Abstract

本发明公开了一种蚓激酶制备工艺,将蚯蚓经生理盐水处理,采用匀浆、冻融法制备蚯蚓粗提物,然后通过超滤、亲和层析、离子交换及分子筛等技术组合,从蚯蚓中提取纯化三种有纤溶活性的蚓激酶,其分子量分别为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种蛋白质,其纯度分别达到SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)显示一条带,高效液相色谱(HPLC)呈现一个峰的水平,其纤溶活性均为每mg含相当于尿激酶活性1000单位以上。体外溶血栓试验证明有良好的直接溶栓作用;动物体内试验证明,其体内溶血栓活性优于尿激酶。

Description

一种蚓激酶制备工艺
本发明公开一种蚓激酶制备工艺,涉及生物化学制药技术领域。
蚯蚓中含有溶解血栓作用的成分早有报道,有的用蚯蚓的粗提物在体外试验证明有血栓溶解作用。也有的用蚯蚓粗提物治疗血栓性疾病,并具有一定的疗效。但该粗提物成分复杂,其中有效成分为几种,是什么成分,分子量多大,均不清楚。也有的从蚯蚓中提纯了某种有一定纤溶活性的成分,为含有两个亚基的分子量约为45000的蛋白质,所含两个亚基分子量为26000与18000,该蛋白质的纤溶活性较低,其分离后的两个亚基则基本无活性。虽然有人用高效液相色谱法纯化获得了一种蚓激酶成分,但该法不适应规模化生产。总之,目前尚未见用规模化生产的工艺技术从蚯蚓中纯化出分子量在2-3.2万之间的高活性蚓激酶的报道。
本发明的任务是公开一种蚓激酶制备工艺,从蚯蚓中提取纯化蚓激酶,其分子量在3万以下,且工艺技术适合规模化生产。
本发明的任务是通过以下方案实施的:将蚯蚓用清水洗净泥土,加入生理盐水冲洗处理,按(V/V)1∶3~5的比例加入0.2MpH4.6~4.8的柠檬酸钠制成匀浆,经1000~4000rpm离心20~40分钟,取上清液,通过超滤法去除大分子蛋白,滤液用亲和层析法初步纯化,收集活性组分,再通过离子交换柱进一步纯化,取其活性组分,再经分子筛层析,分段收集各个活性组分,取分子量为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种活性蛋白质;既得本发明。
本发明的积极效果在于:以上三种蚓激酶纯度分别达到SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)显示一条带,高效液相色谱(HPLC)呈现一个峰的水平。其纤溶活性均为每mg含相当于尿激酶活性1000单位以上。体外溶血栓试验证明有良好的直接溶栓作用;动物体内试验证明,其体内溶血栓活性优于尿激酶。经临床200例血栓性患者使用,总有效率为100%,治愈率达40%。
下面结合实施例进一步说明本发明:
实施例1
取蚯蚓100g用清水洗净泥土,加入生理盐水冲洗三次,放入300ml的0.2MpH4.6的柠檬酸钠制成匀浆,经3000rpm离心30分钟,取上清液用50,000dt膜超滤,滤液上GSAC柱,用0.1MpH6.6柠檬酸钠洗脱,收集活性组分,再上DEAE-52纤维素柱,仍用0.1MpH6.6柠檬酸钠洗脱,收集活性组分,将活性组分通过Sephacryl S-200柱,用0.1MpH6.6 PBS梯度洗脱,分段收集各个活性组分,取分子量为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种活性蛋白质,测定活性为1700u/mg。
实施例2
取洗净的蚯蚓150g用生理盐水冲洗三次,加入600ml的0.2MpH4.7的柠檬酸钠制成匀浆,经3500rpm离心20分钟,取上清液用50,000dt膜超滤,滤液上GSAC柱,用0.1MpH6.7柠檬酸钠洗脱,收集活性组分,再上DEAE-52纤维素柱,仍用0.1MpH6.7柠檬酸钠洗脱,收集活性组分,将活性组分通过Sephacryl S-200柱,用0.1MpH6.7 PBS梯度洗脱,分段收集各个活性组分,取分子量为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种活性蛋白质,测定活性为1500u/mg。
实施例3
取洗净的蚯蚓150g用生理盐水冲洗三次,加入600ml的0.2MpH4.8的柠檬酸钠制成匀浆,经4000rpm离心20分钟,取上清液用50,000dt膜超滤,滤液上GSAC柱,用0.1MpH6.8柠檬酸钠洗脱,收集活性组分,再上DEAE-52纤维素柱,仍用0.1MpH6.8柠檬酸钠洗脱,收集活性组分,将活性组分通过Sephacryl S-200柱,用0.1MpH6.8 PBS梯度洗脱,分段收集各个活性组分,取分子量为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种活性蛋白质,得本发明蚓激酶,测定活性为1400u/mg。

Claims (2)

1、一种蚓激酶制备工艺,包括以下工艺制备:将蚯蚓用清水洗净泥土,加入生理盐水冲洗处理,按(V/V)1∶3~5的比例加入0.2MpH4.6~4.8的柠檬酸钠制成匀浆,经1000~4000rpm离心20~40分钟,取上清液,通过超滤法去除大分子蛋白,滤液用亲和层析法初步纯化,收集活性组分,再通过离子交换柱进一步纯化,取其活性组分,再经分子筛层析,分段收集各个活性组分,取分子量为3.0±0.2万、2.5±0.2万及2.2±0.2万的三种活性蛋白质。
2、根据权利要求1所述的蚓激酶制备工艺,其特征在于:亲和层析法采用GSAC柱,洗脱液为0.1MpH6.5~6.8柠檬酸钠;离子交换采用DEAE-52纤维素柱,洗脱液为0.1MpH6.5~6.8柠檬酸钠;分子筛为Sephaeryl S-200柱,洗脱液采用0.1MpH6.5~6.8PBS。
