Abstract
在合成的、热加工的小吃食品中,在食品的配方中添加一种可选的氨基酸来抑制热加工过程中丙稀酰胺的形成。所述氨基酸可选自以下的组:半胱氨酸、赖氨酸、甘氨酸、组氨酸、丙氨酸、蛋氨酸、谷氨酸、天冬氨酸、脯氨酸、苯丙氨酸、缬氨酸和精氨酸,它们可以是以商业上可获得的形式或是以食品添加成分中的游离形式。将氨基酸添加到合成的食品中可以是在混合阶段,或是通过将未加工的食品浸泡在含有一定浓度氨基酸的溶液中来进行。

发明背景
1.
技术领域
本发明涉及一种在热加工的食品中降低丙烯酰胺量的方法。本发明能够在制备食品过程中使丙烯酰胺的量显著降低。这种方法依赖于在小吃食品的制作中使用一种或多种经选择的氨基酸。
2.
现有技术的描述
化学制品丙烯酰胺一直以聚合物的形式应用在工业中,它可用于水处理、浓缩油回收、造纸、絮凝剂、增稠剂、矿石处理和免烫纤维制品。丙烯酰胺作为一种白色结晶固体是无味的,并具有高水溶性(在30℃时为2155g/L)。丙烯酰胺的同系物包括2-丙烯酰胺、乙烯基羰基酰胺、丙烯酸酰胺、乙烯酰胺、和丙烯酸酰胺。丙烯酰胺的分子量为71.08,熔点为84.5℃,以及在25mmHg下沸点为125℃。
最近,许多食品被检测出对丙烯酰胺单体呈阳性。特别是在被高温加热或加工的碳水化合物食品中发现有丙烯酰胺。经检测对丙烯酰胺呈阳性的食品的例子包括咖啡、谷类食品、曲奇、薯片、饼干、油炸马铃薯条、面包和面包卷以及沾滚上面包屑的炸肉。通常,在加热处理的蛋白质丰富的食品中,丙烯酰胺的含量相对低,而在碳水化合物丰富的食品中,丙烯酰胺的含量相对高,相对而言,在未加热和煮制食品中其含量小到无法测出。据报道,在各种以类似方法处理的食品中,所发现的丙烯酰胺含量为:对于马铃薯片,丙烯酰胺含量的范围为330-2,300(μg/kg);对于炸土豆片,丙烯酰胺含量的范围为300-1100(μg/kg);对于玉米片,丙烯酰胺含量的范围为120-180(μg/kg),在早餐谷物食品中,丙烯酰胺的量可以由小到无法测出到1400(μg/kg)。
目前相信丙烯酰胺的形成是由于氨基酸和还原糖的存在。例如,据信多数油炸食品中发现的丙烯酰胺是由游离的天冬酰胺(asparagine)与游离的还原糖反应而产生的,天冬酰胺是一种通常存在于生蔬菜中的氨基酸。在生的马铃薯中,天冬酰胺的量占游离氨基酸总量的大约40%,在高蛋白黑麦中大约为18%,在小麦中大约为14%。
丙烯酰胺还可能由天冬酰胺以外的氨基酸形成,但是还不确定。例如,据报道,通过将谷氨酸,蛋氨酸,半胱氨酸以及天冬氨酸作为前体进行试验发现有一些丙烯酰胺。这些发现很难被证实,然而,这可能归因于在原始氨基酸中天冬酰胺不纯。尽管如此,天冬酰胺已被认定为是氨基酸前体,在丙烯酰胺的形成中起主要作用。
由于丙烯酰胺在食品中存在是最近发现的现象,所以其形成机理还尚未确定。然而,据信丙烯酰胺形成最有可能是由于麦拉德(Maillard)反应。在食品化学中,麦拉德反应一直以来被认为是食品处理过程中的最重要的反应之一,它可以影响到食品的风味、色泽以及营养价值。麦拉德反应需要热量、湿度、还原糖以及氨基酸。
麦拉德反应是一个涉及许多中间物质的复杂反应,但是通常来说包括三个步骤。第一个步骤包括将一种游离氨基(来源于游离氨基酸和/或蛋白质)和一种还原糖(如葡萄糖)化合,然后形成阿马道莱(Amadori)或汉斯(Heyns)重排产物。第二个步骤包括通过不同的备选方式降低阿马道莱或汉斯重排产物的量,所述方式包括降解、裂变、或是施特雷克尔(Strecker)降解。一系列复杂的反应包括脱水、消去、环化、裂变、以及分解,最后聚集成风味中间产物和风味化合物。第三个步骤的特征在于形成褐色的含氮聚合物和共聚物。使用麦拉德反应作为形成丙烯酰胺的可能路径,图1表示的是由天冬酰胺和葡萄糖开始而形成丙烯酰胺的简化的可能路径。
丙烯酰氨对于人类是否有害还不确定,但是它存在于食品中,尤其是以高的含量存在是不希望的。以前的记载中,在经加热或热加工的食品中发现有相对较高浓度的丙烯酰氨。在上述食品中可以通过以下方式降低丙烯酰胺的量:降低或除去可形成丙烯酰胺的前体物;在制备食品过程中抑制丙烯酰胺的形成;破坏或反应掉食品中形成的丙烯酰胺单体;或在消费之前排除食品中的丙烯酰胺。可理解的,为了完成上述任一种方式,每种食品都存在特定的问题。例如,如果不使用物理方法破坏细胞结构,以通过烹制赋予食品独特的性质,那么粘在一起的切片或烹制食品就可能不太容易与其它的添加物混合。对于特定食品的其他生产要求也同样与降低丙烯酰胺的量形成矛盾或是十分困难。
举例,图2表示的是现有技术中公知的将生马铃薯制备为炸薯片的方法,含有大约重量百分比80%或更多水的生马铃薯,首先经过去皮步骤21,生马铃薯皮被去除以后,被送到切片步骤22。切片步骤22中每一个薯片的厚度取决于最终产品所需的厚度。现有技术中一个实例是将马铃薯切成大约0.053英寸厚。上述切片被运送到清洗步骤23,其中每一个切片的表层淀粉被水洗去。洗后的马铃薯切片又被送到烹制步骤24。烹制步骤24一般包括在一个连续的油炸器内炸这些切片,例如在177℃下炸大约2.5分钟。所述烹制步骤通常可使薯片的水分含量降至低于大约重量百分比2%。例如,离开油炸器的典型的油炸后的薯片的水分含量大约为重量百分比1.4%。所述烹制后的薯片然后被送至调味步骤25,其中调味料被放到一个转鼓中。最后,调味后的薯片进行包装步骤26。所述包装步骤26通常包括将调味后薯片送到一个或多个称重器称重,然后将薯片导向一个或多个垂直式,填充,和密封机械以包装在一个软袋中。一旦包装完成,产品将进入销售并被消费者购买。
对如上所述的薯片制备过程中的一些步骤的微小调整可以显著改变最终产品的性质。例如,在清洗步骤23中,延长切片在水中的时间将导致切片内的化合物浸出,而这些化合物赋予最终产品马铃薯的风味、颜色和质感。在步骤24中,增加烹饪时间或增加加热温度会增加薯片中麦拉德褐变水平,同时降低水分含量。如果希望在油炸之前将其他成分混合入马铃薯切片,那就需要建立一些机制,以使加入的成分被吸收到切片内部,而不会破坏薯片的细胞结构或是浸出切片中有益的化合物。
作为加热食品的另一实例,对降低最终产品中的丙烯酰胺的量具有特别的要求的是,小吃也可以由面团制备。术语“合成小吃”意为一种小吃食品,其原料是其他成分,而不是原始的和未经处理的淀粉原料。例如,合成小吃可以是利用脱水马铃薯作为原料的合成薯片和利用了湿润粉糊(masa)面粉作为原料的玉米片。所述脱水马铃薯可以是马铃薯粉、马铃薯薄片、马铃薯粒、或其它任何以脱水形式存在的马铃薯。当上面任何一个术语在本申请中应用时,应该理解所有这些变化形式都被包括在内。
再参照图2,一种合成马铃薯片可以不需要去皮步骤21、切片步骤22、或是水洗步骤23。而是,合成马铃薯片可由,例如,马铃薯薄片开始,所述马铃薯薄片与水和其他辅料混合而形成的一个面团。这个面团其后被压片以及在进行烹制步骤之前被切断。烹制步骤包括炸或焙烤。然后所述切片送至调味步骤和包装步骤。马铃薯面团的混合通常使它容易加入其它成分。相反,对于一个生的食品来说,如马铃薯切片,加入这种成分要求找到一种机制,这种机制允许这些成分渗透进入产品的细胞结构中。然而,在混合步骤中的任一成分的添加必须要考虑到这些成分可能会对面团的压片特性,以及最终薯片的特性产生负面的影响。
需要开发一种或多种在加热或热加工的食品的最终产品中降低丙烯酰胺含量的方法。理想地,这样的方法应该实质上降低或去除最终产品中丙烯酰胺的量,而不会对最终产品的质量和特性产生负面影响。另外,这种方法应该是容易实施的,优选地,几乎不或完全不增加生产成本。
发明概述
在本发明的方法中,一种或多种经选择的氨基酸在烹制食品之前被添加到食品中,以降低丙烯酰胺的形成。氨基酸可以在磨粉、干燥混合、湿混合或其他混合过程中添加,使得氨基酸可以均匀存在于食品中。通过使生食品成分暴露在氨基酸中,例如浸泡,也可使氨基酸加入到生食品中。所述氨基酸可以以商业上可获得的化学制品的形式存在,或可以以食品方式存在,在所述食品中氨基酸以游离的形式存在。在本发明的两个实施例中,显示了添加半胱氨酸或赖氨酸来减少丙烯酰胺的形成。也显示了用经选择的其他氨基酸来减少丙烯酰胺的形成。
添加一种或多种经选择的氨基酸可有效地降低被加热或热加工的食品的最终产品中的丙烯酰胺的量,同时最低限度地影响最终产品的质量和特性。另外,这种降低丙烯酰胺的方法一般是容易实施的,并几乎不或完全不增加生产成本。

发明详述
氨基酸在丙烯酰氨形成中的作用
在热加工过程中形成丙烯酰胺需要一种碳源和一种氮源。假设碳源是由碳水化合物源提供,而氮源是由蛋白质源或氨基酸源提供。很多植物性食品成分例如大米、小麦、玉米、大麦、大豆、马铃薯和燕麦都含有天冬酰胺,且大多的碳水化合物都含有少量氨基酸成分。典型地,这种食品成分具有一个小的氨基酸组,它除了天冬酰胺以外还含有其他氨基酸。
“热加工过程”意为食品或食品成分在至少80℃的温度下被加热,其中的食品成分如食品成分的混合物。食品或食品成分的热加工过程优选地在大约100℃到205℃之间的温度下进行。食品成分可以在最终食品产品形成之前,在较高的温度下分别处理。一个热加工的食品成分的实施例是马铃薯片,这种薯片是由生的马铃薯形成,形成方法是将马铃薯暴露于170℃的高温下制得。(术语“马铃薯片”、“马铃薯粒”、和“马铃薯粉”在此可以互换,而且意指任何基于马铃薯的脱水产品)。其他热加工的食品成分的例了包括经加工的燕麦、部分煮制的和干燥的大米、烹制的大豆食品、玉米湿润粉糊、焙烤咖啡豆和焙烤可可豆。可选地,生的食品成分可以用来制备最终的食品,其中制备最终产品的过程包括加热步骤。由加热步骤获得最终食品产品的一个原料加工的例子是,由生的马铃薯片通过在大约100℃到大约205℃下的油炸步骤来生产马铃薯片,或在相似温度下油炸以生产炸土豆片。
但是,根据本发明,当还原糖存在时加热天冬酰胺会明显生成丙烯酰胺。当有还原糖例如葡萄糖存在时,加热其他氨基酸例如赖氨酸和丙氨酸不会导致丙烯酰胺的形成。但是,令人意外的是,将其他的氨基酸添加到天冬酰胺-糖的混合物中会增加或减少丙烯酰胺的生成量。
已经确定,在还原糖存在的情况下,加热天冬酰胺会迅速形成丙烯酰胺,通过使天冬酰胺失活,可以减少热加工的食品中的丙烯酰胺。“失活”是指从食品中除去天冬酰胺或使天冬酰胺在丙烯酰胺形成的过程中不反应,使天冬酰胺在丙烯酰胺形成的过程中不反应是通过将其转化或与其他的化学物质结合以阻碍由天冬酰胺形成丙烯酰胺。
I:半胱氨酸、赖氨酸、谷氨酸(Glutamine)和甘氨酸(glycine)在丙烯酰胺形成中的作用
由于天冬酰胺与葡萄糖反应生成丙烯酰胺,增加其它游离氨基酸的含量可能会影响天冬酰胺与葡萄糖的反应,从而减少丙烯酰胺的形成。实验时,准备PH7.0的磷酸钠缓冲液,溶解天冬酰胺(0.176%)和葡萄糖(0.4%)。其它四种氨基酸:甘氨酸(GLY)、赖氨酸(LYS)、谷氨酸(GLN)、半胱氨酸(CYS)以与葡萄糖相同的摩尔浓度添加其中。采用正交试验测定了所有的添加氨基酸的所有可能的组合的情况。溶液在120℃下加热40分钟,然后测定丙烯酰胺。下表1显示氨基酸含量和结果。

如上表所示,在无其他氨基酸的情况下,葡萄糖和天冬酰胺会生成1679ppb的丙烯酰胺。添加的氨基酸产生了三种类型的作用:
1)半胱氨酸几乎消除丙烯酰胺的生成。所有添加半胱氨酸的反应产生的丙烯酰胺少于大约25ppb(减少98%)。
2)赖氨酸和甘氨酸可以减少丙烯酰胺的生成,但效果不如半胱氨酸。所有添加赖氨酸和/或甘氨酸,但无谷氨酸和半胱氨酸的反应生成的丙烯酰胺小于大约220ppb(减少85%)。
3)令人惊奇地,谷氨酸使丙烯酰胺的生成量增加到5378ppb(增加20%)。谷氨酸和半胱氨酸一起不生成丙烯酰胺。在谷氨酸中添加甘氨酸和赖氨酸则减少丙烯酰胺的生成。
这些实验显示了半胱氨酸、赖氨酸、甘氨酸在减少丙烯酰胺方面的作用。然而,由谷氨酸的结果可以看出不是所有的氨基酸都可以减少丙烯酰酸的生成。半胱氨酸、赖氨酸、或甘氨酸与另一种单独的氨基酸混合可加速丙烯酰胺的生成(如谷氨酸),就像减少丙烯酰胺生成的程度一样。
II.半胱氨酸、赖氨酸、谷氨酸和蛋氨酸在不同浓度和温度下的作用
如上所述,当添加与葡萄糖相同摩尔浓度的半胱氨酸和赖氨酸时会减少丙烯酰胺的生成量。一个后续实验用来回答如下问题:
1)浓度降低的半胱氨酸、赖氨酸、谷氨酸和蛋氨酸对丙烯酰胺的生成会如何产生作用?
2)在加热温度为120℃和150℃时,添加的半胱氨酸和赖氨酸所产生的作用是否相同?
准备PH7.0的磷酸钠缓冲液溶解天冬酰胺(0.176%)和葡萄糖(0.4%)。添加两种浓度的氨基酸(半胱氨酸(CYS)、赖氨酸(LYS)、谷氨酸(GLN)或蛋氨酸(MET))。这两种浓度为0.2摩尔氨基酸/摩尔葡萄糖和1.0摩尔氨基酸/摩尔葡萄糖。在一半的试验中,2ml的溶液在120℃下加热40分钟;在另一半的试验中,2ml的溶液在150℃下加热15分钟。加热后用GC-MS测定丙烯酰胺,结果显示在表2中。以不添加氨基酸的天冬酰胺和葡萄糖的溶液作为对照组。

在使用半胱氨酸和赖氨酸的实验中,在120℃下40分钟后,对照组生成的丙烯酰胺为1332ppb,在150℃下15分钟后为3127ppb。半胱氨酸和赖氨酸在120℃和150℃下均可减少丙烯酰胺的生成,减少的程度大致与添加的半胱氨酸或赖氨酸的浓度成正比。
在使用谷氨酸和蛋氨酸的实验中,在120℃下40分钟后,对照组生成的丙烯酰胺为1953ppb,在150℃下15分钟后为3866ppb。谷氨酸在120℃和150℃下均可增加丙烯酰胺的生成量。蛋氨酸在0.2mol/ml葡萄糖的浓度下不影响丙烯酰胺的生成。蛋氨酸在1.0mol/ml葡萄糖的浓度下减少大约小于50%的丙烯酰胺的生成。
III.十九种氨基酸在葡萄糖和天冬酰胺的溶液中对丙烯酰胺的形成的作用
上面已经描述了四种氨基酸(赖氨酸、半胱氨酸、蛋氨酸和谷氨酸)对丙烯酰胺的形成的作用。也测试了另外15种氨基酸。准备一个PH7.0的磷酸钠缓冲液溶解天冬酰胺(0.176%)和葡萄糖(0.4%)。分别添加15种与葡萄糖摩尔浓度相同的氨基酸。以不添加其它氨基酸的天冬酰胺和葡萄糖的溶液作为对照组。溶液在120℃下加热40分钟后用GC-MS测定丙烯酰胺的含量。结果如以下的表3。

如上表所示,所有15种氨基酸中没有任何一种在减少丙烯酰胺生成方面如半胱氨酸、赖氨酸或甘氨酸一样有效。其中9种可将丙烯酰胺减少到相当于对照组的22-78%之间的水平,而6种氨基酸可增加丙烯酰胺的生成到相当于对照组的111-150%之间的水平。
下表4总结所有氨基酸的结果,按氨基酸的有效性大小来进行排序。半胱氨酸、赖氨酸和甘氨酸是有效的抑制剂,可使丙烯酰胺的生成量减少到小于对照组的15%。其后的9种氨基酸是较弱的抑制剂,可使丙烯酰胺的生成量减少到对照组的22-78%之间。接下来的7种氨基酸会增加丙烯酰胺的生成。谷氨酸增加的程度最高,显示为对照组的320%。

V.制备焙烤马铃薯片
根据上述结果,本发明的优选实施例是在合成小吃食品的配方中添加半胱氨酸或赖氨酸,本例中的合成小吃食品指的是焙烤的合成马铃薯片。制备该产品的过程如图3所示。在面团制备步骤31中,马铃薯薄片、水和其他成分被混合在一起做成面团(术语“马铃薯薄片”和“马铃薯粉”在这里是可互换的,且包括所有干燥的薄片或粉状物,而不计较其大小尺寸)。在压片步骤32中,将面团送入可以使其扁平的压片机,然后被切成离散的薄片。在烹制步骤33中,切好的薄片要进行焙烤,直到达到预期的色泽和水份含量。然后将得到的薯片在调味步骤34中进行调味,并在包装步骤35中进行包装。
本发明的第一个实施例通过使用上述制备过程来显示。为了详细说明该实施例,还在对照组和试验批次之间进行比较,在所述试验中,添加了三种不同浓度之一的半胱氨酸和一种浓度的赖氨酸。下表6显示了在不同的批次中所使用的成分。

1:预计氨基酸的D-异构体或氨基酸的D-和L-异构体两者的外消旋混合物是等效的,尽管L-异构体可能是最好和最便宜的来源。
在所有批次实验中,首先将干的成分混合,然后分别添加油并混合。半胱氨酸或赖氨酸事先溶解在水中,然后添加到面团里。压片前面团的水份含量以重量计为40~45%。面团被送去压片,制成厚度为0.020到0.030英寸之间的薄片,然后切成薯片尺寸的薄片,并焙烤。
烹制以后,测定水分、油脂,并根据HUNTER L-A-B色差系统测定色泽。对样品进行测试,以获得成品中的丙烯酰胺含量。下表7显示这些分析结果。

在对照薯片中,烹制结束后丙烯酰胺的含量为1030ppb。所有不同含量的测定结果显示,添加半胱氨酸和赖氨酸都可以显著地减少丙烯酰胺的生成量。图4显示了得到的丙烯酰胺含量的图示。
在面团中添加半胱氨酸或赖氨酸可以显著减少成品中丙烯酰胺的含量。添加半胱氨酸的样品结果显示丙烯酰胺的减少量与半胱氨酸的添加量大致成正比。但是必须考虑到的是,在制备过程中添加氨基酸对最终产品的特性(如色泽、味道和质地)会产生间接影响。
还使用添加半胱氨酸、赖氨酸,和所述两种氨基酸中的任一种与CaCl2的混合物来进行另外的试验。这些试验采用与上述试验相同的程序,但所使用的马铃薯薄片中含有不同含量的还原糖和不同含量的氨基酸和CaCl2的混合物。在下表8中,组1的马铃薯薄片的还原糖含量为0.81%(表的这一部分重现了上述试验的结果),组2的还原糖含量为1.0%,组3的还原糖含量为1.8%。

如本表数据所显示,对于被测的每一种不同含量的还原糖,添加半胱氨酸或赖氨酸都会显著增加丙烯酰胺的生成量。而赖氨酸和氯化钙的混合物却几乎完全消除生成的丙烯酰胺,尽管这个结果是在还原糖含量最高的时候测定的。
VI.切片的油炸马铃薯片的试验
使用由马铃薯切片所得到的马铃薯片也可以产生一个相似的结果。然而,这些所希望的氨基酸并不能如同上述实施例那样简单地与马铃薯切片混合,因为这样有可能会破坏切片的完整性。在一个实施例中,马铃薯切片被浸入一个含有所希望的氨基酸添加剂的水溶液中,浸泡足够的时间使得氨基酸可以渗透进马铃薯切片的细胞结构中。这可以,例如,在如图2所示的清洗步骤23期间进行。
下表9显示在如上面图2所示的步骤23中添加1%的半胱氨酸进行清洗处理的结果。所有的清洗都是在室温下和所指示的时间内进行;对照组的处理则没有在水中添加任何物质。所述薯片在178℃下在所指示的时间内于棉花子油中进行油炸。

如上表所示,将厚度为0.53英寸的马铃薯切片浸泡在含有1Wt%半胱氨酸的水溶液中,可以将最终产品的丙烯酰胺的含量显著降低到100-200ppb。
本发明还证实了将半胱氨酸添加到玉米面团(或湿润粉糊)中制备玉米片的作用。在磨粉的过程中将溶解的L-半胱氨酸添加到煮制的玉米中,使得半胱氨酸在磨粉期间均匀的分散在湿润粉糊中。添加600ppm的L-半胱氨酸可以使丙烯酰胺的生成量从对照样品中的190ppb下降到经L-半胱氨酸处理的产品中的75ppb。
任何数量的氨基酸都可以应用到本发明公开的方案中,只要这些附加的成分可以对产品起到间接的调节作用,如改变食品的色泽、口感和质地。尽管所有显示的例子都是使用α-氨基酸(其中NH2基团附着在α-碳原子上),但申请人预期其他异构体如β-氨基酸或γ-氨基酸也能应用其中,虽然β-氨基酸和γ-氨基酸不常用作食品添加剂。本发明的优选实施例是使用半胱氨酸、赖氨酸、和/或甘氨酸。但是,其他氨基酸如组氨酸、丙氨酸、蛋氨酸、谷氨酸、天冬氨酸、脯氨酸、苯丙氨酸、缬氨酸和精氨酸也可以使用。这些氨基酸,尤其是半胱氨酸、赖氨酸和甘氨酸,通常是相对便宜的并且被广泛地用作食品添加剂。这些优选的氨基酸可以单独或是混合使用,以减少最终产品中丙烯酰胺的含量。另外,所述氨基酸可以在加热前通过以下方式添加到食品中:通过添加商业上可获得的氨基酸到食品的原料中,或是添加含有高含量游离氨基酸的另一种食品成分。例如,酪蛋白含有游离的赖氨酸,凝胶含有游离的甘氨酸。因此,当申请人指出一种氨基酸被添加到食品配方中时,应该理解为这种氨基酸可以作为商业上可获得的氨基酸来添加或是作为含有游离氨基酸的食品来添加,所述含有游离氨基酸的食品中的天冬酰胺的含量大于食品中本身的天冬酰胺含量。
添加到食品中以将丙烯酰胺的含量减少到可以接受的水平的氨基酸的用量,可以通过及种方式表示。出于商业需要,与未作处理的产品来比,需要添加的氨基酸的量,要足够将丙烯酰胺的最终生成量减少百分之二十(20%)。较为优选的,丙烯酰胺的减少量要达到百分之三十五到百分之九十五(35-95%)。更为优选的,丙烯酰胺的减少量要达到百分之五十到百分之九十五(50-95%)。一个使用半胱氨酸的优选实施例中,已经确定,添加至少100ppm的半胱氨酸可以有效减少丙烯酰胺的生成。因此,优选的半胱氨酸的用量范围是100-10000ppm,最优选的用量是大约1000ppm。在使用其他有效的氨基酸的优选实施例中,如赖氨酸和甘氨酸,所添加的氨基酸和产品中的还原糖的摩尔比在至少0.1比1(0.1∶1)的范围内可以有效减少丙烯酰胺的生成。更优选地,所添加的氨基酸和还原糖的摩尔比的范围在0.1∶1到2∶1之间,最优选的比率是1∶1。
所选的氨基酸减少丙烯酰胺生成的机制现在还不确定。可能的机制包括前体的反应和烯释之间的竞争,这样会导致丙烯酰胺生成量减少,以及丙烯酰胺的反应机制被打破。可能的机制包括(1)抑制麦拉德反应,(2)消耗葡萄糖和其他还原糖,和(3)与丙烯酰胺反应。具有一个游离硫醇基的半胱氨酸,在麦拉德反应中扮演着抑制剂的角色。由于丙烯酰胺被认为是由天冬酰胺通过麦拉德反应而得,因此,半胱氨酸应该降低麦拉德反应的速率并减少丙烯酰胺的生成。赖氨酸和甘氨酸会与葡萄糖和其他还原糖快速反应。如果还原糖被赖氨酸和甘氨酸消耗掉,那么就有较少的还原糖去与天冬酰胺反应来生成丙烯酰胺。氨基酸的氨基可以与丙烯酰胺的双键反应,这是一个麦拉德的副反应。而半胱氨酸的游离硫醇基也可以与丙烯酰胺的双键反应。
应该理解添加氨基酸也可对最终产品的特性产生不利影响,如色泽、口感和质地。根据本发明,这些特性的改变可以通过许多其他的方式进行补偿。例如,马铃薯片的色泽可以通过控制原料中糖的量来进行调整。可以在最终产品中添加不同的风味剂来改善其风味。产品的物理质地也可以调整,如,添加发酵剂或是各种乳化剂。
参照一些实施例,本发明已经进行了具体的描述,对于本领域技术人员来说,可以不偏离本发明的精神和范围,而提出其他通过使用一种氨基酸添加到需热加工的食品中米减少丙烯酰胺的生成量的方法。例如,本发明的方法已经公开了可应用于马铃薯和玉米制品,那么这种方法也可以应用于由大麦、小麦、黑麦、大米、燕麦、粟米和其他淀粉谷物制备食品的方法中,以及另外包括其他含有天冬酰胺和还原糖的食品,例如,甜土豆、洋葱和其他蔬菜。此外,本发明的方法已经示例用于马铃薯片和玉米片中,但也可以应用到其他许多食品的制备中,例如其他类型的小吃、谷物食品、曲奇、饼干、硬脆饼干、面包和面包卷以及粘滚上面包屑的肉。在许多这些食品中,氨基酸可以在用于制备食品的面团的混合期间添加,使得氨基酸在食品的烹制过程中起作用,从而减少丙烯酰胺的生成量。另外,添加氨基酸的方法也可以与其他能减少丙烯酰胺的方法结合起来使用,从而使产生的丙烯酰胺的量可以被接受,而且不影响食品的口味、色泽或气味,其他特性。
Claims (49)
1.一种降低含有游离的天冬酰胺和单糖的热加工的食品中的丙烯酰胺含量的方法,所述方法包括以下步骤:
(a)添加一种含有游离氨基酸的成分于食品产品中,其中所述成分的添加量要足以使所述热加工的食品中的丙烯酰胺的最终含量减少到一个可以接受的水平;
(b)热加工所述食品产品。
2.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)中所述成分的添加量足以将所述热加工的食品中的丙烯酰胺的最终含量降低至少20%。
3.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)中所述成分的添加量足以将所述热加工的食品中的丙烯酰胺的最终含量降低至少35%。
4.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)中所述成分的添加量足以将所述热加工的食品中的丙烯酰胺的最终含量降低至少50%。
5.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)中所述成分的添加量足以将所述热加工的食品中的丙烯酰胺的最终含量降低至少65%。
6.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)添加至少100ppm的半胱氨酸。
7.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)添加大约1000ppm的半胱氨酸。
8.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)添加多至10000ppm的半胱氨酸。
9.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)在所述食品中添加至少0.1摩尔氨基酸/每摩尔还原糖。
10.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)在所述食品中添加大约1.0摩尔氨基酸/每摩尔还原糖。
11.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)在所述食品中添加多至2.0摩尔氨基酸/每摩尔还原糖。
12.根据权利要求1所述的方法,其特征在于,所述游离氨基酸选自以下的组,包括半胱氨酸、赖氨酸、甘氨酸、组氨酸、丙氨酸、蛋氨酸、谷氨酸、天冬氨酸、脯氨酸、苯丙氨酸、缬氨酸和精氨酸。
13.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)在所述食品产品中添加一种商业上可获得的氨基酸。
14.根据权利要求1所述的方法,其特征在于,所述添加步骤(a)在所述食品产品中添加一种含有游离氨基酸的食品。
15.根据权利要求1所述的方法,其特征在于,在所述添加步骤(a)中,所述食品被浸泡在含有所述成分的溶液中。
16.根据权利要求1所述的方法,其特征在于,所述成分与其他成分混合制成面团。
17.根据权利要求1所述的方法,其特征在于,所述烹制步骤(b)包括炸所述食品产品。
18.根据权利要求1所述的方法,其特征在于,所述烹制步骤(b)包括焙烤所述食品产品。

(b)热加工所述混合物。
23.根据权利要求22所述的方法,其中所述游离氨基酸选自以下的组,包括半胱氨酸、赖氨酸、甘氨酸、组氨酸、丙氨酸、蛋氨酸、谷氨酸、天冬氨酸、脯氨酸、苯丙氨酸、缬氨酸和精氨酸。
24.根据权利要求22所述的方法,其中所述添加步骤(a)将商业上可获得的氨基酸添加到所述混合物中。
25.根据权利要求22所述的方法,其中所述添加步骤(a)将含有游离氨基酸的食品添加到所述混合物中。
26.根据权利要求22所述的方法,其中所述烹制步骤(b)包括炸所述食品产品。

(c)热加工所述切片以形成薯片。
