Abstract
本发明描述了一种用于制备富含对映体的α-羟基羧基酸和酰胺的酶促方法,该方法包括,在一个步骤中通过羟腈的中间阶段,将羰基化合物转化成相应的酸/酰胺。本发明还提供了一种这样操作的反应体系以及一种优选用于此反应的全细胞催化剂。
Description
本发明涉及一种制备富含对映体的α-羟基羧酸及酰胺的方法。具体而言,本发明涉及一种方法,其中在第一步骤中,在醇腈酶的存在下由氰化物给体、醛和酮生成羟腈,而所述羟腈通过腈水解酶或腈水合酶在第二步骤中进一步转变成相应的酸。本发明还涉及以此方式操作的反应体系,及可以实现上述二步骤反应的新有机体。
在有机化学领域中,富含对映体的α-羟基羧酸及酰胺是重要的合成产品。这些化合物可成功作为用于配体合成的前体分子、作为手性外消旋体-拆分试剂、或作为制备生物活性物质的中间体产品。
这类化合物的传统合成方法通常采取羟腈反应,随后通过形成非对映的盐来进行酸水解和外消旋体的拆分(Bayer-Walter,Lehrbuchder Organischen Chemie,S.Hirzel Verlag Stuttgart,第22版555页)。在酰胺阶段可任选地停止水解反应,或对于酸可全过程执行。
迄今为止,光学活性的α-羟基羧酸的制备也可以通过以下方法实现:或者在手性催化剂如诸如醇腈酶之类的酶的存在下,以氰化物给体对醛的不对称加成反应的形式实现羟腈的形成,然后再进行“传统的”水解;或者备选地制备外消旋的羟腈,然后在腈水解酶的存在下进行对映选择性水解。在以醇腈酶为酶的存在下通过氢氰酸与醛的转化形成手性羟腈的以上第一种方法由例如Effenberger等进行过描述(F.Effenberger等,Angew.Chem.1987,99,491-492)。在此所给出的反应发生在由不与水混溶、优选为乙酸乙酯的有机溶剂相和水相组成的两相体系中。在此情况下实现了至少部分具有出色的收率和光学纯度的醛的转化。已有人就羟腈的光学纯度彻底研究了在酶(R)-醇腈酶和(S)-醇腈酶的存在下氰化物给体对醛的酶加成反应。或者,所述反应也可在纯粹的水体系中进行,并优选在低pH值下进行反应(U.Niedermeyer,M.R.Kula,Angew.Chem.1990,102,423)。在这类反应中也使用了固定化酶(DE-PS 1300111)。还尝试过在有机介质中进行酶促反应(P.Methe等,US-PS 5,122,462;J.Am.Chem.Soc.,1999,120,8587;US 5,177,242)。其它的转化方法还见述于:US-PS 5,122,462;Biotechno1.Prog.1999,15,98-104;J.Am.Chem.Soc.,1999,120,8587)。此外,还开发了固定(S)-醇腈酶的方法,在其操作方式下(S)-醇腈酶可与(R)-醇腈酶相媲美。这样,Effenberger等通过将(S)-醇腈酶附着到硝化纤维的载体上获得了其固定化(F.Effenberger等,Angew.Chem.1996,108,493-494)。Andruski等阐述了酶附着到多孔膜来实现固定化(US 5,177,242)。尽管有这些部分前途十分看好的所提议使用固定化酶的解决方案,但最近报道使用非固定化酶的研究再一次不断地出现在出版物上(如EP-A 0927766和US 5,714,356)。
虽然在羟腈的生物催化不对称合成过程中获得了出色的对映选择性,但随后所需的水解步骤有很大的问题,该水解步骤要使用强无机酸通过“传统的”酸水解来实现。这会产生大量的盐废物,造成经济和生态问题。此外,所需的水解条件也不利,原因在于要求的反应时间长达数小时且温度高。在此水解条件下,具有很大的外消旋化风险。
获得要求的光学活性α-羟基羧酸及酰胺的备选方法涉及-如上所述-外消旋羟腈的酶水解。
这种转化可通过腈水解酶来催化。腈水解酶为可以将有机氰基化合物转化为相应的羧酸的酶。它们属于E.C.3.5.5.1类,尤其可用于(+)-异丁苯丙酸合成的商业应用。现有技术已知状态的概况可参见Enzyme Catalysis in Organic Synthesis,VCH,1995,367页及以下几页。Yamamoto等(Appl.Environ.Microbiol.1991,57,3028-32)也描述了使用腈水解酶制备富含对映体的扁桃酸。
腈水合酶属于E.C.4.2.1.84类。它们由α,β-亚基(subunit)组成,可以以具有高达20多个不同基的多聚体多肽的形式存在(Bunch A.W.(1998),Nitriles,in:Biotechnology,8a卷,Biotransformations I,第6章,Eds.:Rehm H.J.,Reed G.,Wiley-VCH,277-324页;Kobayashi,M.;Shimizu,S.(1998)Metalloenzyme Nitrile Hydratase:structure,regulation,and application to biotechnology.Nature Biotechnology 16(8),733-736)。许多文献介绍了从腈到酰胺的酶转化(EP 0362829(Nitto);DE 4480132(Institute Gniigenetika);WO98/32872(Novus);US 5,200,331;DE 3922137;EP 0445646;Enzyme Catalysis in Organic Synthesis,VCH,1995,365页及以下几页)。
然而,这些备选的方法也存在许多缺点。对映选择性经常不能>99%ee,而这在制药要求中尤其是前提。此外,由于存在腈水解酶和腈水合酶对氰化物给体的存在敏感的风险,所以开始时羟腈必须很纯。
所有以前方法中的一个普遍的缺点是所述工艺的两阶段特征,这导致了时空收率和总体工艺效率明显降低。由于必须考虑到酶促羟腈合成和酶促腈皂化的反应条件互不相容,所以必须采用包括两个条件重整阶段的两阶段工艺。
本发明的目的是提供制备富含对映体的α-羟基羧酸/酰胺的另一种方法的说明书。从经济和生态观点来看,本方法应该具有技术上的优势。尤其是在使用的原料成本、可靠性及效率(如:时空收率)上应优于现有技术的方法,并且应能避免现有技术的上述缺点。尤其是应该可以避免源自以前所有方法的两阶段特性。
这些目的以权利要求中指明的方式来实现。
事实上,在制备富含对映体的α-羟基羧酸或富含对映体的α-羟基羧酸酰胺的方法中,起点是氰化物给体、醛或酮,并且在醇腈酶及腈水解酶或腈水合酶的存在下后者以极其惊人的、并且根据本发明是特别有利的方式进行反应,因此,可得到所述目的的解决方案。使用本发明的体系可以得到收率非常好、对映体的富集度特别高的富含对映体的α-羟基羧酸/酰胺。产生本发明时本领域的技术人员绝不可能知道所述酶的多级连用可以以此方式有效应用于现有的反应介质。就此而言,可认为尤其令人惊讶,特别是可用的氰化物数量相当多,却没有产生现有技术所预期的抑制效应,尤其是对腈水解酶或腈水合酶而言。
相应地,本发明的一个具体构想涉及这样的事实,即在制备富含对映体的α-羟基羧酸的方法中,氰化物给体在醇腈酶和腈水解酶的存在下与醛或酮一起转变。
类似地,富含对映体的α-羟基羧酸酰胺可以在醇腈酶和腈水合酶的存在下由氰化物给体、醛或酮得到。
本领域的技术人员所能立即想到的用于此目的的所有酶可用作醇腈酶。从Enzyme Catalysis in Organic Synthesis,Eds.:K.Drauz,H.Waldmann,VCH,1995,580页及下页中可找到选取的酶。在给定的反应条件下使用这些酶带来长的可使用寿命,而充分的转化具有优势。这些酶尤其应该是源自于选自以下组中有机体的那些醇腈酶:两色蜀黍(Sorghum bicolor)、巴西三叶胶和木薯(Mannihotesculenta)。为了制备(R)-羟腈,使用了源自命名的微生物体或杏核的醇腈酶。就此而言应当指出,为了制备(S)-α-羟基羧酸,优选应当利用(S)-系列的醇腈酶,反过来,也可以保证充分转化为最终的分子。
就腈水解酶而言,原则上可以类似地使用所有可获得的腈水解酶,只要在给定的环境条件下其可以保证充分的稳定性和转化率。从Enzyme Catalysis in Organic Synthesis,Eds.:K.Drauz,H.Waldmann,VCH,1995,365页及下页可找到选取的酶。这些酶尤其应该是源自于选自以下组中有机体的那些酶:红球菌菌株或粪产碱菌。通过与起可逆作用的醇腈酶相互作用,腈水解酶使腈官能向羧酸发生不可逆的转化。由此可保证形成的羟腈失去平衡,并导致根据使用过量的组分而使醛或酮或氰化物给体完全转化。为了保证成品中所要求的对映体纯度,腈水解酶应以对映选择性尽量高的方式进行反应。在此情况下,对使用的醇腈酶的对映选择性要求并不是很高。然而,如果使用的腈水解酶的对映选择性不够高,则应强调适当有所不同的醇腈酶的存在。
就腈水合酶而言,原则上可以类似地使用所有可获得的腈水合酶,只要在给定的环境条件下可以保证充分的稳定性和转化率。从Enzyme Catalysis in Organic Synthesis,Eds.:K.Drauz,H.Waldmann,VCH,1995,365页及下页可找到选取的酶。这些酶尤其应该是源自于选自以下组中红球菌菌株的有机体的那些酶,尤其是红球菌物种(R.spec.)、紫红红球菌(R.rhodochrous)和红平红球菌(R.erythropolis)。在本说明书中,可以参考EP 03001715.6专利以及这里所指定并优选使用的腈水合酶。通过与起可逆作用的醇腈酶相互作用,腈水合酶使腈官能向羧酸发生不可逆的转化。由此,可保证形成的羟腈失去平衡,并导致根据使用过量的组分而使醛或酮或氰化物给体完全转化。为了保证成品中所要求的对映体纯度,腈水合酶应以对映选择性尽量高的方式进行反应。在此情况下,对使用的醇腈酶的对映选择性要求并不是很高。然而,如果使用的腈水合酶的对映选择性不够高,则应强调适当有所不同的醇腈酶的存在。应当指出,作为进一步酶促水解或传统水解的结果,由该体系生成的富含对映体的α-羟基羧酸酰胺可以转化成相应的酸。如果就此而言在酰胺阶段产生的对映体纯度不够,则可进一步使用以对映选择方式工作的酰胺酶来改进。合适的酰胺酶可在Enzyme Catalysis inOrganic Synthesis,VCH,1995,367页及以下几页找到。

在给定情况下考虑的酶可以作为经均质纯化的化合物、并以自由体的形式在应用中使用。此外,所述酶也可以作为未受损的寄生生物的成分使用,或者与分解的且任意高度纯化的宿主生物的细胞团一起使用。所述酶还可以以固定化的形式使用(Bhavender P.Sharma,Lorraine F.Bailey和Ralph A.Messing,″ImmobilisierteBiomaterialien-Techniken und Anwendungen″,Angew.Chem.1982,94,836-852)。冷冻干燥法对固定化具有有利的影响(Dordick等.J.Am.Chem.Soc.194,116,5009-5010;Okahata等,Tetrahedron Lett.1997,38,1971-1974;Adlercreutz等,Biocatalysis 1992,6,291-305)。特别优选在诸如以下表面活性物质的存在下进行冷冻干燥:Aerosol OT或聚乙烯吡咯烷酮或聚乙二醇(PEG)或Brij 52(二甘醇单十六烷基醚)(Goto等,Biotechnol.Techniques 1997,11,375-378)。作为CLECs来使用也是可以想到的(St Clair等,Angew Chem Int Ed Engl 2000 Jan,39(2),380-383)。
原则上,本发明的具体方法可以在纯的水溶液中进行。然而,也可以在水溶液中加入任意比例的水溶性有机溶剂,以便例如就少量水溶性基质对反应进行优化。特别考虑以乙二醇、DME或甘油作为这类溶剂。但是以溶剂混合物作为水相的多相体系、尤其是两相体系也可进一步用于本发明方法中。这里,某些不溶于水的溶剂经证实是相当的(DE 10233107)。其中就此所陈述的相应地适用于此。
原则上,本领域的技术人员可自由选择反应期间的主要温度。这种人员优先接受配方(receipt)的指导,并在尽可能短的时间内以尽可能高的收率得到纯度尽可能高的产品。此外,所使用的酶在使用的温度下应具有充分的稳定性,进行反应时的对映选择性应尽可能高。对于衍生自嗜热生物的酶的使用,80-100C的温度可以明确代表反应过程中温度范围的上限。至于水体系中的低限,-15℃的温度显然是合理的。所述温度范围优选调节在10℃-60℃,更优选为20℃-40℃。
反应期间的pH值可由本领域的技术人员根据酶的稳定性及转化率来进行确定,并针对本发明的方法进行适当调整。一般来说,针对酶的pH优选范围可以选择为3-11。优选可得到3.0-10.0,特别是6.0-9.0的pH范围。
在进一步的配置中,本发明涉及酶促反应体系,该体系包含醇腈酶、腈水解酶或腈水合酶、水、氰化物给体及醛或酮。任选地,另外可以存在有机溶剂,如以上所详细描述的。
原则上就该反应体系而言,原先已陈述的有关本发明方法的优点及优选实施方案同样适用。
所述反应体系可以有利地应用于,例如,既可间歇操作又可连续操作的搅拌罐、多级搅拌罐或膜反应器中。
在本发明范围内,术语“膜反应器”应理解为任何一种其中催化剂封装在反应器内,而低分子量物质可以供应给或可以离开所述反应器的反应容器。就此而言,所述膜可以直接集成在反应室中,或可以以独立的过滤模件结合在外面,反应溶液连续地或间歇地流经过滤模件,而滞留物则回流到反应器中。尤其合适的实施方案描述在WO 98/22415和Wandrey等在Jahrbuch 1998,Verfahrenstechnikund Chemieingenieurwesen,VDI 151页及以下几页;Wandrey等在Applied Homogeneous Catalysis with Organometallic Compounds,2卷,VCH 1996,832页及以下几页;Kragl等,Angew.Chem.1996,6,684及下页。
除了间歇和半连续的操作模式外,该装置可以实施连续操作模式,如所希望的,可以按交叉流过滤模式(图1)或以死端过滤(图2)进行。两种方法的变化形式原则上在现有技术中有描述(EngineeringProcesses for Bioseparations,Ed.:L.R.Weatherley,Heinemann,1994,135-165;Wandrey等,Tetrahedron Asymmetry 1999,10,923-928)。

这种有机体的优点是同时表达了两种酶体系,而只需为反应培养一种rec有机体。为了使酶的表达与其转化率相匹配,妥善编码的核酸片段可以以不同的复制份数容纳在不同的质粒上,和/或可以对不同强表达的基因使用不同的强启动子。使用以此方式匹配的酶体系,有利的是不会引起中间体化合物的积聚,因为该中间化合物在合适的情况下可起阻碍反应的作用,从而使所考虑的反应可以以最佳的总速率进行。然而,本领域的技术人员对此已充分了解(PCT/EP 00/08473;Gellissen等,.Appl.Microbiol.Biotechnol.1996,46,46-54)。就微生物而言,原则上可以使用本领域的技术人员能考虑到的可应用于此目的的所有微生物,举例来说,例如:诸如多形汉逊酵母、毕赤酵母、酿酒酵母菌的酵母,诸如大肠杆菌、枯草杆菌的原核生物,或诸如哺乳动物细胞、昆虫细胞的真核细胞。应优选大肠杆菌(E.coli)菌株用于此目的。更详细地应优选:E.coli XL1 Blue、NM 522、JM101、JM109、JM105、RR1、DH5α、TOP10-或HB101。就所述生物而言,尤其优选使用DE 10155928中所命名的微生物。
就醛或酮而言,可以使用含有脂肪族或芳香族/杂芳香族残基的那些醛或酮。只要证明这些残基对实际转化是惰性的,它们就可以任意地支化和/或取代。在所述反应中有利地为使用以下通式(I)的化合物:

就氰化物给体而言,可以考虑本领域的技术人员在给定条件下能得到的所有化合物。尤其应使用可以尽可能便宜地获得的那些,然而应强调这些化合物在本发明反应中的最佳转化率的重要性。根据定义,氰化物给体为在给定反应条件下能释放出CN-离子的化合物。具体而言,这些氰化物为选自以下组中的那些:氢氰酸及诸如碱金属氰化物、三甲基甲硅烷氰化物的金属氰化物。
一般地,在本发明的反应中其过程是这样的:诸如以下(野生型,通过重组手段制备)的生物质、或未损的寄生生物(例如,全细胞催化剂)中的酶与醛或酮一起被注入水性反应基体中,然后加入氰化物给体,举例来说如碱金属氰化物(氰化钠)。在合适的反应条件下,通过中间体直接形成相应的羟腈,并由此形成富含对映体的α-羟基羧酸或酰胺。这些产物可以根据本领域技术人员熟悉的方法从反应混合物中分离开来。该过程优选的实施方法是这样的:通过过滤去除相对高分子量的组分,而酸或酰胺则通过结晶立即直接从混合物中分离,或者,对亲脂性的酸或酰胺而言,应在分离之前插入有机介质中的萃取步骤。也可以通过离子交换色谱法对酸进行重新调理。
这样,例如苯甲醛就可以和氰化钠一起转变成相应的扁桃酸,其高的收率为>80%、优选>85%、进一步更优选>90%、91%、92%、93%、94%,再进一步优选>95%、96%、97%,对映体富集度为>90%、91%、92%、93%、94%,进一步优选>95%、96%、97%,并且极优选为>98%、99%。
从制备本发明的全细胞催化剂的角度来看,本领域的技术人员可以使用以前描述过的现有技术的方法。具体地,腈水解酶或腈水合酶、同时还有醇腈酶被包含在所述全细胞催化剂中。相关的基因序列可从公共资源的基因数据库中搜集到,例如,可从NCBI基因数据库(Internet:
http://www.ncbi.nlm.nih.gov/Genbank/GenbankOverview.html)查到。就此而言特别优选的是酶,尤其是对氰化物具有高耐受性的腈水解酶或腈水合酶。就此而言,应优选这样的过程:相应的序列应与相应要求的基因序列如启动子等一起连接成一个质粒或连接到多个质粒上。此后,所述质粒转变成选定的有机体,后者被复制,然后活性克隆-未损的或压碎的生物质形式的-被插入到所述反应中。在产生本发明时,完全不清楚以这样的方式可以实现上述转化,并取得如此好的结果。
被称为(C1-C8)烷基的是甲基、乙基、正丙基、异丙基、正丁基、异丁基、仲丁基、叔丁基、戊基、己基、庚基或辛基,及其所有键合异构体。这些可以被以下基团单取代或多取代:(C1-C8)烷氧基、(C1-C8)卤烷基、OH、卤素、NH2、NO2、SH、S-(C1-C8)烷基。
术语“(C2-C8)烯基”应理解为如以上所表示的具有至少一个双键的、除了甲基以外的(C1-C8)烷基残基。
术语“(C2-C8)炔基”应理解为如以上所表示的具有至少一个三键的、除了甲基以外的(C1-C8)烷基残基。
术语“(C3-C8)环烷基”应理解为环丙基、环丁基、环戊基、环己基或环庚基残基等。它们可以由一个或多个卤素和/或含N、O、P、S原子的残基所取代,和/或可以具有在环中包含N、O、P、S原子的残基,举例来说,如:1-、2-、3-、4-哌啶基,1-、2-、3-吡咯烷基,2-、3-四氢呋喃基,2-、3-、4-吗啉基。后者可以被以下基团单取代或多取代:(C1-C8)烷氧基、(C1-C8)卤烷基、OH、卤素、NH2、NO2、SH、S-(C1-C8)烷基、(C1-C8)烷基。
术语“(C6-C18)芳基残基”应理解为含6-18个碳原子的芳族残基。这些包括尤其是诸如苯基、萘基、蒽基、菲基、联苯残基的化合物。后者可以被以下基团单取代或多取代:(C1-C8)烷氧基、(C1-C8)卤烷基、OH、卤素、NH2、NO2、SH、S-(C1-C8)烷基、(C1-C8)烷基。
(C7-C19)芳烷基残基是通过(C1-C8)烷基残基与所述分子键合的(C6-C18)芳基残基。
(C1-C8)烷氧基是通过氧原子与所考虑的分子键合的(C1-C8)烷基残基。
(C1-C8)卤烷基是被一个或多个卤原子取代的(C1-C8)烷基残基。
(C3-C18)杂芳基残基在本发明范围内表示5-、6-或7-元的含有3-18个碳原子的芳环体系,该体系具有杂原子,例如在杂环中的氮、氧或硫。考虑到的这类杂芳族化合物有,具体而言诸如以下的残基:1-、2-、3-呋喃基,1-、2-、3-吡咯基,1-、2-、3-噻吩基,2-、3-、4-吡啶基,2-、3-、4-、5-、6-、7-吲哚基,3-、4-、5-吡唑基,2-、4-、5-咪唑基,吖啶基,喹啉基,菲啶基,2-、4-、5-、6-嘧啶基。后者可以被以下基团单取代或多取代:(C1-C8)烷氧基、(C1-C8)卤烷基、OH、卤素、NH2、NO2、SH、S-(C1-C8)烷基、(C1-C8)烷基。
术语“(C4-C19)杂芳烷基”应理解为对应于(C7-C19)芳烷基残基的杂芳香族体系。
所考虑的卤素为氟、氯、溴和碘。
术语“富含对映体的”所表示的实际情况是当混合物中存在一个光学对映体时,其另一个的比例量为>50%。
当存在一个立构中心时,所提出的结构涉及两种可能的对映体,当分子中存在多于一个的立构中心时,所述结构将涉及所有可能的非对映体,而就非对映体而言,其涉及包含以下所讨论化合物中的两种可能的对映体。

Claims (11)
1.一种在醇腈酶和腈水解酶或腈水合酶的存在下从氰化物给体、醛或酮制备富含对映体的α-羟基羧酸或富含对映体的α-羟基羧酸酰胺的方法。
2.一种在醇腈酶和腈水解酶的存在下从氰化物给体、醛或酮制备富含对映体的α-羟基羧酸的方法。
3.一种在醇腈酶和腈水合酶的存在下从氰化物给体、醛或酮制备富含对映体的α-羟基羧酸酰胺的方法。
4.权利要求1-3之一的方法,其特征在于使用了选自以下组中有机体的或植物成分的醇腈酶:两色蜀黍、巴西三叶胶、木薯和杏核。
5.权利要求1或2的方法,其特征在于使用了选自以下组中有机体的腈水解酶:红球菌菌株或粪产碱菌。
6.权利要求1或3的方法,其特征在于使用了选自以下组中有机体的腈水合酶:红球菌物种、紫红红球菌和红平红球菌。
7.前述权利要求之一的方法,其特征在于所述反应在pH值为6.0-9.0的含水介质中进行。
8.前述权利要求之一的方法,其特征在于所述反应在20-40C的温度范围内进行。
9.一种酶促反应体系,该体系含有醇腈酶、腈水解酶或腈水合酶、水、氰化物给体及醛或酮。
10.一种全细胞催化剂,该催化剂具有用于醇腈酶及腈水解酶或腈水合酶的克隆基因。
11.权利要求9的全细胞催化剂,其特征在于当存在腈水合酶时,所述全细胞催化剂同样具有用于酰胺酶的克隆基因。
