Abstract
本发明提供了一种低聚木糖的制备方法,包括下列顺序步骤:将玉米芯粉与水混合,加入弱酸催化剂进行裂解,将玉米芯中的木聚糖溶出;调节木聚糖水溶液pH值为5.0~6.0,加入木聚糖酶进行酶解,高温对木聚糖酶进行灭活;滤除玉米芯粉渣,对木聚糖糖液进行活性炭和离子交换树脂除杂脱色;大分子截留膜截留大分子木聚糖,过滤出低木聚糖糖液;钠滤膜浓缩脱盐;将低聚木糖糖液真空浓缩为浅黄色粘稠状糖浆,低聚木糖糖浆经赋形剂或直接喷雾干燥成低聚木糖粉剂。该方法简单易行,适宜于工业化生产,得到的低聚木糖纯度高,色浅,可广泛应用于各种食品、保健品、药品中作为添加剂。
Description
技术领域
本发明涉及一种低聚木糖的制备方法,具体涉及一种将玉米芯中的半纤维素转化为木聚糖并从中提纯低聚木糖的方法。
背景技术
低聚木糖亦称木寡糖(xylooligosaccharides),由2~7个D-木糖以β-1,4-木糖苷键结合而成,是功能性低聚糖家族中的一个重要成员。它的甜度比蔗糖和葡萄糖均低,约为蔗糖的40%。低聚木糖对pH值及热的稳定性较好,即使是在酸性条件(pH=2.5~7)加热也基本不分解,适合用于酸奶、乳酸菌饮料和碳酸饮料等酸性饮料中。下图为低聚木糖的主要成分及化学结构。
低聚木糖极难被消化吸收,肠道内残存率高,具有极好的双歧杆菌增殖性,其选择利用性高于其它功能性低聚糖。目前业已研究确认的低聚木糖的生理功能主要包括以下几个方面:
(1)提供较低的能量,满足喜食甜品又担心发胖者的要求,还可供糖尿病人、肥胖病人和高血压病人食用;
(2)活化肠道内双歧杆菌并促进其增殖,抑制病原菌,防止腹泻;
(3)防止便秘;
(4)降低血清中胆固醇含量、降低血压、生成营养物质、增强机体免疫力和抵抗肿瘤;
(5)不会引起龋齿,有利于口腔健康;
(6)清除肠内毒素。
低聚木糖具有的独特的生理功能决定了它的应用主要是利用其能赋予肠道内双歧杆菌选择增殖活性的效果,其次,由于其稳定性强,口感好,已被应用于食品上。研究表明,在饮料中添加低聚木糖,25℃下即使保存8个月,仍有约99%的残存率,显示低聚木糖有良好的稳定性。当将低聚木糖添加到奶油中,若将通常配合比例的砂糖的15~50%更换为低聚木糖,可得到提高产品乳化性的良好效果。而在果冻中,即使将低聚木糖和砂糖按30~50%的比例更换,其产品的嗜好性也没有问题,同时还有爽口的效果。在氨基酸共存条件下,低聚木糖被加热时具有较果糖容易着色的特点,将其用于面包的焙烤当中发现,在面包中添加适量的低聚木糖,能获得易于赋予面包良好的色泽、增加香味、避免生霉和容易控制水分等效果。
低聚木糖一般是以富含木聚糖的植物资源,如木屑、玉米芯、棉籽壳、稻壳和菜籽壳等为原料,经过内切型木聚糖酶水解后,再进行分离、精制而制得。
发明内容
本发明的目的是实现玉米芯中半纤维素转化为高品质低聚木糖的工业化生产方法。该方法简单易行,是以于工业化生产,得到的低聚木糖纯度高,色浅,可广泛应用于各种食品、保健品、药品中作为添加剂。这一目的可以通过具体选择对玉米芯原料的预处理、直热汽爆裂解、酶解反应、脱色与除杂等过程的工艺条件,尤其是木聚糖酶选择和制备工艺以及直热汽爆裂解过程中催化剂和反应条件的选择来实现。
本发明公开了一种低聚木糖的制备方法,包括下列顺序步骤:
(1)将玉米芯粉与水按重量比为1∶6~10混合,并加入玉米芯重量百分比为0.2~1.5%的弱酸催化剂,在155℃~180℃蒸煮30min~120min进行裂解,将玉米芯中的木聚糖溶出;(2)调节木聚糖水溶液PH值为5.0~6.0,按照每克木聚糖加入50~85UI单位活性木聚糖酶的比例加入木聚糖酶,在45~60℃酶解反应4h~10h进行酶解,升温至90℃~105℃,保温10min~30min,对木聚糖酶进行灭活;(3)滤除玉米芯粉,得到木聚糖糖液;(4)对木聚糖糖液进行活性炭和离子交换树脂除杂脱色;如E306FG及阴离子交换树脂如D001脱杂;(5)通过大分子截留膜截留大分子木聚糖,过滤出低聚木糖糖液;对大分子继续酶解;如应用大分子截留膜截留分子量1200以上的低聚木糖再次进行酶解,分子量1200以下的低聚木糖通过截留膜,经过钠滤浓缩膜脱盐率为原液的70%,浓缩率为原液的5-6倍;(6)对低聚木糖糖液进行钠滤膜浓缩脱盐,并进一步按照下列步骤(7)、步骤(8)、步骤(9)、或(10)对钠滤膜浓缩脱盐后的木聚糖糖液进行加工;(7)真空浓缩为浅黄色粘稠状低聚木糖糖浆;(8)真空浓缩为55%~75%的糖液,以麦芽糊精、β-环糊精或玉米淀粉为赋形剂,经喷雾干燥得低聚木糖重量百分比为20%~50%的低聚木糖粉剂;(9)钠滤膜分离单糖,真空浓缩为淡黄色粘稠透明状低聚木糖糖浆;(10)钠滤膜分离单糖,真空浓缩为低聚木糖重量百分比55%~75%的糖浆,经喷雾干燥得总糖为70%~98%的低聚木糖粉剂。
钠滤膜是现有技术中一种利用液体的分子量大小不同进行来分离的一种高端技术,在本工艺中用于制作高纯度低聚木糖,纯度可达到95%以上。大分子过滤膜也是钠滤膜的一种,在本工艺中用于截留酶解不彻底的大分子的低聚木糖,再次酶解,保证了产品质量。
上述方法中个,通常对作为原料的玉米芯粉进行预处理,方法为在50℃~80℃保温30min~120min。
考虑到在木聚糖酶水解过程中,酶的过量添加会造成生产成本的增高,因此本发明采取木聚糖酶粗酶液与玉米芯粉的比为1ml/12g~1ml/16g,木聚糖酶粗酶液酶活性为800UI/ml~1000UI/ml,即每克木聚糖中加入50~85UI单位活性木聚糖酶。
本发明采用的木聚糖酶液是由链霉菌sp.E-86菌株经液体发酵制得。
低聚木糖糖液的脱色采用常规的脱色方法,即活性碳和离子柱脱色,为更好的脱除所得低聚木糖糖液的颜色,本发明的活性炭脱色可为多次,通常为两次,脱色温度可根据需要进行调整,通常为80℃,活性炭添加量为糖液量的0.5%~1.5%,脱色时间25min~40min。
本发明离子交换树脂除杂过程为阳、阴、阳离子交换树脂,低聚木糖糖液温度小于45℃,每小时低聚木糖糖液流速为树脂体积的1~2倍,所得糖液的电导率小于50VS/cm,pH为4.5~6.0。
本发明采用真空对木聚糖糖液进行浓缩,浓缩温度为60~80℃,并通过钠滤浓缩膜进行脱盐和浓缩,钠滤膜浓缩在常温下即可进行,通常在15~30℃左右,20℃温度为最佳,通常真空浓缩方式为双效真空浓缩,浓缩过程可采用多次方式,如采用两次方式,第一次浓缩为钠滤膜浓缩,将低聚木糖糖液浓缩到总糖含量8%~12%的糖浆;第二次浓缩为75℃下,将低聚木糖糖液浓缩到总糖含量55%~75%的糖浆。上述技术方案中,双效真空浓缩设备是现有技术中一种利用低温真空蒸发水分达到浓缩液体的设施。
按照本发明的干燥方法,真空干燥时热风的进口温度为130℃~160℃,出口温度为65℃~85℃,物料流量为0.8~1.2m3/h,塔内滞留时间10秒~20秒。

低聚木糖的制备方法的工艺流程,可通过图1说明:
图1提供了自玉米芯中制备低聚木糖的方法,包括下列循序步骤,1、选择玉米芯;2、除杂;3、对玉米芯进行粉碎;4、对玉米芯预处理;5、高温蒸煮裂解;6、木聚糖酶水解;7、过滤除渣;8、活性炭脱色;9、纳滤膜分离大分子;10、纳滤膜浓缩脱盐;11、离子交换净化;12、真空浓缩;13、纳滤膜分离提纯;14、加赋形剂喷雾干燥;15、浅黄色粘稠状低聚木糖糖浆;16、真空浓缩;17、纳滤膜浓缩;18、低聚木糖粉剂、19、浅黄色粘稠状低聚木糖糖浆;20、低聚木糖粉剂;21、杂糖回收。
自玉米芯中制备低聚木糖的具体方法参见下列具体实施方式。
具体实施方式1
称取40目~60目的玉米芯粉500kg,加入4000kg,纯净水调匀,升温80℃加入2.5kg乙酸,在160℃下蒸煮裂解2h后,经汽液分离器进入酶解罐,冷却至55℃。分别用1mol/ml盐酸溶液和1mol/ml的氢氧化钠溶液调节反应液的pH到5.5,按1ml酶液/14.5g玉米芯粉的量加入由sp.E-86菌株通过液体发酵制得的酶活性为900UI/ml的木聚糖酶液,酶解反应8h后,在90℃保温灭活15min,冷却到60℃进行板框过滤得到糖液。在糖液中加入总体积1%的活性炭粉,80℃搅拌脱色30min,过滤,在75℃将糖液浓缩为总糖含量为15%的低聚木糖糖浆。将15%糖浆进行二次脱色,脱色条件与第一次相同。然后将经过两次脱色的糖浆上离子交换树脂柱,按照阳阴阳方式进行脱色脱杂,应用大分子截留膜截留分子量1000以上的低聚木糖再次进行酶解,分子量1000以下的低聚木糖通过截留膜,经过钠滤膜脱盐,脱盐率为原液的70%,浓缩液为原液的5-6倍。再真空浓缩为55%~75%的糖液,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液经真空浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,或经过钠滤膜提取单糖后低聚木糖含量达到95%,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,得率8.8%。
具体实施方式2
称取40~60目的玉米芯粉480kg,加入4000kg纯净水调匀,加入2.0kg柠檬酸,在170℃下蒸煮裂解100min后,经汽液分离器进入酶解罐,冷却至55℃。分别用1mol/ml盐酸溶液和1mol/ml的氢氧化钠溶液调节反应液的pH到5.5,按1ml酶液/1.3g玉米芯粉的量加入由sp.E-86菌株通过液体发酵制得的酶活性为70UI/ml的木聚糖酶液,酶解反应6h后,在100℃保温灭活13min,冷却到60℃进行板框过滤。在糖液中加入总体积1%的活性炭粉,80℃搅拌脱色30min,过滤,在75℃将糖液浓缩为总糖含量为20%的低聚木糖糖浆。将15%糖浆进行二次脱色,脱色条件与第一次相同。然后将经过两次脱色的糖浆上离子交换树脂柱进行脱色脱杂,应用大分子截留膜截留分子量1000以上的低聚木糖再次进行酶解,分子量1000以下的低聚木糖通过截留膜,再经过钠滤膜脱盐,脱盐率为原液的70%,浓缩液为原液的5-6倍。真空浓缩为55%~75%的糖液,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,得率8.5%;或经过钠滤膜提取单糖后低聚木糖含量达到95%,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,得率8.5%
具体实施方式3
称取40目~60目的玉米芯粉500kg,加入3800kg,纯净水调匀,在165℃下蒸煮裂解2h后,经汽液分离器进入酶解罐,冷却至55℃。分别用1mol/ml盐酸溶液和1mol/ml的氢氧化钠溶液调节反应液的pH到5.5,按1ml酶液/1.45g玉米芯粉的量加入由sp.E-86菌株通过液体发酵制得的酶活性为60UI/ml的木聚糖酶液,酶解反应8h后,在90℃保温灭活15min,冷却到60℃进行板框过滤。在糖液中加入总体积1%的活性炭粉,80℃搅拌脱色30min,过滤,在75℃将糖液浓缩为总糖含量为15%的低聚木糖糖浆。将15%糖浆进行二次脱色,脱色条件与第一次相同。然后将经过两次脱色的糖浆上离子交换树脂柱进行脱色脱杂,糖应用大分子截留膜截留分子量1000以上的低聚木糖再次进行酶解,分子量1000以下的低聚木糖通过截留膜,经过钠滤膜脱盐,脱盐率为原液的70%,浓缩液为原液的5-6倍。再真空浓缩为55%~75%的糖液,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,得率7.6%;或经过钠滤膜提取单糖后低聚木糖含量达到95%,糖液电导率小于50VS/cm,pH5.0。最后在75℃下,将糖液浓缩至总糖含量为70%的浅黄色粘稠状低聚木糖糖浆,得率7.6%
具体实施方式4
按具体实施方式1中的条件和步骤,经钠滤膜脱盐,制得100kg 40%的低聚木糖糖浆,加入纯度为98%的麦芽糊精120kg,在进口温度145℃,出口温度85℃,物料流量为1.0m3/h条件下进行喷雾干燥,得到含量25%的低聚木糖粉状产品。
具体实施方式5
按具体实施方式1中的条件和步骤,经钠滤膜脱盐和真空浓缩后、制得100kg 40%浓度的低聚木糖,经纳滤膜分离提纯糖浆,在进口温度145℃,出口温度85℃,物料流量为1.0m3/h条件下进行喷雾干燥,得到含量95%的低聚木糖粉状产品。
Claims (11)
1、一种低聚木糖的制备方法,包括下列顺序步骤:
(1)将玉米芯粉与水按重量比为1∶6~10混合,并加入玉米芯重量百分比为0.2~1.5%的弱酸催化剂,在155℃~180℃蒸煮30min~120min进行裂解,将玉米芯中的木聚糖溶出;
(2)调节木聚糖水溶液PH值为5.0~6.0,按照每克木聚糖加入50~85UI单位活性木聚糖酶的比例加入木聚糖酶,在45~60℃酶解反应4h~10h进行酶解,升温至90℃~105℃,保温10min~30min,对木聚糖酶进行灭活;
(3)滤除玉米芯粉,得到木聚糖糖液:
(4)对木聚糖糖液进行活性炭和离子交换树脂除杂脱色;
(5)通过大分子截留膜截留大分子木聚糖,过滤出低聚木糖糖液;对大分子继续酶解。
(6)对低聚木糖糖液进行钠滤膜浓缩脱盐,并进一步按照下列步骤(7)、步骤(8)、步骤(9)、或(10)对钠滤膜浓缩脱盐后的木聚糖糖液进行加工;
(7)真空浓缩为浅黄色粘稠状低聚木糖糖浆;
(8)真空浓缩为55%~75%的糖液,以麦芽糊精、β-环糊精或玉米淀粉为赋形剂,经喷雾干燥得低聚木糖重量百分比为20%~50%的低聚木糖粉剂;
(9)钠滤膜分离单糖,真空浓缩为淡黄色粘稠透明状低聚木糖糖浆;
(10)钠滤膜分离单糖,真空浓缩为低聚木糖重量百分比55%~75%的糖浆,经喷雾干燥得总糖为70%~98%的低聚木糖粉剂。
2、根据权利要求1所述的低木聚糖的制备方法,其特征在于:
步骤(1)中玉米芯粉蒸煮裂解时温度为160~170℃,蒸煮时间80~120min。
3、根据权利要求1所述的低木聚糖的制备方法,其特征在于:
步骤(5)中通过大分子截留膜截留出的低聚木糖分子量小于1000分子量,分离温度18℃~28℃,截留出的大分子木聚糖重新回到步骤(2)中进行酶解。
4、根据权利要求1所述的低木聚糖的制备方法,其特征在于:
步骤(4)中的脱色为活性炭和离子柱脱色,脱色温度20~45℃,离子柱脱色时,每小时低聚木糖糖液流速为树脂体积的1~2倍。
5、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
所述步骤(2)中的木聚糖酶通过sp.E-86菌株液体发酵制得。
6、根据权利要求1所述的低聚木糖的制备方法,其特征在于:所述弱酸催化剂为乙酸、甲酸、柠檬酸,加入玉米芯重量百分比为0.5~1.2%。
7、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
对步骤(1)中的玉米芯粉在50℃~80℃保温30min~120min进行预处理。
8、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
活性炭脱色为多次,脱色温度70~85℃,活性炭添加量占糖液重量百分比为0.5%~1.5%,脱色时间25min~40min。
9、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
步骤(4)中,离子交换树脂除杂脱色过程为使木聚糖糖液分别经过阳离子交换树脂、阴离子交换树脂和阳离子交换树脂,低聚木糖糖液温度小于45℃,每小时低聚木糖糖液流速为树脂体积的1~2倍。
10、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
步骤(7)、(8)、(9)和(10)中的真空浓缩方式为双效真空浓缩,浓缩温度为60~80℃;步骤(8)和步骤(10)中的喷雾干燥热风的进口温度为130℃~160℃,出口温度为65℃~85℃,物料流量为0.8~1.2m3/h,塔内滞留时间10秒~20秒。
11、根据权利要求1所述的低聚木糖的制备方法,其特征在于:
所述步骤(6)中,钠滤膜脱盐浓缩的温度为15~30℃。
