Abstract

背景技术
光学检测系统常常被用于定性地、定量地、或者半定量地确定测试试样中是否存在分析物或者分析物的浓度。例如,一些常规光学检测系统采用基于反射的检测技术,其中照明源和检测器被设置在测试条的同一侧。其它常规光学检测系统采用基于透射的检测技术,其中照明源和检测器被设置在测试条的相对侧。基于透射的光学检测系统,例如,有时采用和发光二极管(LED)照明源位置相对的光电二极管检测器。尽管这种检测系统可以提供一定的益处,但是它通常存在着照明源(例如,LED)无法提供散射光的问题。因此,为了确保向测试条提供足以获得精确结果的光,该系统需要复杂昂贵的光学元件,比如透镜或漫射片。

发明内容

根据本发明的另一实施方案,公开了用于检测测试试样中是否存在分析物或者分析物的量的光学检测系统。该系统包括测定器件(assaydevice),该测定器件包括和检测探针连通的多孔膜,该检测探针能够产生检测信号。该系统还包括能够提供电磁辐射的电致发光照明源,所述电磁辐射导致检测探针产生检测信号。还提供了能够将检测探针产生的检测信号寄存的检测器。

代表性实施方案的详述
定义

一般而言,本文所用术语“测试试样”是指怀疑含有分析物的生物材料。测试试样可以来自任何生物来源,比如生理流体,包括血液、间隙液、唾液、目镜流体、大脑脊椎流体、汗液、尿液、乳汁、腹水、粘液、鼻液、痰、滑液、腹膜液、阴道液、月经、羊水、精液等等。除了生理流体以外,可以采用其它液体试样,比如水、食物制品等,用以对环境行为或食品生产进行测定。另外,可以采用怀疑含有分析物的固体材料作为测试试样。测试试样可以以得自生物源的原样直接采用,或者经过预处理以改变试样性质后采用。例如,所述预处理可以包括从血液制备血浆、稀释粘性流体、等等。预处理方法也可以涉及过滤、沉淀、稀释、蒸馏、混合、浓缩、干扰组分的失活、添加试剂、溶解等。另外,固体测试试样进行改性以形成液体介质或者释放分析物也可能是有利的。
发明详述
现在详细参考本发明的各种实施方案,下面给出了这些实施方案的一个或多个实施例。每个实施例通过解释本发明而不是限制本发明的方式提供。事实上,对本领域技术人员而言显而易见的是,可以对本发明进行各种修改和改变而不偏离本发明的范围或精神。例如,作为一个实施方案的一部分进行说明或描述的特征可以用在另一实施方案上,以形成更进一步的实施方案。因此,本发明意在覆盖落在所附权利要求和其等同物的范围中的这些修改和改变。
一般而言,本发明涉及采用了电致发光(EL)照明源的光学检测系统。和一些常规光学检测系统采用的照明源不同,EL器件产生相对均匀的漫射辐射,因此可以向测试试样提供均一的照明。另外,EL器件发射的辐射强度可以通过简单地改变驱动信号的电压或频率来容易地控制。同样,通过改变发光材料可以容易地控制辐射波长。EL器件的相对柔性也使其可以很容易地结合到用于检测测试试样中是否存在分析物的、基于色谱法的测定器件中。
1、电致发光(EL)器件
EL器件通常是在电极之间夹有发光材料(例如,磷光体颗粒)的电容器结构,其中至少一个电极透明从而使得光可以逃逸。在电极之间施加电压使发光材料内形成变化的电场,该电场导致发光材料发光。一般而言,任何已知的EL器件都可以用作照明源。例如,采用了“无机”或“有机”发光材料的EL器件可以用于本发明。合适的“有机”EL器件包括低分子量和高分子量的器件。同样,合适的无机EL器件包括分散体和薄膜磷光体。分散性EL器件通常含有夹在电极层之间的粘结剂中的粉末发光材料分散体。另一方面,薄膜EL器件包括夹在一对绝缘薄膜和一对电极层之间的、设置在电绝缘衬底上的发光薄膜。尽管并不一定要求,但是由于分散性EL器件的成本较低、制备容易,所以在本发明的一些实施方案中特别理想。
例如,参见图1,其示出了可用于本发明的分散性EL器件10的一个实施方案。如图所示,EL器件10具有阴极12、介电层14、发光层16、阳极18和膜19。需要时,可以在阴极12和膜19上任选地施加另外地防水保护层(未示出)。引线65电连接到各个阴极和阳极层12和18上。驱动电路(未示出)经由布线连接到所述引线上,驱动电路还连接到电源(未示出)上。驱动电路和电源的细节通常取决于具体EL器件10的要求。例如,对于较小例如发光面积为5英寸×8英寸的EL器件而言,可以采用低压电路和电池电源。另一方面,较大的EL器件可以由更高电压的电路供电。在示例性应用中,驱动电路将DC电压转变成AC电压输出,用以驱动EL器件10。这种AC转换可以获得50-5000赫兹的大约60-300伏AC。适于这个目的的驱动电路可以买到。
阴极12可以由金属(包括准金属)或其合金(包括金属间化合物)。适用于形成阴极12的材料的例子包括但不限于碳;金属,比如铝、金、银、铜、铂、钯、铱和其合金;等等。阴极12的厚度通常可以改变,可以沉积到电绝缘衬底上(未示出)。所述衬底例如可以由陶瓷材料形成,所述陶瓷材料比如氧化铝(Al2O3)、石英玻璃(SiO2)、氧化镁(MgO)、镁橄榄石(2MgO.SiO2)、滑石(MgO.SiO2)、富铝红柱石(3Al2O3.2SiO2)、氧化铍(BeO)、氧化锆(ZrO2)、氮化铝(AlN)、氮化硅(SiN)、碳化硅(SiC)、玻璃、和耐热玻璃等。另外,也可以采用聚合物材料形成衬底,比如聚丙烯、聚对苯二甲酸乙二酯、聚氯乙烯、和聚甲基丙烯酸甲酯等。
介电层14设置在阴极12上。如同本领域技术人员所公知的那样,形成介电层14的材料通常可以改变。例如,合适的材料包括但不限于钙钛矿结构的介电和铁电材料,比如BaTiO3、(BaxCa1-x)TiO3、(BaxSr1-x)TiO3、PbTiO3和Pb(ZrxTi1-x)O3(称作“PZT”);复合钙钛矿弛豫型铁电材料,比如Pb(Mg1/3Nb2/3)O3;铋层化合物,比如Ba4Ti3O12和SrBi2Ta2O9;和钨青铜型铁电材料,比如(SrxBa1-x)Nb2O6和PbNb2O6。其它适用于介电层14的介电材料可以包括介电材料比如SiO2、SiN、SiON、ZrO2、Al2O3、Al3N4、Y2O3、Ta2O5等。在一个特定实施方案中,介电层114由钛酸钡(BaTiO3)形成。
介电层14可以由本领域技术人员公知的各种技术中的任一种形成。例如,可以首先将用于形成层14的介电材料和合适的溶剂预混。所述溶剂可以包括例如乙二醇醚、烷基酮和芳族溶剂。合适的乙二醇醚可以包括丙二醇甲醚、二丙二醇甲醚、三丙二醇甲醚、乙二醇乙醚、和二乙二醇丁醚等。合适的烷基酮可以包括低级烷酮,比如丙酮、甲基乙酮、乙基酮和甲基异丁酮等。合适的芳族溶剂可以包括甲苯和二甲苯等。在一个实施方案中,钛酸钡以约70重量%-约90重量%的量加到溶剂中。随后,钛酸钡和溶剂被搅拌到一起形成均匀的浆料。
介电材料在和溶剂混合时,也可以和粘合剂混合。例如,在一些实施方案中,粘合剂的加入量为浆料的约10份-约30份。合适的粘合剂是公知的,包括例如环氧树脂、聚苯乙烯、聚乙烯、聚乙烯醇缩丁醛、聚氯乙烯、聚乙酸乙烯酯、聚乙烯醇、聚酯、聚酰胺、聚丙烯腈、聚丙烯酸酯、和聚甲基丙烯酸甲酯等。在一些实施方案中,粘合剂是酚和过量表卤代醇的粘合性热塑性反应产物。合适的酚包括双酚A、二氯双酚A、四氯双酚A、四溴双酚A、双酚F、和双酚ACP。反应在乙二醇醚或者其它合适溶剂的存在下进行。向该反应产物中加入树脂比如尿烷或者环氧树脂,加入量为大约1份的表卤代醇/酚反应产物加入大约5-6份树脂。在Kardon的美国专利No.4560902和Kardon等的No.5352951中详细描述了这些粘合剂,出于所有目的,这些专利在此全文引入作为参考。
如果需要,在EL器件10的组装步骤或者随后可以向粘合剂系统中加入水。在去除溶剂之前或之后,可以将水搅拌到浆料中。根据所采用的特定粘合剂可以吸收的水量,在粘合剂中加入的水量可以发生一些变化。例如,水的存在量可以是至少约百万分之一(“ppm”)(0.0001%)直到粘合剂吸收的最大水量。例如,氰乙基聚乙烯醇粘合剂通常吸收最大约40000ppm(4.0%)的水。另一方面,氰烷基化的支链淀粉粘合剂通常吸收大约100000ppm(10.0%)的水。但是,在大多数情况下,向粘合剂中加入的水量是大约500ppm(0.05%)-大约20000ppm(2.0%)。最终钛酸钡/树脂粘合剂层114的厚度通常为大约0.2-大约6密耳。
再次参见图1,EL器件10还包括设置在介电层14上的发光层16。发光层16的材料可以包括磷光体颗粒。合适的磷光体颗粒可以包括各种金属氧化物、硫化物、氟化物、和硅酸盐化合物。例如,所述磷光体颗粒可以包括锰和砷活化的硅酸锌(P39磷光体)、钛活化的硅酸锌、锰活化的硅酸锌(P1磷光体)、铈活化的硅酸钇(P47磷光体)、锰活化的硅酸镁(P13磷光体)、铅和锰活化的硅酸钙(P25磷光体)、铽活化的硅酸钇、铽活化的氧化钇、铽活化的钇铝氧化物、铽活化的钆氧化物、铽活化的钇铝镓氧化物、铕活化的钇氧化物、铕活化的钇钒氧化物、铕活化的钇氧硫化物、锰活化的硫化锌、铈活化的硫化锶、铥活化的硫化锌、钐活化的硫化锌、铕活化的硫化钙、铽活化的硫化锌、和铈活化的硫化钙等。
磷光体颗粒发射的颜色可以在磷光体的生产过程中确定,或者通过混合不同颜色的磷光体来获得复合颜色。合适磷光体的一些具体例子包括锰活化的硫化锌(发射黄橙色光)、铈活化的硫化锶(发射蓝光)、铥活化的硫化锌(发射蓝光)、钐活化的硫化锌(发射红光)、铕活化的硫化钙(发射红光)、铽活化的硫化锌(发射绿光)、和铈活化的硫化钙(发射绿光)。
通常情况下,磷光体颗粒的平均尺寸小于约15微米,在一些实施方案中小于大约10微米,在一些实施方案中小于约5微米。发光层16可以用本领域技术人员公知的各种技术中的任一种形成。例如,如上所述,包封的磷光体颗粒可以和溶剂混合。在溶剂中加入的磷光体颗粒的量可以是例如混合物的约60重量%-约95重量%,在一些实施方案中是约75重量%-约85重量%。同样,在混合后,如上所述也使粘合剂和磷光体颗粒浆料混合。粘合剂通常的含量为大约5-大约40份。如果需要,磷光体颗粒也可以包封在保护性材料中,以形成本领域公知的防水层。用于包封磷光体颗粒的合适保护性材料包括例如液晶、聚合物粘合剂、和陶瓷材料(例如,胶态二氧化硅、氧化铝、等等)等。在Allinikov的美国专利No.4097776、Wary的美国专利No.4513023、Kardon的美国专利No.4560902和Kardon等的美国专利No.5352951中更详细地描述了包封技术,这些专利在此全文引入作为参考。
磷光体颗粒优选通过本领域技术人员公知的各种技术的任一种沉积成光滑均匀的层。所述技术包括沉降技术、浆料方法(比如丝网印刷、旋涂和旋模(spin casting))、电泳、或者撒粉法(比如静电撒粉、“光粘(phototacky)”法、和高压撒粉)。沉降技术和浆料方法涉及将磷光体颗粒在合适液体介质中形成分散体。一种特别理想的沉积方法是丝网印刷。磷光体/粘合剂层16干燥后的合适厚度为大约0.2-大约6密耳。
除了上述层以外,EL器件10还包括在膜19上形成的阳极18,膜和阳极都设置在发光层16上方。理想情况下,层18和19所用的材料光学透明。例如,阳极18可以由无机导电氧化物比如氧化铟、氧化铟锡(ITO)、氧化锡和氧化锑锡形成。在一个实施方案中,采用了厚度为大约0.2-1微米的氧化铟锡(ITO)层。同样,用于形成膜19的合适材料可以是聚合物膜(例如,聚酯)。应该理解的是,上述实施方案仅仅是示例性的,任何其它已知的EL器件一般都可用于本发明。例如,在Rasmussen等的美国专利No.6004686、Terasaki等的美国专利No.6432516、Watanabe等的美国专利No.6602618、Tanabe等的美国专利No.6479930、Nagano等的美国专利No.6723192、和Yano等的美国专利No.6734469、以及Shirakawa等的美国专利申请No.2003/0193289、Takahashi等的美国专利申请2004/0119400、和Nelson等的美国专利申请2004/0070195中描述了其它合适的EL器件,在此全文引入作为参考。
II、测定器件
一般而言,本发明中采用的测定器件可以执行本领域公知的任何类型的测定,包括均匀的和非均匀的免疫测定。均匀测定是其中没有络合的、经标记的物种不和络合的、经标记的物种分离的测定。非均匀测定是其中没有络合的、经标记的物种和络合的、经标记的物种相分离的测定。可以通过物理分离来进行分离,例如,通过将物种之一转移到另一反应容器、过滤、离心、色谱分离、固相捕捉、磁性分离等,可以包括一个或多个洗涤步骤。分离也可以是非物理性的,因为并不进行物种之一或者两者的转移,而是物种在原位互相分离。例如,在一个特定实施方案中采用了非均匀免疫测定。所述免疫测定采用了免疫系统的机制,其中产生了抗体以响应对生物体而言是致病性的或者外来的抗原。这些抗体和抗原,即免疫反应物,能够互相结合,由此形成高度特异性的、可用于确定在流体测试试样中特定抗原的存在性或浓度的反应机制。
参见图2,例如,现在更详细描述经构造以执行非均匀免疫测定的、基于色谱分析的测定器件20。如图所示,测定器件20包含具有第一表面13和相对第二表面15的色谱分析介质23。介质23的第一表面13的位置和载体21相邻。色谱分析介质23通常由能够让测试试样通过的材料制备,比如流体通道、多孔膜等。同样,介质23也由电磁辐射可以通过其传输的材料,比如光散射(例如,半透明)或者透明材料制备。例如,在一个特定实施方案中,色谱分析介质23由光散射多孔膜制备,所述多孔膜由比如但不限于天然、合成、或者经过合成改性的天然材料的材料制备,所述材料比如多糖(例如,纤维素材料,比如纸张和纤维素衍生物,比如醋酸纤维素和硝基纤维素);聚醚砜;聚乙烯;尼龙;聚偏二氟乙烯(PVDF);聚酯;聚丙烯;二氧化硅;和聚合物一起的无机材料,所述无机材料比如失活的氧化铝、硅藻土、MgSO4、或者其它均匀分散在多孔聚合物基质中的无机细粒材料,所述聚合物比如氯乙烯、氯乙烯-丙烯共聚物、和氯乙烯-乙酸乙烯酯共聚物;布料,天然的(例如,棉花)和合成的(例如,尼龙或者人造丝);多孔凝胶,比如二氧化硅凝胶、琼脂糖、葡聚糖、和明胶;聚合物膜,比如聚丙烯酰胺;等等。在一个具体实施方案中,色谱分析介质23由硝基纤维素和/或聚醚砜材料制备。应该理解,术语“硝基纤维素”是指纤维素的硝酸酯,可以是单独的硝基纤维素,或者硝酸和其它酸的混合酯,比如具1-7个碳原子的脂肪羧酸。

如上所述,载体21承载着色谱分析介质23。例如,载体21可以如图2所示设置成直接和色谱分析介质23相邻,或者可以在色谱分析介质23和载体21之间设置一个或多个插入层。无论如何,载体21通常都可以由能够承载色谱分析介质23的任何材料制备。一般而言,载体21由透光材料制备,所述透光材料比如透明的或者光散射(例如,半透明)材料。另外,通常希望载体21不透液体,从而使流过介质23的流体不会通过载体21泄漏。适用于载体的材料的例子包括但不限于玻璃;聚合物材料,比如聚苯乙烯、聚丙烯、聚酯(例如,Mylar膜)、聚丁二烯、聚氯乙烯、聚酰胺、聚碳酸酯、环氧化物、甲基丙烯酸酯、和聚三聚氰胺;等等。为了提供对色谱分析介质23的充分结构支撑,载体21通常选择具有一定的最小厚度。同样,载体21的厚度通常并不大得会对其光学性质产生负面影响。因此,例如,载体21的厚度可以是大约100-大约5000微米,在一些实施方案中是大约150-大约2000微米,在一些实施方案中大约250-大约1000微米。例如,可以从Millipore Corp.of Bedford,Massachusetts获取一种厚度为大约125微米的合适膜条,名为“SHF180UB25”。
如同本领域公知的那样,色谱分析介质23可以流延到载体21上,其中所得的层压材料可以被冲切成所需大小和形状。可替换地,色谱分析介质23可以简单地用例如粘合剂层压到载体21上。在一些实施方案中,硝基纤维素或者尼龙多孔膜被粘合到Mylar膜上。采用粘合剂,比如压敏粘合剂,将多孔膜结合到Mylar膜上。这种类型的层压结构相信可以购自Millipore Corp.of Bedford,Massachusetts。在Durley,III等的美国专利No.5075077中描述了合适的层压材料测定器件结构的其它例子,在此全文引入作为参考。
可以根据多种因素选择用于层压载体21、色谱分析介质23、和/或器件的任何其它层的粘合剂,所述因素包括检测系统的所需光学性质和用来形成测定器件的材料。例如,在一些实施方案中,所选的粘合剂光学透明,并且和色谱分析介质23以及载体21相容。光学透明可以使其它情况下粘合剂对光学检测系统的任何负面影响最小化。合适的光学透明粘合剂可以由例如丙烯酸酯或者(甲基)丙烯酸酯聚合物(比如(甲基)丙烯酸酯)、丙烯酸或者(甲基)丙烯酸单体等形成。示例性的(甲基)丙烯酸酯单体包括非叔烷基醇的单官能丙烯酸酯或者甲基丙烯酸酯,比如丙烯酸甲酯、丙烯酸乙酯、丙烯酸丙酯、丙烯酸正丁酯、丙烯酸异丁酯、丙烯酸2-甲基丁基酯、丙烯酸2-乙基己基酯、甲基丙烯酸2-乙基己基酯、丙烯酸正辛酯、甲基丙烯酸正辛酯、丙烯酸异辛酯、甲基丙烯酸异辛酯、丙烯酸异壬酯、丙烯酸异癸酯、丙烯酸异冰片酯、甲基丙烯酸异冰片酯、乙酸乙烯酯、和其混合物。示例性的(甲基)丙烯酸单体包括丙烯酸、甲基丙烯酸、丙烯酸β-羧乙基酯、衣康酸、巴豆酸、和富马酸等。在Alahapperuma等的美国专利No.6759121中描述了这种光学透明粘合剂的一些例子,在此全文引入作为参考。另外,合适的透明粘合剂也可以得自AdhesivesResearch,Inc.of Glen Rock,Pennsylvania,商品名为Arclear8154,它是无载体的光学透明的丙烯酸压敏粘合剂。其它合适的透明粘合剂可以得自3M Corp.of St.Paul,Minnesota,名称为“9843”或者“8146”。另外,粘合剂施加的方式也可以提高测定器件的光学性质。例如,粘合剂可以提高载体的某些光学性质(例如,漫射性)。因此,在一个特定实施方案中,可以以和需要提高光学性质的区域相应的模式施加所述粘合剂。
再次参见图2,在第二表面15上提供了吸收垫28,该吸收垫通常接收迁移通过整个色谱分析介质23后的流体。如同本领域所公知的那样,吸收垫28也可以有助于促进毛细作用和促进流体流过色谱分析介质23。为了启动对测试试样中分析物的检测,使用者可以直接将测试试样施加到色谱分析介质23的一部分上,测试试样随后通过该介质沿着图2的箭头“L”的方向移动。可替换地,测试试样可以首先施加到和色谱分析介质23流体连通的试样垫(未示出)上。可用于形成吸收垫28和/或试样垫的一些合适材料包括但不限于硝基纤维素、纤维素、多孔聚乙烯垫、和玻璃纤维滤纸。如果需要,试样垫也可以含有一种或多种测定预处理试剂,所述试剂或者散布性地或者非散布性地连接到试样垫上。
在所示的实施方案中,测试试样通过试样垫(未示出)移至和所述试样垫的一端连通的结合物释放垫22。结合物释放垫22由能够通过流体的材料制备。例如,在一个实施方案中,结合物释放垫22由玻璃纤维形成。尽管仅仅示出了一个结合物释放垫22,但是应该理解在本发明中也可以采用其它的结合物释放垫。
为了便于精确检测测试试样中是否存在分析物,可以在测定器件22的不同位置设置预定数量的检测探针。这些检测探针含有直接或间接产生可光检测的信号的物质,比如分子、聚合物、和枝状物等。合适的可检测物质可以包括例如发光化合物(例如,荧光的、磷光的、等等)、放射性化合物、可视化合物(例如,带色染料或者金属物质,例如金)、脂质体或者其它含有产生信号物质的泡囊、酶和/或底物,等等。在Jou等的美国专利No.5670381和Tarcha等的美国专利No.5252459中可能描述了其它合适的可检测物质,在此全文引入作为参考。如果可检测物质带有颜色,则理想的电磁辐射是波长为互补波长的光。例如,蓝色检测探针强烈吸收红光。
在一些实施方案中,可检测物质可以是产生光学可检测信号的发光化合物。例如,合适的荧光分子可以包括但不限于荧光素、铕螯合物、藻胆蛋白、罗丹明、和其衍生物和类似物。其它合适的荧光化合物是通常称作“量子点”的半导体纳米晶体。例如,所述纳米晶体可以含有具有式CdX的核,其中X是Se、Te、和S等。纳米晶体也可以用具有式YZ的外壳钝化,其中Y是Cd或者Zn,Z是S或Se。在Barbera-Guillem等的美国专利No.6261779和Dapprich的美国专利No.6585939中可能也描述了其它合适的半导体纳米晶体的例子,这些专利在此全文引入作为参考。
另外,合适的磷光化合物可以包括一种或多种金属的络合物,所述金属比如钌、锇、铼、铱、铑、铂、铟、钯、钼、锝、铜、铁、铬、钨、和锌等。特别优选的是钌、铼、锇、铂和钯。金属络合物可以含有一种或多种便于该络合物在含水或不含水环境中溶解的配体。例如,配体的一些合适例子包括但不限于吡啶、吡嗪、异烟酰胺、咪唑、联吡啶、三联吡啶、菲咯啉、二吡啶并酚嗪、卟啉、卟吩和其衍生物。所述配体可以例如用烷基、取代的烷基、芳基、取代的芳基、芳烷基、取代的芳烷基、羧酸酯、羧醛、羧酰胺、氰基、氨基、羟基、亚氨基、羟羰基、氨基羰基、脒、胍基、酰脲、含硫基团、含磷基团、和N-羟基-琥珀酰亚胺的羧酸酯取代。
卟啉和卟吩金属络合物具有吡咯基团,所述吡咯基团通过亚甲基桥结合到一起形成环形结构,该环形结构具有螯合金属的内部空腔。许多这些分子在合适溶剂和无氧环境中在室温下都显示出强的磷光性质。能够显示出磷光性质的一些合适的卟啉络合物包括但不限于铂(II)粪卟啉I和II、钯(II)粪卟啉、钌粪卟啉、锌(II)粪卟啉I、其衍生物等。类似地,能够显示出磷光性质的一些合适的卟吩络合物包括但不限于铂(II)四-间-氟苯基卟吩、和钯(II)四-间-氟苯基卟吩。在Schmidt等的美国专利No.4614723、Hendrix的美国专利No.5464741、Soini的美国专利No.5518883、Ewart等的美国专利No.5922537、Sagner等的美国专利No.6004530、和Ponomarev等的美国专利No.6582930中描述了其它合适的卟啉和/或卟吩络合物,在其全文引入作为参考。

在一些情况下,可能具有较长发射寿命的发光化合物可能具有相对大的“斯托克斯频移”。术语“斯托克斯频移”一般定义为发光辐射的谱线或谱带偏移到比激发谱线或谱带长的发射波长处。较大的斯托克斯频移使得发光化合物的激发波长可以远离其发射波长,由于激发波长和发射波长之间的大差别使得更容易从发射的信号中去除反射的激发辐射,所以较大的斯托克斯频移是理想的。而且,斯托克斯频移大也使得来自试样中发光分子的干扰和/或蛋白质或胶体导致的光散射最小,所述蛋白质或胶体存在于某些体液(例如,血液)中。另外,斯托克斯频移大也使得对消除背景干扰所需的昂贵、高精度滤光片的需求最小。例如,在一些实施方案中,发光化合物的斯托克斯频移大于约50纳米,在一些实施方案中大于约100纳米,在一些实施方案中为大约100-大约350纳米。
例如,斯托克斯频移大的荧光化合物的例子包括钐(Sm(III))、镝(Dy(III))、铕(Eu(III))和铽(Tb(III))的镧系元素螯合物。这些螯合物在非常短的波长下被激发后,可以进行强烈红移地、窄带、长寿命地发射。通常情况下,由于在分子中发色团的位置靠近镧系元素,所以螯合物具有强的紫外激发带。在被发色团激发后,激发能可以从受激发色团转移到镧系元素。随后是镧系元素发射荧光。例如,铕螯合物的斯托克斯频移是大约250-大约350纳米,和仅仅约28纳米的荧光素不同。而且,铕螯合物的荧光寿命长,为大约100-大约1000微秒,和其它荧光标记物的大约1-大约100纳秒不同。另外,这些螯合物的发射光谱窄,通常在大约50%发射处的带宽小于大约10纳米。一种合适的铕螯合物是N-(对异硫氰酸根合苄基)-二亚乙基三胺四乙酸-Eu+3。
另外,在水溶液或水悬浮液中为惰性、稳定并且具有固有发荧光性质的镧系元素螯合物也可用于本发明中,以抵消对于成胶束试剂的需求,成胶束试剂常常用于保护在水溶液或水悬浮液中溶解度有限并且出现猝灭问题的螯合物。这种螯合物的一个例子是4-[2-(4-异硫氰酸根合苯基)乙炔基]-2,6-双([N,N-双(羧甲基)氨基]甲基)-吡啶[参见Lovgren,T等,Clin.Chem.42,1196-1201(1996)]。数种镧系元素螯合物也显示出异常高的信噪比。例如,一种这样的螯合物是四配位的β-二酮化物-铕螯合物[参见Yuan,J.和Matsumoto,K.;Anal.Chem.70,596-601(1998)]。除了上述荧光标记物以外,在Mullinax等的美国专利No.6030840、Davidson的美国专利No.5585279、Singer等的美国专利No.5573909、Wieder等的美国专利No.6242268和Hemmila等的美国专利No.5637509中可能描述了其它适用于本发明的标记物,在此全文引入作为参考。
可检测物质,比如上述的那些,可以单独使用或者结合颗粒(有时称作“珠子”或“微珠”)一起使用。例如,可以使用天然颗粒,比如核、支原体、质粒、质体、哺乳动物细胞(例如,红血球影)、单细胞微生物(例如,细菌)、多糖(例如,琼脂糖)。另外,也可以采用合成颗粒。例如,在一个实施方案中,采用了用荧光染料或者带色染料标记的乳胶微粒。尽管在本发明中可以采用任何合成颗粒,但该颗粒通常由聚苯乙烯、丁二烯苯乙烯、苯乙烯丙烯酸-乙烯基三元共聚物、聚甲基丙烯酸甲酯、聚甲基丙烯酸乙酯、苯乙烯-马来酸酐共聚物、聚乙酸乙烯酯、聚乙烯基吡啶、聚二乙烯基苯、聚对苯二甲酸丁二酯、丙烯腈、和氯乙烯-丙烯酸酯等、或者其醛、羧基、氨基、羟基、或者酰肼衍生物制备。在Jou等的美国专利No.5670381和Tarcha等的美国专利No.5252459中可能描述了其它的合适颗粒。可购买的合适荧光颗粒的例子包括Molecular Probes,Inc.以商品名“FluoSphere”(Red 580/605)和“TransfluoSphere”(543/620)销售的荧光羧化微球,以及同样由Molecular Probes,Inc.销售的“Texas Red”和5-和6-羧基四甲基罗丹明。另外,可购买的合适带色乳胶微粒的例子包括Bang’s laboratory,Inc.销售的羧化乳胶珠子。在本发明中也可以采用金属颗粒(例如,金颗粒)。
在采用时,颗粒的形状通常可以变化。例如,在一个具体实施方案中,颗粒为球形。但是,应该理解的是,本发明还考虑了其它形状,比如板状、棒状、盘状、条状、管状、不规则形状等。另外,颗粒尺寸也可以变化。例如,颗粒的平均尺寸(例如,直径)可以为大约0.1纳米-大约1000微米,在一些实施方案中为大约0.1纳米-大约100微米,在一些实施方案中为大约1纳米-大约10微米。例如,常常需要的是“微米级”颗粒。当采用时,这些“微米级”颗粒的平均尺寸可以是大约1微米-大约1000微米,在一些实施方案中是大约1微米-大约100微米,在一些实施方案中是大约1微米-大约10微米。同样,也可以采用“纳米级”颗粒。这种“纳米级”颗粒的平均尺寸可以是大约0.1-大约10纳米,在一些实施方案中是大约0.1-大约5纳米,在一些实施方案中是大约1-大约5纳米。
在一些情况下,可能需要对检测探针进行某种方式的修改使其能够更容易结合到分析物上。在这种情况下,可以用某些特异性结合成分对检测探针进行改性,所述结合成分粘附到检测探针上形成结合探针。特异性结合成分通常是指特异性结合对的一个成员,所述特异性结合对即为其中一个分子化学和/或物理结合到第二个分子上的两个不同分子。例如,免疫活性特异性结合成分可以包括抗原、半抗原、适体、抗体(初级抗体或次级抗体)、和其络合物,包括通过重组DNA方法或者肽合成方法形成的那些。抗体可以是单克隆抗体或者多克隆抗体、重组蛋白或者其混合物或者片段、以及抗体和其它特异性结合成分的混合物。对本领域技术人员而言,所述抗体的具体制备以及用作特异性结合成分的适用性都是公知的。其它常见的特异性结合对包括但不限于生物素和抗生物素蛋白(或者其衍生物)、生物素和抗生蛋白链菌素、碳水化合物和外源凝集素、互补核苷酸系列(包括在DNA杂交测定中采用的探针和捕获核酸序列,用以检测靶向核酸序列)、互补肽系列(包括通过重组方法形成的那些)、效应物和受体分子、激素和激素结合蛋白、酶辅因子和酶、酶抑制剂和酶等等。另外,特异性结合对可以包括是原始特异性结合成分类似物的成分。例如,可以采用分析物的衍生物或者片段,即分析物-类似物,只要它具有至少一个和分析物相同的抗原决定基即可。
特异性结合成分通常可以采用各种公知技术的任一种连接到检测探针上。例如,可以采用下列能够实现蛋白质偶联反应的基团来实现特异性结合成分和检测探针(例如,颗粒)的共价连接:羧基、氨基、醛、溴乙酰基、碘乙酰基、硫醇、环氧基和其它的反应性或者连接官能团、以及残余自由基和自由基阳离子。由于检测探针的表面可以含有较高表面浓度的极性基团,所以也可以结合表面官能团作为官能化的共聚单体。另外,尽管检测探针在合成后常常经过官能化,比如用聚(苯硫酚)官能化,但是检测探针能够直接和蛋白质共价连接,无需进一步改性。例如,在一个实施方案中,结合的第一步是采用碳化二亚胺活化探针表面上的羧基。在第二步中,活化的羧酸基团和抗体的氨基反应形成酰胺键。所述活化和/或抗体偶联可以在缓冲液比如磷酸盐缓冲的盐水(PBS)(例如,pH为7.2)或者2-(N-吗啉代)乙烷磺酸(MES)(例如,pH为5.3)中进行。随后,将得到的检测探针和例如乙醇胺接触,以封闭所有残余的活性位点。总体而言,该过程形成了结合检测探针,其中抗体共价连接到探针上。除了共价键合以外,本发明中也可以采用其它连接技术,比如物理吸附。
再次参见图2,色谱分析介质23还限定了检测区31,在该检测区内固定了能够结合到结合检测探针上的受体材料(receptivematerial)。例如,在一些实施方案中,所述受体材料可以是生物受体材料。所述生物受体材料是本领域公知的,可以包括但不限于抗原、半抗原、蛋白质A或G、中性抗生物素蛋白、抗生物素蛋白、抗生蛋白链菌素、captavidin、初级或次级抗体(例如,多克隆抗体、单克隆抗体等)、和其络合物。在许多情况下,希望这些生物受体材料能够结合到在检测探针上的特异性结合成分(例如,抗体)上。受体材料充当在分析物和结合检测探针之间形成的络合物的固定结合位点。具体而言,分析物比如抗体、抗原等通常具有两个或多个结合位点(例如,抗原决定基)。当到达检测区域31时,这些结合位点之一被结合探针的特异性结合成分占据。但是,分析物的自由结合位点可以结合到被固定的受体材料上。当结合到被固定的受体材料上时,络合的探针形成新的三元三明治型络合物。
检测区31通常可以提供任意数量的不同检测区,以使使用者可以更好地确定测试试样中具体分析物的浓度。每个区可以含有相同的受体材料,或者可以含有不同的受体材料以捕获多种分析物。例如,检测区31可以包括两个或多个不同的检测区(例如,线、点等)。可以沿着和测试试样流过测定器件20的方向基本垂直的方向,以线的形式设置检测区。同样,在一些实施方案中,可以沿着和测试试样流过测定器件20的方向基本平行的方向,以线的形式设置检测区。
尽管检测区31提供了检测分析物的精确结果,但是有时候在实际测试条件下难以确定分析物在测试试样中的相对浓度。因此,测定器件20也可以包括校准区32。在这个实施方案中,校准区32设置在检测区31的下游。但是,可替换地,校准区32也可以设置在检测区31的上游。校准区32可以具有受体材料,所述受体材料能够和在色谱分析介质23长度上通过的校准探针或者未络合的检测探针相结合。当采用时,校准探针可以用和检测探针相同或不同的材料制备。一般而言,校准探针的选择方式应使其不会结合到检测区31的受体材料上。
校准区32的受体材料可以和检测区31中所用的受体材料相同或者不同。例如,在一个实施方案中,受体材料是生物受体材料。另外,也可能需要采用各种非生物材料作为校准区32的受体材料。聚电解质可以具有净正电荷或负电荷,以及通常为中性的净电荷。例如,一些具有净正电荷的聚电解质的例子包括但不限于聚赖氨酸(可购自Sigma-Aldrich Chemical Co.,Inc.of St.Louis,Missouri)、聚乙烯亚胺;表氯醇官能化的多胺和/或多氨基胺,比如聚(二甲胺-共-表氯醇);聚二烯丙基二甲基氯化铵;阳离子型纤维素衍生物,比如用季铵水溶性单体接枝的纤维素共聚物或者纤维素衍生物;等等。在一个具体实施方案中,可以采用CelQuatSC-230M或者H-100(得自NationalStarch & Chemical,Inc.),它们是含有季铵水溶性单体的纤维素衍生物。而且,具有净负电荷的聚电解质的一些例子包括但不限于聚丙烯酸,比如聚(乙烯-共-甲基丙烯酸,钠盐),等等。也应该注意的是,在本发明中还可以采用其它聚电解质,比如两性聚电解质(即,具有极性和非极性部分)。例如,合适的两性聚电解质的一些例子包括但不限于聚(苯乙烯基-b-N-甲基2-乙烯基吡啶碘化物)和聚(苯乙烯基-b-丙烯酸),两者都可得自Polymer Source,Inc.of Dorval,Canada。在Song等的美国专利申请公开No.2003/0124739中更详细描述了采用聚电解质的内校准体系的其它例子,在此全文引入作为参考。
在一些情况下,色谱分析介质23也可以限定控制区(未示出),所述控制区向使用者发送测定正在正确进行的信号。例如,控制区(未示出)可以含有通常能够和探针或者探针上固定的受体材料形成化学和/或物理键的固定受体材料。所述受体材料的一些例子包括但不限于抗原、半抗原、抗体、蛋白质A或G、抗生物素蛋白、抗生蛋白链菌素、次级抗体和其络合物。另外,也可能需要采用各种非生物材料作为控制区受体材料。例如,在一些实施方案中,控制区受体材料也可以包括可以和未捕获的探针相结合的聚电解质,比如上述聚电解质。由于控制区的受体材料仅仅对于探针具有特异性,所以无论分析物是否存在,都会产生信号。控制区可以沿着介质23设置在任何位置,但是通常设置在检测区31的上游。
当采用检测器件20时,可以采用各种格式来检测是否存在分析物。例如,“三明治”格式通常涉及将带有对分析物的特异性结合成分(例如抗体)的结合检测探针和测试试样混合,从而在分析物和结合探针之间形成络合物。随后,使这些络合物可以和固定在检测区中的受体材料(例如,抗体)相接触。在分析物/探针结合络合物和固定的受体材料之间发生结合,由此使“三明治”络合物被定位下来,该“三明治”络合物可以被检测出来以表明存在分析物。这种技术可用于获得定量或者半定量结果。在Grubb等的美国专利No.4168146和Tom等的美国专利No.4366241中描述了所述三明治型测定的一些例子,在此全文引入作为参考。在竞争性测定中,标记的探针通常和与被确定为分析物或者其类似物的分子结合。因此,标记的探针和目标分析物竞争可用的受体材料。竞争性测定通常用于检测分析物比如半抗原,每种半抗原都是单价的并且能够仅仅结合一种抗体分子。在Deutsch等的美国专利No.4235601、Liotta的美国专利No.4442204和Buechler等的美国专利No.5208535中描述了竞争性免疫测定器件的例子,在此全文引入作为参考。在Lambotte等的美国专利No.5395754、Jou等的美国专利No.5670381和Malick等的美国专利No.6194220中也公开了各种其它器件构造和/或测定格式,在此全文引入作为参考。
III、光学检测系统
当用作光学检测系统的照明源时,EL器件可以提供多种益处。例如,和许多常规光学检测系统采用的点光源(例如,LED)不同,EL器件发射较均匀的漫射光,因此可以提供均匀的照明。这样可以消除对点光源照明系统中通常需要的另外漫射器的需求。另外,可以简单地通过改变驱动信号的电压或频率来很容易地控制EL器件发射的光强。因此,EL器件使得可以采用较简单的、便携的、便宜的光学读数器。
采用了所述EL器件的光学检测系统的实际构造和结构通常可以变化,这是本领域技术人员很容易知道的。例如,可以采用的光学检测技术包括但不限于发光(例如,荧光、磷光等)、吸收(例如,荧光性的或者非荧光性的)、衍射等。通常,光学系统能够发射光,也能寄存检测信号(例如,透射的或者反射的光、发射的荧光或磷光等)。例如,在一个实施方案中,可以采用反射分光光度计检测能显示出可见颜色的探针(例如,染色的乳胶微粒)是否存在。例如,在Kaylor等的美国专利申请公开No.2003/0119202中描述了一种合适的反射分光光度计,其在此全文引入作为参考。在另一实施方案中,可以采用反射模式的分光荧光光度计来检测能发出荧光的探针是否存在。合适的分光荧光光度计和相关的检测技术描述在例如Song等的美国专利申请公开No.2004/0043502中,在此全文引入作为参考。同样,也可以采用透射模式的检测系统来检测是否存在检测探针。
再次参见图2,例如,示出了基于透射的光学检测系统的一个实施方案,该方案采用了电致发光(EL)照明源52和检测器54。检测器54通常可以是本领域公知的、能够传感光信号的任何器件。例如,检测器54可以是经构造进行空间分辨的电子成像检测器。所述电子成像检测器的一些例子包括高速、线性电荷耦合器件(CCD)、电荷注入器件(CID)、互补性金属氧化物半导体(CMOS)器件等。这些图像检测器例如通常是电子光传感器的二维阵列,但是也可以采用包括单线检测器像素或者光传感器的线性成像检测器(例如,线性CCD检测器),比如例如用于扫描图像的那些。每个阵列包括一系列已知的、唯一的、可以称作“地址”的位置。图像检测器中的每个地址被覆盖一定面积(例如,通常为盒子形式或者矩形的面积)的传感器占据。所述面积通常称作“像素”或者像素面积。检测器像素例如可以是CCD、CID、或者CMOS传感器、或者任何检测或测量光的其它器件或者传感器。检测器像素的大小可以变化巨大,在一些情况下直径或长度可以低至0.2微米。
在其它实施方案中,检测器54可以是缺乏空间分辨能力的光传感器。例如,所述光传感器的例子可以包括光倍增器件、光电二极管比如雪崩光电二极管或者硅光电二极管、等等。硅光电二极管有时是有利的,因为其便宜、灵敏、能够高速操作(上升时间短/带宽大)、并且容易集成到大多数其它半导体技术和单片电路中。另外,硅光电二极管体积小,使其能够很容易结合到系统中,和基于膜的器件一起使用。如果采用了硅光电二极管,则发射信号的波长范围可以落在其灵敏度范围(400-1100纳米)之内。另一种合适的检测器是CdS(硫化镉)光电导管,其优点在于其光谱灵敏度和人视力相似,从而可以更容易滤去经反射的发射辐射。
再次参见图2,检测器54位置和载体21相邻,EL照明源52的位置和色谱分析介质23的第二表面15相邻。同样,检测器54的位置可以和色谱分析介质23的第二表面相邻,而EL照明源的位置可以和载体21相邻。因此,EL照明源52可以同时发射光到检测区31和校准区32,检测器54同样也可以同时接收到来自检测区31和校准区32的检测探针的检测信号。可替换地,EL照明源52可以被构造成连续发射光到检测区31和校准区32上。另外,也可以为校准区32采用独立的照明源和/或检测器(未示出)。还应该理解的是,EL照明源52和检测器54可以设置在测定器件20的同一侧,比如都和色谱分析介质23相邻。
为了提高光学检测系统的信噪比但无需一定类型的复杂昂贵光学元件,例如透镜或其它光导元件,可以使EL照明源52和/或检测器54距测定器件20的距离最小化。例如,如图3a所示,传输较大距离的光(方向箭头指示)容易扩散,由此导致一些光子不能到达测试试样或者检测器54。为了减少光散射,可以采用透镜将光聚焦在所需方向,如图3b所示。但是,如图3c和3d所示,通过简单地将EL照明源52和/或检测器54移至更靠近测定器件20,就可以降低对所述昂贵复杂设备的需求。采用更短的光程使得光更少扩散。例如,图3c举例说明了其中EL照明源52的位置更靠近测定器件20的实施方案,而图3d举例说明了其中EL照明源52和检测器54的位置都更靠近测定器件20的实施方案。因此,在一些实施方案中,EL照明源和/或检测器54距离测定器件20的位置可以小于大约5毫米,在一些实施方案中小于大约3毫米,在一些实施方案中小于大约2毫米。例如,EL照明源52可以直接层压到载体21上。同样,如同下面将更详细讨论的那样,EL照明源52和/或检测器54在有些情况下可以直接接触色谱分析介质23。例如,EL照明源52可以携带所述介质23,由此也充当介质的载体。但是,在其它情况下,可能需要将EL照明源52和/或检测器54保持在大得足以防止任何生物试剂被污染的距离。例如,EL照明源52和/或检测器54有时可以设置在距离测定器件20大约1-大约3毫米处。
在图2中,所示EL照明源52是和测定器件20相分离的元件。但是,本发明也考虑了其中照明源和测定器件20一体化的实施方案。例如,在一些实施方案中,载体21可以同时充当色谱分析介质23的物理载体,同样充当光学检测系统的EL照明源。采用EL器件作为载体21由于不再需要另外的、通常成本很高并且导致复杂的、耗空间的系统的光源,而为最终光学检测系统带来了明显的益处。也即,EL器件可以层压到色谱分析介质23上,并同时充当载体21和光学检测系统的光源。可以选择具有一定程度挠性的EL器件,从而使其可以很容易操纵和/或切成测定器件20所需的理想形状和尺寸。一个强度和挠性足以用作载体21的市售EL器件是购自Graphic Solutions Int’l,LLC ofBurr Ridge,IIIinois的灯具,商品名为“Proto-Kut”。
图4示出了其中EL器件用作测定器件的载体的本发明的一个具体实施方案。具体而言,所示测定器件220包括色谱分析介质223、EL器件221、吸收垫228、和结合物释放垫222。介质223具有第一表面212和第二表面214,其中第一表面212的位置和EL器件221相邻。检测区231和校准区232由介质223限定,用于提供检测和校准信号。另外,检测器254的位置和介质223的第二表面214相邻。在本具体实施方案中,EL器件221同时充当照明源和介质223的载体。EL器件221的引线256经由布线连接到驱动电路260上,该驱动电路进而又连接到电源266上。驱动电路260和电源266的详细情况依赖于具体EL器件的要求。例如,由于为了和测定器件220的小尺寸相应使得EL器件221可以较小,所以可以采用低压电路和电池电源来降低系统的成本和复杂性。但是,也可以采用更高压的电路,比如将DC电压转变成AC输出以驱动EL器件221的驱动电路。所述AC转换器可以产生50-5000Hz的大约60-300伏AC电压。
如上所述,本发明的EL照明器件可以提供漫射照明。以此方式,可以基本消除对某些外部光学元件比如漫射器的依赖性。无论如何,所述光学元件可以用于本发明的一些实施方案中。如果采用,则可以采用分离的光学元件作为EL照明源52和检测器54,或者它们可以分享共同的光学元件。例如,在本发明中可以采用光学漫射器来向某方向散射光,比如朝向和/或背离检测区。合适的光学漫射器可以包括沿着各个方向散射光的漫射器,比如毛玻璃、乳色玻璃、不透明塑料、化学蚀刻的塑料、加工的塑料等等。乳色玻璃漫射器含有用于均匀漫射光的乳白色“乳色”涂层,由此形成类朗伯光源。其它合适的散光漫射器包括含有散光材料比如二氧化钛或硫酸钡颗粒的聚合物材料(例如,聚酯、聚碳酸酯等)。在其它实施方案中,可以采用全息漫射器,使照明源发射的光线均匀化并同时赋予其预定的方向性。所述漫射器可以含有控制光传播方向的微雕刻的表面结构。在Petersen等的美国专利No.5534386中更详细描述了所述全息漫射器的例子,在此全文引入作为参考。
滤光片(未示出)也可以设置在EL照明源52和/或检测器54附近。滤光片可以在所需波长范围具有高透射率,在一个或多个不需要的波段可以具有低透射率,从而滤去来自EL照明源52的不需要的波长。例如,在发光检测系统中,不需要的波长范围可以包括如下这些波长:产生可检测的试样自发荧光和/或落在激发最大波长的大约25-大约100纳米范围内并因而是潜在的、来自被散射的激发照明的背景噪声源。可用于本发明的滤光片的一些例子包括但不限于染色的塑料树脂或者明胶滤光片、二色性滤光片、薄的多层膜干涉滤光片、塑料或玻璃滤光片、环氧树脂或者固化的透明树脂滤光片。在一个实施方案中,检测器54和/或EL照明源52可以嵌入到或者封装到滤光片中。
另外,也可以采用透镜收集和会聚光。本发明的一个具体实施方案采用微透镜将光会聚向测试试样和/或检测器54。合适的微光学透镜包括但不限于梯度折射率(GRIN)透镜、球面透镜、和Fresnel透镜等。例如,梯度折射率透镜通常是圆柱形,折射率沿着径向以抛物线形式变化。球面透镜通常是球形,折射率沿径向是常数。由于其尺寸较小,所述微透镜在本发明中尤其有利。可以采用任何各种公知的技术来制备微透镜。例如,微透镜可以如下制备:将衬底(例如,硅或石英)浸没到碱盐溶液中,使得在衬底和盐溶液之间通过衬底上形成的掩模发生离子交换,由此获得折射率的分布和掩模图案相应的衬底。另外,可以用紫外光照射光敏单体以使光敏单体被照射的部分聚合。因此,在被照射部分和未照射部分之间的渗透压的作用下,被照射部分膨胀成透镜构造。在另一实施方案中,光敏树脂可以被图案化成圆形,加热到高于其软化点的温度,以使每个圆形图案的周边部分由于表面张力而下陷(sag)。这种方法称作“热下陷方法”。另外,透镜衬底可以被简单地机械加工成透镜形状。在Watanabe等的美国专利No.5225935、Guerra的美国专利No.5910940和Nishikawa的美国专利No.6411439中描述了用于制备微透镜或其它微光学元件的其它合适技术,其在此全文引入作为参考。
另外,可以采用掩模比如黑色涂层或染料来防止光穿过测定器件20的一个或多个部分。也可以采用光导元件使光指向所需方向,比如单根光纤、光纤束、分叉光纤束的片段、大直径光管、平面波导、衰减全反射型晶体、二色镜、平面镜或者其它光导元件。在Golden的美国专利No.5827748、Bruno等的美国专利No.6084683、Catt等的美国专利No.6235241、Rushbrooke等的美国专利No.6556299和Bentsen等的美国专利No.6566508中描述了可用于本发明的其它光学功能材料的例子,其在此全文引入作为参考。
如果需要,测定器件自身的光学性质可以根据检测系统的光学要求进行选择性定制。例如,再次参见图2,本发明的一个实施方案采用选择性控制载体21以使光学检测系统的性能最优化。例如,在一个特殊实施方案中,载体21可以透光,从而使得光可以从EL照明源52传播到检测器54。另外,载体21可以充当EL照明源52和/或检测器54的漫射器,以提高光学检测系统的信噪比。载体21也可以充当检测系统的滤光片。因此,在示例的实施方案中,来自EL照明源52的光被检测区31和/或校准区32中的探针(未示出)吸收。该探针产生的信号在到达检测器54之前被滤光片衰减。滤光片可以例如在发射波长范围具有高透射率,在一个或多个不需要的波段可以具有低透射率,从而滤去来自检测器54的不需要的波长。光学检测系统也可以包括设置在EL照明源52和色谱分析介质23之间的另外的滤光片(未示出)。该另外的滤光片可以在激发波长范围具有高透射率,在一个或多个不需要的波段可以具有低透射率。可替换地,可以将另外的滤光片集成到EL照明源52和/或检测器54中。载体21也可以具有其它理想的光学质量。例如,载体21可以含有掩模、光导元件、透镜等。在一些情况下,当用于载体21中时,需要采用“微光学”元件。微光学元件通常的尺寸小于大约2毫米,并且以一维或者二维排列。维光学元件由于尺寸小,所以更容易用于载体21中。
当载体21针对具体光学性质进行最优化后,可以选择用于形成载体21的材料使其具有所需的光学性质。可替换地,可以在形成测定器件20之前和/或之后,将所需的光学功能材料简单施加到载体21上。所述光学功能材料可以以各种方式施加到载体21上。例如,光学功能材料可以简单地被染色到或者涂覆到载体21的一个或多个表面上。当以此方式施加时,光学功能材料可以覆盖载体21的仅仅一部分或者整个表面。例如,在一个实施方案中,光学功能材料施加到载体21上和检测区31和/或校准区32相应的部分。以此方式,光学功能材料可以强化测定器件20在使用过程中产生的检测信号或者校准信号。可替换地,光学功能材料也可以结合到载体21的结构中。例如,可以采用已知的技术比如印花、压印、模塑等形成内光学元件。
根据本发明的一些实施方案,光学检测系统也可以采用各种其它的提高分析物检测灵敏度的组件。例如,检测系统有时可以为测定器件采用试样架。例如,参见图5-8,更详细描述了采用所述试样架的光学检测系统的各种实施方案。例如,图5举例说明了可用于本发明光学检测系统的试样架400的一个实施方案。如图所示,试样架400包括下部分402和上部分403。上部分403能够绕着折页404移动,从而使其可以处于打开位置(图5A和5B)和关闭位置(图5C)。而且,试样架400可以包括和下闩锁415相配的上闩锁413,用于将试样架400固定在其关闭位置。也可以提供手柄417,使得使用者可以更容易抓住试样架400。
如图所示,可以在试样架400中限定在下部分402和上部分403之间的内部放置一个或多个测定条405。在该具体实施方案中,测定条405的承载片(未示出)也被层压到EL器件412上。这样使得在使用过程中EL器件412的位置可以靠近测定条405,从而优化光学检测系统的信噪比。EL器件412可以以多种不同方式和引线电接触。例如,EL器件的下表面(例如,阴极侧)可以和八个(8)孔409相邻,但是当然可以采用任何数目的孔。参见图6和7,这些孔409的位置可以与卡盘300的八(8)根相应引线313(图6和7中仅仅示出3根)相邻。具体而言,使用者可以将试样架400的一端419和由卡盘300的主体部分310限定的试样端口315对齐,然后将试样架400经由平行导轨319滑动通过试样端口315,直到孔409位于引线313上方为止。以此方式,EL器件的下侧(例如,阴极侧)和引线313电接触。尽管没有具体示出,但EL器件412的上表面(例如,阳极侧)也可以延伸到测定条405以外并和引线电接触。例如,引线(未示出)可以设置在试样架400的上部分403的内表面(图5),从而在试样架关闭时引线和EL器件412的上表面的延伸部分相接触。因此,在使用过程中,El器件412发光,所述光和测定条405上的检测探针相接触。检测探针产生检测信号,该检测信号通过试样架400的上窗口406和卡盘300的上窗口326到达照相机500的透镜501。
如图8所示,如果需要,上述元件可以容纳在不能透射EL器件发射的或者检测器寄存的电磁辐射的外罩600中,从而使该系统在光学上隔离。例如,在所示实施方案中,试样架400(图5)、卡盘300(图6)和照相机500(图7)被设置在外罩600内。虽然所示的为卵形,但是应该理解的是可以采用任何其它合适的形状和/或尺寸,比如圆形、方形、矩形等。另外,如同本领域技术人员很容易了解的那样,其它光学元件也可以采用并任选地容纳在外罩600中,比如电子电路、微处理器、显示器、镜子、滤光片、透镜等。
无论形成光学检测系统的具体方式如何,根据本发明可以实现对分析物存在与否或浓度的定性、定量或半定量确定。例如,在一个实施方案中,通过将在检测区31中捕捉的探针的信号强度Is和预定的分析物浓度建立关系,可以定量或者半定量地确定分析物的量。在一些实施方案中,信号强度Is也可以和在校准区32中捕捉的探针的信号强度Ic相比较。信号强度Is可以和信号强度Ic比较。在该实施方案中,校准区中探针的总量是预定的和已知的,因此可以用于校准。例如,在一些实施方案(例如,三明治型测定)中,分析物的量和Is∶Ic成正比。在其它实施方案(例如,竞争性测定)中,分析物的量和Is∶Ic成反比。基于检测区31落入的强度范围,可以确定分析物的大致浓度范围。
可以任选地采用微处理器将检测器54的测量值转变成定量或者半定量表明分析物存在性或浓度的结果。微处理器可以包括存储能力,使得使用者可以重新调用最近几次的结果。本领域技术人员会认识到,本发明可以采用任何合适的计算机可读存储器件,比如RAM、ROM、EPROM、EEPROM、闪存卡、数字视频盘、Bernoulli盒等等。也可以采用光学密度(灰度)标准来简化定量结果,这是本领域所公知的。另外,可以任选地采用任何公知的软件来收集数据。例如,Logitech照相机软件可以用来收集基于Logitech照相机的检测器所获得的数据。在保存图像后,可以采用任何已知的商业软件包比如得自Molecular Dynamics of Sunnyvale,CA的ImageQuant进行分析。如果需要,可以采用液晶(LCD)或者LED显示器将结果传递给使用者。
参见下列实施例可以更好地理解本发明。
实施例1
验证了形成具有电致发光照明源的光学检测系统的能力。首先,提供层压到Mylar膜载体上的硝基纤维素膜(SHF-120,Millipore Corp.of Bedford,MA)。采用得自Adhesives Research of GlenRock,PA的名称为“ARclear 8154”的透明粘合剂将Mylar膜直接连接到电致发光(EL)器件上。小心确保不出现气泡、灰尘和污染物。EL器件由BKL,Inc.of Burr Ridge,IL制备,大小为60毫米×300毫米。另外,EL器件也具有在482纳米和580纳米处的双宽发射峰,以发出“白”光。
将GoldlineTM(得自British Biocell International的聚赖氨酸溶液)以条状形式施加到所述膜上形成校准区。在所述多孔膜上固定对C-反应蛋白(BiosPacific,Inc.,浓度为1mg/ml)显示活性的单克隆抗体,以形成检测区。将所述卡片在37.5℃的温度干燥1小时。在将卡片从烘箱中取出后,在膜上靠近校准区的一端连接纤维素芯吸垫(wickingpad)(Millipore Co.)。去除试样的通常用来连接结合物释放垫和试样垫的另一端。然后将卡片切成条(大小为4mm×60mm)。将羧化的蓝色乳胶珠子(0.3mm,Bang’s Laboratories)结合到对C-反应蛋白(BiosPacific,Inc.,浓度为1mg/ml)显示活性的单克隆抗体上。所述结合体和不同浓度的C反应蛋白(CRP)血清标样(Kamiya)混合,置于微孔板中,用半截测试棒(half sticks)测试。在一分钟内显现蓝色的检测和对照线。
在环境条件下干燥1小时后,将侧流条一次四个加载到图5所示的试样架中。试样架在关闭时固定所述测流条,固定方式使得EL器件底面上露出的电极和试样架中的孔对齐。如图7和8所示,随后将试样架插入容纳有照相机系统的外罩中。外罩的目的是将该系统和外部环境在光学上隔离,并确保照相机和测定器件之间正确对齐。照相机是得自Logitech Inc.of Fremont,CA的Logitech QuickCam 3000,并采用USB连接到标准台式电脑上。照相机配有得自Edmund IndustrialOptics of Barrington,NJ的Finite Conjugate Micro Video Imaging Lens(焦距8mm,NT54-853),其位置使得硝基纤维素膜的表面和CCD的成像面相距47毫米。EL器件用400Hz、100V的AC电源(ACM-500,得自Behlman,Hauppauge,NY)供电。在外罩的内侧安装了加载弹簧的触头(70AD/公/4头,Bourns,Riverside,CA),通过试样架中的孔发生电接触。
收集经过照射的测定器件的图像,并用Visual Basic(VB)软件分析。VB软件采用ActiveX模块(QCSDK1)控制Logitech Twain器件驱动器,该驱动器进而控制CCD照相机。控制各种图像获取参数,包括亮度、曝光度、增益、饱和度和白平衡。每个参数的值是:亮度=204(总数256),曝光度=1/300秒,增益=0(总数256),饱和度=120(总数256),白平衡=120(总数256)。另外,连续获取10个图像,经过平均化以降低噪声。在获取了平均图像后,在特征和代表性背景区域周围放置矩形框并调整矩形框的大小,来确定分析的目标区域(ROI)(即,波带和其周边)。计算背景区域中的像素平均值,用以使ROI中的像素归一化。另外用来自空白条的图像的校准数据来校正该数据。采用不规则四边形方法计算目标区域中像素的平均强度和像素的面积。结果显示在屏幕上,并用强度数据画图。图9示出了不同浓度的C反应蛋白的响应曲线。
实施例2
验证了形成具有电致发光照明源的光学检测系统的能力。首先,提供层压到Mylar膜载体上的硝基纤维素膜(SHF-120,Millipore Corp.of Bedford,MA)。采用得自Adhesives Research of GlenRock,PA的名称为“ARclear 8154”的透明粘合剂将Mylar膜直接连接到电致发光(EL)器件上。小心确保不出现气泡、灰尘和污染物。EL器件由BKL,Inc.of Burr Ridge,IL制备,大小为60毫米×300毫米。另外,EL器件也具有在525纳米处的最大发射,以发出“绿”光。
将对C反应蛋白(BiosPacific,Inc.,浓度为1g/ml)有活性的单克隆抗体结合到尺寸为40纳米的胶体金颗粒上。随后,将所述结合物在2mmol水合硼酸钠(Borax,pH7.2)和50%蔗糖(最终10%的蔗糖)中稀释。采用Kinematic 1600分布器将结合物喷洒到5毫米宽的玻璃纤维条(Millipore GF33),喷洒率为5微升/厘米,床速度为5厘米/秒。喷洒后的结合物条在室温、小于20%的相对湿度下干燥过夜。然后,将结合条和干燥剂一起热密封到不透性的袋子中。将羊抗鼠抗体(GAM)在磷酸盐缓冲的盐水(PBS)中稀释成0.1毫克/毫升,并采用Kinematic1600分布器以条形分布到硝基纤维素膜(HF120,Millipore)上,分布率为1微升/厘米,床速度为5厘米/秒。在GAM测试线下方,另外精确地分布上条形的Biogenesis CRP(KC202004A,2.59毫克/毫米)。卡片在37℃干燥1小时。在上面将芯吸和结合物带连接(3mm重叠)并以条形结合到硝基纤维素膜上。将CRP标样(Scipac)在PBS中稀释,得到下列最终CRP浓度:100、20和0微克/毫升。将200微升的每种标准溶液施加到条上。在环境条件下干燥1小时后,如实施例1所述对数个侧流条进行分析。结果如下表1所示。

实施例3
验证了形成具有电致发光照明源的光学检测系统的能力。EL器件由BKL,Inc.of Burr Ridge,IL制备,大小为60毫米×300毫米。另外,EL器件也具有在525纳米处的发射峰,以发出“绿”光。EL器件用400Hz、100V的AC电源(ACM-500,得自Behlman,Hauppauge,NY)供电。EL器件切成4mm×60mm的条,并采用加载弹簧的触头(70AD/公/4头,Bourns,Riverside,CA)插入到图5-8所示的外壳中,从而通过试样架中的孔发生电接触。外罩容纳有两个蓝光增强硅光电二极管(PDB-V601,Photonic Detectors of Simi Valley,California)。外罩的目的是使该系统和外部环境在光学上隔离,并且确保光电二极管和测定器件之间正确对齐。光电二极管的位置使其在插入时距离膜表面为100微米。两个光电二极管的引线的连接使得所述二极管串联,但互相反相(即,两个光电二极管的阴极被焊接在一起)。两个阳极线连接到万用表(123 Industrial Scopemeter,Fluke,Everett,WA)的探针引线上。
虽然对本发明已经参考其具体实施方案进行了详细描述,但是应该理解的是,本领域技术人员在理解了前述内容时,可以很容易地理解这些实施方案的替换、改变和等同方案。相应地,本发明的范围应该由所附权利要求和其任何等同方案评价。
Claims (24)
1.用于测试测试试样中分析物的存在性或量的光学检测系统,所述系统包括提供电磁辐射的电致发光照明源,所述电磁辐射能够导致产生与所述分析物的存在性或量相关的检测信号。
2.权利要求1的光学检测系统,进一步包括:
测定器件,包括和检测探针连通的色谱分析介质,所述检测探针能够产生所述检测信号;和
检测器,能够寄存所述检测探针产生的所述检测信号。
3.权利要求2的光学检测系统,其中所述电致发光照明源和所述检测器置于所述测定器件的同一侧。
4.权利要求2的光学检测系统,其中所述电致发光照明源和所述检测器置于所述测定器件的相对侧,使得色谱分析介质被置于限定在所述电致发光源和所述检测器之间的电磁辐射路径中。
5.权利要求2的光学检测系统,其中所述色谱分析介质是多孔膜或者流体通道。
6.权利要求2的光学检测系统,其中受体材料被固定在由所述色谱分析介质限定的检测区内,所述受体材料经构造以结合所述检测探针或其络合物的至少一部分。
7.权利要求2的光学检测系统,其中所述色谱分析介质由所述电致发光照明源承载。
8.权利要求7的光学检测系统,其中所述电致发光照明源被层压到所述色谱分析介质的载体上。
9.权利要求7的光学检测系统,其中所述电致发光照明源被层压到所述色谱分析介质上。
10.权利要求9的光学检测系统,其中采用光学透明粘合剂将所述电致发光源层压到所述色谱分析介质上、层压到所述色谱分析介质的载体上、或者层压到其结合上。
11.用于检测测试试样中分析物的存在性或量的光学检测系统,所述系统包括:
测定器件,包括和检测探针连通的多孔膜,所述检测探针能够产生检测信号;
电致发光照明源,能够提供使所述检测探针产生所述检测信号的电磁辐射;和
检测器,能够寄存所述检测探针产生的所述检测信号。
12.权利要求11的光学检测系统,其中所述电致发光源和所述检测器置于所述测定器件的同一侧。
13.权利要求11的光学检测系统,其中所述电致发光照明源和所述检测器置于所述测定器件的相对侧,使得所述多孔膜被置于限定在所述电致发光源和所述检测器之间的电磁辐射路径中。
14.权利要求11的光学检测系统,其中所述多孔膜由所述电致发光照明源承载。
15.前述任一权利要求的光学检测系统,其中所述电致发光照明源是分散体或者薄膜器件。
16.前述任一权利要求的光学检测系统,其中所述电致发光照明源包含夹在至少两个电极之间的发光层。
17.权利要求16的光学检测系统,其中至少一个所述电极包含碳或者选自铝、金、银、铜、铂、钯、铱和其合金的金属。
18.权利要求16的光学检测系统,其中至少一个所述电极包含无机氧化物。
19.权利要求18的光学检测系统,其中所述无机氧化物选自氧化铟、氧化铟锡、氧化锡、氧化锑锡和其组合。
20.权利要求16的光学检测系统,进一步包括在所述发光层和至少一个所述电极之间的介电层。
21.权利要求16的光学检测系统,其中所述发光层包含磷光体颗粒。
22.前述任一权利要求的光学检测系统,其中所述测定器件容纳在试样架中。
23.权利要求22的光学检测系统,其中所述电致发光照明源也置于所述试样架中。
