Abstract
本发明提供一种猪繁殖与呼吸综合征病毒nsp12蛋白的抗原表位、单克隆抗体及应用,本研究利用表达和纯化后的nsp12蛋白为抗原免疫小鼠,通过间接ELISA和IFA等免疫学检测方法成功地筛选获得了5株能够稳定分泌nsp12mAb的阳性杂交瘤细胞株。结果表明,单抗3G11和9C2识别抗原表位为93TWGFESDTAY102,2E3识别抗原表位为115DYNDAFRARQ124,10G6和2A4识别抗原表位为142PGPVIEPTL150。而且,新鉴定出的三个抗原表位均位于nsp12蛋白表面,属于免疫显性抗原表位,在PRRSV‑2不同毒株之间的nsp12蛋白中三个抗原表位高度保守,可以作为PRRSV‑2检测的靶标,有助于PRRSV检测方法的开发。

技术领域
本发明属于生物技术领域,具体涉及猪繁殖与呼吸综合征病毒nsp12蛋白的抗原表位、单克隆抗体及应用。
背景技术
猪繁殖和呼吸综合征(PRRS)是由猪繁殖和呼吸综合征病毒(PRRSV)引起猪的一种重要传染病。PRRSV感染可损害妊娠母猪的生殖系统,导致流产、早产、产死胎和木乃伊胎等,而保育猪和育肥猪感染主要表现咳、喘等呼吸系统疾病,对世界各国的养猪业造成了严重危害。PRRSV是一种具有囊膜的单股正链RNA病毒,属于Nidovirales目,动脉病毒科的动脉炎病毒属成员。基因组的全长约为15kb,在mRNA加工过程中,其5'末端形成帽状结构,在3'末端有poly-A尾部结构。PRRSV基因组包含10个已知的开放阅读框(ORF1a,ORF1b,ORF2a,ORF2b,ORF3,ORF4,ORF5a和ORF5-ORF7),其中,ORF1a和ORF1b编码两个大的多蛋白pp1a和pp1ab,这两个多蛋白最终被ORF1a编码的4四种蛋白酶加工成至少14种与基因组复制和转录相关的非结构蛋白。在这些非结构蛋白中,位于ORF1b编码多蛋白C末端的nsp12是动脉病毒所特有的蛋白,但在PRRSV编码的非结构蛋白中nsp12的具体结构和功能目前仍然不十分清楚。最近有研究发现nsp12是一种膜相关蛋白,也是连接PRRSV其他nsps的相互作用的枢纽,推测nsp12可能是复制和转录复合物(RTC)的主要成分。同时,通过PRRSV反向基因操作技术,研究证实nsp12参与正链和负链亚基因组mRNA的合成,但不参与基因组RNA的合成,表明nsp12对PRRSV复制至关重要。由于PRRSV感染会导致促炎细胞因子表达水平显著升高,因此有研究表明nsp12可能是诱导pSTAT1-S727升高以及IL-1β和IL-8的表达的原因。推测nsp12可能通过调节细胞炎症因子的表达参与PRRSV的发病机制。但是目前关于PRRSVnsp12生物学作用的相关报道有限,其蛋白特性和抗原性尚不清楚,还需要通过更多的研究来解析nsp12在PRRSV感染中的确切功能。
发明内容
为了解决上述问题,本研究利用表达的nsp12蛋白为抗原,制备了5株特异性单克隆抗体,并鉴定其抗原表位,为PRRSV的诊断方法开发提供有效工具。
具体的,本发明提供一种PRRSV编码的非结构蛋白中nsp12蛋白的三个抗原表位,其特征在于所述的三个抗原表位分别为:93TWGFESDTAY102、115EDYNDAFRARQ124和142PGPVIEPTL150;
进一步的,本发明提供能够编码上述抗原表位的核酸,所述核酸为能够编码上述PRRSV编码的非结构蛋白中nsp12蛋白的三个抗原表位的基因序列;
进一步的,本发明提供了一种重组表达载体,所述重组表达载体携带上述核酸。
进一步的,本发明提供了一种重组细胞,所述重组细胞含有上述的重组表达载体,或者在基因组中整合有上述的核酸。

有益效果
在本研究中,我们制备了5株抗nsp12蛋白的单克隆抗体,这些单抗都与nsp12蛋白具有良好的反应性。随后,我们鉴定出5株单抗能够识别nsp12蛋白中的三个抗原表位(93TWGFESDTAY102、114EDYNDAFRAR123和142PGPVIEPTL150)。结果验证,属于保守性抗原表位,靶向这些抗原表位中的所有单抗都可以识别PRRSV-2毒株的nsp12蛋白,表明针对上述三个抗原表位的单抗具有作为区分PRRSV-1和PRRSV-2毒株的鉴别诊断的潜力,总之,本研究为基于抗原表位的PRRSV检测提供了新的选择靶点。

利用上述抗体可以实现对现有PRRSV病毒无法准确定量、预期表达效果无法验证的技术难题,解决了配制疫苗的关键问题。同时,含所述抗体的药物组合物,可以不分种属地预防和/或治疗猪源所引发的PRRSV疾病,不仅可解决PRRSV经典株导致的猪繁殖和呼吸综合征还可解决猪繁殖和呼吸综合征病毒变异株导致的猪繁殖和呼吸综合征,具有广谱性;弥补无商品化疫苗的缺憾,可用于紧急预防和/或治疗,减少了发病率、降低了死亡率。

具体实施方式
为使本发明更加容易理解,下面将结合实施例来进一步详细说明本发明,这些实施例仅起说明性作用,并不局限于本发明的应用范围。本发明中所使用的原料或组分若无特殊说明均可以通过商业途径或常规方法制得。
本申请中所使用的细胞、病毒及实验动物
SP2/0细胞、HEK293T细胞、Marc-145细胞用含有10%胎牛血清的DMEM在37℃5%CO2条件下培养;PRRSV-2毒株包括HuN4株、CH-1a株、ZJhz021株以及针对PRRSV-2毒株的阳性猪血清均由本实验室制备和保存[12]。pCAGGS-Nsp12-HA质粒由本实验室构建并保存;6-8周龄BALB/c小鼠购自上海杰思捷实验动物有限公司。
实施例1nsp12蛋白表达质粒的构建
根据PRRSV HuN4株(GenBank:EF635006)基因组中nsp12序列,设计正向引物pColdI-nsp12-F:5’-ACCCTCGAGGGATCCGAATTCATGGGCCGCCATTTTACC-3’(SEQ ID NO.1,下划线处为Ec oRI酶切位点)和反向引物pColdI-nsp12-R:5’-GGTAAAATGGCGGCCCATGAATTCGGATCCCTCGAGG GT-3’(SEQ ID NO.2,下划线处为EcoRI酶切位点),然后以PRRSV HuN4株cDNA为模板通过RT-PCR扩增nsp12基因。反应体系为50μl:包括Q5超保真预混酶(2×)25μl、上游、下游引物各1μl、cDNA模板50ng,ddH2O补至50μl。PCR反应条件:98℃预变性30s;98℃变性10s,58℃退火20s,72℃延伸1min30s,共35个循环;最后,72℃延伸10min;同时,将pColdI质粒经EcoRI酶切处理,然后,将PCR产物和酶切产物经琼脂糖凝胶电泳和胶回收试剂盒纯化后,用以同源重组方式克隆至pColdI质粒中,并用PCR扩增和测序鉴定阳性克隆。
nsp12重组蛋白的表达与纯化
将转化于BL21(DE3)感受态细胞的pColdI-His-nsp12阳性菌株接种于200ml含氨苄抗性的LB液体培养基中,37℃,200r/min振荡培养,当光密度OD600nm达到0.6时,加入终浓度为1mol/L的IPTG,在16℃,200r/min诱导蛋白表达16h。在4℃下以8000g离心5min收集菌体,用20mL缓冲液(50mM Tris-HCl,250mM NaCl,pH 8.0)重悬,并在冰浴条件下进行超声破碎裂解细胞15min,然后将裂解产物在4℃条件下以12000g离心30min,利用QIAGEN Ni-NTA亲和层析树脂纯化带有His标签的nsp12蛋白,并进行SDS-PAGE分析,同时,分别利用PRRSVHuN4阳性血清和His标签抗体进行Western blot鉴定。
结果显示,在大约19KDa左右可观察到His-nsp12相应的蛋白条带,且表达蛋白主要存在于超声裂解产物的沉淀中,经用Ni SepharoseTMexcel亲和柱纯化后,得到的nsp12蛋白纯度可达到95%以上(图1A)。为了进一步鉴定目标蛋白,分别用抗His单克隆抗体和抗PRRSV阳性猪血清作为一抗,通过Western blot鉴定,结果显示两种抗体的检测结果一致,均在19KDa处检测到nsp12蛋白特异性条带(图1B),表明表达纯化后的蛋白可作为免疫小鼠使用的抗原。
实施例2针对nsp12的单克隆抗体的筛选和制备
小鼠免疫和杂交瘤细胞制备
采用100μg纯化的nsp12重组蛋白与等量的弗式完全佐剂(首次免疫)或不完全佐剂混合乳化,免疫接种3只6~8周龄雌性BALB/c小鼠,每隔14天以相同剂量加强免疫一次,共免疫四次,最后一次免疫后3天,取免疫小鼠的脾细胞与SP2/0细胞使用PEG-1450进行细胞融合。将融合的细胞在含有HAT和和20%FBS的DMEM的选择培养基中进行培养。当细胞融合10天后,收集融合细胞的上清液,通过间接ELISA检测针对nsp12蛋白的抗体,筛选阳性杂交瘤细胞,并通过有限稀释法对获得的阳性杂交瘤细胞株进行三次亚克隆。为了获得抗Nsp12蛋白的单克隆抗体,用纯化的融合蛋白His-nsp12免疫小鼠,加强免疫3天后进行细胞融合。通过间接ELISA筛选后,发现有5株杂交瘤细胞分泌的抗体对融合表达的His-nsp12蛋白的反应性较强,其OD450值均大于1.5,将获得的5株分泌抗PRRSV Nsp12蛋白抗体的阳性杂交瘤细胞株,分别命名为3G11、9C2、2E3、10G6和2A4(图2A)。同时,用5株单抗分别对His-nsp12蛋白进行Western blot检测,结果它们特异性识别nsp12蛋白条带(图2B)。利用PRRSV毒株感染的Marc-145细胞进行IFA检测,结果显示,5株mAbs均能特异性识别PRRSV感染的细胞(图2C)。为了鉴定获得的5株mAbs识别PRRSV nsp12蛋白的特异性,分别利用真核表达质粒pCAGGS-Nsp12-HA转染HEK293T细胞48小时后收获的细胞样品,以及用PRRSV HuN4株感染Marc-145细胞48小时后收获的细胞样品,对5株单抗进行Western blot鉴定。结果显示,5株mAbs不仅能特异性识别真核表达的nsp12蛋白,而且也能够特异性识别HuN4株感染Marc-145细胞所表达nsp12蛋白。
nsp12重叠多肽的表达
利用nsp12蛋白分段截短表达的方式鉴定单克隆抗体识别的抗原表位,将nsp12设计分为17个重叠肽段,以PRRSV HuN4 cDNA为模板,使用表1中的引物扩增编码17个肽段的基因片段,并分别将各基因片段克隆至原核表达载体,转化至BL21(DE3)感受态细胞中进行诱导表达。

将收集的蛋白表达样品,与5×蛋白上样缓冲液混合,煮沸10min,用10%的SDS-PAGE电泳分离样品中的蛋白质,并用考马斯亮蓝R-250染色。同时,将电泳的蛋白样品转印至NC膜上,将NC膜用含5%脱脂乳的TBS溶液室温封闭2h,再将NC膜与用筛选出的单克隆抗体或标签抗体、阳性血清等在室温孵育1h,经含有0.5%吐温-20(TBST)的洗涤3次后,用HRP标记的山羊抗鼠IgG(H+L)作为二抗(1:10000)在室温下孵育1h,经TBST洗涤3次后,使用ECL显影,通过天能成像系统检测免疫印记结果。结果显示,5株mAbs不仅能特异性识别真核表达的nsp12蛋白,而且也能够特异性识别HuN4株感染Marc-145细胞所表达nsp12蛋白。
间接免疫荧光(IFA)检测
用0.1MOI的PRRSV HuN4、CH-1a、ZJhz021毒株分别感染Marc-145细胞,感染24h后,用提前预冷的80%乙醇固定感染后的Marc-145细胞。用PBS洗涤细胞3次后,加入筛选获得的单克隆抗体,于37℃作用1h,PBS洗涤3次后,加入1:1000稀释的FITC标记的山羊抗小鼠IgG(H+L),于37℃避光孵育30min,经用PBS洗涤后,在倒置荧光显微镜下记录检测结果。
氨基酸序列分析和结构预测
从GenBank数据库中获得26株PRRSV-2分离株的全长基因,选取每个毒株的nsp12基因,利用DNASTAR MegAlign软件对比分析氨基酸序列的保守性。利用SWISS-MODEL构建PRRSV nsp12蛋白的空间结构,通过PyMOL软件分析可视化抗原表位在NSP12蛋白中的空间结构位置。
结果显示,为了确定5株mAbs识别nsp12蛋白中的抗原表位,我们分别利用重叠表达的17个nsp12及其截短肽段为抗原(图3A),对5株单抗进行了Western blot和ELISA检测,其中,针对克隆于pGEX-6p-1载体表达的A(1~68aa)、B(52~102aa)和C(95~154aa)三个相互重叠的截短体GST融合蛋白,5株单抗均不能识别截短体A(1~68aa),而单抗9C2和3G11只识别截短体B(52~102aa),不识别截短体C(95~154aa);另外三株单抗2E3、2A4和10G6只能识别截短体C(95~154aa),不识别B(52~102aa)(图3B和3C)。基于此,进一步将截短体B(52~102aa)分为4个重叠截短肽段,将截短体C(95~154aa)分为10个重叠截短肽段,分别与GST融合表达后,进行Western blot与ELISA鉴定,结果显示,3G11和9C2不识别B1(52~82)抗原区域,可同时识别B2(83~102)、B3(88~102)和B4(93~102)(图3D),据此,初步确定3G11和9C2三株单抗识别的抗原表位为93TWGFESDTAY102。而单抗2E3可同时识别C1(95~132aa)和C3(95~127aa),但不识别C2(125~154aa)、C4(95~122aa)、C5(95~117aa)和C6(95~112aa),因此推测其识别的抗原区域存在于C3的C末端,在此基础上,对C3的C末端继续分为5个肽段进行截短表达和鉴定,结果显示,单抗2E3仅识别C3.4(115-124aa),但不识别C3.1(118~127)、C3.2(117-126aa)、C3.3(116-125aa)和C3.5(114-123aa)(图3E),因此推测2E3三株单抗识别的抗原表位为115DYNDAFRARQ124。对单抗2A4和10G6检测结果显示,二者可同时识别C2(125~154)、C7(130~154)、C8(135~154)和C9(140~154),但是不识别C10(145~154),为此,继续将上述最小识别区域C9截短为5个肽段进行表达和鉴定,结果显示,2A4和10G6可同时识别C9.2(141~150)和C9.3(142~151),但不识别C9.1(140~149)、C9.4(143~152)和C9.5(144~153)(图4F),据此推测10G6和2A4识别的抗原表位为142PGPVIEPTL150。
选择其中效果较好的三株单抗进行测序分析,其中,
单克隆抗体3G11,其重链可变区氨基酸序列如SEQ ID NO.3所示(EVQLQESGAELVKPGASVKLSCKASGYTFTSYYMFWVKQRPGQGLEWIGEINPSNGGTNFNERFKSKATLTVDKSSSTACMQVSGVASEDCAVYYCTRV),轻链可变区氨基酸序列如SEQ ID NO.4所示(SLGERVTITCKASQDINNYLGWFQQNPGKSPKTLIYRSNRLVDGVPSRFSGSGSGQDYSLTISSLEYEDMGIYYCLQYDEFPPTFGAGTKLGLKR);
单克隆抗体2E3,其重链可变区氨基酸序列如SEQ ID NO.5所示(SLSSPSTVTGYSITSDYAWNWIRQFPGNKLEWMGYISYSGSTSYNPSLKSRISITRDTSKNQFFLQLNSVTTEDTATYYCAREISLLRLYYFDYWGQGTTVTVSS),轻链可变区氨基酸序列如SEQ ID NO.6所示(WNQQKPGQPPRLLIYLVSNLQSGVPARFSGSGSGTDFTLNIHPVEEEDAATYYCQHIRELTRSEGGPSWS);
单克隆抗体10G6其重链可变区氨基酸序列如SEQ ID NO.7所示(EVQLKESGAELVKPGASVKLSCKASGYTFTSYYMFWVKQRPGQGLEWIGEINPSNGGTNFNERFKSKATLTVDKSSST AYMQLSGLTSEDSAVYYCTRDLFAYWGQGTMVTVSS),轻链可变区氨基酸序列如SEQ ID NO.8所示(WNQQKPGQPPRLLIYLVSNLESGVPARFSGSGSGTDFTLNIHPVEEEDAATYYCQH)。
为了分析本研究获得单抗所识别抗原表位的保守性,收集了26株2型PRRSV中L1、L3、L5和L8四个谱系(Lineage)参考毒株。采用DNASTAR MegAlign软件比对分析四个主要流行谱系参考毒株的nsp12编码的氨基酸序列,结果显示,表位93TWGFESDTAY102在四个谱系中高度保守性;表位115DYNDAFRARQ124在L8、L5和L3之间高度保守,在L1中仅存在1个氨基酸的差异(118D-118E);表位142PGPVIEPTLQ150在L8、L5和L3中高度保守,在L1中存在两个氨基酸的差异(144PV145-144HT145)(图4)。通过SWISS-MODEL服务器构建了PRRSV nsp12的三维结构,利用PyMOL软件对新鉴定的三个线性表位在空间结构中的位置进行了可视化分析,结果发现,表位93TWGFESDTAY102可形成结构稳定的β折叠,表位115DYNDAFRARQ124以α螺旋形式存在,表位142PGPVIEPTL150以无规则卷曲形式存在,三者均暴露在结构表面,有利于抗原表位的识别。
nsp12单抗对不同PRRSV检测应用
为了分析上述单抗在不同PRRSV检测中的应用,将5株单抗分别与高致病性PRRSV强毒株HuN4、经典毒株CH-1a和NADC30类毒株ZJhz021等三种不同PRRSV-2分离株感染的Marc-145细胞进行IFA鉴定,结果显示,5株mAbs与上述三种PRRSV毒株均能产生特异性荧光(图5),其中存在于表位115DYNDAFRARQ124中118D/118E的氨基酸突变不影响单抗对该抗原表位的识别。表明获得的5株单抗对PRRSV-2中的HP-PRRSV-like毒株、CH-1a-like毒株和NADC30-like毒株均具有较好的反应性,可用于上述毒株的检测。
综上,从上述结果中我们可以看出,我们鉴定出5株单抗能够识别nsp12蛋白中的三个抗原表位(93TWGFESDTAY102、115DYNDAFRARQ124和142PGPVIEPTL150),通过对26株PRRSV-2不同谱系参考毒株的nsp12氨基酸序列比对分析,表明抗原表位93TWGFESDTAY102在目前国内普遍流行的4个谱系PRRSV-2分离株中是完全保守的;而抗原表位114-123aa(115DYNDAFRARQ124)和表位142-150aa(142PGPVIEPTL150)在L8、5和3之间完全保守,而在Lineage1中仅存在1个或2个氨基酸的差异(118D/E或144PV145/144HT145)。基于不同毒株的IFA检测结果证实,三株单抗均能够特异性识别属于Lineage1的NADC30-like毒株的ZJhz021株,表明尽管在Lineage1中存在突变氨基酸,但118D/E突变的氨基酸并不能影响nsp12蛋白的抗原性变化,属于保守性抗原表位,靶向这些抗原表位中的所有单抗都可以识别PRRSV-2毒株的nsp12蛋白。
上述对实施例的描述是为了便于该技术领域的普通技术人员能理解和使用本发明。熟悉本领域技术人员显然可以容易的对这些实施例做出各种修改,并把在此说明的一般原理应用到其他实施例中,而不必经过创造性的劳动。因此,本发明不限于上述实施例。本领域技术人员根据本发明的原理,不脱离本发明的范畴所做出的改进和修改都应该在本发明的保护范围之内。
Claims (10)
1.一种PRRSV编码的非结构蛋白中nsp12蛋白的抗原表位,其特征在于所述的抗原表位分别为:93TWGFESDTAY102、115EDYNDAFRARQ124和142PGPVIEPTL150。
2.一种能够编码上述抗原表位的核酸,所述核酸为能够编码权利要求1所述的PRRSV编码的非结构蛋白中nsp12蛋白的抗原表位的基因序列;
3.一种重组表达载体,所述重组表达载体携带权利要求2所述的核酸。
4.一种重组细胞,所述重组细胞含有权利要求3所述的重组表达载体,或者在基因组中整合有权利要求2所述的核酸。
5.一种融合蛋白,所述融合蛋白是由权利要求1中的nsp12蛋白的抗原表位中的一个或多个的重复或者组合,或者上述重复或者组合与载体蛋白或其他抗原表位蛋白融合所形成。
6.一种特异性识别权利要求1所述的nsp12蛋白的抗原表位的单克隆抗体。
7.如权利要求6所述的单克隆抗体,所述单克隆抗体分别为:单克隆抗体3G11,其重链可变区氨基酸序列如SEQ ID NO.3所示,轻链可变区氨基酸序列如SEQ ID NO.4所示;单克隆抗体2E3,其重链可变区氨基酸序列如SEQ ID NO.5所示,轻链可变区氨基酸序列如SEQID NO.6所示;单克隆抗体10G6其重链可变区氨基酸序列如SEQ ID NO.7所示,轻链可变区氨基酸序列如SEQ ID NO.8所示。
8.权利要求1中的nsp12蛋白的抗原表位,和/或权利要求6所述的单克隆抗体在制备检测PRRSV感染或检测PRRSV抗体的试剂或试剂盒中的应用。
9.权利要求1中的nsp12蛋白的抗原表位,和/或权利要求6所述的单克隆抗体在制备防治PRRSV感染的药物或疫苗中的应用。
10.权利要求1中的nsp12蛋白的抗原表位在制备PRRSV抗体中的应用。
