本发明产生的酶能在原位作用于乳腺底物,因此,可以看出本发明包括一种生产酶反应产物的方法,该方法是通过在成年的雌性哺乳动物乳腺中表达DNA顺序,将用酶编码的DNA顺序掺入用乳剂乳清蛋白编码的哺乳动物基因中,然后由一种或多种酶作用物来催化制备反应产物,该反应产物一般从成年的雌性哺乳动物乳剂中回收。
最好在试管中,将用肽编码的DNA顺序(“所需的DNA顺序”)掺入到乳剂乳清蛋白质基因中形成融合基因,这种蛋白基因可以在成年的雌性哺乳动物中表达。通过向已受精的哺乳动物卵注射融合基因,从而将融合基因插入种系中,此后,注射过的受精卵发育为成年的雌性哺乳动物。人们预计并非所有注射过的卵都能发育成所需的DNA顺序表达的成年雌性哺乳动物。此外,将近一半的动物是雄性,在下一代生育雌性动物。
而关于外来DNA顺序可以插入到哺乳动物种系中的有关技术已用于小鼠。目前,最有效的途径需要将几百个直链DNA分子直接显微注射到受精卵细胞的原核中,这是本发明的优选方法。显微注射过的卵然后转移到假孕养母的输卵管中,进行培养。Brinster et al在Proc Natl Acad Sci 82(1985)4438-4442中报道培养的小鼠大约有25%继承了一个或多个显微注射的DNA拷贝。从商业观点出发,显然在本发明中,最好将具有高乳剂产率的哺乳动物作为宿主哺乳动物。因此,优先选用家畜,例如绵羊、猪和羊,下面将进一步详细讨论。
研究大量家畜需要相当大的投资,尤其是在所需动物的数目及其成本方面。每个超数排卵的小鼠产生30个卵是正常的,而母羊排卵数只有3至5个。所遇到的另一个问题是农场动物的卵还不透明,这是由于它存在大量的囊,这就使得鉴别及向其原核进行成功注射更为困难。然而,向兔子、绵羊和猪的卵中注射DNA已由Hammer et al在Nature 315(1985)680-683中作了报道,他将金属硫固生长激素融合基因注入这三种动物种系中。这2035个猪卵进行注射并转移,有192只小猪出生,其中20只带有融合基因。该结果对绵羊却并不如此令人鼓舞,对1032个羊卵进行注射并转移,但只有73只羊出生,其中只有一只羊含有外来DNA顺序。
只要将外来DNA并入种系,外来DNA就可以在高选择性的细胞组织内表达,生成一种功能蛋白质,这种蛋白质可以很容易地从动物中获取。要进行这个过程,一般将用所需的肽编码的DNA顺序融合到DNA顺序中,或介导在合适组织内表达的顺序。尽管对小鼠进行的最初实验表明加入DNA的细胞组织的专一表达是略有不同的(例如,Lacey et al,Cell 34(1983)343-356),但是,现在一般的相符性是大多数转染基因的标准细胞组织专一的表达可得到的。参阅Brinster et al Cell 27(1981)223-231;Swift et al Cell 38(1984)639-646;Shani Nature 314(1985)283-286;和Magram et al Nature 315(1985)338-340。关于标准的细胞组织特异性,它对于去除所有原核生物的载体顺序显然是重要的,在显微注射前,它用于所需的DNA顺序的无性繁殖方面(Krumlauf et al Mol.Cell Biol 5(1985)1639-1648)。
这些出版物指出的是将某些不同的细胞组织在基因转移动物中当作目标,而没有提出需要任何特殊的细胞组织来生产含多肽的物质,也没有提出任何用给定的细胞组织表达的,并用来作为引入DNA表达的目标的特殊基因。
关于含肽物质和其直接表达的细胞组织,必须考虑到许多因子。为了显示充分的生物活性,许多蛋白质需要广泛的转译后修饰。例如因子Ⅸ需要将用于生物活性的谷氨酸残余物的特定子集进行γ-羧化(De Seipio and Davie、Biochemistry 18 899-904(1979))。肝是天然合成因子Ⅸ的部位,它能熟练地进行这种修饰,尽管效率较低。成纤维细胞能够进行因子Ⅸ的γ-羧化:de la Salle et al Nature 316 268-270(1985)。然而,只有当在特异组织中合成时才可恰当修饰某种蛋白质,有时必须使表示的部位适合生产蛋白质的需要。可以认为使用乳腺作为细胞组织来表达,不论满意程度如何,这是一个潜在的困难因素。
与固体细胞组织相反,蛋白质需要从体液中获得,因为该途径一般说来是可以重复的,并且大部分生物医学上重要的蛋白质本身是分泌到体液中的。从肝或β淋巴细胞分泌到血流中是一个可行的途径,但是血液的凝固性质以及生物活性肽的抗原分子的存在是进行后续过程的障碍。
根据本发明,可以通过使用乳腺表达细胞组织来克服以上困难,乳剂很容易大量收集,并且有较好的生物化学特性,此外,主要的乳剂蛋白质以高浓度存在于乳剂中(大约1~15g/l)。
世界上许多国家的农场动物一般有四种:猪、山羊、绵羊和牛,也就是说Suidae科、Capra属、Ovis属和Bos属,家畜一般是Taurus种。
当本发明从广义上不限制任何特殊哺乳动物种类时,猪、山羊、绵羊和牛优先,在本发明的范围内它们各有优缺点。这些顾虑起源于实际情况,根据本发明的优选方面,用多肽编码的DNA顺序于试管内掺入可用成年雌性哺乳动物乳腺表达的乳清蛋白基因中,形成融合基因,将这种基因注射到哺乳动物的受精卵中,从而使注射过的受精卵发育为成年雌性哺乳动物。
尽管从牛中获得高乳剂产率是建议优先选用牛的因素,但是,例如控制牛的胚胎比控制羊胚胎有更多的技术困难。然而包括牛在内的绝对实验成本意味着其他三种优选的家庭、农场动物是待选的。

原核可见性的意义在于它是本发明将含有用肽编码的DNA顺序的融合基因注射到受精卵的原核中的一个优选特征。
猪的乳剂产率数据不易得到,但是羊的乳剂生产率为每天1~3升,这取决于繁殖情况。羊的挤奶专用设备可以从供应厂商处买到,该设备对牛也适用。因此羊成为选择的动物,特别是诸如East Frieslands奶羊。由在黑羊内交叉合适的乳剂生产线产生的菌株被看作是可行的替换物。
分泌乳汁的乳腺是一个非常特殊的器官,它包括排出分泌细胞复合体小叶的广泛输管系统。乳腺细胞适合以多种方式高速分泌,例如,它们有特定的运输机制确保有效地吸收血液中的前体,还具有一个广泛的细胞内膜系统(粗面内质网、高尔基氏体等),能高速进行合成蛋白质、转译后修饰和从细胞中输出。
乳腺的生长和作用的各个方面取决于激素,关于乳腺的综述包括Topper and Freeman,Physiol Rev 60 1049~1106(1980)和Forsyth in“The Biochemistry of Lactation”Mepham(Ed),Elsevier(1982)309~349。影响乳腺的激素介导怀胎和喂奶期间发生的显著变化。例如对于羊,这将导致细胞总数增加一倍,以及改变了各种细胞之间的比例和分泌细胞的最终差别。
乳腺将许多不同的蛋白质分泌到乳剂中,不同种乳剂组合物存在定性和定量差别,尽管在酪蛋白和可溶的(乳清)蛋白质之间可以找到一般的差别(参见Jenness in“Developments in Dairy chemistry,I”Fox(Ed)Elsevier(1982)87~109)。在乳腺中合成的反刍乳清蛋白质主要是α-乳清蛋白和β-乳球蛋白。
有三种主要的酪蛋白,它们是α-酪蛋白、β-酪蛋白和χ-酪蛋白,并以多种特征出现,在乳剂中将它们以微胞形式聚集来隔离钙。反刍动物中主要的乳清蛋白质,β-乳球蛋白质的作用是未知的,尽管它似乎与κ-酪蛋白相互作用(Brunner.J.Dairy Sci.64 1038-1054(1981))。α-乳清蛋白在葡萄糖和半乳糖转化为乳糖的过程中是一基本辅因子(Brew,Nature 223 671-672(1969))。表2列出了各种乳剂的蛋白质组合物。

通常人们认为单一复制基因可对不同的乳剂蛋白质进行编码(Mercier and Gaye in“The Biochemistry of Lactation”、Mepham(Ed)Elsevier(1983)177-225)。酪蛋白在小鼠中似乎是双连锁基因(Rosen et al Biochem Soc Trans,9,112(1982)),在奶牛中也如此(Grossclaude Proc、16th Inti,Conf Animal Blood Groups、Biochem、Polymorphy,1 54-59(1979))。因为酪蛋白基因与乳清蛋白基因相比似乎表达水平更高,所以很自然地认为在掺入用于表达的外来DNA时应选择酪蛋白基因。然而,出乎意料的是,本发明则规定了将用所需的肽编码的DNA顺序掺入用乳清蛋白质编码的基因中。
大鼠的α-乳清蛋白基因包含四个外显子,并且包含2.5kb染色体DNA(Qasba和Safaya Nature 311 377~380(1984)),牛的α-乳清蛋白基因似乎是类似的编码。现已发现羊的β-乳球蛋白最可能是单一复制基因,它在4.9kb的转录单位中包含7个外显子,这种基因是本发明中使用的优选基因。
已经确定在大鼠(Nakhasi和Qasba.J.Biol Chem 254 6016~6025(1979))、兔(Shuster et al Eur.J.Biochem.71 193~199(1976))和小鼠(Pauley et al Nature 275 455~457(1978))的乳房发育期间,在试管内利用转译和互补DNA杂化来改变乳剂蛋白信使RNA的含量,乳腺收集了80,000~100,000个由协调机制产生的乳剂蛋白质专一的信使RNA,这种协调机制包括高速合成乳剂蛋白质信使RNA和有效地稳定这些分子(Mercier和Gaye,op.Cit、和Guyette et al Cell 17 1013-1073(1979)),然而也有例外。对于母羊,β-酪蛋白占蛋白质总数的45%,然而仅17%的互补DNA克隆,它从与相应这个基因的互补DNA库分离出来(Mercier et al Biochimie 67 959-971(1985))。对于兔子,在微囊中隔离起来的α-乳清蛋白的量大大低于没有微囊时根据蛋白质合成速率预计的数目,这样就可以假定,单肽没有最佳构型(Gaye et al Biochimie 64 173~184(1982))。许多乳剂蛋白质基因的最初转译产物有很大改变,例如α-乳清蛋白是N-糖基化的,κ-酪蛋白是O-糖基化的、以及α-和β-酪蛋白是O-糖基化的。除了它在转译后修饰中的作用之外,在冷凝和将酪蛋白包裹在微团的过程中还包括高尔基复合体,酪蛋白和乳清蛋白有可能通过不同的途径从细胞中排出。关于这点,人们有趣地注意到,在中部和内部特异比较中,酪蛋白信号肽(与大量的其它分子比较)被很好地保存,预计它在新生的肽作为正确的分泌途径目标时起作用(Mepham et al op Cit)。如上所述,乳剂蛋白质基因在分泌乳汁的乳腺中大量地表达,例如,对于母羊,在多聚腺核苷酸+RNA(Mercier et al((1985)op Cit)中,αS1-酪蛋白占30%左右,β-乳球蛋白占5%左右。给定这些来源于单一复制基因的转录本,于是,这些含量显示了转译的高速率和有效的信使RNA稳定作用(Teyssot和Houdebine.,Eur J.Biochem 110 263~272(1980))。可以通过存在于成熟的信使RNA中的顺序介导特异的信使RNA的稳定作用,在本发明的优选方案中,对引入羊种系中的DNA顺序可以进行类似程度的表达。将它连接到与乳剂乳清蛋白质基因有关的DNA顺序中来完成该过程,从而维持高水平的组织专一表达及信使RNA稳定作用:β-乳球蛋白是优选的基因。
将新基因转入小鼠中是现在惯用的方法,在基因转移的小鼠中显示了对外来基因的高水平的组织专一表达,外来的DNA不仅包括结构基因,而且还包括5′和3′邻接顺序。尽管有大量的证据提出许多重要的调控元素位于信使RNA帽点的5′端(例如参见Mcknight和Kingsbury、Science 217 316~324(1982);Payvar et al Cell 35 381~392(1983);Renkawitz et al Cell 37 503~510(1984);Karin et al Nature 308 513~518(1984)),另外,很明显,重要的调节顺序,特别是介导组织专一表达的调节顺序可以保留在结构基因中,乃至在结构基因的3′端(Charnay et al Cell 38 251-263(1984);Gillies et al Cell 33 717-728(1983))。然而外来顺序(即存在于成熟的信使RNA顺序中)可以含有维持信使RNA稳定性的顺序。表3详细说明了小鼠中外来基因的组织专一表达。
鉴于以上理由,直接用于表达用所需的肽编码的DNA顺序的上述优选融合基因包括一个或多个启动子;转录初始位置;一个或多个(假定的)乳剂蛋白质基因的远5′端调节顺序;结构的乳剂蛋白质基因顺序;邻接乳剂蛋白质基因的3′顺序。最优的融合基因包括所有这三种类型的顺序。
选择的融合基因包括用插入乳清蛋白质基因的第一外显子的所需的肽编码的互补DNA顺序,优先的是将乳清蛋白质基因的5′端邻接顺序的数个kb包含在这样一个融合基因中。在这些构成中,最好通过自身的信号肽介导所需肽的分泌,因而对所需的肽,融合基因最好含有一个信号肽。然而,组织专一的信号肽对于将新生的肽作为进入正确分泌路径的目标是重要的(如上讨论)。因此,特别优先的是将用乳清蛋白质基因信号肽编码的DNA顺序准确地融合到用成熟蛋白质的N端氨基酸编码的插入DNA顺序。插入的3′端最好在其终止密码子之后、但在信息切断和聚腺苷位点之前终止。在插入点下流一般保留其余的结构基因,以及一些3′端邻接顺序。
在基因转移的动物中,这种构成最有可能获得提高水平的乳腺专一表达,最初的转录应进行适当的多腺苷酸化,并在保留乳清蛋白质基因处联接。成熟的信使应含有用于有效稳定信息RNA的顺序,在构成中使用乳剂基因信号肽,转译成熟的信使RNA得到一个融合的前肽,其中从乳剂蛋白质基因中得到的信号肽有效地引导用插入的互补DNA编码的成熟肽的分泌。
如上讨论,羊的β-乳球蛋白是选择的乳清蛋白质基因,它含有用所需的肽编码的互补DNA。对于羊来说,β-乳球蛋白的经常表达为乳腺中的乳清蛋白,其信使RNA包括大约80%的总多聚腺核苷酸+RNA。因子Ⅸ和人体α1抗胰蛋白酶编码的互补DNA顺序的融合构成。
根据本发明的第二个方面,提供一种遗传构成,它通过在成年雌性哺乳动物乳腺中表达DNA顺序,将用肽编码的顺序掺入用乳剂乳清蛋白编码的哺乳动物基因中。
根据本发明的第三个方面,提供一种如上所述的遗传构成的动物细胞,该动物细胞可以是胚胎细胞。
根据本发明的第四个方面,提供一种包括如上所述的遗传构成的质粒。

实施例1
A.复合DNA步骤
复合DNA步骤是在Maniatis et al(“Molecular Cloning”Cold spring Harbor(1982))和“Methods in Enzymology”Vols 68,100和101(Wr.Grossman and Moldave,Eds)、Academic press之后出现的,在这里没有详细说明。除非特别提出,所有的化学品可从BDH Chemicals Ltd.Poole、Dorset、England或the Sigma Chemical Company、Poole、Dorset、England买到。羊的β-乳球蛋白融合基因的构成
羊脾DNA的制备
从刚屠宰的Black face/Suffolk羊中取出羊脾,基本上按照Rurch和Weintraub Cell 33 65(1983)介绍的方法隔离细胞核。将核溶解在0.3M NaCl、10mM Tris HCl、10mM EDTA、1% SDS pH7.4和400mcg/ml蛋白酶K(Sigma Co.Fancy Road,Poole,Dorset BH17 7NH)中,在37℃下保温2小时。用苯酚/氯仿反复萃取,直到完全脱去蛋白质,用乙醇沉淀DNA,使用玻璃棒将其取出,用70%EtOH/30%TE(TE=10mM Tris HCl、1mM EDTA,pH值为8.0)洗涤,在空气中干燥,并重新悬浮在TE中,其浓度为1mg/ml。
羊脾DNA λ融合基因的构成
用噬菌体EMBL 3(Frischauf et al J.Mol.Biol170 827(1983))构成基因库,在生产厂家建议的条件下用过量5倍的限制性内切酶Eco RI和Bam HI(由Amersham Intgernational plc,Lincoln place,Green End.Ayles-bury、Bucking ham shire、England提供)消化30mcg的噬菌体DNA。消化后,加入氯化精胺盐达到5mM,沉淀出λDNA,在冰上保温1小时后,在台式微型离心机中,在10000g作用下分离15分钟,将DNA沉淀成片,在70%EtOH、300mM NaAc、100mM MgCl2中洗涤,反复压成片,最后重新悬浮在1mg/ml浓度的TE中。
用限制性内切酶Sau 3A(Amersham)部分消化羊DNA,在37°下20分钟内,用变量的Sau 3A〔从5~40单位〕消化羊DNA的100mcg整份。加入15mM EDTA终止该反应,消化程度通过在0.6%琼脂糖酸上的电泳进行评价,将适当消化的样品汇集起来,加到38.0ml10~40%的pH为8.0的1M NaCl、20mM Tris HCl、5mM的EDTA构成的蔗糖梯度中,这些梯度在Backmann SW28回旋器中以26,000rpm离心分离24小时,从顶部精馏出蔗糖梯度,收集1ml的馏分。每个馏分其DNA分子的粒径分度由琼脂糖酸电泳确定,依大小收集含有粒径14~25kb的DNA分子的馏分。在TE溶液中稀释2倍后,加入2体积的EtOH,在-20℃沉淀DNA一夜,然后DNA重新悬浮在TE中,浓度为300mcg/ml。
将7.5mcg由Bam HI/Eco RI切割的EMBL3和2.5mcg已用Sau 3A部分消化的羊脾DNA,在50mcl含有60mM Tris HCl、6mM MgCl2、10mM DTT、0.01%明胶、0.25mM rATP和25单位的T4DNA连接酶(Boehringer Company、Boehringer Mannheim House、Bell Lane、lewes、East Sussex)的溶液中混合,在14℃下保温一夜。
连接后,使用从Amersham买到的用具根据生产厂商建议的步骤,在试管内将1mcg DNA包裹起来。包裹后的库用E.coli菌株ED 8654测定滴度,预计该库的大小为5.7×106的空斑形成单位(pfu′s),测定滴度完成后立即将未放大库的整份放在10×22cm2的Petri平皿上(大平板),该过程在大约50,000pfu′s/板密度下使用E.coli菌株ED8654。屏蔽λ基因库
根据Benton和Davis(Science 196 180(1977))的方法,挑取大平板中的噬菌体放到20cm2的硝基纤维素膜上(Schieicher和Schull、Postfach 4,D-3354,West Germany)。通过Rigby et al(J.Mol.Biol.113,237(1977))介绍的方法,将β-乳球蛋白互补DNA克隆(p931-gift of J.C,Mercier,INRA,Jouey-en-Josas,Paris)通过32p dCTP制品转译为比活性>108dpm/mcg,β-乳球蛋白互补DNA按照Mercier et al在Biochimie 67 959-971(1985)的描述进行无性繁殖,由Gaye et al在Biochimie 68,1097-1107(1986)给出p931无性系顺序。
根据Maniatis et al在Cell 15 687(1978)的方法,将滤层预杂化、杂化和洗涤,最后的洗涤是在1×SET中于68℃下进行的(SET是pH为8.0的0.15M NaCl,2mM EDTA、0.03M Tris HCl)。对滤层吸滤干燥,在X-射线底片曝光之前点32p标记对它们定位。参考32p标记将含有阳性杂交空斑区域在大平板上定位,使用巴氏移管的无菌钝园端将其取出。用E.coli ED8654对取出的最初空斑进行滴度测定,并放置到空斑密度将近500/板的15cm直径的Petri平皿上,通过如2介绍的方法将这些板重新屏蔽起来,使用牙鉴将单个的阳性杂交空斑取出,加入1.0ml的噬菌体缓冲液中(噬菌体缓冲液是pH为7.4的10mM Tris HCl、10mM MgCl2、0.01%白明胶)。
无性繁殖的β-乳球蛋白DNA的制备
将0.4ml再悬浮的噬菌体溶液加入到E.coli ED8654(Borg et al Mol.Gen.Genetics 146 199~207(1976))中,放到9cm直径的Petri平皿上,得到细菌菌台的汇合溶解,得到汇合板,将顶部平板琼脂刮到10ml的噬菌体缓冲液中,并加入几滴氯仿保温一夜,在5000rpm下离心分离5分钟,将细菌碎片沉淀成片,噬菌体原种在4℃下保存,用E.coli ED8654测定原种滴度,测出pfu/ml。
在37℃ 10ml的10mM MgSO4中将8×107pfu′s吸收到7×109E.Coli细胞上,15分钟后,将2.5ml整份加入到1升蒸馏瓶内的100mlL Broth/10mM MgSO4中,使劲震动细菌悬浮液几小时,每小时监测OD540。由OD540降落测定,几小时后产生溶菌,完成后,向每个100ml培养物中加入0.2ml氯仿,该培养物在4℃下放置一夜。
在10,000rpm下离心分离15分钟除去细菌碎屑,将10mcg/ml RNA酶A和10mcg/ml RNA酶I加入到上清液中,然后在37℃保温1小时。保温后加入NaCl到40g/l,并加入聚乙二醇(PEG)至10%,将该制备物冷却到4℃,放置至少2小时沉淀噬菌体。在10,000rpm下在15分钟内将噬菌体沉淀成片,并重新悬浮在16.0ml的噬菌体缓冲剂中。在14.0ml的超离心分离管内,在含有1.5ml56%CsCl、1.5ml45%CsCl和2.5ml31%CsCl(溶于噬菌体缓冲液)的梯度中,将8.0ml的这种悬浮液分层。在20℃35,000rpm下,使振动回旋器中离心分离梯度1.5小时,使用一根针和注射器移去噬菌体层,提纯噬菌体颗粒,进行第二梯度离心分离。
将提纯的噬菌体颗粒分解到pH为8的0.1mM NaCl、10mM Tris HCl、1ml pH为8的EDTA中,然后使用苯酚和氯仿连续萃取脱去蛋白质,加入NaCl到最终浓度0.3M,然后,添加2体积的EtOH沉淀噬菌体DNA,在10,000rpm离心分离成20分钟,将DNA沉淀成片,用70%EtOH、30%TE洗涤、干燥、然后重新悬浮在TE中,最终浓度200~400mcg/ml。
复合β-乳球蛋白克隆的说明
用多种限制性内切酶限制如上描述的0.5mcg的DNA制备物,单消化产物和双消化产物在0.6%和1%琼脂糖胶上进行电泳分析,通过Southern(J.Mol.Biol.98 503(1975))的方法,将这些胶上的DNA转移到Hybond膜(Amersham International、Little cha(font、Bucks)上的硝基纤维素滤层上,杂交到32p标记P931。使用的方法基本如上所述,杂交过的滤层通过放射自显影术分析,用多种限制性内切酶P9315′端和3′端的特殊探查物,制成一个限制图,从而测出β-乳球蛋白基因大小和方向(参见图1)。
β-乳球蛋白克隆的同一性和基因5′端与3′端的准确位置直接通过DNA顺序确认。使用合适的限制单位,碎片再次无性繁殖成质粒载体及M13载体,使用Sanger et al(PNAS 74 5463(1977))的二次脱氧方法进行顺序化。
β-乳球蛋白融合基因的加工
加工含有β-乳球蛋白以及在乳腺中表达所需基因的融合基因使用的加工方法在图1-4中概括,该方法使用从λ克隆得到的顺序,它的隔离和特征化如上所述。该方法包括将所需的DNA顺序插入相应于5′端未转译的β-乳球蛋白信使RNA区域的DNA区域,根据这个基因的信使RNA转录酶转译的蛋白质将含有目标蛋白质的分泌肽。
按照图1所示,将1.4kbλ噬菌体SS-1 sphI-Eco RI碎片连接到载体质粒pPoly上,构成亚基克隆pSS-1tgSE,其中载体质粒pPoly被SphI+Eco RI E,coli菌株DH′切割,后者按照Hanahan和Meselson的方法(Gene10 63(1980))进行适当的转化。将抗克隆的氨苄青霉素隔离起来,用Birnboim和Doty的方法(Nuc.Acid Res.7 1513(1979))制备DNA。
在图1中,顶端箭头代表方向和存在于λSS1内的β-乳球蛋白转录单位的大小(大约4.9kb);比例始终相同。一般说来,在图1至图4中,应当注明只示出有关限制部位。大的开口方框代表λ EMBL2臂;窄的开口方框代表pPoly;窄的阴影方框代表将要表达的目标顺序;线代表相应于β-乳球蛋白基因的无性繁殖的羊的顺序以及它的邻接顺序。
通过用限制性核酸内切酶pvuⅡ(图2)的消化作用对pSS-1tg SE线性化,限制性核酸内切酶pvuⅡ在相应于β-乳球蛋白5′端未转译的信使RNA顺序的DNA范围内的质粒中在单拷贝部位作切割,将5mcg完全消化后的质粒溶解在pH为9.0的0.5M Tris HCl、10mM MgCl2、1mM MnCl2、10mM精胺中,用0.01~0.04单位的牛肠磷酸酶(Boehringer)在56℃下处理30分钟,牛肠磷酸酶在0.5%的SDS中失活,用苯酚/氯仿萃取和EtOH沉淀的方法回收DNA。
根据Dr.G.Brownlee Sir William Dunu School of Pathology、University of Oxford、Oxford的方法制造因子Ⅸ DNA克隆p5′G3′CVI,这个克隆在从PAT153中获得的质粒中含有1579bp插入物(Twig et al Nature 283,216~218(1980)),它从离假定的信使RNA初始点-7的TagⅠ位作用到+1572位,并含有用人体因子Ⅸ完整编码的顺序(Arson et al Embo J.3,1053~1060(1984))。
将含因子Ⅸ顺序的1553bp的NheⅠ-HindⅢ碎片切除,用如下描述的方法从载体中提纯。根据Maniatis et al描述的技术(“Molecular Cloning”Cold Spring Harbor(1982)),使用Kle now多聚酶削弱它的HindⅢ端和NheⅠ端,碎片连接到PvuⅡ限制的、磷酸酶的pSS1tg SE(如上描述),在将E.coli DH-1转化为氨苄青霉素阻抗后形成pSS1 tg SE-因子Ⅸ。
收集受精卵
方法类似于Gordon和Ruddle在“Methods in Enzymology”(1983)Vol 101(W.Grossman和Moldave,Eds)Academic Press pp 411至pp432中描述的用于小鼠的方法。
收集在发情期经诱导超数排卵的母羊胚胎,其中发情期靠孕激素类控制,已受精的成熟母羊用由60mg medrog proxy-esterone acetate浸润的子鞘内海绵处理12~16天(Veromix,Upjohn Ltd.Crawley)。在孕激素类处理前28小时及在取出海绵时,肌肉注射2次马的促卵泡激素〔3.5~4.3mg水溶液/羊〕,取出海绵后20~72小时,使发情期的羊进行多次交配,在每天8.00、12.00、16.00和20.00小时开始加热观察母羊,在发情期开始36~72小时后,由外科手术收集在1-4细胞发育阶段的胚胎。
通过静脉注射5-乙基-5-(1-甲基丁基)-2-巴比土酸钠〔Intraval,May and Baker〕进行麻醉,保持在半封闭的循环系统中的氧和一氧化二氮混合物中,胚胎的收集依照Hunter et al(J.Agric.Sci.46 143~149(1955))的方法进行。将再生管通过中心断口,以及插入到通过菌毛的输卵管中的尼龙导管,介质通过钝园的18号针引入子宫腔内,并强制通过子宫输卵管接合处,通过输卵管。在含有能源和蛋白质的磷酸盐一缓冲剂盐水中收集胚胎〔Ovum Culture Medium,Flow Labs,Irvine,Scotland〕。在卵的贮存和显微注射期间,这种介质补充20%的胎牛血清。
注射DNA
因子Ⅸ互补DNA顺序以下表示为TARG,见图2。
按照如上所述制备质粒DNA,并通过用合适的限制性内切酶消化检验,用SphⅠ+Eco RⅠ消化质粒DNA,在含有0.5mcg/ml溴化3,8-二氨基-5-乙基-6-苯基菲啶嗡(sigma)的1%蔗糖胶上电泳,用UV灯(Ultra-Violet Products、Inc,San Gabriel,California,USA)照射测出有关的SphⅠ-Eco RⅠ。在区域前部插入一块渗析膜,DNA在膜上电泳,将DNA从渗析膜上洗涤出来,用“Elu-tip”〔Schleicher and Schll,Postfach 4,D-3354,Dassel,W.Germany〕,根据生产厂家建议的方法隔离DNA。
将λSS-1的XbaⅠ-SalⅠ碎片连接到pPoly消化的XbaⅠ-SalⅠ上构成质粒pSSⅠ tgХS(图3),隔离克隆,按前文所述制备质粒DNA,两个DNA碎片分别与这个质粒或由其获得的小亚克隆各自隔离:10.5kb SphⅠ(部分)HindⅢ碎片和1.2kb Eco RⅠ-HindⅢ碎片(图4)。通过如上所述的胶电泳方法隔离这些碎片。
Sph-Ⅰ-Eco RⅠ、SphⅠ-HindⅢ、和Eco RⅠ-HindⅢ碎片以大致相同的比例连接在一起,使用DNA形成DH-1,按照如上介绍的方法制备质粒DNA(pSSⅠ tg ХS-TARG)用XbaⅠ和SalⅠ消化,从载体中切去β-乳球蛋白基因。用胶电泳方法,然后用“Elu-tip”提纯这种碎片,将用乙醇沉淀出的DNA成片,重新悬浮在TE中,进行苯酚/氯仿萃取、沉淀,DNA最终再悬浮于TE中,直接用于显微注射。
B.基因转移动物的构成
将DNA(1-2mcg/ml)注射到单个卵细胞的原核中,或注射到2个和4个卵细胞的一个或多个核中。在充有卵培养介质的盒中操纵卵,该盒包括具有玻璃支架的硅化显微镜玻片(25mm×2mm×3mm),它平行于长侧的滑动边,盖玻片置于支架顶部,连接部分用硅化脂肪密封,盒的开端充满Dow-Corning200流体(50cs)(BDH Chemicals)
待注射的卵用吸管吸住,用Nikon Diaphet翻转显微镜(Nikon(uk)Ltd.Haybrooke,Halesfield 9,Telford,Shropshire)可以看见原核或核,使用从在显微电极拉出器(Campden Instruments,186 Campden Hill Road,London)上的毛细管(硼硅酸盐玻璃-1mm外径,带有丝的薄壁-Clark Electromedical Instruments,POBOx 8,Pangbourne,Reading.RG8 7HU)中伸出的微量移管,将DNA注射到原核或核中,用显微控制器(Leitz Mechanical Micro-manipulators.E.Leitz(Instruments)Ltd,48 Park Street,Luton,England)控制两种显微仪器的位置。含有待注射的DNA的微量移管通过空气密封管连接到100ml玻璃注射器,由原核或核的可见膨胀显示成功的注射,注射后的卵在室温下保温30分钟以上,使损坏的卵进行可见的退变。
将判定为没有注射过的胚胎转移到未交配的受体母羊中,使用孕激素〔Veromix,Upjohn Ltd〕使其发情期周期与卵供体同步,使用细口移管将胚胎转移到输卵管中,向每只母羊转移4个胚胎,这些胚胎分布在输卵管之间。
母羊的身体是用可溶的肌原纤维细丝〔Descon,Davis andGreck〕和带有Michel clips的皮肤包围起来,每只羊在手术时注射抗菌素〔Duphapen L.A.,Duphar,Amersham〕,手术后10~20天除去Michel clips。
发育和成长
母羊从麻醉中恢复过来后,返回小牧场,在那里它们度过整个怀孕时期。根据需要补充秣、芜菁和浓缩食物,受孕第3个月期间,通过超声波扫描测出胎的数目(White et al Vet.Rec.115,140~143(1986))。然后还要注意怀孕母羊中胎数目的变化,当接近预计的生产日期时,将羊圈起来以便监督,以及在生产时给予协助。
基因转移后的羊的分析
生产小羊后至少2周,用皮下注射器通过静脉穿刺移取10ml血样,并收集在加了肝素的管中。从血样中制备DNA如下:向10ml血样中加入30ml溶胞溶液(155mM NH4Cl、10mM KHCO3、1mM EDTA),该混合物在冰上保温15分钟,在1500g作用下,在40℃分离10分钟,分离出白血细胞,并重新悬浮在10mlSE(75mM NaCl、2mM EDTA)中,然后在SE中洗涤一次。加入蛋白酶κ达到100mcg/ml,再加入1ml20%SDS,该制备物保温四小时,用苯酚/氯仿进行反复萃取,直到完全脱去蛋白质。将1/30th Vol的0.3M NaAc-1 Vol异丙醇加到水相中,沉淀DNA,将其取出,在70%EtOH中清洗,并重新悬浮在TE中。
10 mcg整份DNA制备物用合适的限制性内切酶(例如Eco RⅠ)消化,在0.8%的胶上电泳。通过Southern标记分析这些胶,基本上按照描述的方法进行杂交。
阳性杂交动物,即那些含有融合基因的动物(假定在染色体位置积分)长大成熟,雌性动物交配、一旦分泌乳汁,则分析乳汁的找出所需的物质(参见实施例7)。阳性雄性动物交配,用外源DNA顺序屏蔽它们的女儿,随后,分析其乳汁以找出所需的物质。
实施例2
除了根据R.Cortese.EMBL.Meyerhofstrasse 1,D-6900 Heidelberg,West Germany获得用所需多肽编码的DNA顺序(TARG顺序)编码α1-抗胰蛋白外,实施例2重复了实施例1的方法。切除含有1294 bp插入物(参见Ciliberto et al Cell 41 531~540(1985))的Tag 1-Bst NⅠ碎片,除去这个克隆并通过实施例1中描述的方法无性繁殖到pSS1 tg SE的PvuⅡ位点。

基因转移的羊的传代
由质粒pSS1 tg XS-FIX(也称为pSS1 tg XS-TARG,参见实施例1)切下的SalⅠ-XbaⅠ碎片,注射到羊卵中,每个受精卵注射将近200个拷贝,252个卵细胞中,有一个卵细胞被注射,并再移植到受体母羊中,52只小羊出发。分析由血样制备的DNA,表明其中四个动物载有外源β-乳球蛋白因子Ⅸ顺序(表4)。

图5是羊DNA的Southern标记,羊的DNA按照介绍制备,用指出的限制性内切酶消化,在0.8%的琼脂胶上电泳,并转移到Hybond膜上(Amersham International,Little Chalfont,Bucks,UK)。用32p示踪质粒pSS1 tg XS-FIX(泳道1~7),然后(脱去膜后)用质粒P931(泳道1′~7′)操测滤层,pSS1 tg XS-FIX复制对照包含在指示的胶上。泳道1,对照(非基因转移的)羊DNA,泳道2和3对照DNA以及P5′G3′CVI1和5复制当量;泳道4~7,基因转移的羊6LL225、6LL231、6LL239和6LL240中的DNA。每个基因转移的羊产生因子Ⅸ,后者与pSS1 tg XS-FIX中获得的因子大小一致,杂交范围为5.95kb(EcoRⅠ)和6.05kb(BamHⅠ),这表明基因转移的5′端未触动。观察不到与羊的因子Ⅸ的明显杂交,与P931杂交显示了预测的4.4kb Eco RI和2.1kb Bam HI碎片。尽管内源的羊β-乳球蛋白基因促使在这个区域内杂交,但是增加在6LL225、6LL231和6LL240中的样品密度表明这些区域主要来自注射了β-乳球蛋白因子Ⅸ融合基因,证实了构成的3′端是完整的。用HindⅢ切除的DNA:泳道1,从pSS1 tg XS-FIX中切除的12.1kb提纯的SalⅠ-XbaⅠ碎片;泳道2~5,来自6LL225、6LL231、6LL239和6LL240的DNA。探查物是P5′G3′CVI。杂交12.1kb HindⅢ碎片(6LL225、6LL231和6LL240)与注射的碎片大小一致,它显示了从头到尾的排列,同样的羊15.6kb碎片显示存在从头到头的重复。这些数据表明,在羊6LL225、6LL231和6LL240中,将从pSS1 tg XS-FIX(β-乳球蛋白因子Ⅸ融合基因)获得的SalⅠ-XbaⅠ碎片整合,在一连串试样中未探测出重复排列顺序。对于羊6LL239,其数据与这个碎片的单个非重排复制的整合作用一致。
实施例6
因子Ⅸ顺序遗传到F一代
实施例5得到的雄性基因转移后的羊6LL225(载有将近40个从pSS1 tg XS-FIX制备的SalⅠ-XbaⅠ碎片复制)与许多Finn-Dorset和Friesland母羊连续地交配。使用所述的人体因子Ⅸ质粒探查物P5′G3′CVI,根据由血样制备的DNA和Southern标记分析其后代,对它的一些后代进行的分析列于图5a中。标数泳道1、3、4、5、6、7、9、11、43、45、48和49,来自6LL225后代的DNA(称为7R1、7R3等~7R49);泳道C211,来自对照(非基因转移的)羊6LL211的DNA;泳道5~211及1~211,来自羊6LL211的DNA以及pSS1 tg XS-FIX的5和1复制当量。这些数据表明基因转移的羊6LL225已将因子Ⅸ顺序转移到它的12个子孙中的6个中,从而这些顺序插入到种系中。
实施例7
羊的β-乳球蛋白编码基因在基因转移的小鼠中的表达
基因转移的小鼠一般是通过Gordon和Ruddle在Methods in Enzymology Vol 101(1983),(Eds,Wu,Crossman和Moldave)、Academic Press pp411~432描述的技术方法形成,形成几个载有克隆λSS-1的SalⅠ碎片的基因转移的小鼠(图3),其中之一,B-Lac7多次交配,产生了许多遗传了SS-1顺序的后代。
生产一窝小鼠后8~12天,摄取小鼠的乳剂,该过程是这样进行的,先内膜注射0.3IU催产素(Sigma)和7mcl/g动物的Hypnorm/Hypnovel(Fleckneil,Vet,Rec1983年12月10日,P574),在每四个小时周期中,先移走动物幼子后,等待20分钟,然后用手按摩各个乳腺,将乳剂收集到50ml的毛细管中。
将小鼠乳剂用蒸馏水以1∶5稀释,在间歇式离心分离机中粗略分离进行脱脂,添加1NHCl使得pH值为4.6,以沉淀酪蛋白,离心分离后,移去乳清蛋白,用5%的三氯乙酸沉淀,根据Laemmli(Nature 277、680~684(1970)的方法进行聚丙稀酰胺胶电泳分析。(图6表示鼠和羊的乳清蛋白质的SDS PAGE分析,泳道1,标示蛋白质;2,标准的小鼠乳清;3,羊的乳清;4,标准的小鼠乳清;5,B-Lac 7乳清;6,B-Lac 7乳清(2.5×5),相应于标示踪迹和羊乳清中的β-乳球蛋白的区域标有箭头)。使用兔子中抗羊β-乳球蛋白的抗血清,通过用胶电泳分辨样品上的Western标记(Burnett,Anal、Bioches,112,195~203(1981))探测羊的β-乳球蛋白。图7表示Western标记分析。Western标记与兔的抗-β-乳球蛋白血清和抗兔Ig过氧化物酶血清反应。(泳道1,标记蛋白质;2,羊的乳清;3,B-Lac7乳清;4,标准的小鼠乳清;5,提纯的β-乳清蛋白;6,Coomassie染色的羊的乳清(平行作用)。
分析表明,大量的乳球蛋白分泌到小鼠乳剂中,显示出在B-Lac7中高水平地表达了SS-1。这个克隆可能含所有必要的顺序,以确保在基因转移的小鼠的乳腺中高水平地表达,从而预测,即使不是如此,在同源种类中,即在基因转移的羊中也能有效地作用。因此,可以预计由这个克隆得到的融合基因也能在羊的乳腺中表达(有效)。
实施例8
人体因子Ⅸ在基因转移后的母羊中的表达
两只雌性羊,6LL231和6LL240(各载有将近10个从pSS1 tg XS-FIX制备的SalⅠ-XbaⅠ碎片复制)与Fast Friesland公羊进行连续交配。分娩后,使小羊自然哺乳2周,以刺激分泌乳汁,用手将乳剂(每个动物约25ml)收集到无菌的塑料容器中,同样收集对照(非基因转移的)分泌乳汁羊的乳剂,在-20℃冷冻这些样品,运送到位于爱丁堡皇家医院的苏格兰国家输血机构,在那里对人体因子Ⅸ进行放射免疫测定。
基因转移的乳剂和对照乳剂在4℃蒸馏水渗析一夜,然后冷冻干燥。冷冻干燥后的样品再悬浮于蒸馏水中,而后离心分离出乳剂,用RIA检验人体因子Ⅸ的存在,方法如下:
使用在RIA缓冲剂(50mM Tris/HCl,0.25%胶,1%Tween-20,10mM HCl,pH值7.2)中稀释的标准人体血浆,稀释度1/10-1/1280,建立标准曲线(参见图8)。为了消除试样中乳剂的任何干扰,将标准血浆同样稀释于对照乳剂中,作出标准曲线分析来自6LL231和6LL240冷冻干燥乳剂样品的稀释液,RIA中的每个试管包括:50mcl RIA缓冲液,50mcl样品稀释液,在1/30000稀释度,50mcl兔的聚无性繁殖的抗因子Ⅸ抗体(Dako-patts),以及50mcl 1125示踪Ⅸ。预备包含100mcl RIA缓冲液,50mcl抗体和50mcl 1125因子的用于最大结合的对照Ⅸ,还预备包含150mcl RIA缓冲液,50mcl 1125痕迹的非专一结合的对照。
保温一夜后,50mcl琼酯糖-S1000配以驴抗兔IgG在试管中混合、蔗糖分离回收小球、RIA试样对0.125国际单位(iu)/dl敏感,含有该含量以上因子Ⅸ的样品抑制在RIA中的最大结合。
对下列样品将RIA中的结合百分数与因子的稀释度例数作图。
1.标准血浆。
2.标准血浆加乳剂。
3.来自6LL231(T1)的冷冻干燥的乳剂样品。
4.来自6LL240(T2)的冷冻干燥的乳样样品。
这些结果示于图8。
来自6LL231(T1)和6LL240(T2)的乳剂,分别在2.5iu/dl和8.0iu/dl时可探测出因子Ⅸ的含量。在试验的灵敏度含量时,在对照乳剂亦未测出活性,这些数据表明载有β-乳球蛋白-因子Ⅸ融合基因(特别是从pSS1 tg XS-FIX(通常也称为pSS1 tg XS-TARG)中得到的SalⅠ-XbaⅠ碎片)的基因转移后的母羊表达这个基因,并将人体蛋白质分泌到乳剂中,这就为以这种方式生产人体蛋白质建立了基础。
