技术领域

-在试样中设置标记;
-用第一辐射束照射试样,作为照射的结果在试样中形成激励区,激励区中的标记被激励;
-检测标记响应激励而发出的荧光辐射;以及
-借助第二辐射束阻止对部分激励区中的荧光辐射的检测。
本发明还涉及实施所述方法的装置。
背景技术
从美国专利文本No.6,259,104已知这种方法。
在生物学和组织学中,需要确定生物试样中某些分子,例如蛋白质,抗体和核苷酸的位置,以便清楚了解在例如生物试样中某些过程是在何处以及如何发生的。
荧光显微镜法(FM)是可以做到这一点的在生物学和组织学中通常采用的方法。为此,将所谓的荧光标记(以下简称为”标记”)附着到重要的分子上。这些标记的特点在于它们非常特别地将它们自已附着到例如某些类型的抗体上,而不附着到其它类型的抗体或其它分子上。而且,这些标记的特点在于它们响应激励(例如光子激励)而发出荧光辐射。
这样,通过以下方法确定试样中例如某种类型抗体的位置的可能性提高熬了:将标记附着到这种类型的抗体上,随后利用标记发出的荧光辐射来确定标记的位置。
应当指出,用于FM的标记可以分类为有机标记和无机标记。
有机标记通常包括芳族环。实例有FITC(异硫氰酸荧光素),TRITC(四甲基异硫氰酸罗丹明)和DAPI(bisbenzimide)。许多类型的有机标记都有市售,每一种有其自身的附着特性。这种有机标记如今由例如Molecular Probes,Inc.,Eugene,Oregan,USA销售。
这些有机标记的一个典型特性就是它们的寿命有限:在光化学反应的影响下,这种标记的寿命限于例如105个发射的光子。这对于标记可被激励的时间来说就很重要。这一点以下再作说明。此外,这些标记中的荧光作用会受环境的影响,例如它们所在的试样中的酸度(pH)、温度、溶剂的存在等等。
无机标记含有一种无机材料例如CdSe的纳米晶体,其直径从几个纳米到数十纳米,这种纳米晶体能显示荧光。将这种纳米晶体结合到有机反应基上,有机反应基将标记附着到待加标记的分子(例如蛋白质)上并且确定了标记的具体附着特性。这类无机标记如今由例如Biocrystal Ltd.,Westerville,Ohio,USA销售。
无机标记的特点是比有机标记的寿命长得多且对环境影响的敏感性较低。
在传统FM的情况下,标记利用光束(例如聚焦的激光束)来照射其中存在荧光标记的试样。在被照射区,即所谓激励区中的标记因此被激励,并对此激励作出响应而发出荧光辐射通量。通过检测是否对激励作出响应而发出荧光辐射,就可以确定标记是否位于激励区。这种方法中的空间分辨率由光束焦点的大小确定。一般来说,所述焦点不会小于所采用的光的波长。所采用的波长不能自由选择,因为只有在给定波段的波长内的光才会激励某种类型的标记,结果,传统FM的空间分辨率限于大约500nm。
在FM的用户中,希望继续降低FM的空间分辨率,最好降到分子等级,即几个纳米。
在所述专利文本中,说明了一种方法,相对于传统FM而言可以提高分辨率。
利用第一辐射束短时间地照射试样,在试样中形成激励区,然后第二辐射束阻止对荧光辐射的检测。第一辐射束是激光束,具有适合于激励标记的颜色。第二辐射束由一种或多种激光束组成,具有将标记去激励(“猝熄”)的某种不同(更红些)的颜色。第一辐射束的强度分布应具有中心最大值,而第二辐射束的强度分布应显示中心最小值。去激励最好在激励后数十微微秒内发生。
通过将激励区的一部分中的标记去激励,就可以阻止这些标记的荧光辐射通量被检测,因而可以实现仅仅检测一部分激励区中的标记,于是得到较好的空间分辨率。
发明内容
本发明旨在提供一种方法,通过这种方法可以利用上述美国专利文本所述的方法来获得更好的空间分辨率。
为此,按照本发明的方法的特征在于:
-第二辐射束是在试样上扫描的聚焦粒子束;
-所述粒子束焦点的截面积小于激励区的截面积;
-粒子束中粒子的能量足以由于损坏的结果而降低被粒子冲击的标记所发出的荧光辐射通量;
-在通量降低了至少预定阈值的瞬间记录下聚焦粒子束相对于试样的位置。
对激励作出响应,从试样中发射出荧光辐射通量。所述通量是由位于激励区中的一个或多个标记产生的。如果因激励而产生的通量降低了至少预定的阈值,这必定是因为激励区中的标记被破坏。
本发明基于以下概念,即,通过有意地造成对标记的破坏,通过测定辐射通量因破坏而降低的瞬间粒子束位于何处,来确定标记的位置。可以通过利用聚焦粒子束射击标记来实现这种有意的破坏。
聚焦粒子束的产生是已知技术,并且在例如聚焦离子束(FIB)装置、扫描电子显微镜(SEM)装置和环境扫描电子显微镜(ESEM)装置中都已采用。这些装置可以将粒子束聚焦到试样上,粒子束焦点的截面积例如为1nm,并可使所述射束在试样上扫描。在这些类型的装置中,粒子束中粒子能量一般可在从小于200eV到大于20keV的范围内调节。所以,利用这些装置就有可能产生其能量足以破坏标记的粒子。
应当指出,所需的破坏不仅会通过标记和聚焦粒子束中粒子的相互作用而发生,也会因标记与二次粒子(例如x射线光子和二次电子,系由相互作用区中的聚焦束所引发的)的相互作用而发生。结果,甚至在距聚焦束有一定距离处标记也可能被破坏,导致位置的测定不甚精确。电子显微镜领域的专业技术人员已知,通过适当选择粒子的能量可将相互作用区的尺寸保持在足够小的范围内。
在按照本发明的方法的实施例中,激励是通过光子束产生的。
通过用光子照射试样,标记被激励,随后标记对激励作出响应发射光子。这种照射可以用激光束,也可以用具有宽带频谱的光束(“白光”)进行。
应当指出,使用白光对于同时检测数种类型的标记特别有吸引力,因为各种标记不仅它们所附着的分子类型(例如抗体)不同,而且它们可被什么颜色的光激励,以及它们所发射的荧光辐射的颜色也都不同。如果试样中有数种类型的标记,本发明的方法不仅能用来测定标记的位置,也可测定标记的类型。这是因为,当以下述方式实现用于检测荧光辐射的检测器时,即,对于每种颜色(或换句话说,每种类型的标记),检测器都能测定所述颜色(所述类型的标记)的荧光辐射通量的下降时,就有可能不仅测定标记的位置,也能测定其类型。
在本发明方法的另一实施例中,聚焦粒子束是聚焦电子束。
电子特别在使有机材料发生化学改变方面非常有效。在例如有机标记发生这种化学改变的情况下,标记就会被破坏而不再发射荧光辐射。
在本发明方法的又一实施例中,聚焦粒子束是离子束。
如专业技术人员已知,能量为数keV的离子具有去除它们所碰撞的材料表面的特性(溅射)。这种溅射发生在试样上,也发生在位于试样中的标记上。所以在使标记产生所需的破坏方面离子也非常有效。使用离子的一个附带优点是:原先位于试样深处的标记会缓慢出现在试样表面,随后标记被破坏,其位置即可测定。这样,就有可能对试样中的标记进行三维位置测定。
在本发明方法的另一实施例中,在利用聚焦粒子束照射时引入气体,以便能够进行蚀刻。
将气体与离子束或电子束结合使用以便从试样中去除材料是一种已知技术。此技术应用于例如FIB装置中。在利用聚焦粒子束照射试样时使用此技术,在粒子束扫描试样时试样表面被蚀刻掉。于是,原先位于试样深处的标记会缓慢出现在试样表面,随后标记被破坏,其位置即可测定。这样,就有可能对试样中的标记进行三维位置测定。

具体实施方式
图1示出执行本发明方法的装置的示意图。粒子-光柱(particle-optical column)1,例如离子柱,产生带电荷粒子的聚焦粒子束3。将粒子束3聚焦到位于试样托架5上的试样4上。借助偏转单元2使聚焦粒子束3扫描试样4。
从试样发射的带电荷粒子(例如二次电子)由粒子检测器8(例如Everhardt-Thornley检测器)检测。
聚焦粒子束3所需的真空这样获得,即,借助密封装置6将真空室7密封,然后用真空泵(未示出)将真空室抽空。
应当指出,如果是生物试样,保持很小的水蒸汽剩余压力是有利的,可使试样不会过于干燥。此技术是一种已知技术,应用于例如ESEM(环境扫描电子显微镜)中。
气体注入器9可以将蚀刻气体引入真空室7,在聚焦粒子束3的作用下,可以从试样上去除材料。
试样托架5由透明材料例如玻璃构成。为了防止试样4因聚焦粒子束3的照射而带电,例如可以在玻璃上涂敷导电材料的透明涂层。
激光器10产生光束11,光束11经过半透明镜面12和透镜13,并经过透明试样托架5聚焦到试样4上。聚焦粒子束3和光束11射到试样上基本相同的位置14。从试样4中标记发射的荧光辐射通过试样托架5、透镜13和半透明镜面12由检测器16检测。为了防止因激光器10而产生的散射光和反射光也被检测,在半透明镜面12和光检测器16之间设置过滤器15,所述过滤器不允许具有激光器颜色的光通过,但允许荧光辐射通过。为了尽可能多的检测到试样4发射的光,利用镜面17将沿着离开检测器16的方向发射的光反射到检测器16上。
控制单元20发送控制信号到偏转线圈2,借助于偏转线圈2使粒子束扫描试样4。这样粒子束的偏转通常正比于提供到偏转单元上的控制信号。
在某一瞬间从粒子检测器发出的信号,是从粒子束3在所述瞬间正射到试样上的那个位置发出的。现在使光栅中的粒子束3扫描试样4,同时在监控器21上建立图像22,其亮度正比于粒子检测器8的信号,试样4中被扫描区域的图像22就在监控器21上可见。图像的建立也由控制单元20完成。
此外,控制单元20控制激光器10,从而接通或断开光束。
控制单元20还接收来自光检测器16的信号,并根据所述信号确定荧光辐射通量是否在显著下降。此处”显著”应理解为是指至少下降了预定阈值。在发生这种显著下降的瞬间,粒子束3的位置已知,因为这个位置是经由扫描线圈2的驱动而受控的。控制单元20此时可以使一种指示(例如亮点或不同颜色的点)出现在屏幕21的相应位置上。
如果标记存在于射束扫描的区域中,那么在扫描时这些标记就会受到粒子束的冲击。结果,标记就会被破坏。
图2A示意地示出从试样发射的荧光辐射通量。在扫描开始时,即在T=T0的瞬间,许多标记都发荧光,此处为6个标记。扫描时,这些标记分别被破坏。由于这种破坏的结果,通量下降。图2A中示出在40-i瞬间的这种情况,其中i是第i个被破坏的标记。通过在发生这种通量变化的瞬间在图像22中(图像22显示从粒子检测器8发出的信号)显示亮点或不同颜色的点,很容易看出标记位于试样上的什么地方。
应当指出,利用目前的计算机技术,也可以在图像22中用另一种形式的符号,例如十字形、矩形等,来指示荧光标记的位置。
图2B示意地示出具有粒子检测器8和光检测器16所产生的信息的图像。
粒子检测器8(Everhardt-Thornley检测器)能够产生图像22,图中产生不同数量二次粒子的区域用不同的亮度来成像(31,32)。在此图像22中,还示出在粒子束的哪个位置30-i测到通量的显著降低,或者换句话说,标记位于何处。这样,就很容易看见标记位于试样上的什么地方。
应当指出,这样的实施例是可能的,按照所些实施例,检测器16能够区别并分别处理不同标记发出的以不同荧光颜色为特点的光。
这样,就有可能同时检测不同类型的标记,而且例如在图像22中使用不同颜色的亮点或符号,以便区分不同类型的标记。
还应当指出,如果标记停止发射光子的原因不是由于被带电粒子束3所破坏,那么,这会导致在不正确的位置显示所述标记。这是因为光检测器16会检测到荧光辐射在下降,且控制单元20会在图像22上在所述瞬间粒子束3所对应的位置上显示所述标记,即使射束位置与所述位置上标记的存在毫无关系。
所以,极其不希望出现标记不是由于被粒子束3轰击而停止发光的情况。
一些标记,特别是一些有机标记的寿命,例如被限制在发射105个光子。所以需要在标记发射105个光子的时间范围内扫描整个激励区。
应当指出,重要的是将激励区和扫描区互相对中。当这两个区域互相对中时,它们一般不会很快就偏心。所以这种对中可以作为定期调节进行。可以想出各种方式来确定这种对中。
一种可能的调节方法包括借助粒子束和粒子检测器将可滑动膜片上的孔成像,孔的直径大致等于激光束焦点的直径。使膜片滑动,直到它与粒子束扫描的区域对准中心。随后通过将激光束导向膜片,并使激光器有最大量的光通过小孔,这两个区域就互相对准中心了。检测通过的光量可以借助例如真空室中的光检测器(未示出)进行。
应当指出,激光束11对试样4的照射可以是连续照射,但也可由一系列脉冲组成。
还应当指出,虽然在实现本方法的图示实施例中,粒子束3和光束11从相反方向射到试样4上,但也可以这样实现所述装置,即,射束(3,11)从实际上同一方向射到试样4上。对于比较厚的试样,这样做特别有利,因为激励激光束不能穿透整个试样。
Claims (7)
1.一种确定在待研究的试样中可以由辐射激励的标记的位置的方法,所述方法包括:
-在所述试样中设置标记;
-用第一辐射束照射所述试样,作为照射的结果在所述试样中形成激励区,在所述激励区中的标记被激励;
-检测响应所述激励作从标记发出的荧光辐射;以及
-借助第二辐射束阻止检测在部分所述激励区中的荧光辐射,
其特征在于:
-所述第二辐射束是在所述试样上扫描的聚焦粒子束;
-所述粒子束的焦点的截面积小于所述激励区的截面积;
-所述粒子束中粒子的能量足以由于损坏的结果而降低被所述粒子冲击的标记所发出的荧光辐射通量;
-在所述通量降低了至少预定阈值的瞬间记录下所述聚焦粒子束相对于所述试样的位置。
2.如权利要求1所述的方法,其中所述激励区的截面积是所述粒子束的截面积的至少10倍。
3.如上述权利要求中的一项所述的方法,其中所述激励是利用光子束发生的。
4.如上述权利要求中的一项所述的方法,其中所述聚焦粒子束是聚焦电子束。
5.如权利要求1-3所述的方法,其中所述聚焦粒子束是聚焦离子束。
6.如上述权利要求中的一项所述的方法,其中利用所述聚焦粒子束照射时引入气体,以便进行蚀刻。
7.一种配备成执行根据上述权利要求中的一项的方法的装置,所处装置包括:
-用于产生激励标记的第一辐射束的装置;
-用于检测从受激励的标记发射的荧光辐射通量的检测装置;
-用于产生第二辐射束的装置;
其特征在于:
-用于产生所述第二辐射束的所述装置产生聚焦粒子束,所述粒子束扫描所述试样,以及
-具有记录装置,以便根据先前确定的所述检测的通量的下降来确定所述聚焦粒子束相对于所述试样的位置。
