Abstract
本发明提供口腔护理组合物,含有:作为主要研磨清洁剂的碳酸钙颗粒和作为抗龋剂的含氟化合物;在口腔护理组合物中含有完全中和的聚丙烯酸以降低由碳酸钙和含氟化合物之间相互作用引起的可用氟的损失。
Description
本发明涉及含有颗粒碳酸钙作为主要研磨清洁剂的口腔护理组合物。
在口腔护理组合物例如牙膏中,碳酸钙颗粒是公知的研磨清洁剂,含有这种研磨清洁剂的牙膏已经进入市场。这种研磨清洁剂从其研磨特性上看是可接受的研磨清洁剂,但它有一个主要缺点:它和牙膏中含氟活性成分(例如碱金属氟化物)不兼容,然而为了达到抗龋的效果,通常需要在牙膏中加入含氟成分。
长期以来就已认识到这个问题,在GB-A-907,417(Ericsson)中,提出用碱金属一氟磷酸盐代替碱金属氟化物作为抗龋剂以克服上述缺陷。
在FR-A-2,251,309(Beecham)中,提出应用碱金属氟化物和碱金属一氟磷酸盐的混合物以克服碳酸钙使氟化物失活的问题。然而,应用这些系统确实可以使碳酸钙对含氟试剂的失活更少一些,但对于提供可接受的有效抗龋用途来说,仍然导致活性氟的大量损失。
现在已经发现:在含有作为主要研磨清洁剂的碳酸钙颗粒与含氟抗龋试剂的口腔护理组合物中,含有完全中和的聚丙烯酸盐能够显著提高碳酸钙和含氟试剂的兼容性、降低失活作用和因此减少可用活性氟的损失。
在此方面,在GB-A-1,476,063(Boots)中观测到:如果牙膏中含有碳酸钙和阴离子粘合剂例如羧甲基纤维素钠、藻酸、黄原胶、角叉菜素(carragheenin)和Carbopol_,则其容易产生贮存稳定性不足的问题,例如冷却和/或存放时成团。“Carbopol”商品包括丙烯酸均聚物和共聚物。在GB-A-1,476,063中提出应用硅铝酸盐来克服稳定性不足的问题。
我们已经发现:含有完全中和的聚丙烯酸盐完全不会引起贮存稳定性不足的问题,因此不必应用硅铝酸盐。
在ZA-A-86/0599(Blendax)中记载的牙膏含有碳酸钙作为研磨清洁剂。加入酸调节这些牙膏的pH值为7.5-8.5,以提供钙离子贮藏来补充牙釉质中的矿质。特别地,这些牙膏含有增稠剂例如聚丙烯酸的碱金属盐。然而,本发明的口腔护理组合物的pH大于8.5而且不含有酸,以避免游离钙离子和含氟抗龋剂之间不受欢迎的相互作用。
本发明所用的完全中和的聚丙烯酸盐是完全被下列物质中和的聚丙烯酸:合适的碱金属、铵离子或(烷醇)氨化合物,例如钠和钾的氢氧化物或碳酸盐、氨和三乙醇氨等。优选的完全中和的聚丙烯酸盐为钠盐。聚丙烯酸是丙烯酸的均聚物,分子量为1000-250,000、优选2500-100,000,更优选4000-70,000。特别优选的完全中和的聚丙烯酸盐为分子量8000的聚丙烯酸的钠盐,该产品可以购自BASF,商品名为Sokalan PA 30 CL。
在本发明的口腔护理组合物中,完全中和的聚丙烯酸盐的含量为组合物的0.05-5%重量、通常为0.1-3%重量。自然,还可以应用完全中和的聚丙烯酸盐的混合物。
尽管在最终的口腔护理组合物中可以通过提供足够量碱金属以完全中和聚丙烯酸从而就地产生完全中和的聚丙烯酸盐,但这并不是本发明的优选方案,因为在不破坏组合物其他成分的情况下很难控制就地进行的中和反应。优选地,把完全中和的聚丙烯酸加入到最终口腔护理组合物中,即中和反应独立地进行之后加入到组合物中。
本发明所用的碳酸钙颗粒研磨清洁剂可以是已知用于本领域口腔护理组合物中的任何合适类型的碳酸钙。其可以是天然来源例如霰石或方解石,如精细研磨的大理石和白云石等,或者它可以是合成来源的,例如微细的碳酸钙沉淀。其通常的平均粒子大小为1-60μm、一般为1-40μm。本发明的碳酸钙颗粒的用量为所述口腔护理组合物的10-60%重量、优选20-50%重量、更优选25-45%重量。
本发明所用的含氟抗龋剂为碱金属氟化物和/或碱金属一氟磷酸盐,其中后者是优选的。它们的通常用量足以提供50-1500ppm的可用氟。
所述口腔护理组合物可以制备成任意的合适应用形式,例如凝胶、牙粉和牙膏。它们可以制备成单剂型,或者它们可以制备成含有不同制剂的多隔室容器,例如一个容器含有碳酸钙及其可兼容成分,另一个容器含有其余成分。
本发明的口腔护理组合物还可以含有任选的常规成分例如可药用载体如淀粉、蔗糖、水或水/醇系统等。还可以含有少量表面活性剂例如阴离子、非离子、阳离子和两性表面活性剂。
当制备成牙粉时,这样的制剂可以含有任意的常用牙粉成分。
因此,它们可以含有其它的颗粒研磨材料例如硅酸盐、氧化铝、磷酸二钙、焦磷酸钙、羟磷灰石、三偏磷酸酯和不溶性六偏磷酸酯等,其用量低于碳酸钙的用量。研磨材料的总含量不高于组合物的60%重量。
而且,该牙粉制剂可以含有湿润剂例如甘油、山梨糖醇、丙二醇、木糖醇和乳糖醇等等。
还可以含有粘合剂和增稠剂例如羧甲基纤维素钠、黄原胶和阿拉伯树胶等。
还可以含有调味剂例如薄荷油和绿薄荷油以及防腐剂、不透明剂、调色剂、pH调节剂、甜味剂、牙齿增白剂和过氧化物漂白剂等等。还可以有益地含有过氧化物漂白剂的稳定剂例如吡啶二羧酸或锡酸钠。
还可以含有抗菌剂例如三氯苯氧氯酚、洗必太、铜盐、锌盐和亚锡盐,例如柠檬酸锌、柠檬酸锌钠和焦磷酸亚锡、血根硷提取物和甲硝唑。抗菌剂的其他例子为季铵化合物例如氯化十六烷基吡啶鎓;双胍类例如洗必太双葡萄糖酸盐、氨己嘧啶、奥替尼淀、阿来西定;氯化双苯酚化合物例如2,2'-亚甲基双-(4-氯-6-溴苯酚)。
还可以含有能够增强活性成分例如抗菌剂释放的聚合体化合物。这些聚合物的例子如:聚乙烯基甲基醚和顺丁烯二酸酐的共聚物,和其他类似增强释放的聚合物,例如:在DE-A-3942643(Colgate)中描述的那些聚合物。
而且,还可以含有抗炎试剂例如布洛芬、氟比洛芬、阿斯匹林和消炎痛等。
还可以含有其它的抗龋剂如氟化亚锡、氟化铵、plaque缓冲剂例如脲、乳酸钙、甘油磷酸钙和聚丙烯酸锶等。其它任选的成分包括维生素(例如维生素C)和植物提取物。还可以含有脱敏剂例如柠檬酸钾、氯化钾、酒石酸钾、碳酸氢钾、草酸钾、硝酸钾以及锶盐等。
还可以含有缓冲剂和盐以缓冲组合物的pH值和离子强度,例如正磷酸一钠或正磷酸三钠。为了提高活性成分的释放和稳定性,还可以应用脂质体和其它胶囊成分。
而且,所述口腔组合物可以含有抗结石剂例如碱金属焦磷酸盐、含次磷酸盐的聚合物、有机磷酸酯(盐)和含磷柠檬酸酯(盐)(phosphocitrates)等。
另外,所述组合物可以含有生物分子官能团例如细菌素、抗体和酶等等。
还可以含有的其它任选的成分例如:漂白剂,如在EP-A-0,545,594中所述,起泡系统例如碳酸氢钠/柠檬酸系统和颜色改变系统等。
下列实施例更详细地说明本发明。
实施例1
在一模型系统中,完全中和的聚丙烯酸盐对颗粒碳酸钙和一氟磷酸钠之间相互作用的影响的测试如下:
把20g碳酸钙颗粒(Pacal H)和0.057g一氟磷酸钠分散在25g去离子水中,加入0.04g福尔马林和27g山梨糖醇(70%)。再把各种含量的聚丙烯酸钠的水溶液(pH10)(聚丙烯酸的MW为8,000)加入到上述分散液中,得到的混合物在37℃下储存6天,然后,过滤上清液,测定溶液中剩余的一氟磷酸钠的含量和游离氟的含量。
应用气相色谱和阴离子选择电极分析分别测定总氟含量和游离氟离子的含量。
结果如下:

按照实施例2的方法,但应用不同量的聚丙烯酸钠(0.25%、0.75%和1.30%)制备制剂。同时还制备下列对照品试剂:不含聚丙烯酸钠、含0.5%正磷酸三钠和0.5%正磷酸一钠的制剂。把这些制剂在20℃和40℃下储存3个月。测定pH(20g蒸馏水中含5g糊剂,搅拌10分钟,然后测定pH)。按照实施例1的方法确定可提取氟的含量。得到下列结果。

Claims (8)
1、口腔护理组合物,含有:作为主要研磨清洁剂的碳酸钙颗粒和作为抗龋剂的含氟化合物;其特征在于:组合物的pH值大于8.5,该组合物还含有分子量为1000-250,000、完全中和的聚丙烯酸。
2、权利要求1的组合物,其特征在于分子量为2500-100,000。
3、权利要求2的组合物,其特征在于分子量为4000-70,000。
4、权利要求1-3的组合物,其特征在于:聚丙烯酸被碱金属、铵或(烷醇)胺化合物完全中和。
5、权利要求4的组合物,其特征在于:完全中和的聚丙烯酸为聚丙烯酸钠。
6、权利要求1-5的组合物,其特征在于:组合物中完全中和的聚丙烯酸含量为组合物重量的0.05-5%。
7、权利要求6的组合物,其特征在于:含量为组合物的0.1-3%重量。
8、含有分子量为1000-250,000的聚丙烯酸的、完全中和的聚丙烯酸在制备口腔护理组合物中的应用,其中所述的口腔护理组合物含有:作为主研磨清洁剂的碳酸钙颗粒和作为抗龋剂的含氟化合物,以降低可用活性氟的损失。
