发明背景
黄体素和玉米黄质通常存在于叶黄素(植物色素,是类胡萝卜素中的一小类)中,其中叶黄素可以在一般的消耗食物中找到,如菠菜,羽衣甘蓝,玉米和橙子。人类不能合成黄体素和玉米黄质,因此,人体中的黄体素和玉米黄质只能来源于饮食。与β-胡萝卜素不同,黄体素和玉米黄质不能转化为维生素A。黄体素和玉米黄质的化学结构相似,只能通过专门的分析方法来对彼此进行区别。这种混合物可以被共同称为“黄体素+玉米黄质”,以反映这种混合物不能在化学分析中被分离。
黄体素和玉米黄质是在人类视网膜中唯一发现的与日常饮食有关的类胡萝卜素。目前对黄体素和玉米黄质在眼中的选择性累积机制的了解还很贫乏。然而,黄体素和玉米黄质在人类眼睛中累积的事实表明它们可能具有生物学功能。黄体素和玉米黄质可以作为抗氧化剂或者滤出蓝光中的有害波长而保护视网膜不受损害。另外,黄体素和玉米黄质可以通过滤出没有完全由晶状体聚焦的蓝光来提高视敏度(Hammond,et al,Arch Biochem Biophys(2001),385,p.41-46)。这些特性可能对初生婴儿尤其重要,因为婴儿的视敏度在生命的头几个月内发育缓慢(McCulloch,Opthal Physiol Opt(1998),18,p.140-146),而且他们的眼睛不能象成人一样滤出有害光线(Graillard,et al,IOVS(2000)41,p.1454-1459)。

1994年5月,在美国华盛顿州西雅图召开的儿科研究协会的会议中,发表了一篇名为“富含β胡萝卜素(βC)的配方中βC对足月婴儿个体和全体血清类胡萝卜素的贡献(Contribution of Beta Carotene(βC)from βC EnrichedFormulae to Individual and Total Serum Carotenolds in Term Infants)”的文章,这篇文章比较了配方喂养与母乳哺育的足月婴儿之间类胡萝卜素的血浆浓度。这一发现证明了配方喂养的婴儿的血清类胡萝卜素水平(包括叶黄素水平)比母乳哺育的婴儿低。
已经有三项研究报导了在验尸解剖的婴儿眼部黄体素和玉米黄质的化学分析,虽然这些研究都没有区分母乳哺育与配方喂养的婴儿。Handelman等人(IOVS(1988),29,p.850-855)报导了两个婴儿的视网膜黄体素和玉米黄质水平,同时Bone等人报导了一项研究中五个婴儿的黄体素和玉米黄质水平(Bone et al.,Exp.Eye Res.(1997)64,211-218)和另一项研究中四个婴儿的黄体素和玉米黄质水平(Bone et al.,IOVS(1988),29,p.843-849)。总的来说,婴儿的黄体素和玉米黄质的视网膜总浓度比儿童或成人低,但是婴儿中黄体素与玉米黄质的比率比成人要高很多。因而,就眼中的黄体素和玉米黄质而言,婴儿与年长的孩子和成人明显不同。
几项研究已经测量了人乳样品中黄体素+玉米黄质的混合含量。例如,见Canfield等,Eur.J.Nutr.,40:30-38(2001);Schweigert等,Int.J.Vitam.Nutr.Res.,70(3)p.79-83(2000);Canfield等,Am.J.Clin.Nutr.,67:276-283(1998);Canfield等,Anal.Chem.69,p.1873-1881(1997);Giuliano等,Methods InImmunology,Vol.213 p.381-399(1992);和Patton等,LIPIDS,Vol.25,No.3,p.159-165(1990)。一项研究已经分别报导了人乳中黄体素相对于玉米黄质的水平(Khachik et al.Anal.Chem.(1997),69,1873-1881)。由此可见,人乳中含有黄体素和玉米黄质。然而,目前没有任何婴儿配方中添加了黄体素和玉米黄质。
除了其它成分外,包含类胡萝卜素,如黄体素和玉米黄质的配方是已知的。U.S.Patent No.6,261,598公开了含有β-胡萝卜素,番茄红素和黄体素的类胡萝卜素配方,及其在人类食物中的应用。
WO 200137781公开了一种用于饮料和化妆品的营养物水分散体,它选自于(a)异黄酮,(b)番茄红素,(c)黄体素和(d)一种辅酶。
EP774251公开了玉米黄质的3R-3R立体异构体在制备药物中的用途,这种药物作为营养补充物用于治疗或预防人体的黄斑变性。
EP1155620公开了作为饮食补充物的营养组合物,它含有矿物组分,至少40%的鱼油和维生素组分,其中的维生素组分选自β胡萝卜素,复合维生素B,维生素C,维生素D,维生素E,叶酸和生物素,以及上述维生素的衍生物。
然而,至今都没有黄体素和玉米黄质在婴儿配方组合物中应用的描述。因此,需要这样一种配方。
发明概述
本发明涉及包含黄体素和玉米黄质的婴儿配方组合物。本发明还进一步涉及一种喂养婴儿的方法,包括给予婴儿包含黄体素和玉米黄质的组合物。
发明详述
本配方中使用的黄体素和玉米黄质组分可以是游离叶黄素、叶黄素酯形式或者所属技术领域已知的一种或多种不同方法制得的黄体素和玉米黄质的其它化学形式。例如,可以通过从万寿菊或其它富含叶黄素原料中提取,化学合成,发酵,或其它生物技术衍生和富含叶黄素的来源中获取黄体素和玉米黄质。本发明使用的黄体素和玉米黄质的适当形式是由Roche Vitamins,Inc.,(Parsippany,NJ)商供的“Lutein 5%TG”。
本发明的婴儿配方组合物可以包含大约6至约230mcg/L的黄体素和玉米黄质。本婴儿配方组合物优选为包含大约15至约44mcg/L,更优选为包含大约20至约30mcg/L,最优选为包含大约25mcg/L的黄体素和玉米黄质。这里公开的黄体素和玉米黄质的含量是指两者混合物的总含量,而不是其单独的含量。因此,只要配方包含黄体素和玉米黄质的总含量同上所述即可,对单一组分的含量没有严格要求。
本发明使用的黄体素和玉米黄质的含量与全球人乳中发现的这类混合物的含量近似。如2000国际人乳和哺乳研究组织会议所报导,一项研究分析了9个国家的450名妇女的母乳样品中黄体素和玉米黄质的总量。测得黄体素和玉米黄质的个体浓度最低为6mcg/L,最高为230mcg/L。

本发明的婴儿配方组合物可以通过使用普通牛奶和基于蛋白质的配方如S-26或基于大豆蛋白的配方如来自Wyeth Nutrition的Nursoy制得。
本发明的婴儿配方组合物还可以包含被认为日常饮食必需的维生素和矿物质。这些维生素和矿物质应当以营养显著的量存在于婴儿配方组合物中。适当的维生素和矿物质的例子包括维生素A,复合维生素B,维生素C,维生素D,维生素E,维生素K,钙,镁,钠,钾,磷,铜,锌,碘,硒,铁,烟酸,叶酸,泛酸,生物素,胆碱,肌醇和锰。
本婴儿配方组合物还可以进一步包含一种或多种所属技术领域已知的脂类原料。此外,本组合物还可以包含其它有有益作用的原料,如核苷酸,免疫球蛋白和多不饱和脂肪酸。

将4500mg的黄体素和玉米黄质(Roche Vitamins提供的Lutein 5%TG)与上述脂溶性维生素混合制得例1所述的配方。上述混合物再加入到脂类混合物中得到黄体素和玉米黄质终浓度为225mg/kg的最终配方粉末。其余组分混合后得到最终混合物,将其干燥得到粉末。
在不背离本发明基本内容的前提下,可以以其它具体形式实现本发明,因此,应参照所批准的内容和权利要求,而不是前述说明书来说明本发明的范围。
Claims (10)
1.一种婴儿配方组合物,其中包含黄体素和玉米黄质。
2.权利要求1的婴儿配方组合物,其中包含6~230mcg/L黄体素和玉米黄质。
3.权利要求2的婴儿配方组合物,其中包含15~44mcg/L黄体素和玉米黄质。
4.权利要求3的婴儿配方组合物,其中包含20~30mcg/L黄体素和玉米黄质。
5.权利要求4的婴儿配方组合物,其中包含25mcg/L黄体素和玉米黄质。
6.权利要求1所述的营养组合物,其中所述组合物是粉末形式的。
7.权利要求1所述的营养组合物,其中所述组合物是液体形式的。
8.权利要求1所述的营养组合物,其中进一步含有维生素A,维生素D,维生素E和复合维生素B中的至少一种。
9.权利要求1所述的营养组合物,其中进一步含有钙,镁,钠,钾,磷,铜,锌,碘,硒和铁中的至少一种。
10.一种喂养婴儿的方法,包括给予所述婴儿营养上足够量的权利要求1的配方。
