt und mehr
mals mit Dichlormethan gewaschen. Die wässrige Phase wurde mit
10%-iger H3PO4 auf einen pH-Wert von 6-7 angesäuert und noch
mals mit Dichlormethan ausgeschüttelt. Die organische Phase
wurde mit Na2SO4 getrocknet, eingedampft und aus Ethanol aus
kristallisiert. Es wurden 865 mg der im Titel genannten Ver
bindung gewonnen.

Beispiel 128 beschrieben aus den genannten Ausgangsstoffen

0,2 g (0,5 mmol) der Verbindung T69 aus Beispiel 30 wurden in
30 ml THF/Methanol (1 : 1) gelöst und auf 0°C abgekühlt. Dann
wurden in 30 ml Wasser gelöste 2,3 g (3,7 mmol) Oxon® (Kalium
monopersulfat-Tripelsalz = 2KHSO5 × KHSO4 × K2SO4) zugetropft.
Das Reaktionsgemisch wurde über Nacht gerührt. Dann wurde das
Oxon® abfiltriert, die organischen Lösungsmittel abgezogen und
die wässrige Phase mit Ethylacetat und Dichlormethan ausge
schüttelt. Die vereinigten organischen Phasen wurden mit Na2SO4
getrocknet und eingedampft. Der Rückstand wurde aus Ethyacetat
(wahlweise kann auch CH2Cl2/n-Hexan verwendet werden) auskris
tallisiert. Es wurden 140 mg der im Titel genannten Verbindung
gewonnen.

1,0 g (2,5 mmol) der Verbindung T79 aus Beispiel 35 wurden mit
6,1 g (10 mmol) Oxon® wie in Beispiel 134 beschrieben umge
setzt. Es wurden 0,9 g der im Titel genannten Verbindung ge
wonnen.

500 mg (1,26 mmol) der Verbindung T6 aus Beispiel 24 wurden in
DMF gelöst. Dann wurden 48 mg (2,0 mmol) NaH portionsweise
zugegeben. Die Mischung wurde 10 min gerührt und dann wurden
358 mg (2,52 mmol) Methyliodid zugetropft. Die Reaktion er
folgte 2 Std. bei RT. Zur Inaktivierung des überschüssigen NaH
wurde die Reaktionsmischung anschließend mit 10 ml Methanol
versetzt. Nach abgeschlossener Gasentwicklung wurden die
Lösungsmittel abgezogen und der Rückstand über eine Kieselgel
säule mit Ethylacetat gereinigt.

Die nachstehenden Verbindungen wurden analog zu dem in
Beispiel 136 beschriebenen Verfahren hergestellt:
Ausgangsstoffe: 400 mg (0,8 mmol) T18 aus Beispiel 50; 284 mg
(2,0 mmol) Methyliodid; 96 mg (4,0 mmol) NaH
Ausbeute: 30 mg

Ausgangsstoffe: 340 mg (0,7 mmol) T28 aus Beispiel 29; 213 mg

2,0 g (6,4 mmol) der Verbindung Z11 aus Beispiel 11 wurden in
trockenem THF suspendiert und auf -50°C abgekühlt. Dann wurden
unter Stickstoff 4,2 ml (6,7 mmol) n-Butyllithium zugespritzt.
Nach 5 min Rühren bei der niedrigen temperatur wurden 0,9 g
(6,4 mmol) 1-Brom-2-chlorethan zugetropft. Das Kältebad wurde
entfernt und das Reaktionsgemisch 3 Std. bei RT gerührt. An
schließend wurde das Gemisch wieder auf -50°C abgekühlt und es
wurden 0,9 g (6,4 mmol) N-Methylbenzamid zugesetzt. Es wurden
weitere 4,2 ml (6,7 mmol) n-Butyllithium zugespritzt und das
Reaktionsgemisch über Nacht bei RT gerührt. Das Produkt wurde
über eine Kieselgelsäule chromatographiert, wobei mit Ethyl
acetat/Petrolether (3 : 1) eluiert wurde. Es wurden 200 mg der
im Titel genannten Verbindung gewonnen.

Claims (7)
1. Verbindungen der Formel I
worin
R1 die folgenden Bedeutungen besitzt:
R2 für H, C1-C6-Alkyl oder (CH2)oCOOH steht,
R3 und R4, die gleich oder verschieden sein können, für H, OH, OC1-C6-alkyl, Halogen oder C1-C6-Alkyl, das mit 1, 2 oder 3 Halogenatomen substituiert ist, stehen,
n für 0, 1 oder 2 steht und
o für 0, 1, 2, 3 oder 4 steht,
und die optischen Isomere und physiologisch verträglichen Salze davon.
worin
R1 die folgenden Bedeutungen besitzt:
- - CONR5R6, worin R5 und R6 unabhängig voneinander für H oder C1-C6-Alkyl stehen oder zusammen mit dem Stickstoffatom, an das sie gebunden sind, einen gesättigten heterocyclischen Rest mit 5 oder 6 Ringatomen und ein oder zwei Heteroatomen, die unabhängig voneinander ausgewählt sind unter N und O, bilden;
- - A-CONR5R6, worin A für C1-C6-Alkylen, das gegebenenfalls durch C1-C3-Alkyl-CO substituiert ist, steht und R5 und R6 unabhängig voneinander für H, C1- C6-Alkyl oder Phenyl, das gegebenenfalls durch ein oder 2 Halogenatome substituiert ist, stehen;
- - C1-C6-Alkylen-R7, wobei R7 für NR5R6, einen aromatischen heterocyclischen Rest mit 5 oder 6 Ringatomen und ein oder zwei Heteroatomen, die unabhängig voneinander ausgewählt sind unter N, S und O, wobei der heterocyclische Rest gegebenenfalls mit einem Benzolring kondensiert sein kann, oder COOR8 steht, R5 und R6 unabhängig voneinander für H oder C1-C6-Alkyl stehen und R8 für H oder C1-C6-Alkyl steht;
- - C1-C6-Alkylen-CO-R9, wobei R9 für Phenyl, das gegebenenfalls durch Halogen substituiert ist, steht;
- - C1-C6-Alkylen-NR10-CO-R11, wobei R10 für H oder C1-C6- Alkyl steht,
- - Phenyl, das gegebenenfalls durch 1, 2 oder 3 Substituenten substituiert ist, die unabhängig voneinander ausgewählt sind unter Halogen, CN, NO2, CF3, OC1-C6-Alkyl und C1-C6-Alkyl,
- - Naphthyl,
- - C1-C6-Alkyl, das gegebenenfalls durch 1 oder 2 Phenylgruppen substituiert ist,
- - C2-C6-Alkenyl,
- - CH=CH-Phenyl,
- - einen aromatischen, heterocyclischen Rest mit 5 oder 6 Ringatomen und 1 oder 2 Heteroatomen, die unabhängig voneinander ausgewählt sind unter N, O und S, oder
- - NR5R6 steht, wobei R5 und R6 unabhängig voneinander für H oder C1-C6-Alkyl stehen;
- - C1-C6-Alkylen-NR10-SO2-R12, worin n für 0, 1 oder 2 steht,
- - Phenyl, das gegebenenfalls 1, 2 oder 3 Substituenten aufweist, die unabhängig voneinander ausgewählt sind unter Halogen, NO2, CF3, OC1-C6-Alkyl, C1-C6-Alkyl, NH2 und NHCOC1-C3-Alkyl,
- - C1-C6-Alkyl, das gegebenenfalls durch ein oder zwei Phenylgruppen substituiert ist, oder
- - Naphthyl, steht
R2 für H, C1-C6-Alkyl oder (CH2)oCOOH steht,
R3 und R4, die gleich oder verschieden sein können, für H, OH, OC1-C6-alkyl, Halogen oder C1-C6-Alkyl, das mit 1, 2 oder 3 Halogenatomen substituiert ist, stehen,
n für 0, 1 oder 2 steht und
o für 0, 1, 2, 3 oder 4 steht,
und die optischen Isomere und physiologisch verträglichen Salze davon.
2. Verbindung der Formel I nach Anspruch 1, wobei mindes
tens einer der Reste R3 und R4 für eine Hydroxy- oder
C1-C6-Alkoxygruppe steht.
3. Verbindung der Formel I nach Anspruch 1, wobei beide
Reste R3 und R4 unabhängig voneinander für eine Hydroxy-
oder C1-C6-Alkoxygruppe stehen.
4. Verbindung der Formel I nach einem der vorhergehenden
Ansprüche, wobei R1 für CONR5R6, Phenyl-CO-NR10C1-C6-
Alkylen, Pyridyl-C1-C6-alkylen, C1-C6-Alkyl-CONR10-C1-C6-
alkylen, Carboxy-C1-C6-alkylen steht, wobei R5 und R6
für C1-C6-Alkyl, R10 für H oder C1-C6-Alkyl stehen.
5. Pharmazeutische Mittel, enthaltend wenigstens eine Ver
bindung nach einem der Ansprüche 1 bis 5, gegebenen
falls zusammen mit ein oder mehreren pharmazeutisch
akzeptablen Träger- und/oder Zusatzstoffen.
6. Verwendung wenigstens einer Verbindung nach einem der
Ansprüche 1 bis 4 zur Herstellung eines pharmazeu
tischen Mittels zur Behandlung von Erkrankungen, die
mit einer Störung des Immunsystems im Zusammenhang
stehen.
7. Verfahren zur Behandlung von Erkrankungen, die mit
einer Störung des Immunsystems im Zusammenhang stehen,
wobei man einer Person die einer derartigen Behandlung
bedarf, eine immunmodulierend wirkende und/oder die
Cyclooxygenase-hemmende Menge einer Verbindung der
Formel I nach Anspruch 1 verabreicht.
